Soluble CD30: A Possible Serum Tumor Marker for Primary Effusion Lymphoma

  • Background: The serum level of soluble CD30 (sCD30) is known to be increased with several lymphomas and to correlate with prognosis. Primary effusion lymphoma (PEL) is a highly aggressive malignant lymphoma with poor prognosis, but the existence and significance of sCD30 in PEL have not yet been investigated in detail. Objectives: Since the membrane type of CD30 is frequently expressed on the surface of PEL cells, we compared the expression of the membrane type of CD30 and the production of sCD30 among PEL cell lines as well as other lymphomas. Methods: The expression of surface CD30 in various lymphoma cell lines was analyzed with flow cytometry ans sCD30 was quantified by ELISA. Results: Both surface and sCD30 were detected on PEL cell lines as well as on Hodgkin's lymphoma and adult T-cell leukemia/lymphoma cell lines. Surface CD30 and sCD30 levels of each cell lines correlated with each other. Conclusion: The serum level of sCD30 appear to be a useful biological tumor marker for the diagnosis and management of CD30-positive PEL.

Plasma Soluble CD30 as a Possible Marker of Adult T-cell Leukemia in HTLV-1 Carriers: a Nested Case-Control Study

  • Elevated levels of soluble CD30 (sCD30) are linked with various T-cell neoplasms. However, the relationship between sCD30 levels and the development of adult T-cell leukemia (ATL) in human T-cell leukemia virus type 1 (HTLV-1) carriers remains to be clarified. We here investigated whether plasma sCD30 is associated with risk of ATL in a nested case-control study within a cohort of HTLV-1 carriers. We compared sCD30 levels between 11 cases (i.e., HTLV-1 carriers who later progressed to ATL) and 22 age-, sex- and institution-matched control HTLV-1 carriers (i.e., those with no progression). The sCD30 concentration at baseline was significantly higher in cases than in controls (median 65.8, range 27.2-134.5 U/mL vs. median 22.2, range 8.4-63.1 U/mL, P=0.001). In the univariate logistic regression analysis, a higher sCD30 (${\geq}30.2U/mL$) was significantly associated with ATL development (odds ratio 7.88 and the 95% confidence intervals 1.35-45.8, P = 0.02). Among cases, sCD30 concentration tended to increase at the time of diagnosis of aggressive-type ATL, but the concentration was stable in those developing the smoldering-type. This suggests that sCD30 may serve as a predictive marker for the onset of aggressive-type ATL in HTLV-1 carriers.

  • A mechanistic study by which Cadmium-tolerant P.Putida C1 accumulates high conc of Cd in its cell body was performed. Approximately 57% Cd accumulated was distributed on the cell wall and the other 43% portion was in cytoplasm. 84% Cd of the Cd in the cell wall fractions present in the polyphosphate-polysaccharide fractions, but most of Cd in the cytoplasm fraction was in protein and nucleic acid. Cadmium affected the protein synthesis in P. Putida. The intracellular protein content was decreased by cadmium addition, but the soluble protein precipitated by ammonium sulfate($30{\sim}75%$ satruation) was increased as compared to that from the cells grwon without cadmium. Furthermore, in the cells grown with of cadmium, high-molecular-weight soluble protein was increased, with of cadmium, high-molecular-weight soluble protein was increased, compared with the cells grown without cadmium, but low-molecular-weight soluble protein was decreased. These results indicate that Cd inhibited the intracellular protein biosynthesis but enhance biosynthesis of the high-molecular-weight soluble protein precipitate by ammonium sulfate($30{\sim}75%$ saturation).

A Study on the Heavy Metal Contents of Soil and Rice in the Kum River Basin (금강유역 논토양과 현미의 중금속 함량에 관한 연군)

