Study of the Production of Alkaline Keratinases in Submerged Cultures as an Alternative for Solid Waste Treatment Generated in Leather Technology

  • Six nonpathogenic fungal strains isolated from alkaline soils of Buenos Aires Province, Argentina (Acremonium murorum, Aspergillus sidowii, Cladosporium cladosporoides, Neurospora tetrasperma, Purpureocillium lilacinum (formerly Paecilomyces lilacinus), and Westerdikella dispersa) were tested for their ability to produce keratinolytic enzymes. Strains were grown on feather meal agar as well as in solid-state and submerged cultures, using a basal mineral medium and "hair waste" as sole sources of carbon and nitrogen. All the tested fungi grew on feather meal agar, but only three of them were capable of hydrolyzing keratin, producing clear zones. Among these strains, P. lilacinum produced the highest proteolytic and keratinolytic activities, both in solid-state and submerged fermentations. The medium composition and culture conditions for the keratinases production by P. lilacinum were optimized. Addition of glucose (5 g/l) and yeast extract (2.23 g/l) to the basal hair medium increased keratinases production. The optimum temperature and initial pH for the enzyme production were $28^{\circ}C$ and 6.0, respectively. A beneficial effect was observed when the original concentration of four metal ions, present in the basal mineral medium, was reduced up to 1:10. The maximum yield of the enzyme was 15.96 $U_c/ml$ in the optimal hair medium; this value was about 6.5-fold higher than the yield in the basal hair medium. These results suggest that keratinases from P. lilacinum can be useful for biotechnological purposes such as biodegradation (or bioconversion) of hair waste, leading to a reduction of the environmental pollution caused by leather technology with the concomitant production of proteolytic enzymes and protein hydrolyzates.

The Effect of Cultural Factors on Anther Culture in Spring Wheat (Triticum aestivum)

  • Effects of shaking, medium consistency and anther density on polyhaploid production in two wheat cultivars, Pavon and Chris, were studied using a modified 85D12 medium. Pavon produced more calli in shaking and more albino plants tban Chris. However, Chris produced threefold more green plants than Pavon in non-shaking treatment. More calli and green plants were derived from non-shaking treatment than those from shaking treatment. Anthers were cultured on both liquid and semi-solid 85D12 media, using two anther densities, 48 and 96 anthers per plate. Although Pavon generally produced more calli and albino plants than Chris, Chris produced more green plants than Pavon. More green plants were derived from semi-solid medium than those from liquid medium. A factor that may affect plant regeneration from anthers is the length of time on initiation medium. Most of the calli for both genotypes were transferred during the first two time periods. Fertility, as measured by seed set, was determined for all surviving regenerated plants. About 24% of Chris and Pavon anther-derived green plants in the experiment of medium consistency and anther density produced seed.

The optimal medium for the production of KM100l mutant cell mass is instructed to be composed of 3% glucose, 2% yeast extract, 0.1 % $(KH_2PO_4$, 0.05% $MgSO_4{\cdot}7H_2O$, 0.2% L-asparagine, pH 4.5, and the optimal inoculum size and shaking speed were $1.5{\times}10^6$ spores/50 m1 medium and 150 rpm, respectively. On optimal conditions, 4.1 g/l of the cell mass was obtained at 28$^{\circ}C$ for 3 days. The mycelium were inoculated on 500 g of steamed rice using vinyl bag ($30.6{\times}44$ cm) and incubated at $30^{\circ}C$, 85% humidity for 21 days. Lactone form monacolin K was rapidly increased for 2 days and reached highest concentration of monacolin K (2,930 mg/kg) for 15 days, and monacolin K was decreased after 15 days.

  • In this paper, we deal with a surface discharge that caused an aggravation of the dielectric strength in the $N_2/O_2$ mixture gas, When composit dielectrics were formed from the use of a solid dielectric. It was found from this study that the surface discharge voltage was deeply involved in the mixture ratio of $O_2$, the electrical property of the solid dielectric, kind of the solid dielectric, an electric field at the triple junction and a medium effect. These results expect basic data that will be used to transmission and distribution power system equipment using the $N_2/O_2$ mixture gas.

Improvement of Fungal Cellulase Production by Mutation and Optimization of Solid State Fermentation

  • Spores of Aspergillus sp. SU14 were treated repeatedly and sequentially with $Co^{60}$ ${\gamma}$-rays, ultraviolet irradiation, and N-methyl-N'-nitro-N-nitrosoguanidine. One selected mutant strain, Aspergillus sp. SU14-M15, produced cellulase in a yield 2.2-fold exceeding that of the wild type. Optimal conditions for the production of cellulase by the mutant fungal strain using solid-state fermentation were examined. The medium consisted of wheat-bran supplemented with 1% (w/w) urea or $NH_4Cl$, 1% (w/w) rice starch, 2.5 mM $MgCl_2$, and 0.05% (v/w) Tween 80. Optimal moisture content and initial pH was 50% (v/w) and 3.5, respectively, and optimal aeration area was 3/100 (inoculated wheat bran/container). The medium was inoculated with 25% 48 hr seeding culture and fermented at $35^{\circ}C$ for 3 days. The resulting cellulase yield was 8.5-fold more than that of the wild type strain grown on the basal wheat bran medium.

