Management of Defects on Lower Extremities with the Use of Matriderm and Skin Graft

  • Background The reconstruction of large skin and soft tissue defects on the lower extremities is challenging. The skin graft is a simple and frequently used method for covering a skin defect. However, poor skin quality and architecture are well-known problems that lead to scar contracture. The collagen-elastin matrix, Matriderm, has been used to improve the quality of skin grafts; however, no statistical and objective review of the results has been reported. Methods Thirty-four patients (23 male and 11 female) who previously received a skin graft and simultaneous application of Matriderm between January 2010 and June 2012 were included in this study. The quality of the skin graft was evaluated using Cutometer, occasionally accompanied by pathologic findings. Results All 34 patients showed good skin quality compared to a traditional skin graft and were satisfied with their results. The statistical data for the measurement of the mechanical properties of the skin were similar to those for normal skin. In addition, there was no change in the engraftment rate. Conclusions The biggest problem of a traditional skin graft is scar contracture. However, the dermal matrix presents an improvement in skin quality with elastin and collagen. Therefore, a skin graft along with a simultaneous application of Matriderm is safe and effective and leads to a significantly better outcome from the perspective of skin elasticity.

Effect of Skin Elasticity Improvement and Anti-oxidant Activity of Stem Cells Extract Derived from Cambium of Aloe (알로에 형성층 유래 줄기세포 추출물의 항산화 활성 및 피부탄력 개선 효과)

  • In this study, stem cells were extracted from the callus derived from the aloe cambium, and the antioxidant activity and effect of skin elasticity improvement were assessed. The aloe cambium-derived stem cell extract, AloStem and callus showed the survival rate of each 98.27% and 71.31%. In the results of the DPPH antioxidant activity of AloStem and aloe extract, it was confirmed that the antioxidant effect of AloStem was more than twice that of Aloe extract. AloStem did not affect the cytotoxicity of normal human dermal fibroblasts (NHDF) cells up to 0.25% concentration. Also, AloStem increased elastin, COL1A1 and HAS2 mRNA expression levels dose-dependently. Furthermore, to examine lifting effect of skin elasticity using a sheet mask containing AloStem, 21 adult men and women applied the sheet mask in the face, once a day for 2 weeks. As a result, after 2 weeks the skin length was 116.75 ± 5.58 mm before the use of the sheet mask, but after 2 weeks of use, it was confirmed that 0.59% increased to 117.44 ± 5.17 mm. Thus, we concluded that the sheet mask containing AloStem can help the lifting effect of skin elasticity.

Effect of Essence Containing Abelmoschus manihot on Facial Skin (금화규 함유 에센스가 안면피부에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the efficacy of Abelmoschus manihot (A-M) on the facial skin by observing the changes in the skin caused by continuous use of the essence containing A-M. Methods : Fifty-one participants were enrolled, 38 were classified as experimental group and 13 were classified as control group. Before the beginning of the test, the skin condition of the participants was investigated with the facial analyzer. An essence containing A-M was provided to the experimental group, and an essence base not containing Abelmoschus manihot was provided to the control group. Participants were asked to use the product in the morning and evening after washing their face. We checked the skin change status every 2 weeks and performed the test 3 times for 6 weeks. Results : In the case of t-zone oil and pores, there was a significant decrease in the experimental group. In the case of moisture, a significant increase was observed in the control group, but the significance was significantly increased when the essence containing A-M was used. In the case of elasticity, it was found that the experimental group continued to increase with the period of use. Conclusion : It is thought that the essence containing A-M will be effective in preventing skin aging through the reduction of t-zone oil, the size of the pores, and the increase in moisture and elasticity.

Clinical Evidence of Effects of Lactobacillus plantarum HY7714 on Skin Aging: A Randomized, Double Blind, Placebo-Controlled Study

  • The beneficial effects of probiotics are now widely reported, although there are only a few studies on their anti-aging effects. We have found that Lactobacillus plantarum HY7714 (HY7714) improves skin hydration and has anti-photoaging effects, and in the present study, we have further evaluated the anti-aging effect of HY7714 via a randomized, double blind, placebo-controlled clinical trial. The trial included 110 volunteers aged 41 and 59 years who have dry skin and wrinkles. Participants took 1 × 1010 CFU/day of HY7714 (probiotic group) or a placebo (placebo group) for 12 weeks. Skin hydration, wrinkles, skin gloss, and skin elasticity were measured every 4 weeks during the study period. There were significant increases in the skin water content in the face (p < 0.01) and hands (p < 0.05) at week 12 in the probiotic group. Transepidermal water loss decreased significantly in both groups at weeks 4, 8, and 12 (p < 0.001 compared with baseline), and was suppressed to a greater extent in the face and forearm in the probiotic group at week 12. Volunteers in the probiotic group had a significant reduction in wrinkle depth at week 12, and skin gloss was also significantly improved by week 12. Finally, skin elasticity in the probiotic group improved by 13.17% (p < 0.05 vs. controls) after 4 weeks and by 21.73% (p < 0.01 vs. controls) after 12 weeks. These findings are preliminary confirmation of the anti-aging benefit to the skin of L. plantarum HY7714 as a nutricosmetic agent.

