향나무추출물의 광손상으로부터 피부세포 보호와 자극완화 효과에 대한 연구 (Protection of UV-derived Skin Cell Damage and Anti-irritation Effect of Juniperus chinensis Xylem Extract)

  • 태양광선과 산소는 피부세포에 자유라디칼(free radical)과 활성산소종(Reactive oxygen species, ROS)을 생성시켜 DNA의 손상과 세포막 지질의 과산화를 유발한다. 또한 콜라겐과 같은 세포 외 기질을 파괴하는데 관여하는 효소(matrix metallo-proteinhse, MMPs)의 발현에 영향을 주어, 결합조직을 손상시키며, 피부 염증을 유발시켜 조직을 파괴하고 노화를 가속화시킨다. 본 연구는 자외선과 SLS에 의한 피부손상에 대한 향나무추물물의 피부 보호 효과에 대한 것이다. 실험 결과 프리라디칼 및 슈퍼옥사이드 라디칼 소거 효과가 우수하게 나타났으며, 피부 콜라겐과 같은 메트릭스를 분해하는 효소인 MMP-1의 활성 및 발현 저해 효과는 섬유아세포에서 UVA 조사에 의한 실험에서 우수하게 나타났으며, 피부세포 배양액에 대해 zymography를 실시하여 활성이 감소됨을 확인하였다. 피부 각질형성세포에서 자외선에 의해 유도된 염증관련 사이토카인인 인터루킨 6의 발현량 실험에서도 무처리군에 비해 향나무추출물이 인터루킨 6를 30% 정도 저해효과가 나타났으며, 염증반응 중 cyclooxygenase (COX)에 의한 경로에서 생성되는 프로스타글란딘 E$_2$의 생성량도 감소시켰다. 사람 피부에서 SLS (0.5%)첩포로 유발된 자극성 피부염의 항염증 효과 평가 시 SLS에 의해 유발되는 자극정도가 피검자의 피부상태에 따라 다양하게 나타났으며, 향나무추출물 함유 에멀젼 제품 도포 실험에서는 피부 홍반 완화 효과와 피부장벽 회복 효과가 우수하게 나타났다. 따라서 향나무추출물은 자외선 조사 및 SLS에 의한 피부손상에 대한 보호 효과를 가진 화장품에서 우수한 소재로 적용될 수 있다. 녹두추출물을 화장품에 사용하였을 경우 우수한 항자극 효과를 보임을 확인하였다.완화시키는데 기여하였다. 이러한 결과를 종합하여 한국에서 삼림쇠퇴를 유발할 수 있는 스트레스요인들의 잠재적 상호작용을 모식화하였다. 나아가 그것을 완화할 수 있는 복원방안을 토양개량과 식생복원의 측면에서 제시하였다. mg/l)치 농도가 높을지라도 이것이 낮은 cinnamic acid와 protocatechuic acid에서 현저한 억제현상을 보였다. 서양등골나물 임상의 토양보다 서양등골나물이 분포하지 않는 토양의 중금속 함량이 전반적으로 높았다. 특히 Al, Fe 및 Mn의 함량이 높았으며 이들 중금속은 total phenolic compound 함량이 높은 잎에 대부분이 축적되었다." separating himself from the real world. 감마선조사에 의한 돼지 피부장애에 cyclooxygenase-2의 발현변화 (Gamma-ray-induced skin injury in the mini-pig: Effects of irradiation exposure on cyclooxygenase-2 expression in the skin)

