Persistence of IBP and Isoprothiolane in Rice Plant (수도체(水稻體)중 IBP 와 Isoprothiolane의 잔류소장(殘留消長))

  • Effect of the application time on the persistence of IBP and isoprothiolane in rice plant was studied in the field and effects of the water depth and soil texture on their persistence were also tested as a pot experiment. When granules were applied to the rice paddy water, two fungicides were readily absorbed through the root system and rapidly translocated to the upper parts of the plant. The concentrations of two fungicides in rice plant reached to the maximum within 24 hr regardless of the application time. When applied at the maximum tillering stage, the persistence pattern of two fungicides in plant showed similar trends; that is, residue levels of two compounds declined rapidly upto 7 days after application but more slowly thereafter. When applied at the heading stage, the persistence pattern of IBP in plant was similar to the maximum tillering stage while isoprothilane was quite different; 3 ppm reached on 3rd days after application was maintained almost constant for further 25 days. There was no effect of the water depth on the persistence of two compounds in plant and IBP concentration in plant was also not affected by soil texture. However, isoprothiolane in plant was higher in sandy loam than in loam and clay loam. Isoprothiolane residues in plant were much higher than those of IBP.

Characteritics of Toluene and $H_2S$ Removal in a Biotrickling filters with Plastic & Woodchip composite Media (복합플라스틱계 담체를 이용한 Biotrickling filters의 Toluene과 황화수소 제거특성)

  • This study developed composition-plastic media with woodchips and plastic as main materials, and examined the performance of media. Compared to the existing commercial media, the media had similar performance in removal efficiency and microbes attaching characteristic, and was evaluated that they are distinguished from economic side. Performance test of media was conducted to examine the removal capacity of toluene and hydrogen sulfide in a gas stream by using a lab-scale biotrickling filter systems packed with them. At a volumetric loading of $1.5\;m^3/hr$ with inlet concentration 260 ppm and empty bed residence time (EBRT) 42s, the toluene removal efficiency was shown over 90%, and the maximum elimination capacity of toluene in the biotrickling filter was $77g/m^3{\cdot}hr$. Effective co-treatments of $H_2S$ and Toluene were observed in the lab-scale biotrickling filters. The maximum elimination capacity of $H_2S$ was $100\;g-S/m^3{\cdot}hr$. Up to 100 ppm, the concentration of $H_2S$ did not have an effect on toluene removal efficiency, but the removal efficiency of toluene decreased with increasing inlet $H_2S$ concentration.

Evaluation of Distribution Characteristics for Petroleum Hydrocarbon in Groundwater by TPH Fraction Analysis (석유계 총 탄화수소(Total Petroleum Hydrocarbons, TPH) 분획분석법을 이용한 지하수 중 유류오염물질 분포특성 평가)

  • Total petroleum hydrocarbon (TPH) is a mixture of various oil substances composed of alkane, alkene, cycloalkane, and aromatic hydrocarbons (benzene, toluene, ethylbenzene, xylene, etc.). In this study, we investigated 92 groundwater wells around 36 gas stations to evaluate distribution characteristics of petroleum hydrocarbons. Groundwater in the wells was sampled and monitored twice a year. The fraction analysis method of TPH was developed based on TNRCC 1006. The test results indicated aliphatic and aromatic fractions accounted for 28.6 and 73.8%, respectively. The detection frequencies of TPH in the monitoring wells ranged in 21.6 - 24.2%. The average concentration of TPH was 0.11 mg/L with the concentration range of 0.25~0.99 mg/L. In the result of TPH fraction analysis, in aliphatic fractions were 19% (C6-C8 : 0.2%, C8-C10 : 0.4%, C10-C12 : 0.4%, C12-C16 : 0.5%, C16-C22 : 1.0%, C22-C36 : 16.6%), and aromatic fractions were 81% (C6-C8 : 1.1%, C8-C10 : 0%, C10-C12 : 2.9%, C12-C16 : 0.3%, C16-C22 : 4%, C22-C36 : 66.8%). Fractions of C22-C36 were detected in about 83% of the monitoring wells, suggesting non-degradable characteristics of hydrocarbons with high carbon content.

