Yersinia enterocolitica, which causes yersiniosis, is a bacterium that produces biofilms effectively. The inhibition of biofilm formation provides a method for preventing infections with Y. enterocolitica. In this study, the inhibitory activity of Y. enterocolitica biofilm formation was investigated in a library of 140 edible plant methanol extracts including forest products. It was identified that the biofilm formation could be inhibited by 12 extracts of plants, Agastachis Herba, Agrimoniae Herba, Diospyros kaki leaves, Elsholtziae Herba, Ginkgonis Semen, Lycopi Herba, Melonis Pedicellus, Menthae Herba, Mori Radicis Cortex, Polygoni Multiflori Radix, Prunellae Spica, and Schizonepetae Spica. After changing the solvent to ethanol and water, the greatest inhibition of biofilm formation was produced by a 50% ethanol extract of Polygoni Multiflori Radix. A method to effectively prevent yersiniosis can be developed using the edible plant extracts identified in this study.
인삼시료에 있는 quintozene, tolclofos-methyl 그리고 procymidone 등의 농약성분을 추출하기 위한 방법으로서 microwave를 이용할 수 있을 지의 여부와 인삼을 가공제품으로 만들기 전에 microwave를 이용하여 잔류농약 성분을 신속히 추출하여 제거시킬 수 있는지 알고자 하였다. 인삼분말 시료에 hexane을 추출용매로 이용한 microwave 추출의 최적 조건은 power 150 watts, 추출시간 2분이었고 그 추출 효율은 Soxhlet으로 7시간 동안 가열 환류 추출한 결과를 지수 100으로 하였을 때 quintozene 100, tolclofos-methyl 92, procymidone 100으로 거의 동일한 수준으로 나타났다. 그리고 microwave 추출법을 사용함으로써 실제 Soxhlet 추출법에서 사용된 용매양의 최소 7배 이상 감소시킬 수 있었고 추출 시간 또한 7시간에서 2분으로 훨씬 단축되었다. 인삼 엑기스 제조 전 잔류농약 성분의 제거 실험에 있어서 45%나 60% ethyl alcohol을 용매로 사용하고 microwave로 power 150 watts 추출시간 2분 이내로 1회 추출하여 quintozene 100%, tolclofos-methyl 69%, procymidone 83%정도의 제거효과를 얻었다.
Objectives : This study was performed to find best extraction solvent for application of Ulmi cortex to food or herbal medicine as an antioxidant only using water, ethanol and their mixtures. Methods : The Ulmi cortex extracts were prepared using water and 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% and 100% (v/v) ethanol, and were evaluated yields, total polyphenol contents, DPPH and ABTS radical scavenging activities, lipid peroxidation activities, and catechin and epicatechin contents. Results : Among the Ulmi cortex extracts, the yield was highest in water extract (8.9%) and lowest in ethanol extract (3.8%). The yield of 30% ethanol extract (8.5%) also was very high to similar with water extract. The total polyphenol content was highest in the 30% ethanol extract ($253.6{\mu}g/mg$ extract) and lowest in water extract ($109.0{\mu}g/mg$ extract). The DPPH radical scavenging activity was highest in ethanol extract (IC50, $8.53{\mu}g/ml$), ABTS radical scavenging activity was highest in 60% ethanol extract (IC50, $3.08{\mu}g/ml$), and the inhibition of lipid peroxidation was highest in 70% ethanol extract (IC50, $7.96{\mu}g/ml$). As ethanol content of extraction solvent increased from 0% to 30%, the antioxidant activities were remarkably increased whereas from 30% to 100%, the antioxidant activities were increased or decreased a little. Conclusions : The findings of the present study suggest that 30% ethanol is best solvent for extraction of Ulmi cortex, considering yield, polyphenol content, and antioxidant activities with extraction cost.

The extracts of Pinus densiflora sawdust by n hexane, ethyl acetate and methanol solvent were investigated to identify their mycelial growth inhibition against Lentinus edodes. The yields of n hexane soluble fraction, ethyl acetate-soluble fraction, and methanol soluble fraction from P. densiflora sawdust were obtained 1.36%, 2.21% and 4.03% using organic solvent, respectively. The mycelial growth inhibition of L. edodes was the greatest for n hexane extract, ranging from 36.5% to 47.6% at concentrations of 125 ppm to 1,000 ppm, with the values for all concentrations significantly different from one another. After direct extraction of P. densiflora sawdust using n hexane, ethyl acetate and methanol, each extract was separated into three fractions by silica gel column chromatography and then the fractions were isolated on the values of $R_f$ by thin layer chromatography. The mycelial growth inhibition against L. edodes was recognized in the fractions II (33.5%) and III (37.6%) of n hexane extract, the fraction II (21.4%) of ethyl acetate extract and the fraction II (26.4%) of methanol extract. The fractions III of n-hexane extract showed the highest growth inhibition among the nine fractions of the organic solvent extract.
