Study on the molecular weights of radioprotective ginseng proteins by HPLC method

  • Partialy purified ginseng proteins were either treated with sodium dodecyl sulfate (SDS) and .betha.-mercaptoethanol to denature the proteins or not, and subjected to Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC) to compare the components of each fraction. Standard proteins of known molecular weights (MW) were also either treated with SDS and .betha.-mercaptoethanol or not, and subjected to HPLC to obtain regression lines for MW determination. From the retention times obtained from samples in eiether case by HPLC, the MW were estimated as following in SDS treated condition, Gl fraction showed three peaks each with MW of above 100, 000, 51, 000 and 19, 000. Gll showed one original peak with MW of 21, 000 and Gll, two peaks each with MW of 19, 000 and 14, 000. On the other hand, in non-SDS treated condition, GI fraction showed two peaks each with MW of above 200, 000 NA 52, 000. Gll showed one original peak with MW of 41, 000 and Gill, three peaks each with MW of 28, 000, 19, 000 and 14, 000.

Voltammetric Study of Anionic Surfactant-Modified Glassy Carbon Electrode for Bis(2,2'-Bipyridyl)Copper(II) in 200 mM NaBr

  • When a hydrophobic and hydrophilic environments of bis(2,2'-bipyridyl) copper(II) ($Cu(bpy)_2{^{2+}}$) are produced in the presence of anionic surfactant sodium dodecyl sulfate (SDS), cyclic voltammetry is used to investigate the microscopic environments which occurs at the glassy carbon electrode. In order to see the relation between ${\Delta}E_p$ and a critical micelle concentration (CMC), ${\Delta}E_p$ vs. -Log[SDS] for the redox couples are plotted. The concentration at the intersection of two lines is 2.57 mM SDS, and this concentration can be determined as the CMC (relative error: below 0.03%; 2.63 mM SDS by surface tensiometry).

  • 질트리코모나스의 항원 분석을 시행하기 위하여 병원성이 화인된 질트리코모나스(Trichamonas vaginnlis) 6개주(HY-1,2,3,9,10,13)의 항일을 sodium dodecyl sulfate - polyacrlyamide gel electrophoresis(SDS-PAGE) 한 결과 Coomassie brilliant blue 염색상에서 peptide의 밀도 차이를 제외하고는 주간에 동일한 단백질 분포양상을 보였으며, 12kDa에서 170kDa까지 약 35개 정도의 분획이 관찰되었다. 또한 질트리코모나스 HY월주를 항원으로 하고 질트리코모나스 6개주에 대해 면역된 마우스의 항혈청을 이용하여 enzyme-linked immunoelectrotransfer blot(EITB)을 시행한 결과 각 주마다 서로 다른 반응양상을 보였으며, 51kDa과 96kDa에서 질트리코모나스의 특이한 공통 반응대가 관찰되었고, 각기 다른 6개주의 항원에 대해 HY-1주의 항혈청으로 ETTB한 결과 HY-1 함원(homologous antigen)과의 반응 양상과 타항친(heterologous antigen)과의 반응 양상간의 특이한 차이는 관찰할 수 없었다. 이상의 결과로 보아 질트리코모나스 각 주의 항원은 항인-항체 반응에서 주간에 이종(antigenic heterogeneity)을 형성하고 있는 것으로 보였으며, 41, 47, 55, 74 및 94kDa에서 질트리코모나스애 특이한 공통반응대를 보었으며, 이 부분이 숙주-기생충의 상 호관계에 있어서 중요한 의의를 내포하고 있을 것으로 생각된다.

카본나노튜브에 흡착된 휴믹산의 탈착에 관한 연구 (Desorption of Adsorbed Humic Acid on Carbon nano Tubes)

  • Concerns have been raised over the impact of nano materials on soil and groundwater environment with the increasing attention to the potential applications of carbon nano materials in various fields. Particularly, carbon nano materials introduced into water environment readily make complexes with humic acid (HA) due to their hydrophobic nature, so there have been increasing numbers of studies on the interaction between HA and carbon nano materials. In this study, we investigated the solubility of HA and multiwalled carbon nanotubes (MWCNT) in three different surfactant solutions of sodium dodecyl sulfate (SDS), Brij 30 and Triton X-100, and evaluated whether the HA can be effectively desorbed from the surface of MWCNT by surfactant. The objective of this study was to determine the optimal adsorption condition for HA to MWCNT. Futhermore, sodium dodecyl sulfate (SDS), Brij 30, Triton X-100 were used to elucidate the effect of desorption and separation on adsorbed HA on MWCNT. As a result, HA solution with 12.7 mg of total organic carbon (TOC) and 5 mg of MWCNT showed the highest adsorption capacity at pH 3 reacted for 72 hrs. Weight solubilizing ratio (WSR) of surfactants on HA and MWCNT was calculated. HA had approximately 2 times lower adsorption capacity for the applied three surfactants compared to those of MWCNT, implying that the desorption of HA may occur from the HA/MWCNT complex. According to the results of adsorption isotherm and weight solubilizing ratio (WSR), the most effective surfactants was the SDS 1% soluiton, showing 53.63% desorption of HA at pH 3.