  • This study was carried out to investigate the heavy metal contents and their correlationship between paddy soil and brown rice near the Kum-River area. In this study, eighty soil samples and forty brown rice samples were taken from the paddy soil. The contents of heavy metals were measured by flame atomic absorption spectrophotometry. The results were as follows: 1. The average contents of soluble heavy metals in surface soil were Cd 0.19, Cu 15.31, Zn 18.10 and Pb 9.08 ppm. The average contents of soluble heavy metals in subsurface soil were Cd 0.19, Cu 14.52, Zn 17.75 and Pb 8.11 ppm. There was no statistically significant difference between the two layers. 2. The contens of Cu, Zn and Pb of Taejeon(S6) and Cd of Sinbyung(S5) in surface soil were higher than those of other areas. The contents of Cd and Cu of Taejeon(S6) and Zn and Pb of Kumnam(S3) in brown rice were higher than those of other areas and four heavy metals in soil and brown rice of Simchon(S7) were lower than those of other areas. 3. The ratio of soluble contents(Cd:Cu:Zn:Pb) in surface soil was 1:79:93:47, that of soluble contents in subsurface soil was 1:76:94:43, and that of total contents in brown rice was 1:84:294:12. 4. The correlationship of the content between soluble heavy metals in surface(0-15 cm depth) soil total heavy metals in brown rice was found to be order of Cd>Cu>Zn>Pb. The correlationship of the content between soluble heavy metals in subsurface(20-30 cm depth) soil and total heavy metals in brownricewasfoundtobeorderofcu>Cd>Zn>Pb.

T Regulatory Cell Responses to Immunization with a Soluble Egg Antigen in $Schistosoma$ $mansoni$-Infected Mice

  • The aim of the study is to characterize the phenotypes of $CD4^+$ $CD25^+$ T regulatory cells within the liver granulomas and association with both Foxp-3 gene expression and splenic cytokines. Naive C57BL/6 mice were intravenously injected with multiple doses of the soluble egg antigen (SEA) 7 days before cercarial infection. The immunized and infected control groups were sacrificed 8 and 16 weeks post-infection (PI). Histopathology, parasitological parameters, splenic phenotypes for T regulatory cells, the FOXP-3 expression in hepatic granuloma using real-time PCR, and the associated splenic cytokines were studied. Histopathological examination of the liver revealed remarkable increase in degenerated ova within hepatic granuloma which decreased in diameter at weeks 8 and 16 PI ($P$<0.01). The percentage of T regulatory cells ($CD4^+$ $CD25^+$) increased significantly ($P$<0.01) in the immunized group compared to the infected control at weeks 8 and 16 PI. The FOXP-3 expression in hepatic granulomas increased from 10 at week 8 to 30 fold at week 16 PI in the infected control group. However, its expression in the immunized group showed an increase from 30 at week 8 to 70 fold at week 16 PI. The splenic cytokine levels of pro-inflammatory cytokines, IFN-${\gamma}$, IL-4, and TNF-${\alpha}$, showed significant decreases ($P$<0.05) compared to the infected control group. In conclusion, the magnitude and phenotype of the egg-induced effects on T helper responses were found to be controlled by a parallel response within the T regulatory population which provides protection in worm parasite-induced immunopathology.

Elimination of Heavy Metals(Pb, Cd) by Steaming and Roasting Conditions of Polygonatum odoratum Roots (둥굴레 근경의 증자 및 볶음조건에 따른 중금속(Pb, Cd) 제거 특성)

  • Response surface methodology(RSM) was applied to monitor the elimination rate of heavy metals(Pb, Cd) and soluble solids depending on the steaming and roasting conditions of Polygonatum odoratum roots. Experiments of 16 different steaming and roasting conditions based on a central composite design for steaming time($60{\sim}180$ min), roasting temperature($110{\sim}150^{\circ}C$), and roasting time($10{\sim}50$ min) were conducted, thereby predicting the steaming and roasting conditions for the maximal responses; soluble solids($71.47\%$) at 65.24 min, $126.93^{\circ}C$ and 37.58 min; Pb removal rate($18.87\%$) at 71.23 min, $119.81^{\circ}C$ and 24.35 min; Cd removal rate($50,85\%$) at 160.89 min, $126.43^{\circ}C$ and 15.81 min, respectively. The optimum conditions estimated by RSM for the maximized values of soluble solids and heavy metal elimination rates were $165{\sim}180$ min of steaming time, $120{\sim}135^{\circ}C$ of roasting temperature, and $30{\sim}45$ min of roasting time, respectively. These estimated values were in agreement with those measured by real experiments.

