Buckwheat sprout is used as vegetable, and also flour for making noodles, and so on. Currently, information about tissue culture in buckwheat is limited and restricted to micro-propagation. We carried out somatic embryogenesis and plant regeneration using hypocotyl segments as explant of the cultivated buckwheat species, Fagopyrum esculentum which differs from existing studies in the growth regulator combinations used. Maximum callus regeneration was induced on MS medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) $2.0mg{\cdot}L^{-1}$, benzyladenine (BA) $1.0mg{\cdot}L^{-1}$ and 3% sucrose. Friable callus was transferred to solidified MS media containing BA ($1.0mg{\cdot}L^{-1}$) with various concentrations of 2,4-dichlorophenoxyacetic acid for the induction of embryogenesis. The optimum concentrations of growth regulators (for regeneration of plantlet) were indole-3-acetic acid ($2.0mg{\cdot}L^{-1}$), Kinetin ($1.0mg{\cdot}L^{-1}$), BA ($1.0mg{\cdot}L^{-1}$). Only 2,4-D did not show any significant effect on callus induction or embryogenesis. Regeneration of embryonic callus varied from 5% to 20%. Whole plants were obtained at high frequencies when the embryogenic calli with somatic embryos and organized shoot primordia were transferred to MS media with 3% sucrose. The main objective of this research was to develop an efficient protocol for plant regeneration for common buckwheat, and to apply in future for genetic transformation.

Culture conditions were established for high frequency plant regeneration via somatic embryogenesis from cell suspension cultures of Nymphoides coreana. Zygotic embryos formed pale-yellow globular structures and calluses at a frequency of 85.6% when cultured on half-strength Murashige and Skoog (MS) medium supplemented with 0.3 $mg\;l^{-1}$ of 2,4-D. However, the frequency of pale-yellow globular structures and white callus formation decreased slightly with an increasing concentration of 2,4-D up to 10 $mg\;l^{-1}$ with the frequency rate falling to 16.7%. Cell suspension cultures were established from zygotic embryo-derived calluses using half-strength MS medium supplemented with 0.3 $mg\;l^{-1}$ of 2,4-D. Upon plating onto half-strength MS basal medium, over 92.3% of cell aggregates gave rise to numerous somatic embryos and developed into plantlets. Regenerated plantlets were successfully transplanted into potting soil and achieved full growth to an adult plant in a growth chamber. The high frequency plant regeneration system for Nymphoides coreana established in this study will be useful for genetic manipulation and cryopreservation of this species.

The use of liquid-medium-based procedure relative to the solid media led to a 4.5-fold increase in the number of cotyledon-stage embryos. The most efficient system for multiplication and regeneration of somatic embryos was CP6 procedure with the media MSD40/MSD20/MSM6AC/FNL0S3S3GM. However, the rate of regeneration was lower. About 71% of the embryos with dicotyledon were continued to develop the roots after desiccation treatment and 92% of the germinated embryos produced shoots in 10 days. Of the four morphologically different types of embryos, dicotyledonous ones showed a high frequency of conversion, while only a few with fused and horn type cotyledon developed shoots. Mature somatic embryos were desiccated in empty petri dishes for 12-72 h. Embryo survival rate was the highest after 12 h of desiccation, but maximal germination was observed at 24 h. After desiccation, they were placed on MS medium without growth regulators for germination. Germinating embryos were transferred to small pots with vermiculite for plant regeneration. The etiolating the plants during the growth was resolved to add 1% activated charcoal on hormone-free MS medium.
Effective micropropagation system via somatic embryognesis was established for a Phytophthora resistant Aralia elata cultivar. Different kinds of growth regulators were needed to induce embryogenic callus with different explant sources. When leaf explants were used, a combination of 2,4-D, TDZ and L-glutamine was needed, whereas when petiole and root explants needed only 1.0 mg/L 2,4-D. Embryogenic callus induction rate under the optimum culture condition was 75.0%, 67.0% and 83.0% from leaf, petiole and root segment, respectively. Somatic embryo germination and plantlet conversion rate appeared to be influenced greatly by various osmoticums. More than 90% of embryos germinated when treated with sucrose, glucose and maltose. However, the highest conversion rate (72%) was recorded on medium with 2% sucrose only. The converted plantlets grew normally on 1/2MS basal medium, were acclimatized on artificial soil mixture and survived more than 95% in the greenhouse condition. The results suggest that the species can be clonally propagated through in vitro culture system via somatic embryogenesis.

