참당귀로부터 분리한 Demethylsuberosin의 멜라닌 생성 억제 효과 (Inhibitory Effects on Melanin Production of Demethylsuberosin Isolated from Angelica gigas Nakai)

  • The anti-melanogenic substance was isolated from the root of Angelica gigas Nakai by silica gel column chromatography, preparative HPLC and TLC. As a result of the structure analysis by mass, $^1H$-NMR, and $^{13}C$-NMR spectrometry, the compound was identified as demethylsuberosin. Demethylsuberosin reduced melanin contents of B16F1 melanoma cells in a dose-dependent manner and decreased to about 74% at a concentration $5{\mu}g/ml$. Demethylsuberosin inhibited the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2) in melanocytes. These results suggest that the whitening activity of demethylsuberosin may be due to the inhibition of the melanin synthesis by down-regulation of MITF, tyrosinase, TRP-1 and TRP-2 expression. Thus, our results provide evidence that demethylsuberosin might be useful as a potential skin-whitening agent.

  • 비타민 C는 강력한 환원제로서 멜라닌 색소의 합성을 저해하는 것으로 알려져 있다. 그러나 일반적인 화장품 제형에서는 낮은 안정성과 경피 흡수의 문제점으로 만족할 만한 효과를 나타내지 못한다. 본 연구에서는 안정성이 개선된 비타민 C의 유도체인 ascorbyl glucoside (AsAG)을 유효성분으로 하여 경피흡수를 증가시키고자 하였다. 본 실험에서는 유연하면서도 박막 형태의 배터리를 장착한 패치 화장품을 제조하고 안정성과 경피흡수성을 평가하였다. 피부에 낮은 전류를 증가하는 이온토포레시스를 활용하여 피부에 적용하는 전류의 세기를 증가시키면 물질의 경피흡수는 증가한다. 그러나 전류의 세기를 증가시키면 피부 부작용이 증가하기 때문에 본 연구에서는 한국인의 피부에 맞는 적절한 전류를 선택하여 피부 부작용을 최소화하였다. 이런 결과들을 바탕으로 유연하면서도 가벼운 박막 배터리를 개발하였으며, 2%의 AsAG을 함유한 이온토포레시스 패치의 안전성, 경피흡수정도, 미백효과 등을 검토한 결과 피부에 가하는 최적의 전류는 1.5 V의 배터리를 사용하여 피부 부작용과 경피흡수를 고려하여 평균 0.1 mA이었다. 또한 패치의 임상실험 결과 유의한 수준의 미백효과를 보였으며 피부 자극도도 통상의 화장품 수준을 나타냈다.

Radio-Iodinated arbutin for tumor imaging

  • Arbutin is a hydroquinone derivative with a glucose moiety. As a tyrosinase inhibitor, it is widely used as a skin-whitening cosmetic agent for the treatment of cutaneous hyperpigmentary disorders, such as melasma and freckles. In the medical field, many studies have addressed the use of arbutin in various tumors, but the mechanism for tumor uptake of arbutin is still unclear. In this paper, we radiolabeled arbutin using radioiodine and studied its pharmacokinetics and tumor uptake via biodistribution experiments and single-photon emission computed tomography (SPECT) imaging. Radiolabeled $^{131}I-arbutin$ was stable for up to 24 h in PBS and serum. Biodistribution studies and SPECT imaging indicated high uptake of the compound in the bladder and kidneys shortly after injection. Twenty-four hours post-injection, significant deiodination was observed. Apart from high thyroid uptake, selective tumor uptake was clearly observed. The tumor-to-muscle and tumor-to-blood ratios were 26 and 9, respectively.

Inhibition of Melanogenesis by Abietatriene from Vitex Trifolia Leaf Oil

  • Vitex trifolia L. has been used traditionally to treat various illnesses, such as inflammation, headache, migraine, and gastrointestinal infections. We analyzed and evaluated the composition of V. trifolia leaf oil. Based on the results, we isolated abietatriene from V. trifolia leaf oil and investigated the effect of V. trifolia leaf oil and its active compound abietatriene on melanogenesis in B16F10 melanoma cells. They significantly decreased melanin contents and melanogenic factors, such as tyrosinase, TRP-1, TRP-2, and MITF dose-dependently in both protein and mRNA levels. Protein and mRNA expressions were determined by Western blot analysis and quantitative real time RT-PCR. Findings indicate that V. trifolia leaf oil and abietatriene reduce melanogenesis by regulating the expression of melanogenic factors. These results suggest that V. trifolia leaf oil and abietatriene could comprise a useful therapeutic agent for treating hyperpigmentation and used as effective skin-whitening agents.

