Effects of Seed Disinfectant and Soaking Time on Germination and Disease Occurrence of Adlay, Coix lacryma-jobi L. var. ma-yuen Stapf (율무 종자소독과 침종시간이 발아율 및 병발생에 미치는 영향)

  • This study was carried out to investigate the effect of soaking time of seed disinfectant, Fludioxonil on the germination and disease occurrence of adlay (Coix lacryma-jobi L. var. ma-yuen Stapf). The germination rate was higher in the seed disinfectant treatment for 3 days followed by seed soaking for 3 days than that of other treatments under controlled condition of low temperature. The value of two experiments were 90.8%, 96.7%, repectively. And, the time of seed soaking was longer, the more seed rot was proportionly decreased. In the pot and field experiment, germination rates were somewhat low compared to that of petri dish experiment as 88.3%, 84.3%, and occurence of seedling blight showed similar results that of petri dish experiments. There was not significantly different in the yield components among treatments in the field. However, as compared with untreated plot (194kg/10a), unhulled grain yield was increased about 26% on that treatment. Based on these results, it has been thought that improvement of the germonation rate and decrease of disease occurrence of adlay were accompanied with the seed disinfectant during 3 days followed by seed soaking during 3 days before sowing.

Effects of Scarification and Water Soaking Treatment on Germination of Hard-Seeded Legumes (두과 작물의 경실종자 발아촉진에 대한 종피연화처리의 효과)

  • The crops showing hard seed character have high seed viability after seed storage for long period. The low germination rate due to hard seed coat, however, cause a problem of low seedling establishment in field condition. Three legumes used in the experiment, lablab bean (Dolichos lablab L.), asparagus bean (Vigna sesquipedalis L. Fruwirth), and soybean (Glycine max L. Merr.), showed low germination rate (26, 17, and 5%, respectively) due to thick and hard seed coat. In this study artificial treatment for breaking dormancy was tested in hard seeds. The effect of proper treatment was various depending on species. The germination rate of lablab bean was highly improved up to 94% by soaking into water for 24 hours. In the case of asparagus bean, the rate was increased up to 90% by soaking for eight hours near boiling water until it cools. The germination rate of small hard seed soybean was increase to 96% by soaking into concentrated sulfuric acid for 10 minutes. Ultrastructural change revealed by scanning electron microscope (SEM) reflects that the structure of micropyle was changed and water uptake was facilitated with all treatments tested in the experiment. Especially, sulfuric acid treatment resulted in the degradation of micropylar tissue. These results demonstrate that the artificial treatment including sulfuric acid and (hot) water soaking treatment for promoting water uptake can be applied to improve seed germination in legume seed with thick and hard seed coat.

Changes in Ultrastructure of Tissues, Characteristics of Protein and Soybean Curd Yield with Increased Soaking Time during Soybean Curd Processing (대두(大豆)의 수침시간(水浸時間)에 따른 조직(組織)의 미세구조(微細構造), 단백질(蛋白質) 특성(特性) 및 두부수율(豆腐收率)의 변화(變化))

  • This experiment was conducted to investigate changes of soybean curd yield according to the extension of soaking time during manufacturing of soybean curd. To investigate those changes systematically, transmission electron microscopy and disc-gel electrophoresis were used. The soybean curd yield was increased from 45.0% to 50.5% and 55.4% respectively as soaking time is extended from 5 hours to 10 and 24 hours. The solid extraction and soybean milk coagulation were also increased according to the extension of soaking time. From disc-gel electrophoresis patterns of soybean milk protein and soybean curd protein, numbers of band were increased and major band thickened by expending the soaking time. Most of high molecular bands of soybean milk protein were transfered to soybean curd. Crude 7S proteins of soybean milk and soybean curd in dis-gel electrophoresis were appeared to be 4 and 5 bands respectively, and crude 11S proteins of soybean milk and soybean curd were appeared to be 9 and 8 bands respectively. Of soybean milk bands, most of 11S component transfered to soybean curd. Transmission electron photomicrographs revealed that the dimension of each protein body became larger and the numbers of spherosome around the protein bodies in unit area fewer by extending the soaking time of soybean.