Objective: The aim of this study is to optimize the extraction of beneficial substance from Cudrania tricuspidata leaves grown at Jirisan Mountain in South Korea by three different solvents depending on extraction time and at different temperature. Methods: The total phenolic contents were determined by the method reported by $S{\acute{a}}nchez$-Moreno et al. The total flavonoid contents were analyzed by Slinkard and Singleton. The DPPH radical scavenging activity was determined according to the method reported by Blois Results: The extraction yield for each solvent is 9.05-14.1%, 2.17-5.67%, and 2.3-3.9% for D.W., ethanol, and hexane, respectively. The overall results were maximized for the extract obtained with D.W. for 5 min at $100^{\circ}C$. The average phenol contents were 77.11, 45.64, and 0.343 mg/g at $100^{\circ}C$ in water, $78^{\circ}C$ in ethanol, and $68^{\circ}C$ in hexane, respectively. The flavonoid contents were the highest in the materials extracted with D.W., and were increased with increasing temperature, regardless of the extraction solvents, whether water (green), polar organic ethanol, or nonpolar organic hexane. In the ethanol extract, the flavonoid contents are increased gradually from 5.66 mg/g to 7.73 mg/g. The total flavonoid contents were proportional to the concentrations of the water extracts, ranging from 4.14 mg/g to 48.89 mg/g. The antioxidative activities of the water-extracted compounds are generally increased with increasing temperature from 42.5% to 85.5%. Those of the hexane extracts are increased slowly from 3.79% to 8.8%, while those of ethanol extracts are increased from 29.8% to 47.4%. Conclusion: The extraction yields were dependent upon solvents for extraction as well as extraction time and the temperature. The optimal extraction time was 5 min and the extraction yields were increased with increasing temperature excepted hexane. Of the three tested extraction solvents, the greenest solvent of water shows excellent results, suggesting that water is among the most effective solvents for natural sample extractions for general medicinal, pharmaceutical, and food applications.
Sabaek-san has been used for the treatment of inflammatory diseases derived from the cold with high fever, cough, and lung dysfunction in Korea and China. There is no study for the comparison between different solvent extracts of Sabaek-san. We made two samples, one is Sabaek-san water extract (SBSW) and the other is Sabaek-san 30% EtOH extract (SBSE). Both extracts inhibited inducible nitric oxide synthase(iNOS) protein, reduced iNOS-derived nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Also, they reduced tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-$1{\beta}$ (IL-$1{\beta}$) production. These anti-inflammatory effects caused by induction of heme oxygenase (HO)-1. HO-1 enzyme plays an important role of cellular anti-oxidant and anti-inflammatory systems. The induction of HO-1 is primarily regulated at the transcriptional level, and its induction by various inducers is related to the nuclear transcription factor-E2-related factor 2 (Nrf2). However, it is worth taking note that SBSE has more powerful anti-inflammatory effects than SBSW. In this study we suggest that different solvent extraction makes different therapeutic actions.
Eurya emarginata (Thunb.) Makino (Theaceae) is distributed in coastal areas of island. The leaves of Eurya are used in the traditional medicine of the coastal areas of jeju island with the aim of diuresis or to treat ulcers. Nevertheless, there are few reports on the biological activity and constituents of E. emarginata. In this study, we investigated the pharmacological activity of the solvent extracts of E. emarginata on the several inflammatory markers (TNF-$\alpha$, IL-1$\beta$, IL-6, NO, iNOS and COX-2). Also we examined the antioxidizing effect of the solvent extracts by determination of DPPH radical-scavenging activity. Among the solvent fractions, EtOAc and BuOH extracts showed potent radical scavenging activity (RC$_{50}$=10.9 and 12.7 respectively). The subtractions of EF 5-4-6-3-2 and BF 1 potentially inhibited the mRNA expression of pro-inflammatory cytokines (IL-1$\beta$, IL-6 and TNF-$\alpha$) at the concentration of 100 $\mu\textrm{g}$/mι. Also the fractions inhibited the mRNA expression of pro-inflammatory cytokines (IL-1$\beta$, IL-6 and TNF-$\alpha$) and protein expression of iNOS and COX-2 at the concentration of 100 $\mu\textrm{g}$/mι. And then, the inhibition of iNOS was correlated with the decrease of nitrite level. These results suggest that E. emarginata may have anti-inflammatory activity through the inhibition of pro-inflammatory cytokines, iNOS and COX-2.2.
