Effects of the Chestnut Inner Shell Extract on the Expression of Adhesion Molecules, Fibronectin and Vitronectin, of Skin Fibroblasts in Culture

  • The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.

The Effect of Cosmetic on Anti-Wrinkle of Acer mono Sap (우산고로쇠의 향장효과)

  • The purpose of this study was to research for anti-oxidation and anti-wrinkle effects of Acar mono Sap (AM). To cosmetic effect of AM, safety effect (MTT assay), anti-wrinkle effect (elastase, MMP-1 inhibition assay) and anti-oxidant effect (DPPH assay) were measured. When water extract of AM was used for cell viability, it was over 100% at 6% (6 ml/100 ml in phosphate buffer) concentration. AM showed 45.7% elastase inhibition and 23.7% MMP-1 inhibition at 50% (50 ml/100 ml in phosphate buffer) concentration so that it had good anti-wrinkle characteristic. And AM showed 68.9% antioxidation capacity at 50% concentration by using a DPPH assay. Consequently, AM can be used as natural materials or additives for human skin owing to their beneficial biologic functions, including the anti-wrinkle effect, for cosmetic compositions.

In Vitro Cytotoxicity, Skin Regeneration, Anti-wrinkle, Whitening and In Vivo Skin Moisturizing Effects of Oncheongeum (온청음 물 추출물의 세포독성, 피부재생, 주름개선, 미백 및 보습 효과)

  • Objectives: The objective of this study was to evaluate the effects of cytotoxicity, skin regeneration, anti-wrinkle, whitening and skin moisturizing of Oncheongeum (OCE).Methods: The cytotoxicity of OCE lyophilized aqueous extracts (yield=13.82%) was observed against human normal fibroblast cells and B16/F10 murine melanoma cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium Bromide (MTT) assay, and skin regeneration and anti-wrinkle effects were also evaluated through the assay of collagen type I synthesis compared to the transformation of the growth factor (TGF)-β1, hyaluronidase, collagenase and matrix metalloproteinase (MMP)-1 inhibitory assays compared to oleanolic acid (OA), and elastase inhibitory effects compared to phosphoramidon disodium salt (PP). In addition, OCE’s whitening effects were measured by a tyrosinase inhibitory assay and melanin formation test in B16/F10 murine melanoma cells compared to arbutin, and skin moisturizing effects were observed through a mouse skin water content test, respectively. Results: No OCE treatment-related cytotoxic effects appeared on human normal fibroblasts and B16/F10 murine melanoma cells. OCE concentration-dependently increased the collagen Type I synthesis on human normal fibroblast cells, and also effectively inhibited hyaluronidase, elastase, collagenase and MMP-1 activities. In addition, OCE inhibited melanin production of B16/F10 murine melanoma cells and activity of tyrosinase. And significant and dose-dependent increases of skin water content were detected in OCE-treated mouse skin compared to vehicle control skins. Conclusions: OCE showed favorable and sufficient effects in skin regeneration, anti-wrinkle, whitening and skin moisturizing in this experiment. But more detail mechanisms and studies on the skin protective efficiency of in vivo are needed with the screening of active biological compounds in individual OCE herbs.

Corni Fructus Inhibits Wrinkle Formation by Reduced Advanced glycation end product (AGEs) (산수유의 최종당화산물 억제로 인한 주름 개선효과)

  • Objectives : Corni Fructus (CF) is traditional herbal medicine used on polyuria, low back pain, and tinnitus. This study aimed to evaluate inhibits skin wrinkle formation effect of CF. Methods : To evaluate the produce inhibition effect of CF, SD-rats were distributed into four groups; normal rats (Nor), AGEs (advenced glycation end product)-induced rats (Con), AGEs-induced rats treated with 100mg/kg CF (CF). To induce AGEs, streptozotocin (50mg/kg) was administered intraperitoneally and after 3 days oral administrated 100mM methyl glyoxal for 3 weeks. Results : The oral administration of CF suppressed the reactive oxygen specis (ROS) in serum. The AGEs in skin tissues was significantly reduced through treatment of CF. Furthermore, the expressions of AGEs related proteins such as polyclonal anti-$N^e$-(carboxymethyl)lysine (CML), anti-$N^e$-(carboxyethyl)lysine (CEL), AGE receptors (RAGE) were decreased in CF treated group compared with the control group in skin tissues. Inflammation-related proteins such as Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB), monocyte chemoattractant protein-1 (MCP-1), interleukin-6 (IL-6) reduced in CF treatment group than control group. AGE-induced rats exhibited that the significant decreased collagen however, CF treatment (100mg/kg of body weight) up regulated collagen by improved the expression levels of skin fibril-related genes such as Matrix metalloproteinase (MMP-1). Conclusion : Taken together, our study suggests that CF regulates ROS to prevent accumulation of AGEs and inhibits skin wrinkles. Our finding indicate that CF may be an effective agent for inhibits AGEs formation, and improved skin wrinkle.

Biological activities of extracts from Caryopteris incana Miq. (층꽃나무(Caryopteris incana Miq.) 추출물의 생리활성)

  • In this study, extracts from Caryopteris incana Miq. (C. incana) were investigated to assess anti-oxidation, skin-whitening and anti-wrinkle activity. The total phenolic compounds of C. incana extracts with water and 80 % ethanol showed 7.69 and 12.50 mg/g respectively. Antioxidation activity of C. incana extracts was measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS), protection factor (PF), and Thiobarbituric acid reactive substances (TBARs). At concentration of $200{\mu}g/mL$, the DPPH free radical scavenging activity of water and ethanol extracts were 84 and 92 %, respectively. ABTS radical scavenging activity of water and ethanol extracts were both at approximately 99 %. Antioxidant PF of water and ethanol extracts were 1.56 PF and 1.67 PF, respectively. The TBARs of water and ethanol extracts were 62 and 82 %, respectively. In anti-wrinkle and skin-whitening activity, 80 % ethanol extract had more outstanding effect than water extract at concentration of $200{\mu}g/mL$. The levels of elastase and collagenase inhibitory activity related with anti-wrinkle were 58 and 89 % in ethanol extract. The tyrosinase inhibitory activity related with skin-whitening was 13 % in ethanol extract. The astringent effect of ethanol extract was 50 %. Throughout the results, C. incana extracts showed an excellent effect on anti-oxidation, skin-whitening and anti-wrinkle activity. Therefore, C. incana extracts can be used as a new material for cosmetics.

