Distribution of foodborne pathogens in red pepper and environment (고추와 재배환경의 식품매개 병원균 분포)

  • This study was performed to investigate the extent of microbial contamination, the presence of enterotoxin genes, and the antibiotic susceptibility of Bacillus cereus in 58 red pepper plants and 43 environmental samples (soil, irrigation water, and gloves) associated with the plant cultivation. The detected counts of total aerobic bacteria, coliform bacteria, Escherichia coli, Bacillus cereus, and Staphylococcus aureus were lower in these samples, as compared to the regulations of standards for foods; moreover, pathogens, such as E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp., were not detected. Genes encoding hemolysin BL enterotoxins (hblA, hblC, and hblD) as well as non-hemolytic enterotoxins (nheA, nheB, and nheC) were detected in 23 B. cereus specimens that were isolated from the test samples and had β-hemolytic activity. Interestingly, B. cereus is resistant to β-lactam and susceptible to non-β-lactam antibiotics. However, in this case, the isolated B. cereus specimens exhibited a shift from resistant to intermediate in response to cefotaxime and from susceptible to intermediate in case of rifampin, trimethoprim-sulfamethoxazole, vancomycin, clindamycin, and erythromycin. Therefore, the levels of B. cereus should be monitored to detect changes in antibiotic susceptibility and guarantee their safety.

Economic Analysis, Growth and Pests of Wheat (Triticum aestivum L.) in Gelatin·Chitin Microorganisms-treated Organic Culture (젤라틴·키틴분해미생물을 이용한 밀 유기재배와 관행재배의 생육, 병해충 발생조사 및 경제성 분석)

  • This study was carried out to investigate the economic value of organic wheat production using gelatin·chitin microorganisms in Gwangsan-gu, Gwangju city. The soil condition of experiment field was clay loam Jisan series. The organically cultivated fields were sprayed gelatin and chitin degrading bacteria. The test was performed at conventionally cultivated field and organically cultivated field. Emergence of weed on organically cultivated field was significantly higher than conventionally cultivated field which sprayed herbicide before seeding. Weed emergence have a critical impact on grain yield. Occurrence of diseases and insect pests were higher than conventionally cultivated fields. In 2019, the amount of lodging in conventionally cultivated field were higher than conventionally cultivated field. In 2020, lodging and wet injury were occur in both field. Comparing yield element between organically and conventionally cultivated experimental area, grain yield in organically cultivated field was shown slightly higher amount than conventionally cultivated field. However in the actual yield of 2019, organically cultivated field shows 20% deceased yield because of overgrown weed. In 2020, weed emergence and yellow mosaic virus by wet injury cause 30% decease in the grain yield in organically cultivated field. Content of protein, carbohydrates, ash, water and fat in the grain were not different significance. In 2019, net incomes of conventionally cultivated wheat was 461,031 won/0.1 ha while organically cultivated wheat was 443,437 won/0.1 ha. In the rate of income, conventionally cultivated field was 83.0% as against organically cultivated field (73.3%). In 2020, net incomes of organically cultivated wheat was 437,812 won/0.1 ha while conventionally cultivated wheat was 418,281 won/0.1 ha. In the rate of income, conventionally cultivated field was 81.6% as against organically cultivated field (73.0%).

Application Effects of Organic Fertilizer Utilizing Livestock Horn Meal as Domestic Organic Resource on the Growth and Crop Yields (국내산 유기자원 우각을 활용한 유기질비료의 작물 생육 및 수량에 미치는 영향)

