  • The leukocyte NADPH oxidase of neutrophils is a membrane-bound enzyme that catalyzes the production of $O_2^-$ from oxygen using NADPH as an electron donor. Dormant in resting neutrophils, the enzyme acquires catalytic activity when the cells are exposed to appropriate stimuli. During activation, the cytosolic oxidase components $p47^{phox}$ and $p67^{phox}$ migrate to the plasma membrane, where they associate with cytochrome $b_{558}$, a membrane-bound flavohemoprotein, to assemble the active oxidase. The oxidase can be activated in a cell-free system; the activating agent usually employed is an anionic amphiphile such as sodium dodecyl sulfate (SDS). Because $p47^{phox}$ can translocate by itself during activation, the conformational change in $p47^{phox}$ may be responsible for the activation of NADPH oxidase. We show here that the treatment of $p47^{phox}$ with SDS leads to an increase in the reactivity of the sutbydryl group of cysteines toward N-ethylmaleimide, indicating that the conformational change occurs when $p47^{phox}$ is exposed to SDS. We propose that this change in conformation results in the appearance of a binding site through which $p47^{phox}$ interacts with cytochrome $b_{558}$during the activation process.

  • It was hypothesized that NaF induces calcium sensitization in $Ca^{2+}$-controlled solution in permeabilized rat mesenteric arteries. Rat mesenteric arteries were permeabilized with $\beta$-escin and subjected to tension measurement. NaF potentiated the concentration-response curves to $Ca^{2+}$ (decreased $EC_{50}$ and increased $E_{max}$). Cumulative addition of NaF (4.0, 8.0 and 16 mM) also increased vascular tension in $Ca^{2+}$-controlled solution at pCa 7.0 or pCa 6.5, but not at pCa 8.0. NaF-induced vasocontraction and $GTP{\gamma}S$-induced vasocontraction were not additive. NaF-induced vasocontraction at pCa 7.0 was inhibited by pretreatment with Rho kinase inhibitors H1152 or Y27632 but not with a MLCK inhibitor ML-7 or a PKC inhibitor Ro31-8220. NaF induces calcium sensitization in a $Ca^{2+}$ dependent manner in $\beta$-escin-permeabilized rat mesenteric arteries. These results suggest that NaF is an activator of the Rho kinase signaling pathway during vascular contraction.

  • The critical element for sustainable growth in the construction industry is the development of alternative cements. A new technological process called geopolymerization provides an innovative solution, and the presence of aluminum and silicon oxides in fly ash has encouraged its use as a source material. Many previous investigations have involved curing the binder in a heated environment. To reduce energy consumption during the synthesis of geopolymers, the present study investigated the properties of ambient cured geopolymer mortar at early ages. An experimental program was executed to establish a relationship between the activator composition and the properties of geopolymer mortar in fresh and hardened states. Concentrations of sodium hydroxide and sodium silicate were ascertained that are advantageous for constructability and mechanical behavior. Scanning electron microscopy, energy dispersive X-ray spectroscopy and X-ray diffraction techniques were also used to characterize the material. Test results indicate that there is potential for the concrete industry to use fly ash based geopolymer as an alternative to portland cement.

  • This study is interested in identifying the effectiveness of alkali-activated fire protection material compounds including the alkali-activator such as potassium hydroxide, sodium silicate and fly ash as the fire resistant finishing materials. Also, this paper is concerned with change in compressive strength and pore structure of the alkali-activated fire protection material at high temperatures. The testing methods of fire protection materials in high temperature properties are make use of TG-DSC and mercury intrusion porosimetry measurements. This study results show that compressive strength is rapidly degraded depending on a rise of heating temperature. Porosity showed a tendency to increase irrespective of specimen types. This is due to both the outbreak of collapse of gel comprising the cement and a micro crack by heating. However, alkali-activated fire protection material composed of potassium hydroxide, sodium silicate and fly ash has the thermal stability of the slight decrease of compressive strength and porosity at high temperature. These thermal stability is caused by the ceramic binding capacity induced by alkali activation reaction by the reason of the thermal analysis result not showing the decomposition of calcium hydrate.