Centella asiatica (CA) is a vegetable that has been used for medicinal purposes for a long time, but it is not well known in Korea. In this study, the approximate analysis, physicochemical properties, free sugars, free amino acids and minerals of native and improved CA cultivated in Chungju area were measured. And the antioxidant contents and activities of CA solvent extracts were measured. There was no significant difference between native and improved species. As for the characteristics of CA, it was confirmed that the improved species had a large weight, size and a tough texture. Glucose was detected in native CA, fructose, glucose and sucrose were detected in improved species. A total of 15 free amino acids were detected and the content was slightly higher in improved species than in native species. The mineral content in CA was slightly higher in the improved species, and the detected minerals were Na, P, Ca, K, Mg and Fe. Polyphenols, ascorbic acid and flavonoids in each extraction solvent (water, 70% and 100% ethanol) were all higher in the improved species than in native species. And it was found that the antioxidant activities of ethanol extracts were higher than that of water.
우리는 섬오갈피의 70% 에탄올과 열수추출물의 지표성분인 Acanthoic acid과 Kaurenoic acid 함량을 확인하기 위해 시료 내 각 화합물의 정량분석을 HPLC-UVD로 실시하였다. 또한, LC-MS 분석을 통해 시료 내 각 화합물의 성분을 확인하였다. 그 결과, 70% 에탄올 추출물의 Acanthoc acid와 Kaurenoic acid의 함량은 각각 28.84±0.21 mg/g(2.88%), 26.38±1.63 mg/g(2.64%)로 나타났다. 그러나 열수추출물에서는 두 가지 화합물의 Acanthoc acid와 Kaurenoic acid의 함량이 관찰되지 않았다. 결론적으로 섬오갈피 추출물로부터 Acanthoc acid과 그 대사산물을 추출하기 위한 최적의 추출용매로서 70% 에탄올이 열수용매보다 우수함을 알 수 있었다. 또한 DPPH 라디컬 소거능, total polyphenols, flavonoids 함량이 삼채+섬오갈피 복합추출물의 70% 에탄올 복합 추출물이 열수용매보다 더 우수한 효과를 보였다. 삼채와 섬오갈피 등 에탄올복합추출물(SEC) 처리에 의해 유의적 또는 부분적으로 항염증을 감소되었다.
본 연구에서는 전처리 조건(음건, 숙성, 덖음)과 추출 용매 중 에탄올의 혼합비율(30, 50, 70, 90%)에 따른 섬애약쑥 추출물의 이화학적 특성과 항산화 활성을 비교하여, 최적의 전처리 조건 및 추출용매를 확인하고자 하였다. 추출물의 수율은 음건쑥추출물에 비해 숙성쑥과 덖음쑥 추출물이 더 높았고, 에탄올 농도에 따른 경향은 확인되지 않았다. 색도는 L, a, b값 모두 음건쑥 추출물에서 가장 높았고 열처리 시간이 가장 길었던 숙성쑥 추출물에서 유의적으로 낮았다. 가용성 고형분 함량은 덖음쑥, 숙성쑥, 건조쑥 순으로 높고 에탄올의 비율이 증가할수록 그 함량 또한 증가하였다. 총 페놀, jaceosidin, eupatilin 및 총 플라보노이드 함량 또한 덖음쑥, 숙성쑥, 건조쑥 순으로 높았으며, 전반적으로 50%와 70% 에탄올 추출물이 30%와 95% 에탄올 추출물에 비해 유의적으로 높은 함량이었다. DPPH radical 소거 활성과 ABTS 라디칼 소거활성은 덖음쑥 추출물이 음건쑥과 숙성쑥 추출물에 비해 높았는데, DPPH radical 소거 활성은 덖음쑥 70% 에탄올 추출물이 77.71%로 유의하게 가장 높았으며, ABTS radical 소거 활성은 덖음쑥 30%, 50% 및 70% 에탄올 추출물이 96% 이상의 높은 활성을 나타내었다. 이상의 결과로부터 섬애약쑥은 음건한 것을 덖음 처리를 한 후 50%~70% 에탄올로 추출할 때 유효성분의 함량과 라디칼 소거활성이 높음을 확인하였다.
