The skin barrier function is one of the vital functions. Horny layer and epidermis among various skin layers play an important role in protecting body from environmental insults. Stratum corneum is completely formed by differentiation processes from the basal layer and newly formed corneocytes push the older ones from skin surface. In this study we investigated changes of various parameters - capacitance, transepidermal water loss (TEWL), pH and amount of removed corneocytes - according to the stratum corneum's depth. We removed the stratum corneum using tape stripping technique. We found that uppermost stratum corneum play an important role in skin surface moisturization and lower stratum corneum in protecting transepidermal water loss. In addition, skin pH is more acidic than surface pH. In the stratum corneum, skin pH is mote acidic in the deeper layer. Furthermore amount of removed corneocyte is also decreased according to depth because of stronger cell-cell union strength. From these results we suggest that stratum corneum has different characteristics depending on their position.
In this study, we evaluated the effect of Lactobacillus plantarum HY7714 on skin hydration in human dermal fibroblasts and in hairless mice. In Hs68 cells, L. plantarum HY7714 not only increased the serine palmitoyltransferase (SPT) mRNA level, but also decreased the ceramidase mRNA level. In order to confirm the hydrating effects of L. plantarum HY7714 in vivo, we orally administered vehicle or L. plantarum HY7714 at a dose of $1{\times}10^9CFU/day$ to hairless mice for 8 weeks. In hairless mice, L. plantarum HY7714 decreased UVB-induced epidermal thickness. In addition, we found that L. plantarum HY7714 administration suppressed the increase in transepidermal water loss and decrease in skin hydration, which reflects barrier function fluctuations following UV irradiation. In particular, L. plantarum HY7714 administration increased the ceramide level compared with that in the UVB group. In the experiment on SPT and ceramidase mRNA expressions, L. plantarum HY7714 administration improved the reduction in SPT mRNA levels and suppressed the increase in ceramidase mRNA levels caused by UVB in the hairless mice skins. Collectively, these results suggest that L. plantarum HY7714 can be a potential candidate for preserving skin hydration levels against UV irradiation.

The root of Paeonia lactiflora has been used in Chinese medicine. We conducted to check the comparative qualities of ethanol solvent extraction (PLE) and supercritical carbon dioxide extraction (PLS) of P. lactiflora root. PLE had higher antioxidant and polyphenol contents than PLS. But, PLS were significantly increased peroxisome proliferator-activated receptor (PPAR)-α. In addition, PLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. These results indicate that P. lactiflora supercritical carbon dioxide extract may serve as a cosmeceutical for improving skin barrier function and the treatment of obesity.

Particulate matters (PM) are small particulate pollution that decrease the function of skin barrier, which causes inflammatory skin diseases and extrinsic aging. In this study, we evaluated the effect of preventing the adherence of PMs from several cosmetic products applied to human skin using iron oxide black. The PM floating chamber consists of skin exposure area, PM inlet, floating power device, and an outlet so that PM can be naturally attached to the skin while floating in the chamber. The change in skin brightness according to the floating concentration of alternative fine dust was checked to confirm the optimal floating concentration conditions. The intensity difference (before-after intensity, Δ) before and after adhesion of iron oxide black was proportional to the amount of PM adhered. The anti-adherence effect of iron oxide black on five cosmetic products were evaluated through 20 each subjects by comparing the amount of iron oxide black adhered on the control and treatment. The difference in brightness before and after the iron oxide black attached to the skin was calculated and compared with the control group(p < 0.05). When over 150 mg of iron oxide black was adhered on the skin, the interference of intensity was low and clearly showed the skin adhered pattern. According to the application of the five cosmetics, the intensity difference was significantly lower than the control group. This means that depending on the product, it prevented the attachment of iron oxide black. This study is a safe and useful method to confirm the prevention of PM skin adherence. In conclusion, cosmetics can prevent the adherence of PM on the skin according to the formulation or ingredients characteristics.