Hydrophobic-Hydrophilic 분체의 계면활성제에 의한 Wettability 연구 (A Study on the Wettability by Surfactant of Hydrophobic-Hydrophilic Powders)

  • 분체 형태의 소수성 고분자 polymethylmethacrylate(PMMA), poly vinyl chloride(PVC)와 친수성 금속 aluminium에 대한 water, glycerol, formamide, diiodomethane, 1,1,2,2-tetrabromoethane, 1-bromonaphthalene, acetone, chloroform, benzene, ethanol과 methanol과의 접촉각을 Washburn식을 기본으로 한 wicking법으로 측정하였다. 또한, 농도에 따른 음이온 계면활성제 SDS(sodium dodecyl sulfate)와 양이온 계면활성제 CTAB(cetyl trimethylamonium bromide) 수용액과의 접촉각을 측정하였다. 유기액체들과의 측정된 접촉각으로부터 Zisman plot을 사용하여 임계표면장력, ${\gamma}_c$를 예측하였으며, 농도에 따른 계면활성제 SDS와 CTAB수용액과의 접촉각으로부터 adhesion tension plot을 사용하여 ${\gamma}_c$를 예측하였다. 유기액체, 계면활성제 SDS와 CTAB수용액에 대해서 분체 PMMA의 ${\gamma}_c$는 각각 43.5, 28.3과 $36.2mNm^{-1}$이며, 분체 PVC의 ${\gamma}_c$는 각각 44.2, 28.0과 $34.8mNm^{-1}$값을 얻었다. 이로부터, 고체 표면의 적심성과 성질이 계면활성제의 첨가에 의해 변화됨을 알 수 있었다. 또한, 측정된 접촉각으로부터 조화평균식을 사용하여 각 고체의 표면자유에너지. ${\gamma}_s$와 그 성분 값들을 결정하였다.

Comparative Study of Emission Quenching of Tris(${\alpha},{\alpha}'$-diimine)-Ruthenium(II) Complexes in Homogeneous and Sodium Dodecyl Sulfate Micellar Solutions

  • Emission quenching of photoexcited tris(${\alpha},{\alpha} '$-diimine)-ruthenium(II) complex cations, $RuL_3^{2+}$ (L: 2,2'-bipyridine, 4,4'-dimethyl-2,2'-bipyridine; 4,4'-diphenyl-2,2'-bipyridine; 1,10-phenanthroline; 5-methyl-1,10-phenanthroline; 5,6-dimethyl-1,10-phenanthroline or 4,7-diphenyl-1,10-phenanthroline) by $Cu^{2+}$, dimethylviologen $(MV^{2+})$, nitrobenzene (NB), and oxygen was studied in aqueous homogeneous and sodium dodecyl sulfate (SDS) micellar solutions. The apparent bimolecular quenching rate constants $k_q$ were determined from the quenching data and life-times of $^{\ast}RuL_3^{2+}$. In homogeneous media, the quenching rate was considerably slower than that for the diffusion-controlled reaction. The decreasing order of quenching activity of quenchers was $NB>O_2>MV^{2+}>Cu^{2+}$. The rate with $Cu^{2+}$ was faster as the reducing power of $^{\ast}RuL_3^{2+}$ is greater. On the other hand, the rates with NB and $O_2$ were faster as the ligand is more hydrophobic. This was attributed to the stabilization of encounter pair by van der Waals force. The presence of SDS enhanced the rate of quenching reactions with $Cu^{2+}$ and $MV^{2+}$, whereas it attenuated the quenching activity of NB and $O_2$ toward $RuL_3^{2+}$. The binding affinity of quenchers to SDS micelle and binding sites of the quenchers and $RuL_3^{2+}$ in micelle appear to be important factors controlling the micellar effect on the quenching reactions.