Purpose: Somatic growth is an important indicator of health in children. Adequate organ growth is essential in growth and directly related to body growth. We consider renal length as a surrogate of organ growth in growing children. Measurement of weight, height, and many anthropometric indices, such as body surface area (BSA), body mass index (BMI), and Rohrer and Kaup indices, are used to evaluate growth status. The aim of this study was to evaluate the association between renal length and somatic parameters and analyze the affecting factors for renal size during growth. Methods: The data for renal length in 66 children (age, $12.9{\pm}15.6$ months; male/female, 34/32) were obtained. Each kidney was measured with ultrasonography and dimercaptosuccinic acid scan. The data on age, sex, height, and weight were obtained from the medical records. BSA, BMI, and Rohrer and Kaup indices were calculated from measured height and weight. BSA was calculated by 2 methods, and is expressed as BSA I and BSA II. Results: There were significant correlations between renal size and age, weight, height, BSA I, BSA II, and Rohrer index. In the regression analysis, the most significant contributing factor to renal growth was height ($R^2$=0.636, $P$ <0.001). Conclusion: Height seems to be the most important factor associated with organ growth in growing children. Further studies to evaluate adequate organ growth should be carried out.
Somatic embryos were induced directly from cotyledon explants of Korean ginseng (Panax ginseng C.A. Meyer) on Murashige and Skoog (MS) medium with 2,4-D, BAP, kinetin or lacking growth regulators. When somatic embryos formed on all media grew to cotyledonary stage, the further development of embryos was ceased and remained in white color. By gibberellic acid (over 1.0 mg/1 $GA_3$) treatment, all the somatic embryos turned rapidly to green and germinated within 3 weeks. Chilling treatment also induced the germination of somatic embryos. The effective temperature regime was $-2^{\circ}C$ for over 8 weeks but more higher temperature than $0^{\circ}C$ did not effective for germination of somatic embryos. Ultrastructural observation revealed that the cotyledon cells of somatic embryos without chilling or $GA_3$ treatment contained numerous lipid reserves, dense cytoplasm, proplastids and non-activated mitochondria with poorly differentiated internal structure, but the cotyledon cells of germinating somatic embryos after chilling or $GA_3$ treatment highly vacuolated and contained well-developed chloroplasts and active state of mitochondria enclosing numerous cristae. The above results indicate that in vitro developed somatic embryos of Panax ginseng may be dormant after mature similar to zygotic embryos.
A method for long-term conservation of somatic embryos of Eleutherococcus senticosos was described. Suspension cultured globular somatic embryos were successfully conservated for 36 months at $4^{\circ}C$. The embryos resumed growth within two weeks when returned to MS liquid medium containing $0.2\;mg/{\ell}$. 2,4-dichlorophenoxy acetic acid. The optimal condition for cell proliferation was achieved when somatic embryos cultured at $32^{\circ}C$ in 1/3 MS liquid medium, and about 1.2 g of embryogenic cell was induced from 150 globular embryos after 6 weeks of suspension culture. The embryogenic cells produced from these somatic embryos exhibited normal plant regeneration on auxin-free medium.
Excised cotyledon segments of ginseng zygotic embryos cultured on MS basal medium without growth regulators produced somatic embryos near the basal excised portion at a high frequency. The frequency of somatic embryo formation on the segments declined along with advancing zygotic embryo maturity. In immature cotyledons, all the cells of the epidermis and subepidermis were smaller and more densely cytoplasmic than those in mature cotyledons, and from which multiple cells participated in embryogenic division to form somatic embryos with multiple cotyledons and fasciated radicles (poly-embryos). But in germinating cotyledons, only the epidermal cells were densely cytoplasmic and singularly competent to develop into somatic embryos resulting in single-embryos with closed radicles. This result means that the origin and development of somatic embryos is determined according to whether the cells participating in embryonic division are in a single state or a massive state relative to cotyledon maturity.
An efficient somatic embryogenesis and plant regeneration protocol was developed for Schisandra chinensis Baill, using embryogenic cell suspensions and optimized media conditions. Friable embryogenic callus was induced from cotyledonary leaf and hypocotyl explants of 7 days old seedlings on MS agar medium supplemented with 1.0 to $4.0\;mg\;l^{-1}$ of 2,4-dichlorophenoxyacetic acid (2,4-D). Fast growing and well dispersed embryogenic cell suspensions were developed within two months when embryogenic calli were transferred to MS liquid medium containing $1.0\;mg\;l^{-1}\;2,4-D$. One third strength of MS medium was the best for both overall growth and development of somatic embryos in liquid culture. Over 3400 viable somatic embryos were produced from each 150 ml flask with an initial cell density of 30 mg in 30 ml medium. Germinated somatic embryos developed in liquid medium converted into plantlets after transferred to half-strength MS semi-solid medium. Approximately 90% of the converted plantlets were successfully transplanted to soil and grew into fertile plants.
The transparisation technology for milk and milk products could be applied widely and very importantly to various determination because transparisation can economize the cost and increase with precision in the milk payment system. Component of butanone or Triton in transparisation solvent would inhibit the growth of bacteria and method. Enzymatic determination of leucocytes were proposed to evaluate milk quality as mastitis in the milk payment system, this can be easily applied to simplify automation of the determation with the lowest investment cost in milk pay system. The significance of this technique, it can be used in the quality control of raw milk and milk products, milk payment system, and programming of national dairy project. Transparisation technology is used in somatic cell counting by enzymic methods. The range of deviation for this method is 16% in 74 samples. But the deviation is increased to 20% when the Infoss method is used. It is affected by the percentage of epithelial cells and white blood cells in somatic cells from different animals and the stages of aging. NAgase activity has an obvious correlation with white-blood cells in milk. In the case of mastitis the white-blood cells is 90-95% in somatic cells in milk, it is showing greater precision in measuring the state of mastitis. In conclusion the enzymic method of somatic cell counting is a relatively simple and easy method of measurement and can be easily practiced. And the importance of this method is also worth utilizing for indirect counting of Somatic cells by use of synthetic substrates to NAgase. In the future, with the further development of the research in this field, it will b possible to automatize the measurement.
