국내산 장미꽃 추출물이 Chinese Hamster Ovary 세포 증식에 미치는 영향 (Effect of Domestic Rose Flower Extracts on the Growth of Chinese Hamster Ovary Cells)

  • 국내에서 새로 육성된 장미의 부가가치를 향상시키고 천연 식품소재로서 기능성을 탐색하는 기초연구의 일환으로 5개 장미 품종을 대상으로 유기용매 추출물을 제조하고 장미꽃 추출물이 CHO 세포의 생육에 미치는 영향을 MTT법으로 조사하였다. 장미 추출물들은 추출용매의 종류와 첨가농도에 따라 CHO 세포의 증식을 촉진하거나 생육을 억제하였으며 품종에 따른 차이는 없었다. Ether로 추출한 장미꽃 추출물을 5 ${\mu}$g/ml 농도로 첨가한 경우 대조군에 비하여 CHO 세포의 증식촉진효과가 가장 높았다. 이러한 결과는 장미꽃은 CHO 세포의 증식을 조절하는 생리활성을 나타내며, 신 기능성 식품소재로서 활용하는 가능성을 시사한다.

홍조류 에탄올 추출물 및 다양한 용매 분획물의 라디칼 소거능 (Radical Scavenging Activity of Ethanol Extracts and Solvent Partitioned Fractions from Various Red Seaweeds)

  • The EtOH extracts of red seaweeds (Symphyocladia latiuscula, Chondrus ocellatus and Carpopeltis affinis) and solvent partitioned fractions were investigated for their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects and the total phenolic contents were correlated with ABTS and DPPH radical scavenging activities. The EtOH extracts and their solvent partitioned fractions exhibited strong ABTS and DPPH radical scavenging activities. Among the solvent partitioned fractions obtained from n-Hexane (HX), methylenchloride (MC), ethylacetate (EA), and buthanol (BuOH), the HX fraction from C. affinis showed higher radical scavenging activities than other fractions. Total phenolic contents showed significant correlation ($r^2$ = 0.709) with ABTS radical scavenging activity. The results of this study suggest that the strong radical scavenging activity of HX fraction from C. affinis is a promising natural antioxidant for healthcare products.

수산폐기물 재활용을 위한 미더덕껍질 용매 추출물의 생리활성 탐색 (Screening of Biological Activity of Solvent Extract from Styela clava Tunic for Fishery Waste Recycling)

  • Styela clava tunic is generated in large amounts as a waste from S. clava processing plants and causes environmental problem. Although biological activities of S. clava were reported by many investigators, study on S. clava tunic was little. In this study, therefore, tyrosinase inhibition and antioxidative activities of extracts from S. clava tunic using different solvent were investigated for recycling of the fishery waste. Among extraction methods tested, autoclaved extraction (25.7%) and hot water extraction (18.2%) appeared to be effective for extraction. The highest total phenolic content was 46.6 mg/g in autoclaved extract while the highest flavonoid content was 23.0 mg/g in chloroform extract. All extracts possessed tyrosinase inhibition activity and the inhibition activity was concentration-dependent. Inhibition concentration ($IC_{50}$) against tyrosinase activity was $0.36{\times}10^4$ mg/ml in ethanol extract, $0.11{\times}10^3$ mg/ml in acetone extract and 0.27 mg/ml in n-butanol extract. Among extracts tested, hot water and autoclaved extracts displayed higher antioxidative activity than organic solvent extracts. Therefore, our data suggest that extract from S. clava tunic may potential candidate for cosmetic product with whitening effect and medicine for diseases caused by various oxidative stresses.

추출방법에 따른 Nutmeg Oil의 물리적, 화학적 성질에 관한 연구 (Studies on the Some Physical and Chemical Characteristics of Nutmeg Oil by Different Extraction Methods)

  • 추출방법을 달리하여 얻은 육두구 정유에 대한 물리적 성질을 측정하고, GC와 GC/MS를 이용하여 분석하였다. 추출수율은 용매추출이 23%로 가장 많았고 SDE 추출, $CO_2$ 추출순이었다. 육두구유에 대한 물리적 성질과 GC분석결과에서 본 실험에 사용한 육두구는 East Indies type에 가까운 것을 확인할 수 있었고 각각 추출물의 분석결과로부터 sabinene은 3.6-14.1%, miristicin은 28.4-48.7% elemicin은 1.2-2.1%이 함유되어 있음을 알 수 있었다. 또 발암물질로 알려진 safrole은 SDE 추출에서 3.8%, 용매추출에서 3.6%, $CO_2$추출에서 5.6% 함유하고 있는 것으로 나타났다. SDE 추출이나 용매추출에서 확인된 camphene은 $CO_2$추출에서 확인할 수 없었고 monoterpene, terpene alcohol 계통의 화합물은 SDE 추출이나 용매 추출에 많았다. 방향족 화합물은 $CO_2$추출에서 많이 추출됨을 확인할 수 있었다.