음이온 계면활성제를 사용한 modified Fenton 반응의 과수안정제 개발에 관한 연구 (A Study about Development of Hydrogen Peroxide Stabilizer in Modified Fenton Reaction Using Anion Surfactant)

  • 본 연구에서는 modified fenton 반응에서 과산화수소를 안정화하여 오염토양 정화의 효과를 증대시키고자 하였다. 오염토양을 모사하기 위하여 PAHs 계열의 대표적인 오염물질인 phenanthrene을 사용하였다. 과수안정제로는 음이온 계면활성제인 SDS(Sodium dodecyl Sulfate)를 사용하였다. Modified Fenton 반응에서 phenanthrene의 제거율을 확인하기 위하여 Fe(II) 4 mM, SDS 5~50 mM 및 $H_2O_2$ 102.897 mM를 phenanthrene 125 mg/kg으로 오염된 토양에 주입하였다. 과수안정제인 SDS가 30 mM이 사용된 경우 phenanthrene의 제거 효율이 95%로 가장 높게 나타났으며 30 mM이싱에서는 시스템에서 SDS가 scavenger로 작용하여 오염물질의 제거효율이 SDS 30 mM 일 때 보다 낮게 나타났다. 과수안정제를 사용한 뒤 과산화수소의 농도변화를 분석한 결과 Fe(II) 2 mM에서 48시간 이후 14.6995 mM 이상 남아있어서 가장 안정적이었지만, Fe(III)을 주입한 경우에는 과산화수소가 안정화되지 않았다. Modified Fenton 반응에서 철과 SDS 농도 사이의 최적의 비율을 찾기 위하여 SDS의 농도는 30 mM로 고정하고 철의 농도를 2~8 mM로 변화시켜 실험한 결과 Fe(II) 4 mM 및 SDS 30 mM에서 약 95%의 가장 높은 제거율을 보였다.

  • 도데실황산나트륨(SDS)이 수식된 유리탄소전극에 의해 철(III) 이온의 정량분석이 선택성 있게 제안되었다. 이것은 SDS와 $Fe^{3+}$의 정전기적 인력으로 착물이 형성되는 데 근거한 것이다. 철(III) 이온의 정량분석은 시차펄스전압전류법(DPV)에 의해 하였고, 그 정량분석을 위한 $(DS^-)_n-Fe^{3+}$의 환원 피크는 +0.466(${\pm}0.002$)volt (vs. Ag/AgCl)였다. 철(III) 이온의 정랑분석을 위한 검량선은 $0.50{\times}10^{-5}{\sim}10{\times}10^{-5}mol/L$의 농도 범위에서 얻었으며, 검출한계는 $0.14{\times}10^{-5}mol/L$였다. $Cu^{2+}$, $Ni^{2+}$, $Co^{2+}$, $Pb^{2+}$, $Zn^{2+}$$Mn^{2+}$는 철(III) 이온의 정량에 거의 영향을 미치지 않으나, $CN^- $$SCN^-$은 철(III) 이온의 정량을 크게 방해하였다.

Study of Kinetics of Bromophenol Blue Fading in the Presence of SDS, DTAB and Triton X-100 by Classical Model

  • In this paper, kinetics of reaction between Bromophenol blue (BPB) and $OH^-$, called fading, has been studied through a spectrophotometric method in the presence of nonionic Triton X-100 (TX-100), anionic sodium dodecyl sulfate (SDS) and cationic dodecyl trimethylammonium bromide (DTAB) surfactants. The influence of changes in the surfactant concentration on the observed rate constant was investigated. The results are treated quantitatively by pseudophase ion-exchange (PPIE) model and a new simple model called "classical model". The binding constants of BPB molecules to the micelles and free molecules of surfactants, their stoichiometric ratios and thermodynamic parameters of binding have been evaluated. It was found that SDS has nearly no effect on the fading rate up to 10 mM, whereas TX-100 and DTAB interact with BPB which reduce the reaction rate. By the use of fading reaction of BPB, the binding constants of SDS molecules to TX-100 micelles and their Langmuir and Freundlich adsorption isotherms were obtained and when mixtures of DTAB and TX-100 were used, no interaction was observed between these two surfactants.

체외성숙 우난포란의 체외수정과 발달에 관한 연구 IV. 난구세포의 생화학적 특성 검토 (Studies on In Vitro Fertilization and Development of Bovine Follicular Oocytes Matured in Vitro)

  • These experiments were undertaken as a basic study to understand the role of cumulus cell on in vitro maturation and fertilization process with identifying the cumulus cell-secreted proteins. By sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) and fast protein liquid chromatography(FPLC), the proteins of cumulus cells were identified. The results obtained in these experiments were summarized as follows ; 1. When the proteins secreted from cultured cell for 30 hours were separated by SDS-PAGE, there were a major band (>94,000) and other minor 2 bands with molecular weight ranging 30,000∼67,000 dalton. 2. In addition, the fractionations of these proteins by FPLC were idirectly shown that three bands were hyaluronic acid, chondroitin sulfate, and heparin.
