Studies on the Relationship Between the Contents of Absorbed Alkaline Elements and Acidity Level in Sand Medium by Some Pine Species (I) -Different pH levels of Sand Medium and Absorbed Sodium Contents- (국내재식(國內栽植) Pinus속(屬) 사경(砂耕)에서의 흡수염기류(吸收鹽基類)와 배지(培地) 산도(酸度)와의 관계(關係)에 대한 연구(I) -Sodium의 흡수축적량(吸收蓄積量)과 배지산도(培地酸度)에 대하여-)

  • This experiment was conducted to determine the responses of some pine species to acidity of cultured soils, and absorption behavior of alkaline elements and to observe the growth characteristics during afforesting period. The results were summarized as follows. 1. Total dry weight indicating growth volume was not much influenced by the acidity of cultured soils (pH 4~8) in all of the 4 species studied. Pinus koraiensis showed more dry weight than any other species, which indicated statistically significant difference. 2. T-R ratio of dry weight was not much affected by the acidity of cultured soils (pH 4~8) and the difference of cultured soils (pH 4~8) and by species. Growth phase also did not show remarkable change through the experiment period. 3. The average sodium content ratio in the plant body had no correlation with cultured soils (pH 4~8), bue significant differences were recognized among species statistically. 4. It was found that sodium content ratio was not much influenced by the cultured soils (pH 4~8), even when the tops and roots were separately analyzed. But, very significant difference was found among the species. Pinus koraiensis especially, showed greater volume growth maintaining low sodium content ratio than any other species, which indicates that Pinus koraiensis has unfavorable characteristics for growing on the seaside.

Effect of Pine Needle and Green Tea Extracts on the Survival of Pathogenic Bacteria (솔잎과 녹차 추출물이 식중독세균의 생존에 미치는 영향)

  • The sensitivity of various pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O157:H7 and Salmonella typhimurium) to the pine needle and green tea extracts was tested. Water extract of pine needle(PNW), 70% ethanol extract of pine needle(PNE), water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activty. Tryptic soy broth(TSB) containing 0∼2%(w/v) of pine needle and green tea extracts were inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacterium and incubated at 35$\^{C}$ for 24 hours. The standard plate count method was used to measure the inhibitory effect of the extracts. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were derived from the survival curves of pathogenic bacteria. Antibacterial activities of the pine needle and green tea extracts were compared with that of sodium benzoate, a preservative, by clear zone test. L. monocytogenes, S. aureus and A. hydrophila were completely inhibited at 0.4∼1.6% level while E. coli and S. typhimurium were very resistant to the pine needle extracts. Green tea extracts completely inhibited all strains tested at 0.2∼1.0% level and bactercidal to all strains except L. monocytogenes at 0.5∼2.0% level. Antibacterial activities of pine needle and green tea extracts were stronger than that of sodium benzoate. The order of antibacterial activities of pine needle and green tea extracts to the pathogenic bacteria was GTE > GTW > PNE > PNW. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.

Evaluation of the Addition of Sodium Lactate and a Fat Replacer in Very Low-fat Bologna (model system) on the Product Quality and Shelf-life Effect during Refrigerated Storage (젖산나트륨과 지방대체제의 첨가가 냉장저장 중 저지방 볼로나 소시지의 품질 및 저장성에 미치는 영향)

  • This study was performed to investigate the effect of sodium lactate (SL, 60%) and a mixed fat replacer (FR) on the product quality and shelf-life effect of low-fat bologna sausage (LFBS) in a model system during refrigerated storage. Low-fat and regular-fat bologna sausages had pH values ranged from 6.15 to 6.30 and water activity values ranged from 0.95 to 0.96. LFBS had a moisture content of 74~76%, <2% fat and 14~15% protein, whereas regular-fat bologna had 60% moisture, 22% fat and approximately 12% protein in the final products. Expressible moisture (%) increased (p<0.05) in all bolognas, resulting in the soft texture, as the storage time (weeks) increased. LFBS manufactured with SL and a FR had lower (p<0.05) the cooking loss (%) and had higher (p<0.05) texture profile analysis (TPA) values than the regular-fat counterpart. As the sodium lactate level increased up to 5% in the formulation of LFBS, vacuum purge and TPA hardness values also increased (p<0.05), but thiobarbituric acid (TBA) values decreased (p<0.05). Total plate counts of LFBS were reduced (p<0.05) when the addition level of 60% SL solution was more than 3.3%. This result indicated that the increased level of SL (>3.3%) in the combination of a FR in the formulation of LFBS improved the product quality and did inhibit the total microbial growth of LFBS during storage, as compared to the control.

