Water deficit is a serious constraint to soybean [Glycine max L. (Merr.)] production in rainfed regions of Asia, Africa, and America. This study was conducted to develop a simple and effective screening method for drought tolerance in soybean. Fifteen soybean cultivars, eight identified to be drought-tolerant and seven drought-sensitive in previous studies, were used for the evaluation of drought tolerance under the new screening conditions. The seedling screening method was consisted of a treatment in a PEG solution and drought treatment in parafilm-layered pots. 5-day-old seedlings were treated in a 18% PEG solution for 4 days and their wilting and hypocotyl browning were recorded. Three seedlings grown in a parafilm-layered pot containing peat moss were drought-stressed by withholding water from the third day after seedling emergence, and root and seedling growth were examined. Degree of drought tolerance were rated based on seedling vigor in the PEG solution and drought-stressed parafilm-layered pots, and also on the penetration ability of roots through parafilm layer. Most of seedlings of the drought-tolerant cultivars showed higher vigour and root penetration than those of the drought-sensitive cultivars under the new screening conditions. Our results indicate that the new method can be used as a simple and effective screening procedure for drought tolerance in soybean breeding programs.
Fifty-nine Korean soybean (Glycine max L. Merr.) landrace accessions were tested for genotype fingerprinting, differentiation and association between morphological traits and SSR profile. Using 8 SSR loci, 59 varieties were divided into 55 groups, and only 4 pairs of varieties were not uniquely identified. The resolving power of SSR for soybean genotyping was much higher than that of the morphological traits that were studied. Identification efficiency also differed among SSR loci. Those loci with higher numbers of alleles distinguished varieties more effectively. Genetic differentiation values of the soybean landraces varied from 0.57 to 0.82 with a mean of 0.68. The number of alleles detected by the 8 loci ranged from 3 to 8. and the effective number of alleles ranged from 2.3 to 5.1. In a study of the association of SSR alleles with morphological traits, some alleles seemed to be related with some specific morphological traits. Comparison of two kinds of dendrograms which were derived from SSR markers and quantitative traits indicated that the dendrograms were not consistent. Considering the correlation between single SSR locus and qualitative traits governed by major genes, the data suggest that alleles of microsatellite loci be more closely related to some traits determined by major genes than those determined by minor genes.
Genome duplication(i.e. polyploidy) is a common phenomenon in the evolution of plants. The objective of this study was to achieve a comprehensive understanding of genome duplication for SNP discovery by Thymine/Adenine(TA) cloning for confirmation. Primer pairs were designed from 793 EST contigs expressed in the roots of a supernodulating soybean mutant and screened between 'Pureunkong' and 'Jinpumkong 2' by direct sequencing. Almost 27% of the primer sets were failed to obtain sequence data due to multiple bands on agarose gel or poor quality sequence data from a single band. TA cloning was able to identify duplicate genes and the paralogous sequences were coincident with the nonspecific peaks in direct sequencing. Our study confirmed that heterogeneous products by the co-amplification of a gene family member were the main cause of obtaining multiple bands or poor quality sequence data in direct sequencing. Counts of amplified bands on agarose gel and peaks of sequencing trace suggested that almost 27% of nonrepetitive soybean sequences were present in as many as four copies with an average of 2.33 duplications per segment. Copy numbers would be underestimated because of the presence of long intron between primer binding sites or mutation on priming site. Also, the copy numbers were not accurately estimated due to deletion or tandem duplication in the entire soybean genome.

The dlm (disease lesion mimic) mutant of soybean (Glycine max L. Merr) shows the similar lesion of a soybean disease caused by a fungus, Corynespora cassilcola. The lesion was examined at cellular and molecular level. Trypan blue staining result indicated that cell death was detectable in the entire region of leaves excluding veins when the lesions had already been developed. We found that the mesophyll cells of palisade layer in the dim mutant appeared to be wider apart from each other. The chloroplasts of the dim mutant cells contained bigger starch granules than those in normal plants. We also found that the lesion development of dlm plant was light-dependent and the starch degradation during the dark period of diurnal cycle was impaired in the mutant. Three soybean pathogenesis-related genes, PR-1a, PR-4, and PR-10, were examined for their expression patterns during the development of disease lesion mimic. The expression of all three genes was up-regulated to some extent upon the appearance of the disease lesion mimic. Although the exact function of DLM protein remains elusive, our data would provide some insight into mechanism underling the cell death associated with the dim mutation.

The growth characteristics and yield per unit area of two cropping systems, corn monoculture and 'wrnsoybean intercropping, were compared and the obtained results were as follows; 1.The two cultivation systems were not significantly different in leaf length, leaf width and length of internode at harvesting time. 2.The weight of stalk and the leaves of a plant were decreased with maturity, while the weight of ear was markedly increased. The patterns of relative ratio of each components were essentialy same in each cultivation systems. In corn monoculture, the percentages of stalk, leaf, and ear at final harvesting stage were 22.9, 13.7, and 63.4 and in corn-soybean intercropping, they were 21.8, 10.9, and 67.3 respectively. 3.Dry matter yield per unit area (kg/lOa) of intercropped corn at yellow stage was similar to that of monocultured corn (1, 483 kg/ 10a vs 1, 509 kg/ 10a). At ripe stage, however, the dry matter yield of intercropped plant was more than that of monocultured (1, 679 vs 1, 660 kg/ 10a). 4.The same pattern was observed in organic matter yield.