  • 방사선 노출에 따른 피부손상의 기본이론들은 정립되어 있지만, 정확한 기전에 관해서는 알려지지 않은 실정이다. 본 연구에서는 감마선 조사 후 용량 및 시간에 따라 돼지 피부의 장애를 육안 및 조직학적인 변화를 통해 평가하고 cyclooxygenase(COX)-2 발현 정도를 비교하고자 하였다. 미니돼지의 등 쪽 피부에 20-70 Gy 감마선을 국소조사 후 12 주 동안 육안적인 변화를 관찰하고 생검을 통해 조직학적인 변화를 관찰하였다. 방사선 조사 후 피부의 기저세포층에서의 세포자멸사와 표피층의 두께 변화를 평가하였고 COX-2 발현 정도를 면역염색을 통하여 비교하였다. 방사선 조사 후 피부 장애는 용량이 증가할수록 피부손상이 더욱 심하였으며 초기에 발적 소견을 보이다가 50 Gy 이상 조사군에서는 미란과 궤양으로 이어졌다. 조직학적인 변화는 육안적인 소견과 일치하였다. 방사선 조사 후 3일부터 기저세포에서 세포자멸사가 관찰되었으며 기저세포수의 감소가 유발되었으며 이러한 변화는 용량이 증가할수록 더욱 증가하였다. 방사선 조사 후 표피층의 두께는 3일 무렵에 일시적으로 증가하다가 점차 감소하였으며 20, 30, 40 Gy 조사 군에서는 다시 회복되는 소견이 관찰되었으나 50, 70 Gy 조사 군에서는 다시 회복되지 못하고 표피창의 두께 소실을 보였다. 방사선 조사 후 피부에서의 COX-2 발현은 피부손상정도와 일치하게 관찰되었다. 방사선 조사 후 COX-2 발현은 방사선의 용량이 증가할수록 발현이 증가하였고 시간이 증가할수록 증가하였다. 이러한 조직학적인 변화와 함께 방사선 손상을 일으키는 신호전달에 관여하는 COX-2 발현이 방사선조사 용량에 비례하여 증가하며 이러한 단백질의 발현은 피부손상과 관련성이 높은 것으로 사료된다.

Effects of EGb 761 and Korean Red Ginseng on Melanogenesis in B16F10 Melanoma Cells and Protection Against UVB Irradiation in Murine Skin

  • These days there is a constant possibility of exposure to UV radiation which can cause abnormal production of melanin and result in skin disease such as hyperpigmentation and melanoma. Many materials were investigated for skin whitening and protection against UV radiation. In this study, we assessed the melanogenesis inhibitory activities of Korean Red Ginseng (KRG, Ginseng Radix Rubra) and Ginkgo (EGb 761 Ginkgo Biloba) in an attempt to develop a new skin whitening agent derived from natural products. B16F10 melanoma cells were treated for 48 hr with KRG and EGb 761. The inhibitory effect on melanogenesis was measured and related cytokines and proteins expression were also investigated by RT-PCR and Western blotting. In addition, we also assessed the effects of these substances on the skin of C57BL/6 mice. Cell growth, melanin content and tyrosinase activity were inhibited effectively in B16F10 melanoma cells treated with KRG and EGb 761. Moreover, tyrosinase mRNA expression was inhibited clearly and melanogenesis related proteins (MRPs) containing tyrosinase, TRP1 and TRP2 were also reduced by KRG and EGb761, while cytokines such as IL-$1{\beta}$ and IL-6 were induced. In the case of UV irradiated mice, we observed induction of cytokine mRNA levels and reduction of MRPs mRNA expression. In addition, a decrease in pigmentation from treatment with KRG and EGb 761 on the skin of mice was observed. These results indicate that KRG and EGb 761 inhibit melanogenesis in B16F10 cells and have display protective activities against UVB. Therefore, we suggest that KRG and EGb 761 are good candidates to be used as whitening agents and UVB protectors for the skin.

Oxidative Stress and Skin Diseases: Possible Role of Physical Activity

  • Background: The skin is the largest body organ that regulates excretion of metabolic waste products, temperature, and plays an important role in body protection against environmental physical and chemical, as well as biological factors. These include agents that may act as oxidants or catalysts of reactions producing reactive oxygen species (ROS), reactive nitrogen species (RNS), and other oxidants in skin cells. An increased amount of the oxidants, exceeding the antioxidant defense system capacity is called oxidative stress, leading to chronic inflammation, which, in turn, can cause collagen fragmentation and disorganization of collagen fibers and skin cell functions, and thus contribute to skin diseases including cancer. Moreover, research suggests that oxidative stress participates in all stages of carcinogenesis. We report here a summary of the present state of knowledge on the role of oxidative stress in pathogenesis of dermatologic diseases, defensive systems against ROS/RNS, and discuss how physical activity may modulate skin diseases through effects on oxidative stress. The data show duality of physical activity actions: regular moderate activity protects against ROS/RNS damage, and endurance exercise with a lack of training mediates oxidative stress. These findings indicate that the redox balance should be considered in the development of new antioxidant strategies linked to the prevention and therapy of skin diseases.