  • Effect of insecticide carbofuran and phenobarbital sodium(PB) or 3-methylcholanthrene(3-MC), they were orally administered by the chemicals, alone or in combination, on activities of several enzymes in rats were investigated. In in vivo test for the effect of this chemicals on activity of enzyme in rat, activities of acetylcholinesterase(AChE) and butyrylcholinesterase(BuCheE) were inhibited by $20{\sim}70%$ for 48 hrs after the oral administration of carbofuran alone of 3.8mg/kg, whereas those were lowered at the beginning, but recovered to the control level after 24 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. The activity of glutathione S-transferase(GST) was inhibited by more than 15 to 35% for an early period of 0.5 to 6 hrs, in the case of the administration of carbofuran alone, whereas that was slightly inhibited at the beginning, recovered almost to the control level after 3 hrs, and raised by mere than 20% above the control after 6 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. When carbofuran was administered alorig with PB or 3-MC, the activities of UDP-glucuronosyltransferase(UDPGT) and cytochrome P-450 were more than 2.6 to 2.8 times higher than that in the case of the administration of carbofuran alone for 6 hrs. These results suggest that a simultaneous application of carbofuran and PB or 3-MC is critical for the enhancentment of activity of GST, UDPGT and cytochrome P450 and the protection of rat from carbofuran toxicity.

Effect of Salinity on Growth and Osmoregulation of Sweetfish, Plecoglossus altivelis (은어, Plecoglossus altivelis의 성장과 삼투압 조절에 미치는 염분의 영향)

  • In order to investigate the effect of salinity on growth and osmoregulation of the sweetfish, Plecoglossus altivelis, the fish (post-larva, juvenile, adult) reared in seawater were directly transferred into the experimental rearing water of the different salinities (post-larva : $0\textperthousand$, $5\textperthousand$, $10\textperthousand$, $15\textperthousand$, $20\textperthousand$, $30\textperthousand$. juvenile : $0\textperthousand$, $\10textperthousand$, $20\textperthousand$, $30\textperthousand$. adult : $0\textperthousand$, $15\textperthousand$, $35\textperthousand$, respectively). Fish samples were taken in time courses after transfer for blood and body fluid collection. The changes of sodium, potassium concentration and osmolality of the body fluid and plasma, muscle water content, gonadosomatic index (GSI) and hepatosomatic index (HSI) in the experimental fish were observed in time courses. Growth rate of the post-larva in $\5~25\textperthousand$ were faster than in $0\textperthousand$ (P<0.05). Survival rate was very low in $0\textperthousand$ (P<0.05). No fish survived more than 50 days after transferred into $0\textperthousand$. The concentration of sodium, potassium and osmolality in body fluid of the juvenile sweetfish were slightly decreased at 0.25 day after the beginning of the maintainable level of sodium, potassium and osmolality were $36.4\pm6.2 mM/\ell, 47.5\pm0.7 mM/\ell, 366.3\pm19.1 mOsm/kg, respectively. Except potassium concetration, fish in $0\textperthousand$ and $10\textperthousand$ displayed the pattern of hypoosmotic regulation. Growth rate and survival rate of juveniles of $30\textperthousand$ are significantly low than other groups (P<0.05). The concentration of sodium, potassium and osmolality in plasma of adult sweetfish were slightly decreased at 1 day after the beginning of the maintainable level of sodium, potassium and osmolality were $159.0\pm2.5 mM/\ell, 5.8\pm0.9 mM/\ell, 338.1$\pm13.3 mOsm/kg respectively. Compared with these values, fish in $0\textperthousand$ and $15\textperthousand$ displayed hypoosmotic regultion. Growth and survival rate of adult in $35\textperthousand$ are significantly low than other experimental groups. GSI was the highest in $15\textperthousand$ after 60 days. HSI was the lowest in $35\textperthousand$. With decreased salinities, oxygen consumption was found to be decreased in the other experimental groups compared to the more than $30\textperthousand$.

