Effect of Different Nutrient Solution and Light Quality on Growth and Glucosinolate Contents of Watercress in Hydroponics (배양액의 종류 및 광질이 물냉이의 생육 및 Glucosinolate 함량에 미치는 영향)

  • Aim of this study was to investigate the effects of different nutrient solutions and various light qualities generated by LED on the growth and glucosinolates contents of watercress (Nasturtium officinale) grown under hydroponics for 3 weeks. The seeds of watercress were sown on crushed rockwool media and raised them for two weeks. They were transplanted in a semi-DFT (deep flow technique) hydroponics system. A controlled-environment room was maintained at $20{\pm}1^{\circ}C$ and $16{\pm}1^{\circ}C$ temperatures and $65{\pm}10%$ and $75{\pm}10%$ relative humidity (day and night, respectively), with a provided photosynthetic photon flux density (PPFD) of $180{\pm}10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and a photoperiod of 16/8h. To find out the best kinds of nutrient solutions for growing watercress, Otsuka House 1A (OTS), Horticultural Experiment Station in Korea (HES), and Netherland's Proefstaion voor Bloemisterij en Gasgroente (PBG) were adapted with initial EC of $1.0-1.3dS{\cdot}m^{-1}$ and pH of 6.2, irradiating PPFD with fluorescent lamps (Ex-1). Either monochromatic (W10 and R10) or mixed LEDs (R5B1, R3B1, R2B1G1, and W2B1G1) were irradiated with a differing ratio of each LED's PPFD to understanding light quality on the growth and glucosinolates contents of watercress (Ex-2). Although significant difference in the shoot growth of watercress was not found among three nutrient solutions treatments, but the root fresh weight increased by 13.7% and 55.1% in PBG and OTS compared to HES, respectively. OTS increased the gluconasturtiin content by 96% and 65% compared to PBG and HES. Compared with the white light (W10), the red light (R10) showed a 101.3% increase in the shoot length of watercress. Increasing blue light portion positively affected plant growth. The content of total glucosinolates in watercress was increased by 144.5% and 70% per unit dry weight in R3B1 treatment compared with R2B1G1 and W10 treatments, respectively. The growth and total glucosinolates contents of the watercress were highest under R3B1 among six light qualities.

  • The genetic defects in human gametes and embryos can cause adverse effects on overall reproductive events. Biopsy of embryos for preimplantation genetic diagnosis (PGD) offers a new possibility of having children free of the genetic disease. In addition, advanced embryo culture method may enhance the effectiveness of embryo biopsy for the practical application of PGD. This experimental study was undertaken to evaluate the effects of coculture on the development in vitro of biopsied mouse embryos as a preclinical model for PGD of human embryos. Embryos were obtained after in vitro fertilization (IVF) from F1 hybrid mice (C57BLfemale/CBAmale). Using micromanipulation, 1, 2, 3 or 4 blastomeres of 8-cell stage embryos were aspirated through a hole made in the zona pellucida by zona drilling (ZD) with acidic Tyrode's solution (ATS). After biopsy of blastomeres, embryos were cultured in vitro for 110 hours in Ham's F-10 supplemented with 0.4% BSA or cocultured on the monolayer of Vero cells in the same medium. The frequence of blastocyst formation were recorded, and the embryos beyond blastocyst stage were stained with 10% Giemsa to count the total number of nuclei in each embryo. There was no significant difference in the blastocyst formation between the zona intact control group and the zona drilling (ZD) only, or biopsied groups. The hatching rate of all the treatment groups except 4/8 group was significantly higher than that of control group. In all the treatment groups, there was a significant reduction in the mean cell number of embryos beyond blastocyst stage ($50.2{\pm}14.0$ in control group vs. $41.2{\pm}7.9$ in ZD, $39.3{\pm}8.8$ in 7/8, $29.7{\pm}6.4$ in 6/8, $25.1{\pm}5.7$ in 5/8, and $22.1{\pm}4.3$ in 4/8 groups, p<0.05). When the same treatments were followed by coculture with Vero cells, a similar pattern was seen in the blastocyst formation and the hatching rate. However, in all the treatment groups, there was a significant increase in the mean cell number of embryos beyond blastocyst stage with coculture, compared with the parallel groups without coculture. In the cleavage rate of biopsied blastomeres cultured for 110 hours after IVF, there was no significant difference between coculture and non-coculture groups (87.2% vs. 78.7%). However, the mean cell number of embryos developed from the biopsied blastomeres was significantly higher in coculture group ($11.5{\pm}4.7\;vs.\;5.9{\pm}1.9$, p<0.05). In conclusion, biopsy of mouse embryos after ZD with ATS is a safe and highly efficient method for PGD, and coculture with Vero cells showed a positive effect on the development in vitro of biopsied mouse embryos and blastomeres as a preclinical model for PGD of human embryos.

