Addition of l00mM $MgSO_4$ to a cell culture after 54 hours resulted in a 2.4-fold increase in the sisomicin titre compared to a control to which no $MgSO_4$ was added, and a considerable amount of intracellular sisomicin was liberated outside the cells. The occurrence of product inhibition in fermentation was confirmed by a reduction in net sisomicin production with increasing amounts of added sisomicin without addition of $MgSO_4$. All added sisomicin was bound to sisomicin-free cells in the absence of $MgSO_4$, whereas approximately 40% of added sisomicin was bound with the addition of l00mM $MgSO_4$. Under conditions of no enzmye synthesis, maintained by adding chloramphenicol to exclude product repression, sisomicin was produced in the presence of 100 mM $MgSO_4$ but little sisomicin was produced in the absence of $MgSO_4$.
Sisomicin, which is one of aminoglycoside antibiotics, was produced by Micromonospora inyoensis. The effects of carbon sources on sisomicin production were studied in batch cultures. Starch, dextrin and maltose were good carbon sources for the production of sisomicin. However, when glucose was used, the antibiotic productivity decreased significantly due to a carbon catabolite regulation. The carbon catabolite regulation depends mostly on carbon catabolite repression, but not on carbon catabolite inhibition. On the other hand, the growth-production curves of batch cultures show that sisomicin is produced most actively during the idiophase.
This study was carried out to enhance the production of sisomicin by Micromonospora inyoensis ATCC 276000. The effect of various sugars and organic acids as the supplements to a basal mineral medium was tested for the sisomicin production by fully grown mycelium. Among the substances tested, acetate and citrate greatly increased the production of sisomicin at the sixth day of culture. Especially, in the basal mineral medium containing 0.1M sodium acetate, sisomicin production was 6.7 times more than that in the same medium with glucose. When 0.1M sodium acetate was added in the fermentation medium initially, the cell growth was inhibited by sodium acetate, although specific productivity was higher than that in the same medium without sodium acetate. On the contrary, when sodium acetate was added to the culture after three days, the sisomicin production and specific productivity were 1.6 times higher than those in the same medium without sodium acetate. The results suggested that sodium acetate was a stimulating substance of sisomicin production by M. inyoensis.
Methylotrophic actinomycetes No. 155 produced an aminoglycoside antibiotics(AG)-resistant inhibitor. We have previously reported that the inhibitor shows strong inhibition to sisomicin-resistant strain. In order to understand the functions of inhibitor and sisomicin-resistance, characterizations and purification of inhibitor were investigated. Strain No. 155 was tentatively identified as Nocardiopsis sp. based on morphological and some physiological characteristics. In the antimicrobial activity test, the addition of inhibitor to sisomicin showed a reduction effect of MIC on the test strains such as Gram(+), Gram(-) bacteria and yeasts. The combination of the inhibitor and various antibiotics revealed synergistic against E. coli K-12 and B. subtilis PCI 219. The induced intracellular proteins from sisomicin-resistant strain exhibited the sisomicin inactivation by invitro test. And the induced intracellular proteins were inactivated by addition of the inhibitor. The inhibitor compound was purified by anion exchange chromatography(Dowex-1) and HPLC using Asahipak ES-502C column. The purified inhibitor compound was detected in a single peak(above 98.5% purity) through the HPLC analysis.
The effects of cobalt chloride, methionine, and various carbon sources on the sisomicin production by M. inyoensis NRRL 3292 were investigated. It was found that both cobalt chloride and methionine exerted a greater stimulatory effect on sisomicin formation. Kinetic studies with various carbon sources revealed thai polysaccharide such as starch or dextrin was found io be better than glucose for sisomicin production Moreover, the relatively low concentration of dissolved carbon dioxide was one of the most important factors In accelerating sisomicin production during idiophase.
A sisomicin-producing strain, Micromonospora inyoensis, was treated with various mutagens. The optimal death rates to obtain the high-producers of sisomicin were 90% for ultraviolet light, 99% for nitrosoguanidine, and 99.3% for nitrous acid, respectively. In place of the method of liquid culture, an agar plug method, which is easy and convenient, was used to measure the antibiotic-producing abilities of the strains isolated from mutagen-treated cells. On the other hand, as the selection method of overproducing mutants after mutagenesis, gradient agar plates which included various antibiotics and salts were used. Among the antibiotics and salts tested, gentamicin and kanamycin as antibiotics, and CuCl$_2$ and HgCl$_2$ as salts, were effective to select the high-producers of sisomicin.
Fermentation patterns were changed by adding magnesium sulfate to the fermentation broth and its effect on enhancement of sisomicin production was investigated. When cell growth was expressed by DNA content, trophophase and idiophase were separated, but not by dry cell weight. On the other hand, addition of magnesium sulfate had the antibiotic accumulated inside the cells be liberated into the outside, and this effect resulted in improving the final antibiotic yield. The maximum antibrotic yield was obtained when 100 mM magnesium sulfate was added after one day of cultivation, and enhanced more than three times compared to that of the control to which it was not added.
Effects of mineral salts on sisomicin fermentation were investigated. The optimal concentration of CoCl$_2$for accomplishing a high antibiotic yield was found to be 16.8 $\mu$M at which it could function as a cofactor. At this level the other mineral salts tested had no effect. On the other hand, at much higher concentration levels (above 1 mM), four mineral salts such as ZnSO$_4$, KH$_2$PO$_4$, FeSO$_4$and MgSO$_4$were used in order to liberate the intracellular sisomicin out-side the cells, because the sisomicin accumulated mostly in cells and it was supposed to limit the improvement of antibiotic yield. ZnSO$_4$and KH$_2$PO$_4$had no effect at all, and FeSO$_4$brought about some improvement. However, by keeping the concentration of MgSO$_4$to be 25 mM or higher in culture broths, the antibiotic yield could be improved by more than 100%, partially due to the enhanced liberation of the intracellular antibiotic.
Effects of temperature on sisomicin fermentation were investigated. From the specific growth rates for logarithmic phase estimated at various temperatures, 8.2 kcal/g-mol was obtained as an activation energy for cell growth. It suggests that cell growth rate was limited by the internal diffusion layers for nutrients or oxygen caused by aggregated cells. Final antibiotic titer was decreased with in-creasing temperature, and it depended highly on the temperature to which cells were exposed during the logarithmic phase of growth. Temperature shifts during fermentation brought about an increase in antibiotic productivity.
In order to predict gas hold-up and oxygen transfer rate during sisomicin fermentation employing air lift fermentor, simulated media similar to fermentation broths in rheological proper- ties were prepared and used. Rheological properties of fermentation broths from 40 hours and 60 hours of cultivation were analyzed by applying to Power's Law equation. Regardless of addition and no addition of MgSO$_{4}$, the tendencies, that n value was decreased and K value was increased as aeration rate was increased, were shown. Simulated media of twelve different fermentation broths were formulated in a range of 0.7 to 2.1% CMC, and the values of gas hold-up and k$_{L}$a depending on superficial air velocity were measured using these simulated media. And the relation- ships, $\varepsilon$=$\alpha$U$_{Gr}$$\beta$, K$_{L}$a=$\gamma$U$_{Gr}$$\delta$ were obtained, and these equations are thought to be used to predict the values of gas hold-up and k$_{L}$a during fermentation.