  • A chemical investigation of Peucedanum praeruptorum has resulted in the isolation of 3 khellactone derivatives, which have inhibitory effects on melanogenesis in B16 mouse melanoma cell lines. The khellactone derivatives were isolated from the crude extract of the roots of Pecedanum praeruptorum by a combination of adsorption chromatography and HPLC. The structures of isolated compounds were identified as 3', 4'-diangeloyl-cis-khellactone, 3'-angeloyl-4'-senecioyl-cis-khellactone and, 3', 4'-disenecioyl-cis-khellactone by $^1$H NMR, $^{13}$ C NMR and mass spectral studies and by comparisons of spectral data with reported literatures. These khellactone derivatives can be a good candidate for new skin whitening agent due to its strong inhibitory activity and safety.

더덕추출물이 멜라닌생성에 미치는 영향 (The effect of Codonopsis lanceolata on the melanogenesis)

  • Acquired pigmentary skin diseases such as abnormal melanogenesis, vitiligo, chloasma and inflammatory pigmentation are related to regulate the melanin production. The aim of this study was to investigate the effect of Codonopsis lanceolata on the melanogenesis of HM3KO human melanoma cells biologically. The cells were treated for 5 days with Codonopsis lanceolata at several concentrations. Treatment with Codonopsis lanceolata suppressed melanin contents as a dose dependent manner without cytotoxicity and morphological change. And the extract of Codonopsis lanceolata also inhibited tyrosinase, a key enzyme forming melanin, in a dose-dependent manner. And it did not affect the DOPAchrome tautomerase(TRP-2) activity. These results suggest that Codonopsis lanceolata is a candidate for an efficient whitening agent which supresses melanogenesis by a Raper-Mason pathway.

목단피로부터 멜라닌 생성 억제성분의 분리 (Isolation of Inhibitory Components on Tyrosinase Activity from the Bark of Paeonia moutan)

  • The MeOH extract of the bark of Paeonia moutan showed potent inhibitory effect on the mushroom tyrosinase activity in vitro. The activity-guided fractionation of t he MeOH extract resulted in the isolation of three active compounds. The chemical structures of these compounds were elucidated by chemical and spectroscopic evidence as catechin, 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose and 1,2,3,4,6-penta-0-galloyl-${\beta}$-D-glucose, respectively. Among them, the inhibitory activity by 1,2,3,6-tetra-galloyl-${\beta}$-D-glucose on mushroom tyrosinase was more potent $(IC_{50}=3.5\;{\mu}M)$ than that of kojic acid $(IC_{50}=8.7\;{\mu}M)$ ,but catechin enhanced the mushroom tyrosinase activity 50% in the concentration of 34.5M.

참나물추출물의 멜라닌 생성저해 효과 (New Whitening Agent From Pimpinella brachycarpa)

  • 천연물로부터 새로운 미백 화장품 소재를 개발하기 위해 참나물(P. brachycarpa)을 선택하여 B16 멜라노마 세포에서 tyrosinase 활성 저해 효과, melanin 생성 저해 효과를 측정하였다. 먼저 참나물추출물의 4가지 극성별 용매 분획화 실험을 실시하였으며, 항산화효과와 티로시나아제 저해효과를 측정하였다. 참나물의 미백 활성 메카니즘을 알아보기 위해 Western blotting과 RT-PCR을 이용하여 tyrosinase, TRP-1, TRP-2의 단백질 발현과 mRNA 변화를 연구하였다. 또한, HaCaT 각질형성세포에서 UVB 조사 후 엔도세린-1(ET-1)의 발현은 사람 ET-1 항체를 이용하여 quantitative enzyme immunoassay(EIA)로 측정하였다. 그 결과 참나물추출물과 4가지 분획(hexane, EtOAc, butanol and aqueous)은 100 ${\mu}g/mL$ 농도에서 각각 87.2, 2.5, 97.2, 80.5, 49.8%의 프리라디칼 소거효과를 나타내었으며, tyrosinase 저해효과는 100 ${\mu}g/mL$ 농도에서 각각 18.3, 15.1, 55.4, 13.1, 0%로 나타났다. 각 극성별 분획 중 EtOAc 분획 100 ${\mu}g/mL$ 농도에서 58% 이상의 가장 우수한 멜라닌 생성 저해 효과가 나타났다. 참나물 EtOAc 분획은 B16 멜라노마 세포에서 티로시나아제 활성 및 발현을 모두 저해하였으며, RT-PCR 결과에서도 tyrosinase, TRP-1의 mRNA 발현 저해효과가 우수하게 나타났다. 또한 HaCaT 각질형성세포에서 UVB 조사 후 생성된 엔도세린-1의 생성 실험에서도 참나물 EtOAc 분획 $12.5{\sim}50{\mu}g/mL$ 농도에서 엔도세린-1의 생성이 컨트롤에 비해 40% 정도로 우수하게 저해되었다. 결론적으로 참나물추출물은 멜라닌 합성과정에서 우수한 tyrosinase 저해효과와 엔도세린-1 발현저해효과를 가지는 새로운 천연 미백 소재로 적용될 수 있을 것이라 기대된다.