  • The purpose of the study is to evaluate the degree of the marginal leakage of class V cavities with 4 brands of esthetic filling materials by means of the dye penetration at the enamel & dentinal margins. 160 cavities of class V were prepared on the buccal & lingual surfaces of 80 extracted premolar teeth, which divided into 4 groups. The four groups of cavities were filled with Durafill$^{(R)}$-Durafill bond$^{(R)}$(KuIzer & Co GmbH), Bisfil M$^{(R)}$-Resin bond$^{(R)}$(BISCO Inc.), Silux$^{(R)}$-Scotchbond$^{(R)}$(3M Co.) and Fuji ionomer$^{(R)}$(type II, G-C Co.) each. All specimens were immersed in 2% methylene blue dye solution for 24 hours at $37^{\circ}C$ after a 30-second thermocycling at $4^{\circ}C$ and $60^{\circ}C$ and longitudinally sectioned with diamond disk into two parts. The results were as follows : 1. At enamel margins, the group filled with Durafill$^{(R)}$-Durafill bond$^{(R)}$, Bisfil M$^{(R)}$-Resin bond$^{(R)}$ and Silux$^{(R)}$-Scotchbond$^{(R)}$ show no significant difference each other(p>0.05), and the above groups show less marginal leakage comparing with the group filled with Fuji ionomer$^{(R)}$(p<0.01). 2. At dentinal margins, the group filled with Silux$^{(R)}$-Scotchbond$^{(R)}$ or Fuji ionomer$^{(R)}$ show less marginal leakage than that with Durafill$^{(R)}$-Durafill bond$^{(R)}$ or Bisfil M$^{(R)}$-Resin bond$^{(R)}$(p<0.01). 3. The enamel margins show less marginal leakage than dentinal margins in all the class V cavities(p<0.01).

Butachlor and Silicate Fertilizer Interaction to Rice (Butachlor와 규산질비료(珪酸質肥料)의 수도(水稻)에 대(對)한 상호작용(相互作用))

  • Pot and laboratory tests were undertaken to investigate the influence of silicate fertilization on butachlor phytotoxicity to rice. Growth of rice seedlings at 150 ppm of $SiO_2$ was stimulated, while adverse effect was observed over 300 ppm of $SiO_2$ and growth reduction was enhanced with combination of butachlor and $SiO_2$ Rice growth in pot trial at 150kg/10a of silicate fertilization was not influenced by recommended amounts of butachlor and nitrofen, however, the growth of Seokwang byeo at 300kg/10a of silicate was markedly retarded by butachlor in the initial stage of growth. Growth reduction of Seokwang byeo caused by combined application of silicate and butachlor was recovered 50 days after herbicide application. Growth reduction from butachlor was not influenced by pH level and also degradation behaviors of butachlor in submerged soil was not altered by silicate fertilization. Adsorbed amount of butachlor on rice root was increased with addition of $SiO_2$ and its amount in Seokwang byeo was higher than that of Jinju byeo. Butachlor absorption by Seokwang byeo was accelerated by 150 ppm of $SiO_2$ applied simultaneously, but those effect was not encountered in Jinju byeo. Butachlor absorption of rice seedlings was also increased by 150 ppm of $K_2O$, while CaO hindered the absorption and $Na_2O$ had no effect on the absorption. Residual level of butachlor in Seokwang byeo treated with combined solution of butachlor and $SiO_2$ was continued higher than that with butachlor alone during 10 days after transplantation to culture solution.

  • Rare earth elements(REEs) have been used as an useful tool in understanding the various geological processes such as evolution and differentiation in the crust. The REEs also have been used as an analog of actinides for radioactive wastes at the water-rock interactions. Using physicochemical properties of the REEs and actinides, we have shown that Eu is an optimum analogue for understanding the behavior of Am in subsurface environments. Factors affecting sorption behavior of radioactive nuclides in groundwater were investigated by batch experiments. Four nuclides such as $^{241}Am,\;^{152}Eu,\;^{160}Tb\;and\;^{60}Co$ were selected to test our hypothesis, and $^{160}Tb$ and $^{60}Co$ were specifically used to compare to the sorption behavior between $^{241}Am-^{152}Eu$ and other radioactive nuclides. Four different rock samples and one groundwater were used in the batch experiments where solution pH for all experiments was fixed at 5.5. Our results demonstrate that $^{241}Am,\;^{152}Eu,\;and\;^{160}Tb$ show similar sorption behavior whereas $^{60}Co$ is different in sorption behavior at the mineral-water interface, suggesting that the sorption behavior of $^{60}Co$ is affected by different rock types. Our results also show that 1) Eu in REEs is optimum analogue of fate and transport of Am in subsurface environments, and 2) mineral compositions such as $SiO_2,\;TiO_2,\;P_2O_5$ and distribution of REEs such as Eu anomaly play key roles in affecting sorption behavior of radioactive nuclides even though physicochemical properties of geological materials such as specific surface area and cation exchange capacity can not be ruled out.