  • Inositol, is a water-soluble crystalline compound. It helps with people’s metabolism and decreases cholesterol levels. It is also known to have anti-cancer results. In order to find out the affects of Inositol on the skin, Inositol skin lotion was produced with each amount of Inisitol: 0, 0.5, 1.0, 2.0, 3.0 wt% and tested on the faces and the arm areas of women in all ages for 7 weeks. The moisture, sebum, change in elasticity, and improvement of wrinkles were measured. Corneometer, Sebumeter, Cutometer, and an image analyzer were used as measuring equipments. There are subtle differences in the subjects when 1-2% of Inositol is used the moisture of the skin improved 19%, elasticity by 17%, and the amount of sebum for dry and oily skin types adjusted to the amount of sebum of the neutral skin types. This influenced the length, width, the number of peak, and the height of the wrinkles to improve 12.4%. It is thought that Inositol would be an effective new raw material in cosmetics.

  • We had tested the effect of amino propane sulfonic acid(APSA) on the human skin with non-invasive technique. It was tested with four parameters that were hydration, elasticity, color and skin wrinkles. In vitro culture systems, APSA stimulated the proliferation of fibroblasts bolt it didn't stimulate that of keratinocyte. Also we obtained the similar effects in the raft culture method. So we concluded that APSA affected the dermal region than the epidermal region. In clinical tests, APSA changed the skin color, pbiomechanical properties(especially elasticity) and reduced skin wrinkles of the volunteers. And we could get the better results of skin wrinkle improvement by use of Skin Visiometer than Silflo Image analysis systems.

Analysis the skin physical properties of periorbital region and cheek based on female's aging pattern by 7 years (한국 성인 여성의 눈가 피부 특성 분석)

  • Objectives : We investigated the skin properties on the periorbital region and cheek of Korean female according to age pattern by 7year based on Sang Go Cheun Jin Ron (上古天眞論) in So Moon (素問,) Methods : 49 Korean females, aged 21-56 years ($39.54{\pm}10.04$ years) were participated in this study. We measured the skin properties of periorbital region and cheek with SkinTouch (Amorepacific, Korea). Skin moisture, sebum, elasticity, lightness, roughness, area of wrinkle, depth of wrinkle were assessed. Results : Differences were noticed depending on the site and age. The level of moisture was higher on the eyelid than crow's feet. The level of lightness and elasticity were lower on the periorbital region than cheek. Skin lightness was highest on group1(under 28y) and lowest on group5(over 49y). Skin roughness was highest on group1(under 28y) and lowest on group4(over 42y under 49y). The level of wrinkle are was lowest on group1(under 28y) and highest on group 5(over49y). The level of wrinkle depth was lowest on group1(under 28y) and highest on group 4(over 42y under 49y). Conclusion : Unique aging pattern by 7 years of female is reflected to diverse skin properties of eyes. It can be possible to apply the theory on Sang Go Cheun Jin Ron (上古天眞論) to practical use in dermatology and cosmetic.

Cosmetics Material Research of FRO Extract

  • Purpose: In this study, skin and lotion containing 10% FRO were prepared to compare the skin improvement effects. Research design, data and methodology: After using cosmetics containing FRO ingredients, analysis was performed for 6 weeks. Results: As a result of measuring the moisture and elasticity of the pores, significant results were obtained only in the pores when used for 2 weeks between the experimental group and the control group (p<.001). Also after 4 weeks of use, moisture (p<.05) showed significant results, and after 6 weeks of use, moisture (p<.01) and elasticity (p<.001) showed significant.

  • Acquired cutis laxa is a rare disease. Owing to few reports on the condition, no statistical data have been produced. Cutis laxa is characterized by drooping skin, caused by decreased levels of dermal elastin, leading to reduced skin elasticity. The disease usually emerges on the neck or trunk and spreads throughout the body; however, it rarely involves the extremities. Moreover, cases localized to the face are rare. The objective of this clinical case report was to highlight this unusual disease in a 24-year-old female, with localization on the face and neck. The patient underwent surgery for treatment of bilateral ear lobe and eyelid skin laxity.

Inhibitory Effects of Saposhnikoviae Radix Extracts on the Melanin Production and Elastase Activity in B16F10 cells (흑색종 세포주에서 멜라닌 생성과 엘라스타제 활성 억제에 미치는 방풍의 효과)

  • Saposhmikoviae Radix can treat various skin disease by anti-pruitus and anti-inflammatory effects. This study was designed to investigate effects of Saposhmikoviae Radix Extracts(SRE) on skin elasticity and whitening using B16F10 cell lines. In this experiment, We observed effect of SRE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. In results, SRE treated group showed lowered proliferation rates significantly compared to non-treated group. More than SRE $125{\mu}g/m{\ell}$ of treated groups were lower levels of melanin synthesis respectively. SRE did not show inhibitory effect on tyrosinase activities in vitro and in B16F10 cells. Finally, SRE suppressed elastse type I and IV activities in dose-dependent manner in vitro. And SRE also slightly suppressed elastase activities in B16F10 cells. In conclusion, these results suggest that SRE can inhibit melanin synthesis and inhibt elastase activity. So, We suggest that SRE can be maintained skin elasticity or whitening.