  • Objective of this study was to develop an organic fertilizer utilizing domestic livestock horn meal and to investigate the application effect of rice and eggplant. The possibility of utilization of livestock horn meal as an organic resource to replace imported expeller cake fertilizer was examined. In order to select domestic organic resources with high nitrogen content, 8 kinds of organic matter such as chicken manure, fish meal, soybean meal, sesame meal, perilla meal, blood meal, livestock horn meal, and beer sludge were analyzed and organic resources with high nitrogen content were selected. In addition, the conditions for the production of organic fertilizers that can be used in organic agriculture were established by mixing of the rice husk biochar and the rice bran as the supplements with the raw materials for mixing ratios. The content of total nitrogen (T-N) in the livestock horn meal was 12.0 %, which was the next low in 13.5 % blood meal. The content of total nitrogen was 5.9 ~ 7.9 % in fish meal and oil cakes. Total nitrogen content of non-antibiotic chicken manure for organic farming was 3 % and nitrogen content in beer sludge was 3.5 %. Organic fertilizer was produced by using biochar, rice bran as a main ingredient of non-antibiotic chicken manure, livestock horn meal and beer sludge. Compared to nitrogen content (4.0 to 4.2 %) of imported expeller cake fertilizer (ECF), the nitrogen content of organic fertilizer utilizing domestic livestock horn meal is as high as 7.5 %. The developed organic fertilizer is met as Zn 400 mg/kg, Cu 120 mg/kg the quality of organic agricultural materials such as or less. To investigate the effect of fertilizer application on the crops, prototypes of developed organic fertilizer were used for the experiment under selected conditions. As a result of application the developed organic livestock horn meal fertilizer (LHMF) for cultivation of the rice and eggplant, the application quantity of the developed organic LHMF 100 % was decreased by 40 % compared to that of the mixed expeller cake fertilizer (MECF). The application of LHMF, which refers to the application rate corresponding to the nitrogen fertilization recommended by the soil test, was reduced by 40% compared to the application rate of MECF, but the same results were obtained in crop growth and yield. The selection of a new high concentration nitrogen source utilizing domestic organic resources and the development of organic fertilizer is the starting point of the research for substitution of imported ECF using domestic local resources at the present time that the spread of eco-friendly agriculture is becoming increasingly important. If it is expanded in the future, it is expected to contribute to the stable production of eco-friendly agricultural products.

A Change of Stream Water Quality by Forest Types (임상에 따른 계류수의 수질변화에 관한 연구)

  • This study was carried out to clarify the change characteristics of stream water quality by type of forest from June to August, 2009 in three stands (Castanea crenata, Pinus densiflora and Plantation Land) of Samgye-ri Naedong-myeon Jinju-si Gyeongsangnam-do. The pH of stream water in three stands was highest in Pinus densiflora (pH 7.18) followed by Castanea crenata (pH 6.90) and Plantation land (pH 6.90) while the electrical conductivity of stream water was highest in Plantation land followed by Castanea crenata stand and Pinus densiflora stand was the lowest. Cations contents of stream water in three stands were high in order of $Ca^{2+}$, $Na^{+}$, $Mg^{2+}$, $K^{+}$, and $NH_{4}{^{+}}$. But anions of stream water in Castanea crenata stand and Pinus densiflora stand were high in order of $SO_{4}{^{2-}}$, $Cl^{-}$ and $NO_{3}{^{-}}$ while those of stream water in Plantation land were high in order of $SO_{4}{^{2-}}$, $NO_{3}{^{-}}$ and $C\lambda^{-}$. The stream water in three stands was significant at pH, EC, $NO{^{3-}}$, $Ca^{2+}$, $Mg^{2+}$, $Na^{+}$, $Cl^{-}$, TNU and Color by duncan test. These results indicate that quality of stream water have a difference among three stands. The level of pH, $NH_{4}{^{+}}$, $Cl^{-}$, $SO_{4}{^{2-}}$ and $NO_{3}{^{-}}$ of stream water in three stands were within the domestic use standard for drinking water. but turbidity and color of stream water were more than that of domestic use standard for drinking water. Therefore, non-point sources like urban forest watersheds which are soil erosion and fertilizer application lands should be taken to the appropriate mitigation measures if they are to be used as source of drinking water.