Optimization for the Post-Harvest Induction of trans-Resveratrol by Soaking Treatment in Raw Peanuts (침지조작에 의한 레스베라트롤 증가조건의 최적화)

  • In this study, the effects of varying the amount of water, soaking time at $25^{\circ}C$ and drying time after soaking at $45^{\circ}C$ on the induction of resveratrol were evaluated to optimize the soaking treatment by response surface methodology (RSM). After response surface regression (RSREG), the second-order polynomial equation was fitted to the experimental data. The analysis of variance showed that the model appeared to be adequate $(R^2=0.9547)$ with no significant lack of fit (p>0.1). From statistical analysis, amount of water and soaking time were found to be significant factors. On the other hand, drying time was not significant. Ridge analysis showed that the optimized parameters were $67.15\%$ for amount of water, 19.58 hr for soaking time, 65.56 hr for drying time. It was confirmed that resveratrol content was increased from $0.1\;{\mu}g/g$ to $4.55\;{\mu}g/g$ under the optimized conditions. In addition, the experimental values at the optimized condition agreed with values predicted by ridge analysis. The analytical method validation parameters such as accuracy, precision, and specificity were calculated to ensure the method's validity.

Effects of Roasting Conditions on Quality and Yields of Barley Tea (보리의 볶음조건이 보리차의 품질 및 수율에 미치는 영향)

  • An investigation was carried out to improve the quality and yield of barley tea(water extracts) by modifying the roasting method. The modified methods employed were crushing the barley into 4-10 parts and soaking in water at room temperature for 20 minutes followed by roasting (light brown or dark brown) at $250^{\circ}C$ before boiling with water. The varieties of barley used for this study were Ol-bori(with hulls) and Youngsan-bori(without hulls) and the qualities measured were the physical, chemical and sensory characteristics of roasted barley and its extracts. The results showed that the higher solid yields in barley tea was obtained with an increase in soaking and roasting time. The solid yields increased rapidly with extraction time in boiling water until 30 minutes and then slowed down thereafter. However the intrinsic viscosity of barley tea was reduced by increase in soaking and roasting time. The color of barley tea expressed as Hunter L, a, b values showed that lower L value and higher a and b values were measured for those prepared without soaking and with more extensive roasting. Organoleptic evaluation clearly showed that the sensory quality of barley tea was significantly improved in intensity of odor and taste by crushing, soaking and dark brown roasting.

Solids Loss with Water Uptake during Soaking of Soybeans (대두의 침지과정중 침출액의 성분변화)

  • Dried soybeans (varieties: Saeal, Kwanggyo, Tanyob) took up water rapidly for first 3hr followed by a slower rate of uptake. The beans took up an equal weight of water (100% hydration)after approximately 3.5hr at $50^{\circ}C$, 5hr at $30^{\circ}C$ and 7.5hr at $20^{\circ}C$ respectively. pH of the soaking solutions decreased during the soaking period. This was undoubtedly caused by the ionization of the cellular components resulting in increased levels of hydrogen ions in the liquor. Soluble solids were leached out of the beans at fairly steady rate throughout the hydration and the amount was greater with higher temperature. This amounted to 0.4-0.7g at $20^{\circ}C$ and 10.2-15.0g at $50^{\circ}C$ per 100g soybeans. Temperature was the most important factor in determining the rate of water absorption and of solid losses. Of the total solids lost, 12-25% was protein. The proportion of protein loss increased as the soaking time and temperature increase. Amount of protein loss was 80-200mg at $20^{\circ}C$ and 440-480mg at $50^{\circ}C$ after 24hr soaking per 100g soybeans. About 5% of soluble sugars, including fructose, sucrose, raffinose, and stachyose, was removed from the beans after 24hr soaking at $20^{\circ}C$.

