Biogas potential estimation for mono- and co-digestion of cow manure and waste grass (우분뇨와 폐잔디의 단독 및 병합소화 잠재량 평가)

  • Biogas production potential was experimentally estimated for mono- and co-digestion of cow manure and waste grass. The two organic wastes were mixed at five different ratios (100:0, 75:25, 50:50, 25:75, 0:100) on the volatile solids basis, and were assessed using biochemical methane potential (BMP) test. Thee reaction temperatures, 25℃, 30℃ and 35℃, were applied as well, resulting in 15 different combinations for the test. The results showed that both higher temperature and waste grass mixing ratio resulted in higher methane yield and maximum methane production rate. Based on the experimental results, a theoretical farm- or community-scale (240 or 2400 ㎥) anaerobic digester was designed to evaluate the energy balance associated with mono- and co-digestion of the wastes at different temperatures. Although the energy production increased as the temperature and the waste grass mixing ratio increased, the net energy gain, energy production subtracted by energy consumption for heating and maintenance, was estimated to be the highest at 30℃, followed by at 35℃ and 25℃. Therefore, it is advised that both the experimental methane production and the detailed design parameters must be considered for the optimization of the net energy gain from these wastes.

Effect of Grafting Methods on Plug Seedling Quality, Growth after Transplanting and Yield of Oriental Melon (Cucumis melo L. var. makuwa Mak.) (참외 접목방법이 플러그 묘 소질과 정식 후 생육 및 수량에 미치는 영향)

  • This study was conducted to investigate the effect of grafting methods on plug seedling quality, growth after transplanting, and yield of oriental melon that those seedlings were raised in the 32 hole plug tray for protected cultivation. Number of leaf and leaf area in approach grafting were higher until 15 days after grafting but lower on 20 days after grafting. Plant height, stem diameter and content of chlorophyll were nearly the same of each grafting methods on 20 days after grafting. Growth of top part, that is, plant height, leaf number, and leaf area on root removed single cotyledon ordinary splice grafting was the highest among the grafting methods. In the underground part, dry weight of root was high and T/R ratio was the lowest. Seedling quality of root removed single cotyledon ordinary splice grafting was the highest among grafting methods before transplanting. Plant height, number of leaf, leaf area and dry weight of root removed single cotyledon ordinary splice grafting in the growth of field were the highest on 30 days after grafting among grafting methods and days required for first flowering was also shorter, 38.4 days among grafting methods. But Fruit weight, content of soluble solids, fruit hardness, and color characteristics in fruit quality were insignificant among each grafting methods. Consequently, grafting methods influenced on the seedling quality, of oriental melon early growth after transplanting and yields. It was concluded that root removed single cotyledon ordinary splice grafting was the best methods in present study. It will be needed to convert grafting methods. Thus new grafting method should be applied.

Effects of Feeding TMR Feed with Dietary Citrus Byproducts on Chemical Properties of Raw Milk from Holstein Cow's Raw Milk (원유의 이화학적 특성에 미치는 감귤 부산물 TMR 사료 급여의 영향)

  • In this study, the effects of feeding providing Holstein cows with with TMR feed including amended with undried citrus byproducts on the properties of their raw milk were investigated. Two samples were used for the experiment: T0 (raw milk produced by dairy cows fed with TMR feed not including citrus byproducts) and T1 (raw milk produced by cows fed with TMR feed including citrus byproducts). The All experiments were conducted with Holstein cows at a dairy farm in the on Jeju island, and were repeated three times, in each, after of which raw milk obtained from 7 cows of each samplein each group was analyzed[ED highlight - please ensure this is correct]. The daily milk yield and somatic cell numbers of T0 and T1 were 24.16 kg and 25.97 kg, and 660,000 thousands and 445,000 thousands, respectively, which means that feeding citrus byproducts to cows increases daily milk yield and reduces somatic cell numbers. There was were no significant differences between T0 and T1 in terms of the raw milk's total plate count, specific gravity, titration acidity, and or the amount of milk protein, lactose, nonfat solids, free amino acids and volatile compounds, while raw milk in T1 showed significantly lower levels of milk fat in raw milk. The antioxidant activity of raw milk was improved by citrus byproducts TMR feed containing citrus byproducts.