Estimation of Carbon Emission and Application of LCA (Life Cycle Assessment) from Rice (Oryza sativa L.) Production System (쌀의 생산과정에서 발생하는 탄소배출량 산정을 위한 전과정평가 적용)

  • LCA (Life Cycle Assessment) carried out to estimate carbon footprint and to establish of LCI (Life Cycle Inventory) database of rice production system. The results of collecting data for establishing LCI D/B showed that organic fertilizer and chemical fertilizer input to 4.29E-01 kg $kg^{-1}$ rice and 2.30E-01 kg $kg^{-1}$ rice for rice cultivation. It was the highest value among input for rice cultivation. And direct field emission was 3.23E-02 kg $kg^{-1}$ during rice cropping. The results of LCI analysis focussed on greenhouse gas (GHG) was showed that carbon footprint was 8.70E-01 kg $CO_2$-eq. $kg^{-1}$ rice. Especially for 80% of $CO_2$ in the GHG and 7.02E-01 kg of its $CO_2$-eq. $kg^{-1}$ rice. Of the GHG emission $CH_4$, and $N_2O$ were estimated to be 13% and 5%, respectively. With LCIA (Life Cycle Impact Assessment) for rice cultivation system, it was observed that fertilizer process might be contributed to approximately 80% of GWP (global warming potential).

The Source Identification of Spilled Oil by Pristane/Phytane Ratio

  • In order to identify the origin and nature of the spilled oil in the potential source, we analyzed the concentrations of specific fuel constituents in fuel standard and environmental samples. The ratios of pristane/phytane are virtually unaltered because these compounds have the same bolatility in environmental samples. These were useful to identify the source of the fuel oil and to assess the effect of microbial degradation and weathering of the fuel oil. We analyzed the ratios of pristane/phytane in neat white kerosene, boiler kerosene, JP-8 and diesel products from L and S gas station. The ratios of pristane/phytane in L-white kerosene and JP-8 was 3.10 $\pm$0.03 and 1.77 $\pm$ 0.01, respectively. Otherwise, the ratios of pristane/phytane in water phase after distribution of fuel oil and water was 2.97 $\pm$0.02 in case of white kerosene and 1.65 $\pm$ 0.02 in case of JP-8. It is apparent from the results that the ratios of pristane/phytane were as product-specific, especially between kerosene and JP-8, and therefore, can also be used for fuel type identification in free products and groundwater samples which were collected in monitoring wells.

Atmospheric Acid Deposition : Nitrogen Saturation of Forests (대기 산성 강하물 : 삼림의 질소 포화)

  • Atmospheric Acid Deposition: Nitrogen Saturation of Forests: Volume weighted annual average wet deposition of nitroge at 33 sites in Korea during 1999-2004 ranged 7.28 to $21.05kgN{\cdot}ha^{-1}{\cdot}yr^{-1}$ with average $12.78kgN{\cdot}ha^{-1}{\cdot}yr^{-1}$, which values are similar level with nitrogen deposition of Europe and North America. The temperate forests that suffered long-term high atmospheric nitrogen deposition are gradually saturated with nitrogen. Such nitrogen saturated forest watersheds usually leach nitrate ion ($NO_3^-$) in stream water and soil solution. It may be likely that Korean forest ecosystems are saturated by much nitrogen deposition. In leaves with nitrogen saturation ratios of N/P, N/K and N/Mg are so enhanced that mineral nutrient system is disturbed, suffered easily frost damage and blight disease, reduced fine-root vitality and mycorrhizal activity. Consequently nitrogen saturated forests decrease primary productivity and finally become forest decline. Futhermore understory species are replaced the nitrophobous species by the nitrophilous one. In soil with nitrogen saturation uptake of methane ($CH_4$) is reduced and emission of nitrogen monoxide (NO) and nitrous oxide ($N_{2}O$) are increased, which gases are greenhouse gas accelerating global warming.

