Enhanced Lovastatin Production by Solid State Fermentation of Monascus ruber

  • The purpose of this study was to optimize the solid state cultivation of Monascus ruber on sterile rice. A single-level-multiple-factor and a single-factor-multiple-level experimental design were employed to determine the optimal medium constituents and to optimize carbon and nitrogen source concentrations for lovastatin production. Simultaneous quantitative analyses of the ${\beta}$-hydroxyacid form and ${\beta}$-hydroxylactone for of lovastatin were performed by the high performance liquid chromatography (HPLC) method with a UV photodiode-array (PDA) detector. The total lovastatin yield ($4{\sim}6\;mg/g$, average of five repeats) was achieved by adding soybean powder, glycerol, sodium nitrate, and acetic acid at optimized levels after 14 days of fermentation. The maximal yield of lovastatin under the optimal composition of the medium increased by almost 2 times the yield observed prior to optimization. The experimental results also indicated that the ${\beta}$-hydroxylactone form of lovastatin (LFL) and the ${\beta}$-hydroxyacid form of lovastatin (AFL) simultaneously existed in solid state cultures of Monascus ruber. while the latter was the dominant form in the middle-late stage of continued fermentation. These results indicate that optimized culture conditions can be used for industrial production of lovastatin to obtain high yields.

A Study on the Subtraction Method and Characteristics as Design Organization found in the Architecture of Aires Mateus (아이레스 마테우스 건축에 나타난 디자인 조직체로서의 서브 트랙션 방법과 특성에 관한 연구)

  • Aires Mateus Architects is one of the most well-known Portuguese architects after $\acute{A}lvaro$ Siza and Eduardo Souto de Moura. The studio shows clear design methods in which the relationship between solid mass and void space are creatively inter-connected. Subtractive transformation of form is the fundamental aspect of this relationship. Most of Aires Mateus' projects were designed based on the subtractive method. Although subtraction can be seen as architectural formal presentation, in Aires Mateus' cases, there are deeper logics, strategies, and meanings. Not only to create a form, subtractive method has been flexibly used to organize and compose different programs and spatial atmosphere. To examine this hidden dimension of Aires Mateus' unique design process, 5 case projects were selected and analyzed in detail based on program, light & visual-perception, and site's placeness. From the case study results, it is found that there are much deeper role of subtraction: relationship between solid-void are constantly changing to generate serviced and servant spaces, there are central spaces that are contrast against the complexity of exterior forms, aiming to build an archetypal form that transcends ever-changing time, ambivalent balance between universality and peculiarity in the genealogy of contemporary architecture. The study concluded that the unique characteristics of subtraction method as design organization have various possibilities that can apply on other architectural and interior projects which would have similar ideas and circumstances.

  • The LB71350,(3S, 4R)-Epoxy-(5S)-[[N-(1-methylethoxy)carbonyl]-3-(methylsulfonyl)-L-valinyl]amino]-N-[2-methyl-(1R)-[(phenyl)carbonylpropyl-6-phenylhexanamide, is a novel HIV protease inhibitor. Its equilibrium solubility at room temperature was less than $40{\mu}g/mL.$ It was speculated that the low aqueous solubility might be due to the high crystalline lattice energy resulting from intermolecular hydrogen bonds. The present study was carried out to learn the solid-state characteristics of LB71350 using analytical methods such as NMR, FT-IR and XRD. $^{13}C$ Solid-state NMR, solution NMR, and FT-IR spectra of the various solid forms of LB71350 were used to identify the conformation and structure of the solid forms. The chemical shifts of $^{13}C$ solid-state NMR spectra suggest that the crystalline form might have 3 intermolecular hydrogen bondings between monomers.

Preparation and Evaluation of Solid Composites Containing Choline Alphoscerate

  • The objective of this study was to evaluate the characteristics of the mixtures of choline alphoscerate (alpha-glycerylphosphorylcholine, alpha-GPC), in the liquid form, and sucrose ester, which formed a solid composite. The choline alphoscerate solid composites were prepared using different ratios of sucrose ester, and different preparation methods, such as air drying and rotary evaporation, were compared for their preparation efficacy. We examined the characteristics of the solid composites by using scanning electron microscopy (SEM), angle of repose, and moisture content. The ideal mixing ratio of choline alphoscerate and sucrose ester was determined as 1:3 and air drying was found to be more suitable for the preparation of solid composites than rotary evaporation. SEM measurements of the degree of dispersion and the size of particles indicated that a high-temperature air method was more suitable. These results demonstrated the successful preparation of choline alphoscerate solid composites that have potential for industrial use.