Antioxidant potential of silk protein sericin against hydrogen peroxide-induced oxidative stress in skin fibroblasts

  • The antioxidant potential of silk protein sericin from the non-mulberry tropical tasar silkworm Antheraea mylitta cocoon has been assessed and compared with that of the mulberry silkworm, Bombyx mori. Skin fibroblast cell line (AH927) challenged with hydrogen peroxide served as the positive control for the experiment. Our results showed that the sericin obtained from tasar cocoons offers protection against oxidative stress and cell viability is restored to that of control on pre-incubation with the sericin. Fibroblasts pre-incubated with non-mulberry sericin had significantly lower levels of catalase; lactate dehydrogenase and malondialdehyde activity when compared to untreated ones. This report indicates that the silk protein sericin from the non-mulberry tropical tasar silkworm, A. mylitta can serve as a valuable antioxidant.

In vitro Effects of the Complex of Korean Citrus junos and Medicinal Herbs on the Cell Growth in CCD-986sk Human Fibroblast Cell Line

  • In order to investigate whether or not CCD-980sk cell line can be affected by Korean Citrus junos and medicinal herbs, we examined the MTT assay when we treated Korean Citrus junos and medicinal herbs in CCD-986sk human fibroblast cell line. The samples that added complex of grinded extracts of Korean Citrus junos and boiling-water extracts from Korean medicinal herbs were tested toy cell proliferation activity by means of a modification of the MTT assay. Among mixture of Citron 3 (Citron 3, less mellowed citron which was ripened for three months) and boiling-water extracts, the group Citron 3+Phellinus linteus showed significantly strong cell proliferation activity. And among mixture of Citron 4 (Citron 4, completely mellowed citron which was ripened for four months) and boiling-water extracts, the group Citron 4+Cordyceps militaris and Citron 4+Phellinus linteus showed significantly strong cell proliferation activity, respectively. These results suggest that complex of Korean Citrus junos and medicinal herbs could be an excellent candidate toy protection of human skin aging.

Comparison of Skin Injury Induced by β- and γ-irradiation in the Minipig Model

  • Background: The effects of radiation on tissues vary depending on the radiation type. In this study, a minipig model was used to compare the effects of ${\beta}$-rays from $^{166}Ho$ and ${\gamma}$-rays from $^{60}Co$ on the skin. Materials and Methods: In this study, the detrimental effects of ${\beta}$- and ${\gamma}$-irradiation on the skin were assessed in minipigs. The histopathological changes in the skin from 1 to 12 weeks after exposure to 50 Gy of either ${\beta}$- (using $^{166}Ho$ patches) or ${\gamma}$- (using $^{60}Co$) irradiation were assessed. Results and Discussion: The skin irradiated by ${\beta}$-rays was shown to exhibit more severe skin injury than that irradiated by ${\gamma}$-rays at 1-3 weeks post-exposure; however, while the skin lesions caused by ${\beta}$-rays recovered after 8 weeks, the ${\gamma}$-irradiated skin lesions were not repaired after this time. The observed histopathological changes corresponded with gross appearance scores. Seven days post-irradiation, apoptotic cells in the basal layer were detected more frequently in ${\beta}$-irradiated skin than in ${\gamma}$-irradiated skin. The basal cell density and skin thickness gradually decreased until 4 weeks after ${\gamma}$- and ${\beta}$- irradiation. In ${\beta}$-irradiated skin lesions, and the density and thickness increased sharply back to control levels by 6-9 weeks. However, this was not the case in ${\gamma}$-irradiated skin lesions. In ${\gamma}$-irradiated skin, cyclooxygenase-2 (COX-2) was shown to be expressed in the epidermis, endothelial cells of vessels, and fibroblasts, while ${\beta}$-irradiated lesions exhibited COX-2 expression that was mostly limited to the epidermis. Conclusion: In this study, ${\beta}$-rays were shown to induce more severe skin injury than ${\gamma}$-rays; however, the ${\beta}$-rays-induced injury was largely repaired over time, while the ${\gamma}$-rays-induced injury was not repaired and instead progressed to necrosis. These findings reveal the differential effects of ${\gamma}$- and ${\beta}$-irradiation on skin and demonstrate the use of minipigs as a beneficial experimental model for studying irradiation-induced skin damage.