A Thermostable Protease Produced from Bacillus sp. JE 375 Isolated from Korean Soil (한국의 토양으로부터 내열성 단백질 분해효소를 생산하는 Bacillus sp. JE 375의 선별)

  • A thermophilic microorganism, strain JE 375, which produces a thermostable protease, was isolated from soil and compost in Korea. This gram-positive, rod-shaped, catalase positive, motility positive, and hemolysis ${\beta}$ containing organism was implicated in glucose fermentation, mannitol fermentation, xylose oxidation, aerobic activity and spore formation. The color of the colony was yellowish white. The temperature range for growth at pH 6.5 was between 55 and $70^{\circ}C$, with an optimum growth temperature of $65^{\circ}C$. This result confirmed the strain JE 375 as a thermophilic microorganism. The enzyme was produced aerobically at $65^{\circ}C$ during 20 hr in a medium (pH 6.5) containing 1% trypton. 1% maltose, 0.5% yeast extract and 1% NaCl. The 16S rDNA of strain JE 375 had 97.6% sequence similarity with the 16S rDNA of Bacillus caldoxyloyticus. On the basis of biochemical and physiological properties and phylogenetic analysis, we named the isolated strain as Bacillus sp. JE 375. The thermostable protease from Bacillus sp. JE 375 had been partially purified and characterized. The molecular weight of the enzyme was deduced from SDS-PAGE and gel chromatography as 55 kDa and its optimal temperature was $60^{\circ}C$. The enzyme showed its highest activity at pH 7.5 and was stable from pH 7.0 to 8.0.

Directions towards sustainable agricultural systems in Korea

  • The question of how to establish sustainable agricultural systems has become as prominent as questions related to water, energy and climate change. High input/high output agriculture has brought with it many adverse effects; the massive deterioration of soil and water in both quantity and quality, increased greenhouse gas emissions and an increased prevalence of unsafe foods. Additionally, urbanization and climate change has worsened the shortage of farmland and reduced the supply of agricultural water. Given these challenges, maintaining, conserving and efficiently using agri-environmental resources, through fostering of sustainable agriculture, have emerged as key tasks in solving these problems. What is needed therefore is research, based on systematic and comprehensive empirical analyses, that can propose plans and methods for establishing an appropriate sustainable agricultural system. The empirical analysis of sustainable agricultural system is approached separately from economic, environmental and social aspects. An analysis of environment effect reveals that the available phosphate level is 1.3~2.1 times greater than the optimal amount in rice paddies, upland fields and orchards. Further examination has revealed that the excess nutrient is polluting both ground water and surface water. Analytical results for economic feasibility show that factors of production have been invested heavily in the rice crop. Under these conditions, sustainable agriculture, including low-input agriculture, appears to be a possible alternative that will facilitate simultaneous improvements in both economic feasibility and environment effects. Analysis results for sociality reveal that social factors include the value of producer, association and interior networks. Social conditions are comprised of leadership, consumers' awareness, education and conflict solutions. In addition, analysis as to the degree investments contribute to improving agricultural value added has revealed that the direct payment program is the most effective instrument. Experts confirm that economic feasibility can be improved by scientific and well-reasoned nutrient management on the basis of soil testing. Farmers pointed to 'economic factors' as being the largest obstacle to switching to the practice of sustainable agriculture. They also indicate 'uncertainty with regards to sustainable agriculture technology' as an impediment to practicing sustainable agriculture. Even so, farmers who believe environmental and regional issues to be the most pressing problems have expanded their practice of sustainable agriculture. The keys to establishing sustainable agriculture system are classified into the following four aspects. Firstly, from an economic aspect, the research indicates that agricultural policy needs to be integrated with environmental policy and that the function of market making based on the value chain needs to be revitalized. Secondly, from an environmental aspect, there is a need for an optimal resource management system to be established in the agricultural sector. In addition, sustainable agriculture practice will need to be extended with attendant environmentally-friendly and sustainable intensive technology also requiring further development. Thirdly, from a social aspect, green agriculture management needs to be fostered, technology and education extended, and social conflict mediated. Lastly, from a governance aspect, it will be necessary to strengthen good governance, assign and share suitable roles and responsibilities, build a cooperation system and utilize community supported agriculture.