Effects of Sodium Sulfite and Extrusion on the Nutritional Value of Soybean Meal in Piglets Weaned at 21 Days

  • A total of 80 weaned piglets (Landrace $\times$ Yorkshire $\times$ Large White) were used in a 28-day growth assay to detennine the optimal inclusion level of sodium sulfite ($Na_{2}SO_{3}$) as an extrusion enhancer of soybean meal for nursery piglets. piglets (21 d of age, 6.04 kg of BW) were grouped into 4 treatments in a completely randomized block design. Treatments were: 1) Extruded SBM (Control), 2) Extruded SBM with 0.5% $Na_{2}SO_{3}$ (0.5 ESBM), 3) Extruded SBM with 1.0% $Na_{2}SO_{3}$ (1.0 ESBM) and 4) Extruded SBM with 1.5% $Na_{2}SO_{3}$ (1.5 ESBM). Each treatment has 4 replicates of 5 heads per pen. In phase I (d 0 to 14), diets supplied 3,400 kcal ME/kg, 23% crude protein, 1.65% lysine, 0.50% methionine, 0.9% Ca and 0.8% P. Phase II (d 14 to 28) diets contained 3,300 kcal ME/kg, 21% crude protein, 1.45% lysine, 0.45% methionine, 0.9% Ca and 0.8% P. For d 0 to 14, piglets fed 1.5 ESBM had greater ADG, ADFI and FCR compared to piglets fed control and 0.5 ESBM diet. ADG was significantly higher in piglets fed 1.5 ESBM diet than other groups (p<0.05) except 1.0 ESBM. In phase II (d 14 to 28), there was no significant differences in production traits among treatments. For overall period (d 0 to 28), piglets fed diets with high sodium sulfite grew faster than piglets fed control and 0.5 ESBM diets. The highest ADG and the best FeR were obtained in piglets fed diets with 1.5 ESBM during the entire period. Piglets fed 1.5 ESBM diet showed significantly higher crude protein digestibility than 0.5 ESBM (p<0.05) at d 14 post-weanling, but not at d 28 post-weanling. There were no significant differences in digestibilities of total amino acids. In conclusion, the addition level of 1~1.5% sodium sulfite for SBM extrusion could be favorable for rate and efficiency of growth in weaning pigs.

Effect of Dietary Zinc Levels on Calcium, Magnesium, Sodium, and Potassium Contents in Rats (아연의 섭취 수준이 흰쥐의 칼슘, 마그네슘, 나트륨, 칼륨 함량에 미치는 영향)

  • The effect of dietary zinc levels on macro mineral contents was investigated in rats. Rats were divided into three groups by zinc levels, such as low (50% of requirement, Zn/L), adequate (100%, Zn/A) and high (200%, Zn/H) levels of dietary zinc. The calcium, magnesium, sodium and potassium contents in serum and organs were measured. The food intake was significantly higher in Zn/L group compared to that of Zn/H group. The body weight gain in Zn/H group was significantly lower than that of other two groups. There were no significant differences in liver, kidney and spleen weights. Also, serum calcium, magnesium, sodium and potassium concentrations were not significantly changed by the zinc diet at any level. In the low zinc diet group, liver calcium content was significantly lower, but the magnesium and potassium contents in spleen were significantly higher than that in other groups. The calcium, magnesium, sodium and potassium contents of kidney were not significantly different for any level of zinc. In conclusion, the macro mineral contents of liver and spleen in the normal rats were changed by zinc levels. Based on this study, the interaction between micro and macro minerals remains to be further studied.

PLASMA CONCENTRATION OF L. THYROXINE (T4). L. TRIIODOTHYRONINE (T3) AND CERTAIN OTHER BLOOD BIOCHEMICAL CONSTITUENTS IN GROWING CROSSBRED (B0S taurus × B0S indicus) CALVES