  • This study investigated the effects of blast furnace slag mortars activated with sodium hydroxide(NaOH) and gypsum in relation to flow, setting time and compressive strength. The parameters in this studied was the gypsum ratio 0 to 50%, 3M and 6M of activator concentration and $20{\pm}2^{\circ}C$ and $35{\pm}2^{\circ}C$ of curing temperatures. The results of flow was increase, setting time was increase as the amount of gypsum increases. But the results of compressive strength was dependent on the gypsum ratio, indicating that the compressive strength increased with the increase of the amount of gypsum until a certain limit, beyond which the strength decreased quickly.

  • Geopolymer is a sustainable concrete, replaces traditional cement concrete using alternative sustainable construction materials as binders and alkaline solution as alkaline activator. This paper presents the strength characteristics of geopolymer concrete (GPC) developed with fly ash and GGBS as binders, combined Sodium silicate ($Na_2SiO_3$) and Sodium Hydroxide (NaOH) solution as alkaline activators. The parameters considered in this research work are proportions of fly ash and GGBS (70-30 and 50-50), curing conditions (Outdoor curing and oven curing at $600^{\circ}C$ for 24 hours), two grades of concrete (GPC20 and GPC50). The mechanical properties such as compressive strength, split tensile strength and flexural strength along with durability characteristics were determined. For studying the durability characteristics of geopolymer concrete 5% $H_2SO_4$ solutions was used and the specimens were immersed up to an exposure period of 56 days. The main parameters considered in this study were Acid Mass Loss Factor (AMLF), Acid Strength Loss Factor (ASLF) and products of degradation. The results conclude that GPC with sufficient strength can be developed even under Outdoor curing using fly ash and GGBS combination i.e., without the need for any heat curing.

  • Geopolymer concrete is environmentally friendly and could be considered as a construction material to promote the sustainable development. In this paper fly ash based geopolymer concretes with different percentages of alccofine were made by mixing sodium hydroxide and sodium silicate as an alkaline activator and cured at ambient as well as heat environment in an electric oven at $90^{\circ}C$. Effects of various parameters such as the percentage of alccofine, curing temperature, a period of curing, fly ash content, was studied on compressive strength as well as workability of geopolymer concrete. The study concludes that the presence of alccofine improves the properties of geopolymer concrete during a fresh and hardened state of concrete. Geopolymer concrete in the presence of alccofine can be used for the general purpose of concrete as required compressive strength can be achieved even at ambient temperature. The 28 days compressive strength of 73 MPa, when cured at 90-degree Celsius, confirmed that it is also very suitable for precast concrete components.

  • Our previous study has shown that sodium selenite can cause apoptosis in acute promyelocytic leukemia-derived NB4 cells in a caspase-dependent manner, but the detailed mechanism is unknown. Here we demonstrate a requirement for extracellular signal-regulated protein kinase (ERK) in mediating sodium selenite -induced apoptosis in NB4 cell. Though no apparent elevation of ERK activity was observed during the apoptosis in NB4 cells caused by 20 μM sodium selenite treatment, PD98059 and U0126, specific chemical inhibitors of the MEK/ERK signaling pathway, were shown to strongly prevent the apoptosis process, while ERK activator TPA enhanced the process. It is also known that p38 MAPK inhibitor SB203580 and JNK inhibitor SP600125 had slight effects on apoptosis. Further study indicated that ERK exerted its proapoptotic effect only at the early stage of apoptosis and played an antiapoptotic role at the later stages. Taken together, our findings suggest that ERK plays an active role in mediating sodium seleniteinduced apoptosis in NB4 cells .

  • Geopolymer Concrete is typically proportioned with activator solution leading to moderately high material cost. Such cost can be enduring in high value added applications especially when cost savings can be recognized in terms of reduction in size of the members. Proper material selection and mix proportioning can diminish the material cost. In the present investigation, a total of 27 mixes were arrived considering the mix parameters as liquid-binder ratio, slag content and sodium hydroxide concentration to study the mechanical properties of geopolymer concrete (GPC) mixes such as compressive strength, split tensile strength and flexural strength. The derived statistical Response Surface Methodology is beleaguered to develop cost effective GPC mixes. The estimated responses are not likely to contrast in linear mode with selected variables; a plan was selected to enable the model of any response in a quadratic manner. The results reveals that a fair correlation between the experimental and the predicted strengths.