Ultraviolet (UV) irradiation from the sun is the primary environmental factor that causes skin damages including skin cancer and premature skin aging. Because, even the most powerful sunscreen can't always afford enough protection, it is necessary to enhance the defensive power of skin against UV. Recently, constitutive photomorphogenic protein-1 (COP1) has shown to contribute to the regulation of UVB response of keratinocytes. In this study, we represent that COP1 and its associated protein, de-etiolated 1 (DET1), might participate in photoaging process in human skin as Arabidopsis COP1 does sun-protective function in plants. After UVB irradiation, the decrease of COP1 and DET1 mRNA expression was followed by the increase of c-Jun total protein. Moreover, transfection with DNA vectors expressing COP1 and DET1 down-regulated the c-Jun total protein. We found that Zanthoxylum piperitum extract (ZE) up-regulated the expression of COP1 and DET1 on human keratinocytes, and inhibited the expression of MMP1 which is one of the genes regulated by c-Jun signal. In addition, ZE has been reported to stimulate PPAR-${\alpha}$ and strengthen the skin barrier. We found that ZE decreased the UVB-induced IL-6 and IL-8 in NHEK cells. In human study, ZE protected skin against UV-B induced erythema and erythema-induced pigmentation. These results indicate that ZE could be useful for the protection against the adverse effects of UV irradiation through various mechanisms.
Transdermal drug delivery offers an alternative method to the conventional oral and injection delivery method. Its advantages include its ability to deliver drugs directly into systemic circulation. However, there have been restrictions in its application to deliver drugs because of the skin's barrier function. In this study, we try to combine a Sonophoresis and oxygen Pressure method in order to increase the Permeability of the skin. we used water as the compound and by utilizing the skin impedance method. we measured the hydration Permeability of skin Ultrasound was applied using a sonicator(Solcare-U1000. Solco, Korea) operating at a frequency of 1MHz. oxygen Pressure was applied using a compressor(Oxyjet-Pointer, Nora Bode. Germany) operating at a pressure of 2Bar/cm2. Experiment was performed in vivo for 42 People. We divided the subjects into four smaller groups. A different transdermal drug delivery method was applied for each group on the back of their hand. We measured the skin impedance variations on the hand. during a 20-minute time Period. The control group did not show any significant increase or variation of skin impedance to water. In comparison to the control group(Passive diffusion) the hydration Permeability of the ultrasound group and the oxygen Pressure group was approximately 25 and 30 times higher consecutively. Futhermore, the hydration permeability of the combination of ultrasound and oxygen Pressure group was about 70-fold higher in comparison to the control group(passive diffusion) . The results reveal that a combination of ultrasound and oxygen Pressure will significantly enhance transdermal water transport compared when only one of them is used.

The measurement of hydration level in the surface layer of the skin, stratum corneum (SC), gives important information on the biophysical properties and function of the skin barrier such as softness, flexibility, and healthiness of the skin. But it is difficult to measure a consistent hydration level from a sample to another sample due to individual variations and environmental changes. The aim of this study was to evaluate objective hydration after using various products in the SC. The SC Hydration was measured by capacitance (Corneometer$^{(R)}$, C+K, Germany) on the ventral site of forearm from 40 healthy volunteers. The skin surface was chronologically measured immediately after application of the test products and 3 and 6 hours later. We analyzed the averages of five measurements of each site and used the hydration increase rate for correction on untreated site variation. We found that most polyols including glycerol and butylenes glycol influenced directly the hydration increase rate in the SC previously. In this study, glycerol was used to prepare the standard products from 0 to 20 percents and applied to the same volunteers. The individual standard curve showed linear relation to glycerol concentrations. Based on the the standard curve, hydration of SC was converted into hydration increase rate to glycerol concentrations. The converted glycerol concentrations of products were repetitive and reproducible. In addition, the individual standard curve was used to relate the skin type of each individual. These results suggest that the hydration of the SC standardized regardless of external variation and individual skin condition can explain detailed skin state variation. Further studies will be conducted with other ingredients such as surfactants, lipids and aqueous materials, and with other methods for noninvasive measurement.