  • A temperature equation which is derived from an enthalpy transport equation by using an assumption of a constant specific heat is very attractive for analyses of heat and fluid flows. It can be used for an analysis of a solid-fluid conjugate heat transfer, and it does not need a numerical method to find temperature from a temperature-enthalpy relation. But its application is limited because of the assumption. A new method is derived in this study, which is a temperature-explicit formulation of the energy equation. The enthalpy form of the energy equation is used in the method. But the final discrete form of the equation is expressed with temperature. It can be used for a solid-fluid conjugate heat transfer and multiphase flows. It is found by numerical tests that it is very efficient and as accurate as the standard enthalpy formulation.

  • In the study, we described on the 3D solid model be made on the AutoCAD haved an effect on the deviation of form as RP manufactures the facetres. The STL file improved the deviation of form as the facetres value but the deviation of form have the difference a little as the RP system. In result, we ruled the correlation with the 3D solid model and the RP sample manufacturing, with manufacture RP sample according to facetres value in complex shape. And we developed the program which recombination the STL file that we make use of the AutoLISP.

Investigating the Leaching Rate of TiTe3O8 Towards a Potential Ceramic Solid Waste Form

  • An important property of glass and ceramic solid waste forms is processability. Tellurite materials with low melting temperatures and high halite solubilities have potential as solid waste forms. Crystalline TiTe3O8 was synthesized through a solid-state reaction between stoichiometric amounts of TiO2 and TeO2 powder. The resultant TiTe3O8 crystal had a three-dimensional (3D) structure consisting of TiO6 octahedra and asymmetric TeO4 seesaw moiety groups. The melting temperature of the TiTe3O8 powder was 820℃, and the constituent TeO2 began to evaporate selectively from TiTe3O8 above around 840℃. The leaching rate, as determined using the modified American Society of Testing and Materials static leach test method, of Ti in the TiTe3O8 crystal was less than the order of 10-4 g·m-2·d-1 at 90℃ for durations of 14 d over a pH range of 2-12. The chemical durability of the TiTe3O8 crystal, even under highly acidic and alkaline conditions, was comparable to that of other well-known Ti-based solid waste forms.

  • Considering the immense applicability of isoprene rubbers, such as natural rubber (NR) and synthetic polyisoprene rubber (IR), attempts are being made to introduce more functionality within the rubber structure, e.g. epoxidation, to widen their technological viability. Epoxidation introduces polar epoxy bonds into the rubber molecular chain, resulting in enhanced intermolecular interactions among the rubber chains, increasing the oil resistance and air impermeability. Although there have been many reports on the epoxidation of NR in its latex form, there has been no such report using its solid form (or gum), which limits the epoxidation in terms of portability. Furthermore, the gum form has longer lifetime, while the latex form has limited lifetime for its efficient use. In this study, the epoxidation of natural rubber and polyisoprene rubber (using meta-chloroperoxybenzoic acid (mCPBA) as the epoxidizing agent) by dissolving their gum in hexane (i.e., the solution method) have been studied and compared. The effects of the amount of mCPBA, reaction time, and reaction temperature were investigated. The present process is easy and facilitates the epoxidation of rubbers in their solid form; therefore, it can be used for industrial upscaling of epoxidized rubber production.

  • A sensitive, selective and rapid method for the determination of nickel based on the rapid reaction of nickel(II) with QADMAA and the solid phase extraction of the Ni(II)-QADMAA chelate with $C_{18}$ membrane disks has been developed. In the presence of pH 6.0 buffer solution and sodium dodecyl sulfonate (SDS) medium, QADMAA reacts with nickel to form a violet complex of a molar ratio of 1 : 2 (nickel to QADMAA). This chelate was enriched by solid phase extraction with $C_{18}$ membrane disks. An enrichment factor of 50 was obtained by elution of the chelates form the disks with the minimal amount of isopentyl alcohol. The molar absorptivity of the chelate was $1.32{\times}10^5L\;mol^{-1}cm^{- 1}$ at 590 nm in the measured solution. Beer's law was obeyed in the range of 0.01-0.6 ${\mu}$g/mL. This method was applied to the determination of nickel in water and biological samples with good results.

The solid angle estimation of acetabular coverage of the femoral head (입체각을 이용한 관골구와 대퇴골두의 접촉영역 측정)

  • We developed a method for the solid angle estimation of acetabular coverage of the femoral head in 3D space. The superior half of the femoral head is modeled as part of a sphere. And the tangent lines connecting from a set of points of the acetabular outline to the center of the fitted sphere are obtained. The lines passthrough the unit sphere whose center is the same as that of the femoral head. The interesecting points form a boundary on the unit sphere. With the points on the unit sphere, we calculate the covered area of the femoral headand estimate the solid angle. Solid angle is defined asthe suface area within the boundary on the unit sphere. In this measurements, the solid angle of normal subjects is on an average 4.3(rad) and the corresponding acetabular coverage is 68%. Unlinke the conventional methods, this solid angle estimation shows real 3D acetabular coverage.