Effects of Rumen Undegradable Protein and Minerals Proteinate on Early Lactation Performance and Ovarian Functions of Dairy Cows in the Tropics

  • A 90 d study was designed to investigate the effects of rumen undegradable protein (RUP) and a mixture of Cu, Zn and Mn proteinate (CZMP) on milk yield and composition and ovarian functions during rainy months. Twenty four Holstein${\times}$ indigenous cows in their 2nd and 3rd lactation were randomly allocated to total mixed rations (TMR) containing soy bean meal (SBM) as a source of rumen degradable protein (RDP), SBM plus CZMP, and formalin treated SBM (FSBM) as a source of RUP. Maximum and minimum temperature humidity index during the experimental period were 83.6-84.7 and 75.4-76.1. There were no differences (p>0.05) in intakes of dry matter, crude protein and net energy and in contents of butterfat, lactose and minerals. Cows on TMR containing FSBM not only lost less weight (-278, -467 and -433 g/d) with more intake of RUP (0.92, 0.58 and 0.59 kg/d) but also produced more milk (19.27, 18.23 and 18.13 kg/d) and 4% fat corrected milk (18.57, 17.57 and 17.51 kg/d) with more protein (3.06, 2.81 and 2.80%), solids-not-fat (8.69, 8.38 and 8.38%) and less milk urea N (9.3, 15.4 and 15.0 mg/dl) compared with those on TMR containing SBM and SBM+CZMP, respectively (p<0.01). However, cows on TMR containing SBM and SBM+CZMP did not differ in these respects (p>0.05). Whereas incidence of cystic ovaries at 20 and 90 d pospartum was less (p<0.01) in cows on TMR containing SBM+CZMP (37.3 and 12.5%) than those on TMR containing SBM (62.5 and 25%), it was nil for cows on TMR containing FSBM. Cows in all three group differed (p<0.01) from each other for the recurrence of first observed estrus with those on TMR containing FSBM having least days (22, 36 and 47 d) compared with their counterpart on TMR containing SBM+CZMP and SBM, repectively. The results suggest that RUP is one of the limiting factors affecting milk yield and its composition and ovarian functions during early lactation of dairy cows in the tropics.

Use of Leucaena leucocephala and Gliricidia sepium as Nitrogen Sources in Supplementary Concentrates for Dairy Goats Offered Rhodes Grass Hay

  • A study was conducted to evaluate the replacement value of Leucaena leucocephala and Gliricidia sepium as nitrogen sources in commercial type supplements for dairy goats. Six crossbred $(Toggenburg{\times}Saanen)$ goats at late stage of lactation were allocated to three dietary treatments in a double $3{\times}3$ Latin square design. The animals were offered rhodes grass (Chloris gayana) hay ad libitum and supplemented with either Leucaena-based concentrate (LBC), Gliricidia-based concentrate (GBC) or commercial based concentrate (CC). Voluntary food intake, milk yield and composition and changes in live weight were measured. The total dry matter (DM) intake was higher (p<0.05) in goats fed GBC than CC (1385 vs 1331 g/d). The DM intake for LBC (1343 g/d) was similar to CC (1331 g/d). The DM intake of hay was also higher (p<0.05) in goats fed GBC (834 g/d) than those receiving LBC or CC (789, 782 g/d, respectively). Animals supplemented with GBC recorded positive (11 g/d) weight gain while the other groups lost weight (13, 19 g/d) for LBC and CC respectively, although these differences were not statistically significant (p>0.05). The composition of milk were: butterfat 58, 49 and 55 g/kg; crude protein 37.0, 35.4 and 36.1 g/kg; lactose 33, 29 and 30 g/kg; Ash 8.5, 8.5 and 7.9 g/kg and total solids 136.5, 121.9 and 129.0 g/kg, for goats fed LBC, GBC and CC respectively. There were no differences in the composition of milk due to these dietary treatments. At the end of performance trial, a digestibility trial was conducted using 6 female goats allocated to the three treatments (LBC, GBC and CC) in an incomplete randomized block design and each goat received a different supplement in each of two successive periods. There were no differences in nutrient digestibility except for DM, which was higher (p<0.05) in CC compared to the other treatments (615, 622, 720 g/kg for LBC, GBC and CC, respectively). Economic analysis showed that CC diet was more expensive (0.20 US$/kg) and had a lower margin over supplementation (0.11 US$) compared to LBC and GBC (0.13 vs 0.12 US$/kg and 01.5 vs 0.12 US$, respectively). It is concluded that the Leucaena and Gliricidia could contribute as nitrogen sources in compounded diet supplements without any detrimental effects on production in dairy goats.