Hairless mice (Skh:HR-1) exposed to single doses (0.5, 1.0 and 3. OMED) of UV-B radiation were displayed remarkable changes of barrier function and surface morphology. Trans- epidermal water loss (TEWL) as an index of barrier function was measured by evaporimeter, and wrinkle density (WD) as an index of morphological alteration was measured by image analyzer. Significant changes of TEWL were not observed in the control and 0. SMED group, but 1.OMED and 3. OMED groups noted significant difference. TEWL of 3. OMED group was rapidly increased to the 3rd day and decreased until the 14th day when it reached nearly to normal level, Time-courses of TBWL for 1. OMEB and 3. OMED groups displayed similar pattern, but different only in the magnitude. WD were significantly decreased during the 3rd-5th day in all of the irradiated groups and then increased during the last period to the 14th day, but did not recover the normal level at the 14th day. Time-courses of WD for all groups exhibited similarity, and were entirely dependent on the exposed doses. We also observed histological changes which included hyperplasia, sunburn cell (SBC) formation, accumulation of polymorphonuclear leukocyte (PMNs), and loss of collagen of UVB- exposed hairless mouse skin. Changes of TEWL and WD are helpful in understanding of epidermal and dermal damages by single exposure of UVB.

We validated the physiological activity of Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan (S. claviflorum) extracts (leaves, stems, fruits, and flowers) as a cosmetic ingredient. Firstly, S. claviflorum extracts removed over 80% of free radicals at various concentrations in antioxidant experiments using the DPPH and ABTS assay. In cytotoxicity experiments using human epidermal keratinocytes, S. claviflorum extracts showed low cytotoxicity. In addition, S. claviflorum extracts significantly increased the expression of keratin (KRT)1, KRT2, KRT9, KRT10, which are differentiation markers of keratinocytes, as well as genes involved in the maintenance of skin barrier function, including involucrin (IVL), loricrin (LOR), filaggrin (FLG), and claudin1 (CLDN1). In particular, the expression of FLG protein, inhibited by interleukin (IL)-4/IL-13 in atopic dermatitis, was restored by S. claviflorum extracts in in vitro experiments. Therefore, S. claviflorum extracts with excellent antioxidant efficacy and skin barrier improvement function will be useful materials for the development of future atopic dermatitis treatments and cosmetics.

Body fluid has been studied for diverse fields like Ringer's solutions, artificial joint fluids, cell growth culture media because it plays a crucial role in controlling body temperature and acts as a solvent for diverse metabolite processes in the body and delivery media of mineral, energy source, hormone, signal and drug from and to cell via blood or lymphatic vessel by osmotic pressure or active uptake. Stratum corneum containing extracellular lipids and NMF (natural moisturizing factor) absorbs atmospheric water residing outside of cells and utilize it to hydrate inside of their own. This process is related to skin barrier function. In this study, we conducted the cell viability test with Cell Bio Fluid $Sync^{TM}$, which mimicks body fluids including amino acids, peptides, and monosaccharides to strengthen skin barrier, and the clinical skin improvement test with cosmetics containing Cell Bio Fluid $Sync^{TM}$. In the cell viability test, HaCaT cell was treated with PBS for 3 hours, followed by the treatment of a cell culture medium (DMEM) and isotonic solution (PBS) and Cell Bio Fluid $Sync^{TM}$ for 3 hours each. Then, MTT assay and image analysis were conducted. In the clinical skin improvement test, twenty-one healthy women participated. Participants applied cosmetics containing Cell Bio Fluid $Sync^{TM}$ on their face for a week and evaluated the skin hydration, skin roughness, brightness and evenness. All measurements were conducted after they washed off their face and took a rest under the constant temperature ($22{\pm}2^{\circ}C$) and constant humidity conditions ($50{\pm}5%$) for 20 minutes. All the data were analyzed by SPSS (version 21) software program. Results showed that Cell Bio Fluid $Sync^{TM}$ improved both the cell viability and in vivo skin conditions such as skin hydration, roughness, brightness and evenness.
