Protective Effect of Sanguisorba officinalis L. Root on Amyloid ${\beta}$ Protein (25-35)-induced Neuronal Cell Damage in Cultured Rat Cortical Neuron

  • Sanguisorbae radix (SR) from Sanguisorba officinalis L. (Losaceae) is widely used in Korea and China due to its various pharmacological activity. The present study aims to investigate the effect of the methanol extract of SR on amyloid ${\beta}$ Protein(25-35) $(A{\beta}\;(25-35))$, a synthetic 25-35 amyloid peptide, -induced neurotoxicity using cultured rat cortical neurons. SR, over a concentration range of $10-50\;{\mu}g/ml$, inhibited the $A{\beta}$ (25-35) $(10\;{\mu}M)-induced$ neuronal cell death, as assessed by a 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and the number of apoptotic nuclei, evidenced by Hoechst 33342 staining. Pretreatment of SR $(50\;{\mu}g/ml)$ inhibited $10\;{\mu}M\;A{\beta}$ (25-35)-induced} elevation of cytosolic calcium concentration $([Ca^{2+}]c)$, which was measured by a fluorescent dye, fluo-4 AM. SR $(10\;and\;50\;{\mu}g/ml)$ inhibited glutamate release into medium induced by $10\;{\mu}M\;A{\beta}(25-35)$, which was measured by HPLC, and generation of reactive oxygen species. These results suggest that SR prevents $A{\beta}$ (25-35)-induced neuronal cell damage in vitro.

  • The misuse of anabolic hormones or illegal drugs is a ubiquitous problem in animal husbandry and in food safety. The ban on growth promotants in food producing animals in the European Union is well controlled. However, application regimens that are difficult to detect persist, including newly designed anabolic drugs and complex hormone cocktails. Therefore identification of molecular endogenous biomarkers which are based on the physiological response after the illicit treatment has become a focus of detection methods. The analysis of the 'transcriptome' has been shown to have promise to discover the misuse of anabolic drugs, by indirect detection of their pharmacological action in organs or selected tissues. Various studies have measured gene expression changes after illegal drug or hormone application. So-called transcriptomic biomarkers were quantified at the mRNA and/or microRNA level by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) technology or by more modern 'omics' and high throughput technologies including RNA-sequencing (RNA-Seq). With the addition of advanced bioinformatical approaches such as hierarchical clustering analysis or dynamic principal components analysis, a valid 'biomarker signature' can be established to discriminate between treated and untreated individuals. It has been shown in numerous animal and cell culture studies, that identification of treated animals is possible via our transcriptional biomarker approach. The high throughput sequencing approach is also capable of discovering new biomarker candidates and, in combination with quantitative RT-qPCR, validation and confirmation of biomarkers has been possible. These results from animal production and food safety studies demonstrate that analysis of the transcriptome has high potential as a new screening method using transcriptional 'biomarker signatures' based on the physiological response triggered by illegal substances.

  • Korea was recognized as a relatively safe area for earthquake. However, due to considerable damage to facilities caused by the earthquake in Gyeongju and Pohang, interest in the maintenance and repair of structures is increasing. So interest in vibration damping technology applicable to existing structures is also increasing. However, vibration damping technology has a problem in that its usability is reduced due to damage of the damping device when a strong earthquake occurs. Recently, in order to solve such a problem, study is being conducted to apply a superelastic shape memory alloys (SSMA) capable of recentering bracing. Therefore, in this study, nonlinear dynamic analysis is performed to evaluate the seismic performance of the buckling-restrained braced frame (BRBF) applied SSMA to bracing.

Also, propriated Wireless sensor network that use Zigbee by solution of home network testbed.   • After harvesting the medicinal parts of Curcuma longa, the remaining underground parts were discarded. From the remaining underground parts of Curcuma longa quercetin-3-O-${\beta}$-D-glucopyranoside-7-O-${\alpha}$-L-rhamnopyranoside (Q37) was isolated. The antioxidant activities in vitro and lifespan-extension effect of Q37 were elucidated using the Caenorhabditis elegans. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect of Q37 showed similar potent activities in comparison with vitamin C. Q37 also showed potent superoxide quenching activities as measured by the riboflavin- and xanthine-originated superoxide quenching activity tests. Q37 prolonged lifespan of worms under normal culture condition. In terms of protective effect of Q37 on the stress conditions such as thermal and oxidative stresses, Q37-treated worms exhibited enhanced survival rate, as compared to control worms. To know the possible mechanism of Q37-mediated increased lifespan and stress resistance of worms, we examined the activities of Q37on superoxide dismutase (SOD), and invested intracellular reactive oxygen species (ROS) levels. The results revealed that Q37 was able to elevate SOD activity of worms and reduce intracellular ROS accumulation in a dose-dependent manner.