Effects of Co-current and Cross Flows on Circular Enhanced Gravity Plate Separator Efficiencies

  • This study compares the effects of flow on oil and suspended solids removal efficiencies in circular enhanced gravity plate separator equipped with coalescence medium. Coalescence medium acts to capture rising oil droplets and settling solid particles and assist in the coalescence of oil and coagulation of solid. The circular separator uses an upflow center-feed perforated-pipe distributor as the inlet. The co-current flow is achieved using 4 increasing sizes of frustum, whereas cross flow uses inclined coalescence plates running along the radius of the separator. The different arrangement gave the cross flow separator a higher coalescence plan area per operational volume, minimal and constant travelling distance for the oil droplets and particles, lower retention time, and higher operational flowrate. The cross flow separator exhibited 6.04% and 13.16% higher oil and total suspended solids removal efficiencies as compared to co-current flow.

  • True collagenolytic enzymes in animal tissues were first demonstrated by Gross and Lapiere (1962), who showed the ability of such an enzyme in the culture medium of living explants of tadpole tissue to degrade a specific substrate of undenatured collagen under physiological conditions. Recently, tumor-associated collagenolytic activity has been demonstrated in human neoplasm and in ascites V Carcinoma. This investigation have been peforme to determine whether or not a collagen lytic enzyme could e found in isolated solid Tawa sarcoma of Donryu female rat obtained the culture medium. The results were as follows. 1. 11.5mg% of hydroxyproline contained in Donryu rat skin collagen, which was extracted by 0.5M acetic acid. 2. Cultivation of solid Tawa sarcoma tissues on reconstituted rat skin collagen gels showed lysis of adjacent gel after 18 hours, and much more extensive lysis after 5 days. 3. Collagen substrate was not attacked by the common proteolytic enzymes, trypsin, pepsin, and pronase.

Nonlinear stability of non-axisymmetric functionally graded reinforced nano composite microplates

  • The nonlinear buckling response of nano composite anti-symmetric functionally graded polymeric microplate reinforced by single-walled carbon nanotubes (SWCNTs) rested on orthotropic elastomeric foundation with temperature dependent properties is investigated. For the carbon-nanotube reinforced composite (CNTRC) microplate, a uniform distribution (UD) and four types of functionally graded (FG) distribution are considered. Based on orthotropic Mindlin plate theory, von Karman geometric nonlinearity and Hamilton's principle, the governing equations are derived. Generalized differential quadrature method (GDQM) is employed to calculate the non-linear buckling response of the plate. Effects of FG distribution type, elastomeric foundation, aspect ratio (thickness to width ratio), boundary condition, orientation of foundation orthotropy and temperature are considered. The results are validated. It is found that the critical buckling load without elastic medium is significantly lower than considering Winkler and Pasternak medium.

  • This work aims to analyze the thermo-viscoelastic interaction in an orthotropic solid cylinder. The medium is considered to be variable thermal conductivity and subjected to temperature pulse. Analytical solution based on dual-phase-lags model with Voigt-type for behavior of viscoelastic material has been effectively proposed. All variables are deduced using method of Laplace transforms. Numerical results for different distribution fields, such as temperature, displacement and stress components are graphically presented. Results are discussed to illustrate the effect of variability thermal conductivity parameter as well as phase-lags and viscoelasticity on the field quantities. Results are obtained when the viscosity is ignored with and without considering variability of thermal conductivity. A comparison study is made and all results are investigated.

Somatic Embryogenesis and Plant Regeneration from Stem Tissues of Orostachys japonicus A. Berger

  • Orostachys japonicus A. Berger is a Perennial herbaceous plant which has been traditionally used as an anti-inflammatory agent to treat hepatitis and as an anticancer agent. The objective of this study was 1) to establish and proliferate in vitro plant of O. japonicus 2) to induce indirect somatic embryogenesis from O. japonicus. General calli and embryogenic calli in all ranges of 2,4-D and BA combination, were induced and were best at 22% (embryogenic cell) in 5.0 mg/L 2,4-D and 0.5 mg/L BA combination. Embryogenic cell line was maintained by subculture at 2 week intervals and transferred to solid and liquid medium for embryo formation. In solid medium culture, globular and heart shaped embryos were observed in MS medium containing 5.0 mg/L 2,4-D and 0.5 mg/L BA combination. The number of embryos was 6.5 per 0.5 g cell, and then the immature embryos transferred to MS basal medium for embryo development. In a suspension culture of embryogenic cells, globular and heart shaped embryos were emerged in MS medium supplemented with 3.0 mg/L 2,4-D and 0.3 mg/L BA combination after 10 days of incubation. The embryo formation rate was about 33% by suspension culture. The ratio of embryo germination was 60.9%, on the other side, the root formation rate was 74.3% in 1/2 MS continuously.