Proliferation Assay of Splenocyte and PBMC by the Evaluation of Alamar Blue Dye Reduction Value in Broiler Chicks (Alamar Blue 색소의 환원량 평가에 의한 급성기 반응중 육계병아리의 비장세포와 PBMC 증식도 측정)

  • In this study, hatched male broiler chicks(Ross) were fed on a basal diet and LPS was administered via intraperitoneal injection three times every other day, on the 9th, 11th and 13th days of the experiment, and then PBMC and splenocytes were isolated on day 14. The degree of alama blue reduction was evaluated at 4, 24, 48, 96 and 120 h in the splenocytes, and at 4, 8, 12, 24 and 48 h for PBMC of incubation after the addition of alama blue solution to the media. The cell numbers used in this experiment were 103, 104 and 105 cells per well, and the con A levels were 0.0, 1.0, 5.0, and 10.0 ㎍ per ml of medium. 1. The degree of alama blue reduction was found to increase in a linear fashion with increasing incubation time and cell numbers, for both splenocytes and PBMC. 2. During acute phase response, the degree to which alama blue was reduced was significantly elevated (p<0.05) at an incubation time of 24 hr for the splenocytes, 4 hr for PBMC, and a cell number of 105 cells per well, respectively. 3. The raised reduction of alama blue to control was linear with Con A levels in medium, and higher reduction in Con A 10.0 ㎍ relative to 1.0 or 5.0 ㎍ in ml medium was shown 4. The medium with autologous serum evidenced a significantly (p<0.05) higher reduction of alama blue relative to FBS. 5. Splenocytes and PBMC from the LPS-injected birds evidenced significantly higher levels of alama blue reduction regardless of incubation time, number of cells, level of Con A added, or serum type, as compared with what was observed in normal birds. The results indicated that the assay conditions for proliferative activity using the alama blue method in birds in which the acute phase response had been activated via intraperitoneal LPS injection requires 4 hrs of incubation for PBMC, 24 hrs of incubation for splenocytes, and 10㎍ of Con A per ml of medium.

Effect of Extracting Conditions on Chemical Compositions of Korean Mountain Ginseng Extract (추출조건이 장뇌삼추출물의 화학성분 조성에 미치는 영향)

  • Korean mountain ginseng roots were freeze-dried at $-70^{\circ}C$ and extracted by different extracting solution conditions to investigate chemical compositions of extracts. The soluble solid content of the extract from $7.04\~13.45\%\;and\;50\%$ EtOH and MeOH extracts were higher than those of other extracts. $100\%\;water\;and\;90\%$ EtOH extracts gave the highest Brix with $19.98\%$\;and\;19.65\%$, respectively. pH of the extracts were ranged from $5.82\~6.60$. Browning color at 470 nm of the extract were high value in 50$\%$ EtOH extract. In case of Hunter's color value, L value of extract was higher in $100\%$ water extract (21.28) than EtOH extract $(17.18\~21.02)$, a and b values of extract were the highest in $100\%$ water (-0.12) and $90\%$ MeOH extract (1.34). The contents of free sugars in the EtOH extract were increased with the ethanol concentration. Sucrose contents of $90\%$ EtOH and MeOH extracts were 6,159 mg/100 g and 5,238 mg/100 g. Major organic acids of the extract were citric and malic acids. Major free amino acids of the extract were L-arginine, L-proline, $\gamma$ -amino-n-butyric acid, alanine and aspartic acid. The highest ginsenoside content was shown to be about $10.50\%\;in\;90\%$ MeOH extract. Major minerals of extract were P, K, Na, Mg and Ca.