Anaerobic Digestion of Fish Offal(II) : Evaluation of Biodegradability Using Biochemical Methane Potential (생선 폐기물의 혐기성 소화 처리(II) : Biochemical Methane Potential을 이용한 생분해도 평가)

  • It is essential to understand the decomposition characteristics for developing the optimum anaerobic digestion system of organic wastes. In this study, BMP (Biochemical Methane Potential) test using serum bottle was conducted to evaluate the anaerobic degradability of fish offal. 3 different groups of fish offal including waste from mackerel and hairtail handling except viscera and fish viscera were chosen for the substrates. Grinded fish offal was transferred anaerobically to serum bottle in amounts of 50 ml, 100 ml and 150 ml, respectively. BMP test was carried out in triplicate. Cumulative methane production and methane production rate depending on incubation time were evaluated. These results varied depending on substrate characteristics. The average values of ultimate methane yield ranged between $420ml{\cdot}CH_4/g{\cdot}VS$ and $490ml{\cdot}CH_4/g{\cdot}VS$, and the methane production and degradation rate of viscera were higher than those of other parts of fish offal. According to the analysis of elemental composition, average C/N ratio of fish offal used in this study was 5.2. Theoretical ultimate methane yield calculated from elemental composition was $522ml{\cdot}CH_4/g{\cdot}VS$. Biodegradability was calculated as 0.847.

The New X-ray Induced Electron Emission Spectrometer

  • The new spectrometer for X-ray Induced Electron Emission Spectroscopy (XIEES) .has been recently developed in KRISS in collaboration with PTI (Russia). The spectrometer allows to perform research using the XAFS, SXAFS, XANES techniques (D.C.Koningsberger and R.Prins, 1988) as well as the number of techniques from XIEES field(L.A.Bakaleinikov et all, 1992). The experiments may be carried out with registration of transmitted through the sample x-rays (to investigate bulk samples) or/and total electron yield (TEY) from the sample surface that gives the high (down to several atomic mono-layers in soft x-ray region) near surface sensitivity. The combination of these methods together give the possibility to obtain a quantitative information on elemental composition, chemical state, atomic structure for powder samples and solids, including non-crystalline materials (the long range order is not required). The optical design of spectrometer is made according to Johannesson true focusing schematics and presented on the Fig.1. Five stepping motors are used to maintain the focusing condition during the photon energy scan (crystal angle, crystal position along rail, sample goniometer rail angle, sample goniometer position along rail and sample goniometer angle relatively of rail). All movements can be done independently and simultaneously that speeds up the setting of photon energy and allows the using of crystals with different Rowland radil. At present six curved crystals with different d-values and one flat synthetic multilayer are installed on revolver-type monochromator. This arrangement allows the wide range of x-rays from 100 eV up to 25 keV to be obtained. Another 4 stepping motors set exit slit width, sample angle, channeltron position and x-ray detector position. The differential pumping allows to unite vacuum chambers of spectrometer and x-ray generator avoiding the absorption of soft x-rays on Be foil of a window and in atmosphere. Another feature of vacuum system is separation of walls of vacuum chamber (which are deformed by the atmospheric pressure) from optical elements of spectrometer. This warrantees that the optical elements are precisely positioned. The detecting system of the spectrometer consists of two proportional counters, one scintillating detector and one channeltron detector. First proportional counter can be used as I/sub 0/-detector in transmission mode or by measuring the fluorescence from exit slit edge. The last installation can be used to measure the reference data (that is necessary in XANES measurements), in this case the reference sample is installed on slit knife edge. The second proportional counter measures the intensity of x-rays transmitted through the sample. The scintillating detector is used in the same way but on the air for the hard x-rays and for alignment purposes. Total electron yield from the sample is measured by channeltron. The spectrometer is fully controlled by special software that gives the high flexibility and reliability in carrying out of the experiments. Influence of Time of Hand-thining on Bitter Pit Incidence and Fruit Quality in 'Gamhong'/M.9 Apple Trees (인력 적과 시기가 '감홍'/M.9 사과나무의 고두증상 발생과 과실품질에 미치는 영향)

  • This study was conducted to investigate the effect of time of hand-thinning on vegetative growth, bitter pit incidence, fruit quality, and return bloom in 'Gamhong'/M.9 apple trees. The time of hand-thinning were 3, 5, 7 and 9 weeks after full bloom, and the primary thinning (leaving only the king fruit on cluster) and secondary thinning (adjusting crop load) were conducted at the same time. The time of hand-thinning was correlated to the vegetative growth, average fruit wight, yield, soluble solids content, bitter pit incidence, and return bloom, negatively, and to the yield of middle grade fruits (fruit weight was 250-299g and none pit on fruit surface) per tree, calcium contents of leaves, and fruit red color, positively. There was no significant effect of time of hand-thinning on fruit firmness, titratable acidity, and total incomes per tree. In conclusion, if the time of hand-thinning of 'Gamhong'/M.9 apple tree was completed at 9 weeks after full bloom, it could produce about 300g of high-quality fruit without bitter pit.