Reinforcing Effects around Face of Soil-Tunnel by Crown & Face-Reinforcing - Large Scale Model Testing (천단 및 막장면 수평보강에 의한 토사터널 보강효과 - 실대형실험)

  • One of the most popular pre-reinforcement methods of tunnel heading in cohesionless soils would be the fore-polling of grouted pipes, known as RPUM (reinforced protective umbrella method) or UAM (umbrella arch method). This technique allows safe excavation even in poor ground conditions by creating longitudinal arch parallel to the tunnel axis as the tunnel advances. Some previous studies on the reinforcing effects have been performed using numerical methods and/or laboratory-based small scale model tests. The complexity of boundary conditions imposes difficulties in representing the tunnelling procedure in laboratory tests and theoretical approaches. Full-scale study to identify reinforcing effects of the tunnel heading has rarely been carried out so far. In this study, a large scale model testing for a tunnel in granular soils was performed. Reinforcing patterns considered are four cases, Non-Reinforced, Crown-Reinforced, Crown & Face-Reinforced, and Face-Reinforced. The behavior of ground and pipes as reinforcing member were fully measured as the surcharge pressure applied. The influences of reinforcing pattern, pipe length, and face reinforcement were investigated in terms of stress and displacement. It is revealed that only the Face-Reinforced has decreased sufficiently both vertical settlement in tunnel heading and horizontal displacement on the face. Vertical stresses along the tunnel axis were concentrated in tunnel heading from the test results, so the heading should be reinforced before tunnel advancing. Most of maximum axial forces and bending moments for Crown-reinforced were measured at 0.75D from the face. Also it should be recommended that the minimum length of the pipe is more than l.0D for crown reinforcement.

Isolation and Characterization of Lactic acid bacteria Leuconostoc mesenteroides DB3 from Camellia japonica Flower (백꽃으로부터 분리한 Leuconostoc mesenteroides DB3의 특성)

  • Lactic acid bacteria (LAB) are widespread in a variety of environments including fermented dairy products, gastroinstetinal and urogenital tracts of human and animals, plant, soil and water. Leuconostoc mesenteroides DB3 was detected by the strongest antibacterial activities among 24 Leuconostoc strains isolated from Camellia japonica flowers. Acid tolerance of L. mesenteroides DB3 existed up to pH 2.5, but the resistance did not show at pH 2.0, which relatively excellent acid resistance existed. Bile acid tolerance was very stable within the test range to 1.2%. L. mesenteroides DB3 exhibited the optimal growth at 30℃, and showed a slight slow growth when compared with L. mesenteroides KCTC3505, which reached a stationary phase at 18 hr. The pH was changed along with the growth curve, but was maintained above pH 3.98. L. mesenteroides DB3 had higher initial antibacterial activities when compared to L. mesenteroides KCTC3505, but it showed similar activities with the standard strain after the latter part of the logarithmic growth phase. Although lactic acid production in L. mesenteroides DB3 was induced by lower amount in the initial part to the standard strain, it was exhibited by similar amounts after the late logarithmic growth phase. Muicin adhesion of L. mesenteroides DB-3 maintained superior to L. mesenteroides KCTC3505. Both strains showed excellent emulsification ability for kerosene. In summary, we evaluate that L. mesenteroides DB-3 has a high potential for application as probiotics owing to its excellent antibacterial activity, acid resistance, bile acid resistance, and muicin adhesion.

Studies on the Rice Yield Decreased by Ground Water Irrigation and Its Preventive Methods (지하수 관개에 의한 수도의 멸준양상과 그 방지책에 관한 연구)