Aerobic Degradation of Tetrachloroethylene(PCE) by Pseudomonas stutzeri OX1

  • Since trichloroethylene (TCE), dichloroethylene (DCE), and vinyl chloride (VC) arise from anaerobic degradation of tetrachloroethylene (PCE) and TCE, there is interest in creating aerobic remediation systems that avoid the highly toxic VC and cis-DCE which predonominate in anaerobic degradation. However, it seemed TCE could not be degraded aerobically without an inducing compound (which also competitively inhibits TCE degradation). It has been shown that TCE induces expression of both the toluene dioxygenase of p. putida F1 as well as toluene-p-monooxygenase of P.mendocina KRI. We investigated here the ability of PCE, TCE, and chlorinated phenols to induce toluene-o-xylene monooxygenase (ToMO) from P.stutzeri OX1. ToMO has a relaxed regio-specificity since it hydroxylates toluene in the ortho, meta, and para positions; it also has a broad substrate range as it oxidizes o-xylene, m-xylene, p-xylene, toluene, benzene, ethylbenzene, styrene, and naphthalene; chlorinated compounds including TCE, 1, 1-DCE, cis-DCE, trans-DCE, VC, and chloroform : as well as mixtures of chlorinated aliphatics (Pseudomonas 1999 Maui Meeting). ToMO is a multicomponent enzyme with greatest similarity to the aromatic monooxygenases of Burkholderia pickettii PKO1 and P.mendocina KR1. Using P.sturzeri OX1, it was found that PCE induces P.mendocina KR1 Using P.situtzeri OX1, it was found that PCE induces ToMO activity measured as naphthalene oxygenase activity 2.5-fold, TCE induces 2.3-fold, and toluene induces 3.0 fold. With the mutant P.stutzeri M1 which does not express ToMO, it was also found there was no naphthalene oxygenate activity induced by PCE and TCE; hence, PCE and TCE induce the tow path. Using P.putida PaW340(pPP4062, pFP3028) which has the tow promoter fused to the reporter catechol-2, 3-dioxygenase and the regulator gene touR, it was determined that the tow promoter was induced 5.7-, 7.1-, and 5.2-fold for 2-, 3-, 4-chlorophenol, respectively (cf. 8.9-fold induction with o-cresol) : however, TCE and PCE did not directly induce the tou path. Gas chromatography and chloride ion analysis also showed that TCE induced ToMO expression in P.stutzeri OX1 and was degraded and mineralized. This is the first report of significant PCE induction of any enzyme as well as the first report of chlorinated compound induction of the tou operon. The results indicate TCE and chlorinated phenols can be degraded by P.stutzeri OX1 without a separate inducer of the tou pathway and without competitive inhibition.

Foams for Aquifer Remediation: Two Flow Regimes and Its Implication to Diversion Process

  • Foam reduces the mobility of gas phase in porous media to overcome gravity override and to divert acid into desired layers in the petroleum industry and to enhance the efficiency of environmental remediation. Recent experimental studies on foam show that foam exhibits a remarkably different flow rheology depending on the flow regime. This study, for the first time, focuses on the issues of foam diversion process under the conditions relevant to groundwater remediation, combining results from laboratory linear-flow experiments and a simple numerical model with permeability contrasts. Linear flow tests performed at two different permeabilities (k = 9.1 and 30.4 darcy) confirmed that two flow regimes of steady-state strong foams were also observed within the permeability range of shallow geological formations. Foam exhibited a shear-thinning behavior in a low-quality regime and near Newtonian rheology in a high-quality regime. Data taken from linear flow tests were incorporated into a simple numerical model to evaluate the efficiency of foam diversion process in the presence of permeability contrasts. The simple model illustrated that foam in the high-quality regime exhibited a successful diversion but foam in the low-quality regime resulted in anti-diversion, implying that only foam in the high-quality regime would be applicable to the diversion process. Sensitivity study proved that the success of diversion process using foam in the high-quality regime was primarily controlled by the limiting capillary pressures (${P_c}{^*}$) of the two layers of interest. Limitations and implications are also discussed and included.
