Incorporation of Tobacco Chloroplasts into Soybean Protoplasts (콩 원형질체내로의 담배 엽록체 이입)

  • Chloroplasts isolated from tobacco (Nicotiana tabacum L. cv. Virginia 115) leaves have been transferred into protoplasts of soybean (Glycine max Merr. cv. Jangyeop) suspension-cultured cells with the help of polyethylene glycol (PEG). The increased yield in protoplasts of chloroplast uptake was depended upon the concentration of both PEG 4,000 and PEG 6,000. The highest yield(36%) occurred at 50% of both PEG, and the yield was decreased above this concentration. The rate of uptake with the incubation time was highest at one hour, then decreased. The process of the chloroplast uptake into the protoplasts was similar with that of a protoplast fusion, except forming invagination during uptake.

Positional mapping for foxglove aphid resistance with 180k SNP array in soybean [Glycine max (L.) Merr.]

  • Foxglove aphid, Aulacorthum solani (Kaltenbach), is a Hemipteran insect that infected a wide variety of plants worldwide and caused serious yield losses in crops. The objective of this study was to identify the putative genes to foxglove aphid resistance in wild soybean, PI 366121 (Glycine soja Sieb. and Zucc.). One hundred and forty-one F4:8 recombinant inbred lines developed from a cross between susceptible variety, Williams 82 and foxglove aphid resistance wild soybean, PI 366121 were used. The two type of resistance response, antibiosis and antixenosis resistance were evaluated through choice and no-choice test, graded by the degree of total plant damage and primary infestation leaf damage; a genome-wide molecular linkage map was constructed with 29,898 single-nucleotide polymorphism markers utilizing a Axiom(R) 180K soyaSNP array. Using inclusive composite interval mapping analysis for foxglove aphid resistance, one major candidate QTL on chromosome 7 was identified. The major QTL on chromosome 7 showed both antixenosis and antibiosis resistance responses. The newly identified major QTL was consistent with previously reported QTL, Raso2, which showed around 5 times narrow down interval range with 8 candidate genes. Furthermore, total 1,115 soybean varieties including Glycine soja and Glycine max were exposed to germplasm screening, and 31 varieties, which showed significant antibiosis type foxglove aphid resistance were identified. This result could be useful in breeding for new foxglove aphid resistant soybean cultivars and developing novel insecticides.

  • The soybean [Glycine max (L.) Merr.] is one of the most important crop resources worldwide as food and forage. It is also important and valuable that to hold crop resources to have high genetic diversities. Recently, a core collection has been constructed in many plants to preserve the genetic resources of various plants. A core collection is small population to represent the genetic diversity of the total collection, and is of strategic importance as they allow the use of a small part of a germplasm collection that is representative of the total collection. Here, we developed the core collection consisting of 816 accessions by using approximately 180,000 (180K) single nucleotide polymorphisms (SNPs) developed in previous study. In addition, we performed genetic diversity and population structure analysis to construct the core collection from entire 4,392 collections. there were excluded sample call rates less than 93% and duplicated samples more than 99.9% according to genotype analysis using 180K SNPs from entire collections. Furthermore, we were also excluded natural hybrid resources which Glycine max and Glycine soja are mixed in half through population structure analysis. As a result, we are constructed the core collection of genetic diversity that reflects 99% of the entire collections, including 430 cultivated soybeans (Glycine max) and 386 wild soybeans (Glycine soja). The core collection developed in this study should be to provide useful materials for both soybean breeding programs and genome-wide association studies.

  • This study was carried out to investigate the specific activity fo peroxidase(POD) in conjunction with the phytotoxic effect of the tolerant soybean(Glycine max Merr.) and the susceptible crabgrass(Digitaria adscendens(L.) Scopol.) to acifluorfen. The POD activity increased during the growth of soybean and crabgrass seedlings. But after the foliar application of acifluorfen, the POD activity in the treated leaves was considerably higher than in controlled leaves, suggesting that soybean had higher tolerance to acifluorfen with growth. All the treated soybeans recovered from herbicidal injury in 14 days after treatment, while all the treated crabgrasses were dead with extreme necrosis and leaf burning. It was measured that the POD activity of crabgrass increased abruptly in 1 to 2 days after treatment and then significantly decreased in 5 days.