Inhibitory Effects of Nude Pack Containing Black Tea Water Extract on Skin Wrinkle Formation in Hairless Mice (홍차추출물 함유 누드팩의 Hairless 마우스 피부주름 형성 억제효과)

  • The aim of this study was to evaluate the inhibitory effect of nude pack containing black tea water extract (NPBT) on skin wrinkle formation in hairless mice. Skin wrinkles were induced by UVB irradiation to the backs of hairless mice for 5 weeks. And at the same time, NPBT was applied topically. Wrinkle formation, histological changes, expression of matrix metalloproteinase-3 (MMP-3) and protein activities of MMP-2 and MMP-9 were observed or analyzed. Wrinkles for the control group were formed as a pattern of deep furrows and thick crests. Whereas wrinkles for the NPBT treated group were formed as a pattern of shallow furrows and thin crests, and their wrinkle areas were significantly (p<0.001) lower than the control group. Collagen fibers were arranged irregularly and sparse in density and some elastic fibers were degenerated in the control group, while they were almost intact in the NPBT treated group. MMP-3 mRNA expression in the control group was significantly (p<0.001) higher than the normal group, and that of NPBT treated group was significantly (p<0.001) lower than the control group. The NPBT treated group showed remarkably lower protein activities of MMP-2 and MMP-9 than the control group. NPBT could have a considerable inhibitory effect on skin wrinkle formation in hairless mice.

Inhibitory Effects of Peonia japonica Water Extract on Skin Aging (II) - Focussed on Inhibitory Effects of Wrinkle Formation- (백작약 열수추출물의 피부노화 억제 효과 (II) -주름형성 억제 효과를 중심으로-)

  • To investigate the inhibitory effects of Peonia japonica water extract(PJWE) on skin wrinkle formation, skin wrinkles were induced by both the irradiation of UVB and the application of squalene monohydroperoxide to the backs of hairless mice for 4 weeks. And at the same time experimental materials were applied topically. Wrinkles for the control (C) group were formed as a pattern of deep furrows and thick crests. Whereas wrinkles for the positive control (PC, 0.01% retinoic acid) and experimental(E, PJWE) groups were formed as a pattern of shallow furrows and thin crests, which were similar to that of the normal(N) group. Collagen and elastic fibers in dermis of the PC and E groups were almost intact with a regular arrangement, which were similar to those of the N group. The activity of xanthine oxidase, the free radical generating enzyme, was significantly lower in the E group than the C and PC groups. The activities of superoxide dismutase and catalase, the free radical scavenging enzymes, were much higher in the E group than the C and PC groups and similar to the N group. As for the amount of matrix metalloproteinase-3(MMP-3) expression, PC and E groups were significantly lower than the C group. Therefore, PJWE could be very effective natural herbal material for the inhibition or improvement of wrinkle formation in hairless mice skin.

  • Mt. Jiri, where is located in southwestern area of Korea, can be a treasury of herbal medicine plants grown wild. More than 1200 species currently classified as herbs are grown or cultivated. Recently, the studies on the safflower which attracts the interests in the inhibition of skin aging in the modern people have been actively investigated, and this study aims to confirm that polyphenol compound, the main ingredient of safflower, can be a natural material to cause the delayed effect on the skin aging, inhibiting the generation of wrinkle in the 30 to 40s middle aged females. Primarily, this study is to confirm the anti-oxidizing effect by DPPH assay in vitro, and to evaluate the efficacy of inhibition or improvement on the wrinkle generation in the face skin by clinical trials, additionally. In addition, we are confident to contribute on the improvement of functional cosmetic industry, developing the sample product of anti-aging mask pack which has the main ingredient with the extract of safflower.

A Study on the Skin Anti-wrinkle Effect of Novel Palmitoyl Tripeptide (신규한 Palmitoyl Tripeptide의 피부 주름개선 효과에 관한 연구)

  • Collagen is the most abundant animal protein in mammals, accounting for about 30 % of all proteins. It is present in connective tissue and contributes to the structural framework of most organs. The tripeptide (glycineproline-hydroxyproline) with the INCI name Tripeptide-29 is main component of collagen type I. The palmitoyl tripeptide (palmitoyl-glycine-proline-hydroxyproline) with the INCI name Paimitoyi Tripeptide-29 is a synthetic material that was designed as a topical agent to stimulate collagen production. We synthesized the palmitoyl tripeptide as a potential anti-wrinkle compound. This compound has been characterized using HPLC. This compound proved, through in vitro tests, to stimulate collagen production and fibroblast proliferation. These results were very promising, so human studies were subsequently performed. We investigated the skin improvement effect of the palrnitoyl tripeptide on human skin by using non-invasive instruments. We measured physiological effects such as skin wrinkles and elasticity after volunteers applied the cosmetic products for 8 weeks. We observed significant improvement in skin wrinkles and elasticity after use of the cosmetic products for 8 weeks. We concluded that the palmitoyl tripeptide had an anti-aging effect on human facial skin.