Antibodies raised against the purified p-subunit of $\beta$-conglycinin were used in immunohistochemical studies to monitor the pattern of $\beta$-conglycinin mobilization in the cotyledons during soybean [Glycine max (L.) Merr.] seed germination. Western blot analysis revealed that the break down of the $\beta$-subunit of $\beta$-conglycinin commenced as early as 2 days after seed imbibition (DAI). Concurrent with the degradation of the $\beta$-subunit of $\beta$-conglycinin, accumulation of 48, 28, and 26 kD proteolytic intermediates was observed from 2 to 6 DAI. Western blot analysis also revealed that the acidic subunit of glycinin was mobilized earlier than the basic subunit. The basic glycinin subunit was subjected to proteolysis within 2 DAI resulting in the appearance of an intermediate product approximately 2 kD smaller than the native basic glycinin subunit. In contrast to the major seed storage proteins, lipoxygenase was subjected to limited proteolysis and was detected even after 8 DAI. The first sign of $\beta$-conglycinin breakdown was observed near the vascular strands and proceeded from the vascular strands towards the epidermis. Protein A-gold localization studies using thin sections of soybean cotyledons and antibodies raised against the $\beta$-subunit of $\beta$-conglycinin revealed intense labeling over protein bodies. A pronounced decrease in the protein A-gold labeling intensity over protein bodies was observed at later stages of seed germination. The protein bodies, which were converted into a large central vacuole by 8 DAI, contained very little 7S protein as evidenced by sparse protein A-gold labeling in the vacuoles.
Four flavonoids 1-4 and one phytosterol 5 were isolated from methanol extract of Taekwangkong, one of the soybean cultivars, and the structures of these compounds were fully characterized by physical and spectral analysis. The content of compounds 1-4 as determined by $C_{18}$ reversed phase HPLC (high-performance liquid chromatography) coupled with diode-array detector were 12.1, 624.6, 18.0, and $219.6\;{\mu}g/g$, respectively, and the total phenolic content of this cultivar was measured as 3.7 mg gallic acid equivalent per g dry material (GAB/g). Also, compound 1 showed strong radical scavenging activity in the 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assay ($IC_{50}\;=\;47.6\;{\mu}M$), five-fold higher than seen in the 1,1-diphenyl-2-picrylliydrazyl (DPPH) assay. These results lead to the conclusion that soybean not only has many phytoestrogens but also has potent antioxidant activity.
Effects of leaf and pod removal on changes in leaf and seed number, and leaf, seed, pod and stem dry weight of soybean [Glycine max (L,) Merr.] cultivar ‘Hwangkeumkong’ were measured at the research farm of Korea University in 1992. The upper 40% and lower 60% of leaves and pods were subjected to treatments at the growth stage of beginning pod(R3). Leaf number and dry weight of lower part were increased by upper leaf-lower pod removal, but seed number and dry weight were decreased. Upper leaf-lower pod removal increased lower stem dry weight and decreased upper pod dry weight. Leaf-seed ratio of the upper leaf-lower pod removal was the highest with 3.54 and harvest index was the lowest with 27% among the treatments. Lower leaf-upper pod removal showed that more assimilates from upper leaves were translocated to lower seeds than from lower leaves to upper seeds in upper leaf-lower pod removal treatment.
The competitive effects of annual weeds on soybeans (Glycine max. (L.) Merr. 'Gwang Gyo') were Studied on silt clay loam soil. Weeds allowed to grow in the row for 2, 4, 6, 8, 10 weeks after soybean planting and full season reduced soybean yields 6.5, 3.2, 10.7, 8, 8.24.4, and 44%. respectively. Reduction in leaf area index (LAI) and crop growth rate (CGR) of soybeans were closely correlated to percent soybean yield reduction. Weed com petition showed to reduce the number of soybean pods per plant, the number of branches per plant, and plant height.
Water deficit is the primary constraint of soybean [Glycine max (L.) Merr.] yield, and a physiological understanding of processes affected by water deficit is a key step in identifying and improving drought tolerance in soybean. The objectives of this research were to evaluate biomass and nitrogen accumulation patterns and water use efficiency (WUE) as possible mechanisms associated with the drought tolerance of Jackson. Biomass accumulation of Jackson was contrasted with the PI416937, which also has demonstrated tolerance to drought. For water-deficit treatment, total biomass accumulation was negligible for PI416937, but biomass accumulation continued at approximately 64 % of the well-watered treatment of Jackson. Transpirational losses for Jackson and PI416937 were approximately the same for the water-deficit treatment, indicating that Jackson had superior WUE. Isotopic discrimination of $^{13}$ C relative to $^{12}$ C also indicated that Jackson had higher WUE. Results indicated that increased WUE for Jackson under water deficit showed it was tolerant to drought rather than had an avoidance mechanism.