  • The purposes of this thesis are to clarify experimentally the variation of ground water temperature in tube wells during the irrigation period of paddy rice, and the effect of ground water irrigation on the growth, grain yield and yield components of the rice plant, and, furthermore, when and why the plant is most liable to be damaged by ground water, and also to find out the effective ground water irrigation methods. The results obtained in this experiment are as follows; 1. The temperature of ground water in tube wells varies according to the location, year, and the depth of the well. The average temperatures of ground water in a tubewells, 6.3m, 8.0m deep are $14.5^{\circ}C$ and $13.1^{\circ}C$, respercively, during the irrigation period of paddy rice (From the middle of June to the end of September). In the former the temperature rises continuously from $12.3^{\circ}C$ to 16.4$^{\circ}C$ and in the latter from $12.4^{\circ}C$ to $13.8^{\circ}C$ during the same period. These temperatures are approximately the same value as the estimated temperatures. The temperature difference between the ground water and the surface water is approximately $11^{\circ}C$. 2. The results obtained from the analysis of the water quality of the "Seoho" reservoir and that of water from the tube well show that the pH values of the ground water and the surface water are 6.35 and 6.00, respectively, and inorganic components such as N, PO4, Na, Cl, SiO2 and Ca are contained more in the ground water than in the surface water while K, SO4, Fe and Mg are contained less in the ground water. 3. The response of growth, yield and yield components of paddy rice to ground water irrigation are as follows; (l) Using ground water irrigation during the watered rice nursery period(seeding date: 30 April, 1970), the chracteristics of a young rice plant, such as plant height, number of leaves, and number of tillers are inferior to those of young rice plants irrigated with surface water during the same period. (2) In cases where ground water and surface water are supplied separately by the gravity flow method, it is found that ground water irrigation to the rice plant delays the stage at which there is a maximum increase in the number of tillers by 6 days. (3) At the tillering stage of rice plant just after transplanting, the effect of ground water irrigation on the increase in the number of tillers is better, compared with the method of supplying surface water throughout the whole irrigation period. Conversely, the number of tillers is decreased by ground water irrigation at the reproductive stage. Plant height is extremely restrained by ground water irrigation. (4) Heading date is clearly delayed by the ground water irrigation when it is practised during the growth stages or at the reproductive stage only. (5) The heading date of rice plants is slightly delayed by irrigation with the gravity flow method as compared with the standing water method. (6) The response of yield and of yield components of rice to ground water irrigation are as follows: \circled1 When ground water irrigation is practised during the growth stages and the reproductive stage, the culm length of the rice plant is reduced by 11 percent and 8 percent, respectively, when compared with the surface water irrigation used throughout all the growth stages. \circled2 Panicle length is found to be the longest on the test plot in which ground water irrigation is practised at the tillering stage. A similar tendency as that seen in the culm length is observed on other test plots. \circled3 The number of panicles is found to be the least on the plot in which ground water irrigation is practised by the gravity flow method throughout all the growth stages of the rice plant. No significant difference is found between the other plots. \circled4 The number of spikelets per panicle at the various stages of rice growth at which_ surface or ground water is supplied by gravity flow method are as follows; surface water at all growth stages‥‥‥‥‥ 98.5. Ground water at all growth stages‥‥‥‥‥‥62.2 Ground water at the tillering stage‥‥‥‥‥ 82.6. Ground water at the reproductive stage ‥‥‥‥‥ 74.1. \circled5 Ripening percentage is about 70 percent on the test plot in which ground water irrigation is practised during all the growth stages and at the tillering stage only. However, when ground water irrigation is practised, at the reproductive stage, the ripening percentage is reduced to 50 percent. This means that 20 percent reduction in the ripening percentage by using ground water irrigation at the reproductive stage. \circled6 The weight of 1,000 kernels is found to show a similar tendency as in the case of ripening percentage i. e. the ground water irrigation during all the growth stages and at the reproductive stage results in a decreased weight of the 1,000 kernels. \circled7 The yield of brown rice from the various treatments are as follows; Gravity flow; Surface water at all growth stages‥‥‥‥‥‥514kg/10a. Ground water at all growth stages‥‥‥‥‥‥428kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥430kg/10a. Standing water; Surface water at all growh stages‥‥‥‥‥‥556kg/10a. Ground water at all growth stages‥‥‥‥‥‥441kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥450kg/10a. The above figures show that ground water irrigation by the gravity flow and by the standing water method during all the growth stages resulted in an 18 percent and a 21 percent decrease in the yield of brown rice, respectively, when compared with surface water irrigation. Also ground water irrigation by gravity flow and by standing water resulted in respective decreases in yield of 16 percent and 19 percent, compared with the surface irrigation method. 4. Results obtained from the experiments on the improvement of ground water irrigation efficiency to paddy rice are as follows; (1) When the standing water irrigation with surface water is practised, the daily average water temperature in a paddy field is 25.2$^{\circ}C$, but, when the gravity flow method is practised with the same irrigation water, the daily average water temperature is 24.5$^{\circ}C$. This means that the former is 0.7$^{\circ}C$ higher than the latter. On the other hand, when ground water is used, the daily water temperatures in a paddy field are respectively 21.$0^{\circ}C$ and 19.3$^{\circ}C$ by practising standing water and the gravity flow method. It can be seen that the former is approximately 1.$0^{\circ}C$ higher than the latter. (2) When the non-water-logged cultivation is practised, the yield of brown rice is 516.3kg/10a, while the yield of brown rice from ground water irrigation plot throughout the whole irrigation period and surface water irrigation plot are 446.3kg/10a and 556.4kg/10a, respectivelely. This means that there is no significant difference in yields between surface water irrigation practice and non-water-logged cultivation, and also means that non-water-logged cultivation results in a 12.6 percent increase in yield compared with the yield from the ground water irrigation plot. (3) The black and white coloring on the inside surface of the water warming ponds has no substantial effect on the temperature of the water. The average daily water temperatures of the various water warming ponds, having different depths, are expressed as Y=aX+b, while the daily average water temperatures at various depths in a water warming pond are expressed as Y=a(b)x (where Y: the daily average water temperature, a,b: constants depending on the type of water warming pond, X; water depth). As the depth of water warning pond is increased, the diurnal difference of the highest and the lowest water temperature is decreased, and also, the time at which the highest water temperature occurs, is delayed. (4) The degree of warming by using a polyethylene tube, 100m in length and 10cm in diameter, is 4~9$^{\circ}C$. Heat exchange rate of a polyethylene tube is 1.5 times higher than that or a water warming channel. Studies on the Cutting Managemente of Pasture during the Mid Summer Season I. Effect of cutting management on tall fescue dominated pasture (고온기 초지의 예취관리에 관한 연구 I. 고온기 예취방법이 tall fescue 우점초지의 재생 , 잡초발생 및 수량에 미치는 영향)

