  • Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). It has been reported to have an anti-inflammatory effect on rat paw edema model. To develope as an anti-allergic drug, genotoxicity of sophoricoside was investigated in bacterial and mammalian cell system such as Ames bacterial test, chromosomal aberration assay and single cell gel electrophoresis (Comet) assay.(omitted)

Antitumor constituents from some Korean medicinal plants

  • For the screening of bioactive natural products, the benzene or methanol extracts from 93 medicinal plants of Korea were prepared, and tested for the cytotoxicity against L1210 cells and for the antitumor action (Bae et al., 1992 and 1996). Of 93 extracts tested, 6 samples showed a cytotoxicity in both benzene and methanol extract, 39 samples in benzene and 13 samples in methanol extract. The benzene extract of the root of Scutellaria indica L., Sophora fIavescens Solander ex Aiton, Carpesium abrotanoides L., Gymnaster koraiensis (Nakai) Kitamura, Pyrola japonica Klenze, and Forsythiae Fructus showed a potent cytotoxic activity. This observation led to isolate active cytotoxic components, some of which demonstrated some antitumor action. In addition, the structure-activity relationship was discussed.

  • Fourteen kinds of medicinal plants were screened for determining inhibitory activities on monoamine oxidase B. The extracts of Artemisia Messer-Schmidtiana (herba), Chrysanthemum indicum(flos), Ericibe obtusifolia (radix et rhizoma) and Sophora japonica (flos) strongly inhibited the enzyme. Among them, Chrysanthemi flos was chosen for elucidating its active principles, and some flavonoids were isolated and identified as acasetin (I), 5, 7-dihydroxy chromone (II), diosmetin (III), apigenin (IV), eriodictyol (V) and luteolin (VI).$IC_{50} were determined as following: 1, 2.46;II, 0.19; III, 2. 11mM, and the others showed weak inhibition.

  • Interleukin (IL)-5 appears to be one of the main proinflammatory mediators among a growing number of cytokines and chemokines that induce eosinophilic inflammation. Sophoricoside and their analogs isolated from Sophora japonica show relatively potent inhibitory activity of interleukin (IL)-5 as a small molecule. Initial attempt to identify the structural requirement of this isoflavonone led to find new chalcones to exhibit the inhibitory activity of IL -5. (omitted)

  • Phytochemical examination of Sophorae Fructus yielded six phenolic compounds. The structures were elucidated as genistein(1), genistin(2) and genistein 7-O-${\alpha}$- L-rhamnopyranoside(3) by phytochemical and spectral evidences. The other compounds(4, 5, 6) are understudied by 2D-NMR. Nitric Oxide and PGE$_2$ production inhibitory activities in INF-${\gamma}$, LPS stimulated RAW 264.7 cell were examined. Compound 2 and 4 showed significant nitrogen monoxide(NO) production inhibitory activity in IFN-${\gamma}$, LPS stimulated RAW 264.7 cell. (omitted)

  • Sophoricoside analogs are natural isoflavonoids isolated from fruits of Sophora japonica L. and exhibited an inhibitory effect on IL -5. Many synthetic variations on isoflavonoids has been reported. but relatively few examples of quinolone analogs have been described. As part of our endeavor to develop novel and effective IL-5 inhibitor, we have synthesized azaisoflavones by cyclization of the key intermediate, 2'-aminochalocone obtained from substituted aniline. The synthesized azaisoflavones were evaluated for their inhibitory activtities on IL-5 comparing with natural Sophoricoside analogs. None of the azaisoflavones showed promixing inhibitory effects In the assay. Nevertheless. assay data Indicated that 5.7-phenolic hycroxy groups on the A-ring and alkyl subsitiuent on N1 seemed to play an importnt role in the IL-5 bioassay.

Sophoricoside analogs inhibit COX isozymes but not iNOS and TNF in LPS-stimulated macrophages Raw264.7

  • Macrophages activated by lipopolysaccharide (LPS) are known to induce several proinflammatory proteins including COX-2. iNOS and TNF which produce chemical mediators involved in inflammatory response. Sophoricoside and its analogs (genistin, genistein and orobol) from Sophora japonica (Leguminosae) showed differential inhibitory effects on COX-1 and 2 activities. Sophoricoside and genistin shwoed IC50 values of 4 uM and 6 uM on COX-2 activity and of 1,497 uM and 135 uM on COX-1 activity, respectively. Genistein and orobol showed IC50 values of 3 uM on COX-2 activity and of 28 uM and 18 uM on COX-1 activity. respectively. Therefore. the legume isoflavonoids to be selective COX-2 inhibitors. However. sophoricoside and its analogs did not show inhibitory effects of COX-2, iNos and TNF transcripts. which were identified by the RT-PCR.

  • The antioxidant activities of six isoflavonoids isolated from Sophora japonica wood and bark were examined by DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging method. This study was focused on the relationship between antioxidant activity of isoflavonoids and their chemical structures. From the results of this study, it could be concluded that the hydroxyl groups that linked at ring B and ring A in isoflavonoids have importance in the antioxidant activity. Additionally, the absence of the 2,3-double bond on the isoflavonoid enhances its antioxidant activity.