Effect of Dietary Fibers Isolated from Tangerine Peels on Lipid and Cadmium Metabolism in the Rat (감귤과피로부터 분리한 식이섬유가 흰쥐의 지방 및 Cadmium 대사에 미치는 영향)

  • This study was performed to investigate the effects of dietary fibers from tangerine peels on lipid and cadmium metabolism. And effects were compared with those of commercial dietary fibers($\alpha$-cellulose, citrus pectin). Sixty male rats of Sprague-Dawley strain weighing 186.7$\pm$2.6g were blocked into 12 groups according to body weight, and were raised for 2 weeks. Cadmium chloride was given at the levels of 0 of 400 ppm in diet. Various dietary fibers were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. In lipid metabolism, insoluble fibers[insoluble dietary fibers from tangerine peels(IDE), $\alpha$-celluolse] increased fecal excretion of lipids by inhcreasing feces weight, and decreased the concentrations of serum triglyceride and liver lipids. Soluble dietary fibers from tangerine peels(SDF) decreased the concentrations of serum cholesterol and liver lipids by increasing fecal lipids, too. In cadmium metabolism, soluble fivers(SDF, pectin) inhibited Cd absorption by increasing fecal Cd excretion and decreased Cd concentrations of intestion, liver and kidney. In conclusion, among the extracted fibers, SDF were more effective on lipid and Cd lowering activity and IDF had effect of increasing fecal lipid excretion. This result is useful to reduce food waste and utilize waste products.

Reaarding Effect of Dietary Fibers Isolated from Tangerine Peels on Glucose, Bile Acid, Cadmium transport In Vitro (감귤과피로부터 분리한 식이섬유의 포도당, 담즙산, 카드뮴 투과억제에 관한 In Vitro 연구)

  • Retarding effects of the dietary fibers from tangerine peels on glucose, bile acid and cadmium transport were evaluated by dialysis method, and were compared with those of commercial dietary fibers(citrus pection, CM-cellulose, guar gum, $\alpha$-cellulose). Yields of total (TDF), insoluble(IDF) and soluble dietary fibers(SDF) from tangerine peels on the fresh matter basis were 2.84%, 1.95% and 0.39% respectively. The amount of insoluble fibers was 5.2 times higher than that of soluble fibers. Soluble fibers(guar gum, CM-cellulose, SDF, pectin) had the retarding effect on glucose transport, while IDF, TDF and $\alpha$-cellulose did not have. Guar gum showed the greatest effect, followed by CM-cellulose, SDF and pectin. Among the extracted fibers, only SDF had the effect on glucose transport retardation. Regarding bile acid dialysis, guar gum had the greatest retarding effect, and all dietary fibers from tangerine peels, especially SDF, showed the effect of bile acid retardation. On cadmium transport retardation, CM-cellulose had the greatest effect, followed by SDF, TDF, IDF, guar gum and pectin. Among the extracted fibers, SDF had the greatest effect on Cd trasport transport retardation. The extracted dietary fibers showed higher retarding effect on Cd transport than glucose and bile acid transport, and the effect of SDF was higher than IDF.

  • The stabilization of Aspergillus sp. $\alpha$-amylase was attained by modification with periodate-oxidized sol-uble starch. The pH stability of modified enzyme was increased at pH 3~4 and 9~11 in the presence of $\alpha$-cyclodextrin($\alpha$-CD) compared with that of native enzyme. Thermal stability of the modified enzyme was increased. After treatment at 6$0^{\circ}C$ for 30min the activity remained 20% for the enzyme modified at pH 9.7 in the presence of $\alpha$-CD and tested in the presence of $\alpha$-CD 10% for the enzyme modified at pH 9.7 in the presence of $\alpha$-CD 0% for the native enzyme. The native enzyme and modified enzyme showed one peak in HPLC. The substrate specificity of the modified enzyme was not changed in HPLC analysis of reaction product.

  • This study was carried out to investigate the effect of mine activity on paddy fields, agricultural water, and plants in the ruined Oksung Cu-Zn mine area. Soil samples collected from paddy fields adjacent to the Cu-Zn mine sites were sequentially extracted and determined the contents of heavy metals such as Cd, Zn, Cu, Pb, Cr and Ni. Distribution of exchangeable heavy metals in soils was 30.2% of Cd, 11.3% of Zn, 2.2% of Cu, 4.6% of Cr, 0.6% of Pb and 3.9% of Ni. Water soluble heavy metals were only detected with Zn. The contents of heavy metals in water collected from the mine were 0.01 of Cd, 27.35 of Zn, 4.86 of Cu, 1.04 of Pb, 0.03 of Cr and $0.08mg\;L^{-1}$ of Ni. while the contents in waler collected out of the mine were 16.67 of Zn, 0.59 of Cu, 0.49 of Pb, 0.05 of Cr and $0.06mg\;L^{-1}$ of Ni. On the other hand, agricultural water near mine area were 1.26 of Zn, 0.05 of Cu, 0.05 of Pb and $0.02mg\;L^{-1}$ of Ni. Both Cd and Cr were not detected in the agricultural water.