Astaxanthin induces migration in human skin keratinocytes via Rac1 activation and RhoA inhibition

  • BACKGROUND/OBJECTIVES: Re-epithelialization has an important role in skin wound healing. Astaxanthin (ASX), a carotenoid found in crustaceans including shrimp, crab, and salmon, has been widely used for skin protection. Therefore, we investigated the effects of ASX on proliferation and migration of human skin keratinocyte cells and explored the mechanism associated with that migration. MATERIAL/METHOD: HaCaT keratinocyte cells were exposed to $0.25-1{\mu}g/mL$ of ASX. Proliferation of keratinocytes was analyzed by using MTT assays and flow cytometry. Keratinocyte migration was determined by using a scratch wound-healing assay. A mechanism for regulation of migration was explored via immunocytochemistry and western blot analysis. RESULTS: Our results suggest that ASX produces no significant toxicity in human keratinocyte cells. Cell-cycle analysis on ASX-treated keratinocytes demonstrated a significant increase in keratinocyte cell proliferation at the S phase. In addition, ASX increased keratinocyte motility across the wound space in a time-dependent manner. The mechanism by which ASX increased keratinocyte migration was associated with induction of filopodia and formation of lamellipodia, as well as with increased Cdc42 and Rac1 activation and decreased RhoA activation. CONCLUSIONS: ASX stimulates the migration of keratinocytes through Cdc42, Rac1 activation and RhoA inhibition. ASX has a positive role in the re-epithelialization of wounds. Our results may encourage further in vivo and clinical study into the development of ASX as a potential agent for wound repair.

DNCB로 아토피피부염을 유발한 NC/Nga mice에서 사이프러스 에센셜 오일의 효능에 관한 연구 (A Study on the Skin Protection Effects of Cypress Essential Oil on the DNCB-induced Atopic Dermatitis in NC/Nga Mice)

  • Objectives : The application of DNCB (1-chloro-2,4-dinitrochlorobenzene) can cause cellular immunity allergic reaction such as erythema or edema on NC/Nga mice and combinational function of cells participating in immunity increase inflammatory mediator. In this study, the effects of cypress essential oil on NC/Nga mice have been assessed. Methods : Male SPF NC/Nga mice aged 8 weeks have been used for atopic dermatitis induction skin. 1% DNCB was applied on ears and backs of which hair was removed using clipper on $1^{st}$ day and 0.4% DNCB was applied three times a week for 3 weeks. In this study, cypress essential oil has been treated 1 time a day for 3 weeks after application of DNCB to induce atopic dermatitis on skin for further experiments. Results : This study shows that inhalation or application of cypress essential oil reduced edema in ears and the thickness of epidermis induced by DNCB treatment. And it can be known that treatment of cypress essential oil inhibited mast cell proliferation and reduced IgE level similar to that of the negative control especially when cypress essential oil was inhaled by the mice. Synthetic oil showed the effects lower than those of cypress essential oil. Conclusions : Inhalation or direct application of cypress essential oil on skin reduced IgE level in blood and prohibits the proliferation of mast cell, from which it can be known that cypress essential oil can be effectively used to reduce the symptoms of atopic dermatitis.

Impact of 0.35 T Magnetic Field on Dose Calculation for Non-small Cell Lung Cancer Stereotactic Radiotherapy Plans

  • Background: We investigated the impact of 0.35 T magnetic field on dose calculation for non-small cell lung cancer (NSCLC) stereotactic ablative radiotherapy (SABR) in the ViewRay system (ViewRay Inc.), which features a simultaneous use of magnetic resonance imaging (MRI) to guide radiotherapy for an improved targeting of tumors. Materials and Methods: Here, we present a comprehensive analysis of the effects induced by the 0.35 T magnetic field on various characteristics of SABR plans including the plan qualities and dose calculation for the planning target volume, organs at risk, and outer/inner shells. Therefore, two SABR plans were set up, one with a 0.35 T magnetic field applied during radiotherapy and another in the absence of the field. The dosimetric parameters were calculated in both cases, and the plan quality indices were evaluated using a Monte Carlo algorithm based on a treatment planning system. Results and Discussion: Our findings showed no significant impact on dose calculation under the 0.35 T magnetic field for all analyzed parameters. Nonetheless, a significant enhancement in the dose was calculated on the skin surrounding the tumor when the 0.35 T magnetic field was applied during the radiotherapy. This was attributed to the electron return effect, which results from the deviation of the electrons ejected from tissues upon radiation due to Lorentz forces. These returned electrons re-enter the tissues, causing a local dose increase in the calculated dose. Conclusion: The present study highlights the impact of the 0.35 T magnetic field used for MRI in the ViewRay system for NSCLC SABR treatment, especially on the skin surrounding the tumors.