  • The major mycotoxins were detected from wheat(Triticum aestivum L.), soybean(Glycine max Merr.) and com(Zea mays L.), infected postharvest phathogens, Penicillium, Aspergillus and Fusarium. Analyses of the major mycotoxins were conducted using HPLC analysis. Detected Penicillium mycotoxins of infected cereals were brefeldin A with amount ranged from 3.1 to 1240 ppm, citreoviridin with amount ranged from 40 to 80 ppm, griseofulvin with amount ranged from 3.6 to 26.0 ppm, citrinin with amount ranged from 0.3 to 4.0 ppm and patulin with amount ranged from 420 to 3800 ppm. Aspergillus toxins of infected postharvest wheat, soybean and corn were ochratoxin A with amount of 730 ppm, 12.4 ppm and 310 ppm, respectively.

Agronomic Performance of G. max x G. soja Hybrid Progenies for Crop Improvement in Soybean

  • Genetic improvement of the cultivated soybean [Glycine max (L.) Merr] may be possible through hybridization with its wild progenitor, G. soja Sieb. & Zucc. Interspecific cross between G. max (Hwangkeumkong) and G. soja (IT.182932) was made in the summer of 1997. In F$_2$ the percentage of plant height, nodes per plant, and pods per plant were high but gradually reduced from F$_2$ to F$_4$. In contrast pod length, seeds per pod, and 100-seeds weight were increased gradually through generations advanced. Wild variation as evident in F$_2$ in plant height, number of branches, pods per plant, and 100-seeds weight. Twenty six percent of the F$_2$, 44 % of the F$_3$ and 60% of the F$_4$ segregants showed more G. max traits. The combination of useful traits from both species is possible through interspecific hybridization. The characters that could be transferred from wild species to cultivated species are more pod number, better capacity, and resistance to disease and insects. The interspecific derivatives offer scope for selection for high grain yield. Therefore, introducing genes from G. soja to G. max could be contribute to greater genetic diversity of future cultivars. And semicultivated soybean had some desired characteristics including tolerance to adverse environments and multi-seed characters. It means the infusing of semicultivated germplasm to the cultivated soybean could increase number of seeds and pods per plant significantly, and consequently could enhance selecting potential on yield.

Isolation of soybean mutants with high and low inorganic phosphorus

  • In soybean (Glycine max (L.) Merr.) seeds, phosphorus (P) is primarily stored in the form of phytate, which is generally indigestible by monogastric animals such as human, pig, poultry, and fish. Thus, this study was conducted to isolate soybean mutants with high available P. Inorganic P content was assessed in a total of 1,266 ethyl methanesulfonate (EMS) $M_4$ lines from the Pungsannamul cultivar. Among the tested lines, four EMS lines (PE379, PE432, PE2205, and PE2503) showed higher mean inorganic P ($1.21-1.56gkg^{-1}$) than did the Pungsannamul cultivar ($0.90gkg^{-1}$). Additionally, six EMS lines (PE718, PE828, PE1466, PE1552, PE3378, and PE3386) showed lower mean inorganic P ($0.38-0.60gkg^{-1}$). The high inorganic P mutants isolated in this study will be further investigated for phytate and total P levels. Moreover, the high and low inorganic P lines will be utilized in a future study of the biochemical pathway of phytate.

A New Soybean Cultivar "Gaechuck#1" : Black Soybean Cultivar with Lipoxygenase2,3-free, Kunitz Trypsin Inhibitor-free and Green Cotyledon

  • Lipoxygenase and Kunitz trypsin inhibitor protein of mature soybean [Glycine max (L.) Merr.] seed are main anti-nutritional factors in soybean seed. A new soybean cultivar, "Gaechuck#1" with the traits of black seed coat, green cotyledon, lipoxygenase2,3 and Kunitz trypsin inhibitor protein free was developed. It was selected from the population derived the cross of "Gyeongsang#1" and C242. Plants of "Gaechuck#1" have a determinate growth habit with purple flowers, brown pubescence, black seed coat, black hilum, oval leaflet shape and brown pods at maturity. Seed protein and oil content on dry weight basis have averaged 39.1% and 16.2%, respectively. It has shown resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and soybean pod borer. "Gaechuck#1" matured on 5-10 October with a plant height of 50 cm. The 100-seed weight of "Gaechuck#1" was 23.2g. Yield of "Gaechuck#1" was averaged 2.2 ton/ha from 2005 to 2007.