With fertilizer ingredients in pig sludge chemical qualities, the content of the nitrogen and the phosphoric acid comes 4.4% to be 6.29%, pH 7.02 and content of the copper and the zinc which is a heavy metal which contains in pig sludge with 805 mg/kg and 1,704 mg/kg, it is a restrictive standard of the fertilizer, 300 mg/kg and 900 mg/kg it sees to be high, it manufactures citric acid 1 hydrate with the organic acid solution, heavy metals of pig sludge where it is a mixture ratio of the organic acid solution, it divides to 25%, 50%, 75% and 100% 4 kind levels, the result which measures the heavy metal exclusion ratio of the copper and the zinc, the mixture ratio of the organic acid solution to be many exclusion ratio of the copper and the zinc is showing a just interrelation, from organic acid solution 100% level content of pig sludge remains copper and zinc 330.03 mg/kg and 41.28 mg/kg, it shows the exclusion ratio of copper 59% zinc 97%. 'Cheonganok' growth characteristics with citric acid 1 hydrate, Treatment 2 and control growth characteristics etc, it exclusion the copper and the zinc it doesn't appear on significant difference statistically but, treatment 3 after only pig sludge in resources disposal where it seeding, growth characteristics of leaf area etc. is badness, it compares in control and treatment 2 the growth characteristics badness, it is appearing, it is caused by with disease and insects occurrence of $Ostrinia$ $furnacalis$ and brown spot, the damage was many. From forage value, Treatment 2 where it exclusion the heavy metal with the citric acid 1 hydrate with control it compares and there are not significant difference from crude protein and ADF and NDF contents etc., seeding only Pig Sludge in resources disposal treatment 3, it is caused by with $Ostrinia$ $furnacalis$ etc., trunk and aging of the leaf to be high ADF content is low. but from crude protein, the nitrogen ingredient which pig sludge has and interrelation it seemed and high numerical value were confirmed.

  • The development of Japanese geoduck, Panopea japonica, grown under culture conditions, has been examined through the morphological characteristics in fertilized egg, larvae and juvenile. Gametes were stripped from ripe broodstock and placed into two separate containers. Eggs were washed through a $40{\mu}m$ sieve and fertilized with dilute sperm solution. Developing larvae were maintained at $19{\pm}1^{\circ}C$. Fertilized eggs with $81.6{\mu}m$ diameter developed to trochophores within 14 h and to D-stage larvae ($116{\mu}m$ shell length) within 27 h. Larvae were spontaneously settled at shell length of $311{\mu}m$ after 20 days. The hatching from fertilized eggs and larval rearing were normally available in $18.5-21.5^{\circ}C$, and the growth was good in a cashmilon substrate, as well as sand. After rearing of day 108 from metamorphosis, the shell length of juvenile P. japonica reached 13 mm, and growth rate of shell length of the juvenile was $117.5{\mu}m/d$.

Bioequivalence of S-napine Tablet 10 mg to Alesion Tablet(Epinastine HCl 10 mg) (알레지온 정(염산에피나스틴 10mg)에 대한 에스나핀 정 10밀리그람의 생물학적동등성)

  • Epinastine is an antiallergic drug effective for bronchial asthma, allergic rhinitis, urticaria and dermatitis. Epinastine is topically active, direct H1-receptor antagonist and an inhibitor of the release of histamine from the mast cell. The purpose of the present study was to evaluate the bioequivalence of two epinastine hydrochloride tablets, Alesion Tablet (Boehringer Ingelheim Korea Ltd.) and S-napine tablet 10 mg(Sam Chun Dang Pharm. Co., Ltd), according to the guidelines of the Korea Food and Drug Administration(KFDA). The release of epinastine from the two epinastine formulations in vitro was tested using KP VIII Apparatus II method with various dissolution media(pH 1.2, 4.0, 6.8 buffer solution and water). Twenty six healthy male subjects, $23.35{\pm}1.57$ years in age and $66.29{\pm}10.61kg$ in body weight, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After two tablets containing 20 mg as epinastine hydrochloride was orally administered, blood was taken at predetermined time intervals and the concentrations of epinastine in serum were determined using HPLC with UV detector. The dissolution profiles of two formulations were similar at all dissolution media. In addition, the pharmacokinetic parameters such as $AUC_t,\;C_{max}\;and\;T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t.\;C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the reference drug, Alesion tablet, were 1.50, 1.46 and -13.48% for $AUC_t,\;C_{max}\;and\;T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log 0.8 to log 1.25(e.g., log 0.95$\sim$log 1.12 and log 0.93$\sim$log 1.10 for $AUC_t\;and\;C_{max}$, respectively). Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating S-napine tablet 10 mg was bioequivalent to Alesion tablet.