  • Optimum pasture management during the summer season is an important factor to maintain good regrowth and persistence of pasture in Korea. This experiment was carried out to investigate the effects of the cutting management on the dead plant, weed appearance, regrowth and carbohydrate reserves in stubble, and dry matter yield of tall fescue dominated pasture during the mid summer season. For the test, a split plot design with 4 replications was treated with 2 different the third cutting times (July 12 and Aug. 4) as the mainplots, and 3 different cutting heights (3, 6 and 9 cm) at the third cut as the subplots, and the experiment was done at the experimental field of the Livestock Experiment Station, in Suweon, 1984. The results obtained are summarized as follows: 1. Considering the meteorological conditions during the experimental period, the temperature was a little higher by $2^{\circ}C$ than that of average year, especially the first and second decade of August were high. And the precipitation of 1984 tended to be low when compared with the average year. 2. Temperature of soil surface and underground tended to increase by $1-3^{\circ}C$ as the stubble height was low during the summer season. 3. Regrowth leaf length and leaf area after the third cut increased significantly with the high cutting height at the third cut. 4. A significant higher total nonstructural carbohydrate (TNC) content in stubble after the third cut was observed in the high stubble cut on July 12. The results indicate that the high stubble height reserves more carbohydrates for early regrowth stage after the third cut when compared with the low stubble. On Aug. 4, however, the recovery of TNC contents after the third cut was not effective due to high temperature and rainfall. 5. The percentage of dead plant after the third cut was found to be high with the low cutting height during the mid summer season (p<0.05). 6. With the low stubble height on July 12 cut, it was appeared that the percentage of weed was significantly increased (p<0.05), and main weeds appeared after the third cut were Echinochloa crusgalli>Digitaria sanguinalis>Cyperus iria>Rumex crispus, and so on. In case of cut on Aug. 4, weed appearance was no difference at three cutting heights. 7. Dry matter yield at the third cut was increased in the plot of cutting on Aug. 4 and stubble height (p<0.05). However, yields at the fourth and fifth cut were increased with high stubble height (p<0.05), regardless of harvest time. 8. In total dry matter yield after the third cut, there was no significant difference between the cutting time and forage yield. However, total yield on July 12 was increased with the high stubble height (p<0.05). 9. From the above results, it is suggested that the 9 cm cutting height during the mid summer season is the most effective for good regrowth, weed control and forage yield of tall fescue dominated pasture.