김치 용매 추출물의 항산화성 (Antioxidant Activity of Various Solvent Extracts from Freeze Dried Kimchi)

  • 동결건조한 김치를 물, 메탄올, 에칠 에테르로 각각 추출한 후 이들 용매추출물들이 지니고 있는 지질산화반응에서의 항산화력에 대하여 고찰하였다. 즉, linoleic acid mixture의 액상산화반응 조건($37^{\circ}C$)에서 과산화물 생성억제에 대한 용매별, 김치발효기간별, 농도별, 그리고 혼합용매종류별로 실험하여 추출물의 항산화능을 살펴보았다. 그 결과 물과 메탄올 추출물이 에틸에테르 추출물 보다 과산화물의 생성을 더 억제 하였다.그리고 첨가농도가 증가할수록 항산화성이 증대되었으며 발효 초일의 김치나 과숙김치보다 잘 숙성된 김치(15$^{\circ}C$에서 7일간 발효)의 추출물에 더 항산화성이 높았다. 또한 물과 메탄올의 비율을 달리 하여 추출한 경우 순수 메탄올보다 50~80% 메탄올 농도에서의 용매추출물에서 항산화효과가 높았다. 비록 75% 메탄올추출물의 항산화성은 BHA와 $\alpha$토코페롤보다는 낮았지만 동 추출물에서 계통적 분획 등에 의한 분획물의 항산화성을 계속 검토하므로서 보다 높은 항산화성 물질이 분리될 수 있음을 알 수 있었다.

Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

Radical Scavenging Activity of Grape-Seed Extracts Prepared from Different Solvents

  • Antioxidant activities of grape seeds extracted with various solvents were evaluated by measuring total phenol and flavanol contents, thiobarbituric acid reactive substances (TBARS) following lipid peroxidation, 2-deoxyribose degradation, SOD-like activity, 2,2'-azino-bis(3-ethylbenzthizaoline-6-sulfonic acid (ABTS) radical-scavenging ability, and electron-donating ability using 1,1-diphenyl-2-pycryl hydrazil (DPPH) method. Total phenol and flavanol contents of mixted-solvent extracts were higher than those of single-solvent extracts, with the mixing ratio of 17:3 (ethyl acetate: water) (EW) showed the highest contents. Antioxidant activities (%) of TBARS following phosphatidylcholine peroxidation were 14, 45, 45, 7, 4, 25, 21, 23, and 20% for ascorbic acid (AA), butylated hyroxytoluene (BHT), quercetin (Q), acetone extract (AT), ethyl acetate (EA) extract, methanol (MeOH) extract, 4:1 (EA) extract, 9:1 (EW)-extract, and 17:3 EW extract, respectively. Antioxidant activities for 2-deoxyribose degradation were 5, 80, 87, 78, 56, 73, 64, 60, and 75% in AA, BHT, Q, AT, EA, MeOH extract, 4:1 EW extract, 9:1 EW extract, and 17:3 EW extract, respectively. MeOH grape seed extract showed distinctly stronger electron-donating activity than other solvent extracts.

프로폴리스 농도가 항균활성에 미치는 영향 (The effect of propolis concentration on the antibacterial activity)

  • The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, $20^{\circ}C$, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensitivity of propolis samples to Staphylococcus aureus was investigated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

넓패(Ishige foliacea) 추출물의 피부노화 억제활성 (Anti-skin Aging Properties of Different Solvent Extracts from Ishige foliacea)

  • In this study, the antioxidant and anti-skin aging properties of the Korean marine algae Ishige foliacea were investigated. Solvent extracts from I. foliacea were prepared with 70% ethanol, 80% methanol, and water. The extraction yields of various solvent extracts ranged from 9.55% to 35.12%. In terms of antioxidant activity, the ethanol extract showed the highest ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging activity, nitrite oxide scavenging activity, reducing power, and FRAP (ferric reducing antioxidant power). Regarding anti-skin aging activity, evaluation of the skin whitening and anti-wrinkle activities revealed that the methanol, water, and ethanol extracts possessed the highest tyrosinase (IC50=0.98 mg/mL), elastase (IC50=0.15 mg/mL), and collagenase (IC50=0.06 mg/mL) inhibitory activities, respectively. These results suggest that I. foliacea holds potential as an antioxidant and anti-skin aging substance in food and cosmetic materials.

다시마 용매추출물과 다시마 분말 첨가에 의한 매작과의 산화안정성 (The Oxidative Stability of Solvent Extracts of Sea Tangle Powder(STP)and Maejakgwa Made with STP)

  • The purpose of this study was to investigate the antioxidative effects of Sea tangle powder(STP) solvent extracts as well as Maejakgwa made with STP. The STP solvent extracts were added to soybean oil at a quantity of 0.05%. The solvents used for extraction were methanol, ethanol, ethyl acetate, and petroleum ether. Soybean oil without added STP was used as a negative control, and soybean oil samples containing 0.02% butylated hydroxy toluene(BHT) and $\alpha$-tocopherol were used as positive control, respectively. Each sample was stored at $50^{\circ}C$ for 30 days. The oxidation levels of these samples were determined by measuring their acid values, peroxide values, and thiobarbituric acid(TBA) values. The soybean oil samples containing the STP extracts had lower oxidation levels than both the negative control and $\alpha$-tocopherol positive control, and the sample containing the 0.05% methanol extracts had the lowest oxidation. According to the Rancimat method, the methanol extract(320 min) and ethanol extract(316 min) demonstrated longer induction periods as, compared to the control(253 min), $\alpha$-tocopherol(255 min), and BHT(309 min) samples. For the Maejakgwa, acid values increased over the storage period, however, the samples made with STP had lower values than the control group. Peroxide values increased rapidly for 30 days and then decreased. The TBA values of the Maejakgwa samples made with 3% and 9% STP were lower than those of the 15% STP sample and the control. In conclusion, the oxidative stability of soybean oil containing solvent extracts of STP and Maejakgwa made with STP were increased.