Physicochemical quality properties of oriental melon pickle with the harvest period after pollination and soaking conditions (수정 후 수확시기와 염지조건에 따른 참외피클의 이화학적 품질특성)

  • This study was conducted to investigate the physicochemical quality properties of oriental melon pickles harvested after pollination (green and fully ripe period) and soaking (cured and ripened). After the increase of the pollination size of melon, at 21 days, melon had a strong yellowish color, but at 24 days, its tip was completely yellow. In addition, 21 days after 5 days soaking, the weights of the harvested melons were reduced. Since then, there were no remarkable changes in weight. At 27 and 30 days, the b values of the melons that were harvested showed no trace of influence by the number of days of soaking and ripening. The melons harvested after 24 and 27 days, and the oriental melon pickles, maintained a constant hardness and texture after five days of soaking. This result indicates that melons harvested between 15 and 27 days after pollination can be used for melon pickles. To produce high-quality melon pickles with a yellowish color and a chewy texture, melons harvested 27 days after pollination and then soaked and ripened in salt water for 15 days should be used.

Effects of Water-soaking and Mechanical and Chemical Scarifications on Seed germination of Hairy Vetch (Vicia villosa Roth) (헤어리베치 종자의 침지처리와 종피 파상 및 황산처리가 발아에 미치는 영향)

  • Seed coat of hairy vetch is very thick and hard, and difficult to absorb water during germination. It requires much time that cotyledon comes out from seed coat. Therefore this experiment was carried out to increase the germination rate by enhancing water absorption through water-soaking of seed, chemical scarification by sulfuric acid, and mechanical scarification on seed coat. Water-soaking for 5 hours seemed to be highly effective in enhancing germination rate. Although not significant, water-soaking resulted in increase of germination rate by 9%. Effect of soaking temperature was not significant, but early germination rate was increased to be 5.0%, 31.7% and 48.3% at $10^{\circ}C$, $20^{\circ}C$ and $30^{\circ}C$, respectively. Mechanical scarification of seed coat led to a germination rate of 97% whereas intact seed showed that of 65%. Chemical scarification by sulfuric acid for 10min, 20min and 30min resulted in a germination rate of 76.7%, 74.7% and 96.0% respectively. It is clear that scarification increased germination rate.

Soaking Properties and Quality Characteristics of Korean white Gruel with Different Blending Time of High-Dietary Fiber Rice'Goami 2' (고아미 2호의 수침특성 및 마쇄 시간을 달리한 흰 죽의 품질 특성)

  • The objectives of this study were to investigate the soaking properties of the high-dietary fiber rice 'Goami 2'and to develop korean white Gruel prepared with Goami 2. With increasing soaking time at room temperature ($20^{\circ}C$), the water absorbing character of Goami 2 and Ilpum significantly increased during the first hour of soaking time, after which it remained constant. On the contrary, the hardness of Goami 2 and Ilpum significantly decreased with increasing soaking time until one hour, after which it remained constant. The properties of Korean white gruel were evaluated using two varieties of rice (Goami 2, Ilpum) and three blending times (10, 20 and 30 seconds respectively). The rice flour of Goami 2 for Korean white gruel showed a greater number of small particles (<20 $\mu$m) than that of Ilpum. Hunter a'and b'values of Korean white gruel prepared with Goami 2 were higher than that of Korean white gruel prepared with Ilpum. The consistency values on Bostwick consistometer of Korean white gruel prepared with Goami 2 were higher than those of Korean white gruel prepared with Ilpum. The texture of Korean white gruel was examined using a back extrusion rig. All of the rheological parameters of Korean white gruel prepared with Goami 2 were decreased with increasing blending time, while in llpum they were increased. In the sensory evaluation results, the overall acceptability of korean white gruel prepared with Goami 2 blended for 20sec showed the highest sensory scores and desirability.