  • The relationship of thyroid status and certain blood biochemical constituents with body weight gains (ADG) and age (13 to 96 weeks) was studied in Holstein Friesian ${\times}$ Hariana ($1/2F{\times}1/2H$) crossbred male calves by assessing their plasma triiodo thyronine ($T_3$), Thyroxine ($T_4$), sodium, potassium, total proteins and cholesterol level at two energy levels. Body weight gains (ADG) were higher during the 50 to 72 weeks of age and declined thereafter, the plasma $T_3$ conc. was aignificantly (p < 0.01) higher during this period compared to all other periods. There was no significant variation due to energy level. Overall mean plasma $T_3$ conc. was $1.19{\pm}0.12ng/ml$. Plasma $T_4$ conc. did not show any significant variation either between the different age periods or between the two energy levels. The mean plasma $T_4$ conc. was $37.34{\pm}1.32ng/ml$. The plasma sodium and potassium concentration did not vary significantly due to energy levels. But amongst the different age periods, sodium concentration was highest ($147.70{\pm}2.29mEq/L$) during 49-60 weeks of age and lowest ($134.70{\pm}1.78mEq/L$) during 13-24 weeks, where as for potassium concentration changes were nonsignificant. There was very little variation amongst other periods. Plasma protein level was higher at 100% energy level than at 75%. Amongst the different age period, it was significantly lower ($6.44{\pm}0.36$) during 13 to 24 weeks of age than at 37 to 48 weeks of age ($7.14{\pm}0.11$). Plasma cholesterol values were higher for 75% energy level than that of 100%. Between the periods it was highest during 61 to 72 weeks of age and the difference amongst the age period were highly significant.

Effect of Cold Exposure on Thyroid Thermogenesis in Rats (한냉에 노출된 흰쥐에서 갑상선 호르몬이 체열 생산인 미치는 영향)

  • It has been well documented that animals exposed to cold show increased activity of thyroid gland. The calorigenic action of thyroid hormone has been demonstrated by a variety of in vivo and in vitro studies. According to Edelman et al., the thyroid thermogenesis is due to activation of energy consuming processes, especially the active sodium transport by the hormone in target tissues. If so, the increase in thyroid activity during cold exposure should induce increased capacity of sodium transport in target tissue and the change in tissue metabolism should be precisely correlated with the change in Na+_K+_ATPase activity of the tissue. This possibility was tested in the present study: in one series, changes in oxygen consumption and Na+_K+_-ATPase activity of liver preparations were measured in rats as a function of thyroid status, in order to establish the effect of thyroid hormone on the tissue respiration and enzyme system in another series, the effect of cold stimulus on the serum thyroid hormone level, hepatic tissue oxygen consumption and Na+_K+_ATPase activity in rats. The results obtained are as follows: 1. The Na+_dependent oxygen consumption of liver slices, the oxygen consumption of liver mitochondria and the Na+_K+_ATPase activity of liver preparations were significantly inhibited in hypothyroidism and activated in hyperthyroidism. Kinetic analysis indicated that the Vmax. of Na+_K+_ATPase was decreased in hypothyroidism and increased in hyperth)'roidism. 2. In cold exposed rats, the serum triiodothyronine (T₃) level increased rapidly during the initial one day of cold exposure, then declined slowly to the control level after two weeks. The serum thyroxine (T₄) level decreased gradually throughout the cold exposure. Accordingly the T₃/T₄ratio increased. The mitochondrial oxygen consumption and the Na+_dependent oxygen consumption of liver slices increased during the first two days and then remained unchanged thereafter The activity of the Na+_K+_ATPase in liver preparations increased during cold exposure with a time course similar to that of oxygen consumption. Kinetic analysis indicated that the Vmax. of Na+_K+_ATPase increased. 3. Once the animal was adapted to cold, induction of hypothyroidism did not significantly alter the hepatic oxygen consumption and Na+_K+_ATPase activity. These results indicate that: 1) thyroid hormone increases capacities of mitochondrial respiration and active sodium transport in target tissues such as liver; 2) the increased T₃level during the initial period of cold exposure facilitates biosynthesis of Na+_K+_ATPase and mitochondrial enzymes for oxidative phosphorylation, leading to enhanced production and utilization of ATP, hence heat production.

  • Effects of addition level of sodium chloride (NaCl) on gel properties of surimi-like pork (SLP) were investigated. Porcine semimembranosus muscle was used to manufacture SIP contained 1, 2, 3 and 4% NaCl to measure moisture content, pH, color, gel strength, micro-structure and sensory evaluation. The pH and moisture content of SLP were decreased as increasing of NaCl level. However, the gel strength of SLP was increased with increasing of NaCl level. Values of yellowness and chroma were lower in SLP of 2% and 3% NaCl compared with those of 1% and 4% NaCl. Amorphus protein particles size in micro-structure of SLP was decreased and coagulated as increasing level of NaCl. SLP of 1% NaCl had a structure formed by aggregates of densely packed globular proteins and arranged in clusters, whereas a well-structured matrix with a highly interconnected network of strand was observed in SLP of 4% NaCl. Result suggested that the increasing gel strength with NaCl level might be due to lower moisture content and denser micro-structure of gel.