  • It is essential to decrease energy consumption and excess sludge to economically operate sewage treatment plant. This becomes more important along with a ban on sea dumping and exhaustion of resource. Therefore, many researchers have been study on energy consumption reduction and strategies for minimization of excess sludge production from the activated sludge process. The aeration cost account for a high proportion of maintenance cost because sufficient air is necessary to keep nitrifying bacteria activity of which the oxygen affinity is inferior to that of heterotrophic bacteria. Also, additional costs are incurred to stabilize excess sludge and decrease the volume of sludge. There were anoxic, aerobic, membrane, deairation and concentration zone in this MBR process. Continuous aeration was provided to prevent membrane fouling in membrane zone and intermittent aeration was provided in aerobic zone through ammonia sensor. So, there was the minimum oxygen to remove $NH_4-N$ below limited quantity that could be eliminated in membrane zone. As the result of this control, energy consumption of aeration system declined by between 10.4 % and 19.1 %. Besides, we could maintain high MLSS concentration in concentration zone and this induced the microorganisms to be in starved condition. Consequentially, the amount of excess sludge decrease by about 15 %.

  • XML(eXtensible Markup Language) has emerged as a prevalent standard for document representation and exchange on the Internet. XML documents contain information of different sensitivity degrees, so that XML Document must selectively shared by user communities. There is thus the need for models and mechanisms enabling the specification and enforcement of access control policies for XML documents. Mechanisms are also required enabling a secure and selective dissemination of documents to users, according to the authorizations which the users have. In this paper, we give an account of access control model and mechanisms, which XML documents can be securely protected in web environments. We make RBAC Based access Control polices to the problem of secure and selective access of XML documents. The proposed model and mechanism guarantee that the secure use for XML documents through definition of authority for element, attribute, link within XML document as well as XML document.

  • In this paper, we propose the progressed interworking method between H.323 and SlP, then explain the improved property. The VolP(Voice over Internet Protocol) technology which is able to use a voice service through internet is more cheaper then existing telephone charges, and is easil)· accept the various of multimedia services from internet. Previous connectionmethod of VoIP used H.323 protocol, but it is very complex to connection establishment. so, the SIP(Session Initiation Protocol) protocol that propose in SIP-Working Group is in use recently. Therefore, we need new interworking methodology between H.323 and SIP Products. In this thesis, the progress interworking method between H.323 and SIP are Propose, then interpret unnecessary packet delay for call setup and improved feature of message exchange.

  • In recent times, due to wire drawing of high carbon steel at a high speed to ensure a high productivity and high strength, axial residual stress are generated because of rapid increase in surface temperature. In the process, the temperatures of the wires increased because of the deformation of the wires and the friction between the die and wire. In particular, in the case of the wire drawing at a high speed, friction leads to a large temperature gradient so that considerable axial residual stress is generated on the surface. In this study, the relationship between axial residual stress and increase in the surface temperature was investigated, and a prediction model of uniform temperature was proposed. Then, a prediction model for residual stress was developed. The proposed model was verified by measuring the residual stress by X-ray diffraction on drawn wires.

Change of Antioxidant Activity and Antioxidant Compounds in Saururus chinensis by Extraction Conditions (추출조건에 따른 삼백초 추출물의 항산화 활성 및 성분의 변화)

  • This study was conducted in order to establish the optimum extraction conditions in obtaining Saururus chinensis extracts. At the optimum extraction solvent (40% ethanol solvent), yield, DPPH, total phenolic compounds and total flavonoid in the extract of Saururus chinensis were 13.50%, 83.50%, 2.60 mg/ml and 2.09 mg/ml, respectively. At the optimum extraction time (4 hours), yield, DPPH, total phenolic compounds and total flavonoid in the extract of Saururus chinensis were 12.79%, 71.13%, 2.83 mg/ml and 2.16 mg/ml, respectively. At the optimum conditions (40% ethanol solvent and 4 hours), quercetrin and quercetin contents were 360.13 mg/100 g and 1379.54 mg/100 g, respectively. From the above results, we suggest that the optimum Saururus chinensis extract conditions are 40% ethanol solvent and 4 hours.