Effects of photoperiod and light intensity on milk production and milk composition of dairy cows in automatic milking system

  • The purpose of this study was to determine the effects of photoperiod and light intensity on milk production, milk composition, hormones levels and blood metabolites indices of Korean Holstein dairy cows in automatic milking system (AMS). A total of 24 Holstein dairy cows were selected and used to four subsequent treatments for the experimental periods of 60 days. The light programs consisted of (1) Control: the natural photoperiod with 14.2 h of the light period and 9.4 h of the dark period (below 10 Lux); (2) T1: 16 h of the long day photoperiod (LDPP) with 50 Lux of light; (3) T2: 16 h of LDPP with 100 Lux of light; and (4) T3: 16 h of LDPP with 200 Lux of light, respectively. Importantly, there was a significant difference in the thurl activity of dairy cows between the different light intensity programs (p < 0.05). Milk yield was higher in T1 and T2 (40.80 ± 1.71 and 39.90 ± 2.02 kg/d, respectively) than those of Control and T3 (32.18 ± 1.51 and 35.76 ± 2.80 kg/d, respectively) (p < 0.05), but DMI was lower in T1, T2, and T3 compared to Control (p < 0.05). Also, milk fat percentage, the contents of milk fat and total solids were higher in T2 than those in the others (p < 0.05). The average daily melatonin level in milk was high to T3 (28.20 ± 0.43 pg/mL), T2 (24.62 ± 0.32 pg/mL), T1 (19.78 ± 0.35 pg/mL), and Control (19.36 ± 0.45 pg/mL) in order (p < 0.05). Also, the cortisol levels in milk and blood were lower in treatment groups than in Control (p < 0.05). The results of this study showed that it will be effective to improve the milk yield and milk composition, and to reduce the stress of dairy cows when the light conditions regulate to extend the photoperiod to 16 h at a light emitting diode (LED) intensity of 100 Lux under the AMS in dairy farm.

Comparison of Characteristics of Acid-catalyzed Hydrothermal Fractionation for Production of Hemicellulose Hydrolyzate from Agricultural Residues (농경잔류물로부터 헤미셀룰로오스 가수분해물 생산을 위한 산촉매 열수 분별공정의 특성 비교)

  • The objective of this work was to investigate the feasibility of acid-catalyzed hydrothermal fractionation for maximum solubilization of the hemicellulosic portion of two typical agricultural residues. The fractionation conditions converted into combined reaction severity (CS) in the range of 1.2-2.9 was used to establish a simple reaction criteria at glance. The hemicellulosic sugar yield of 56.6% was shown when rice straw was fractionated at the conditions at the conditions; 160 ℃ of temperature 0.75% (w/v) of H2SO4, 20 min of reaction time, 1:15 solid/liquid ratio. The hemicellulosic sugar yield of 83.0%, however, was achieved when barley straw was fractionated at the conditions at the conditions; 150 ℃ of temperature 0.75% (w/v) of H2SO4, and 15 min of reaction time, 1:10 solid/liquid ratio. For barley straw, acid-catalyzed hydrothermal fractionation could be effectively performed. After the fractionation process, the remaining fractionated solids were 48.5% and 57.5% from raw rice and barley straws, respectively. The XMG contents in the solid residues decreased from 17.3% and 17.6% to 6.0% and 2.6%, which corresponded to 16.7% and 8.5% on the basis of the raw straws, respectively. In another way, only 5.6% of cellulose and 8.5% of XMG were lost due to excessive decomposition during the acid-catalyzed hydrothermal fractionation of barley straw, compared to cellulose and XMG losses of 6.4% and 26.6% in rice straw. Hemicellulosic sugars from the rice straw were considered more over-decomposed due to the somewhat higher reaction severity at the acid-catalyzed hydrothermal fractionation.