  • 본 연구자들은 백출의 메탄올 추출물로부터 분리, 정제한 selina-4(14), 7(11)-dien-8-one (Selina)이 B16 멜라노마 세포에 대하여 매우 강력한 미백효과가 있음을 밝혀내었다. 본 연구에서는 Selina의 non-tumorigenic melanocyte cell line인 Melan-a 세포에 대한 작용 메카니즘에 대한 보고를 하고자 한다. 덧붙여 Selina를 함유하고 있는 제품의 임상효과도 연구하였다. Selina는 $10 {\mu}g/mL$의 농도에서 세포독성을 보이지 않으면서 Melan-a 세포의 멜라닌 합성을 $50\%$ 저해하였다. 또한 $10 {\mu}g/mL$의 Selina의 처리에 의하여 타이로시네이즈의 활성이 $60\%$ 감소시키지만 타이로시네이즈에 대한 직접적인 저해활성은 없는 것으로 밝혀졌다. 이러한 Selina의 작용 기작을 밝히기 위하여 연구자들은 타이로시네이즈, TRP-1, TRP-2의 3가지 유전자의 mRNA 수준과 단백질의 발현량을 RT-PCR과 western blotting을 이용하여 연구하였다. 그 결과 $10 {\mu}g/mL$의 Selina 처리에 의하여 타이로시네이즈의 mRNA와 단백질 양은 현저히 감소하였지만 TRP-1과 TRP-2의 mRNA 및 단백질 양에는 변화가 없었다. 이러한 결과는 Selina가 그 미백효과를 주로 타이로시네이즈의 발현조절에 의하여 나타낸다는 것을 알 수 있다. $0.2\%$의 Selina를 함유하는 화장품 제형을 이용, 7주간 20명의 피시험자를 대상으로 한 임상시험에서도 어떠한 부작용도 없이 통계적으로 유의한(p<0.05) 수준의 미백효과를 볼 수 있었다. 이상의 결과를 종합해 볼 때 Selina는 깨끗하고 밝은 피부를 위한 유용하면서도 안전한 신규 미백 원료임을 알 수 있다.

Antimelanogenesis and skin-protective activities of Panax ginseng calyx ethanol extract

  • Background: The antioxidant effects of Panax ginseng have been reported in several articles; however, little is known about the antimelanogenesis effect, skin-protective effect, and cellular mechanism of Panax ginseng, especially of P. ginseng calyx. To understand how an ethanol extract of P. ginseng berry calyx (Pg-C-EE) exerts skin-protective effects, we studied its activities in activated melanocytes and reactive oxygen species (ROS)-induced keratinocytes. Methods: To confirm the antimelanogenesis effect of Pg-C-EE, we analyzed melanin synthesis and secretion and messenger RNA and protein expression levels of related genes. Ultraviolet B (UVB) and hydrogen peroxide ($H_2O_2$) were used to induce cell damage by ROS generation. To examine whether this damage is inhibited by Pg-C-EE, we performed cell viability assays and gene expression and transcriptional activation analyses. Results: Pg-C-EE inhibited melanin synthesis and secretion by blocking activator protein 1 regulatory enzymes such as p38, extracellular signal-regulated kinases (ERKs), and cyclic adenosine mono-phosphate response element-binding protein. Pg-C-EE also suppressed ROS generation induced by $H_2O_2$ and UVB. Treatment with Pg-C-EE decreased the expression of matrix metalloproteinases, mitogen-activated protein kinases, and hyaluronidases and increased the cell survival rate. Conclusion: These results suggest that Pg-C-EE may have antimelanogenesis properties and skin-protective properties through regulation of activator protein 1 and cyclic adenosine monophosphate response element-binding protein signaling. Pg-C-EE may be used as a skin-improving agent, with moisture retention and whitening effects.
