보중익기합대칠기탕(補中益氣合大七氣湯) 추출물의 ICR마우스에서 경구 단회투여독성 평가 (Single Dose Toxicity Test of Bojungikkeehapdaechilkitang water extract in Male and Female ICR Mice)

  • An herbal water extract of Bojungikkeehapdaechilkitang(BDT) was prepared to test it for single-dose and repeated-dose toxicity, genotoxicity and reproductive toxicity, and to obtain a 50% lethal dose$(LD_{50})$, approximated lethal dose(ALD), and approximated target organs for BDT. The extract was tested on female and male ICR mice according to KFDA Guideline 1999-61 at doasge level of 2000, 1000, 500, 250 and 125mg/kg/10mL In this study, clinical signs, mortalities and gross findings of principal organs were observed for 14 days of single dosing, and afterwards in some cases. The ALD and $LD_{50}$ of BDT extract obtained in this study was>2000mg/kg for both male and female ICR mice. Also, any possible digestive toxicity of BDT extract was found to be above 1000mg/kg in both male and female ICR mice. The results of this study strongly suggest that BDT extract has no toxic effect at dosage level below 500mg/kg.

  • The immunopotentiating effect of ethanol extract, butanol fraction and petroleum ether extract of Panax ginseng on the immunotoxicity of benzo(a)pyrene were investigated in mice. A single administration of benzo(a)pyrene induced an apparent but relatively transient reduction in HY titer, Arthus reaction, delayed type hypersensitivity, rosette forming cell and natural killer cell activity Ethanol extract very significantly restored HY titer, Arthus reaction. RFC and natural killer cell activity. Butanol fraction have no effect. But petroleum ether extract very significantly restored humoral and cellular immune response and especially natural killer cell activity.

  • The immunopotenciating effects of ethanol extract, butanol fraction and petroleum ether extract of Korean Ginseng on the immunotoxicity of mitomycin C were investigated in mice. A single administration of mitomycin C induced an apparent but relatively transient reduction of spleen weight, hemagglutination titer, Arthus reaction, RFC and natural killer cell activity. Ethanol extract of Ginseng significantly restored spleen weight, HA titer, RFC and natural killer cell activity. Butanol fraction of Ginseng significantly restored HA titer, RFC and natural killer cell activity. Petroleum ether extract of Ginseng very significantly restored spleen weight, Arthus reaction, DTH, RFC and natural killer cell activity.

생부자(生附子) 및 삼부탕(蔘附湯)의 aconitine 함량 분석 및 삼부탕 단회투여 독성시험 (Single Oral Dose Toxicity Study and Aconitine Content Analysis of Raw Aconiti Tuber and Sambu-tang)

  • Objectives: This study investigated the aconitine contents analysis of Buja extracts (raw material of Buja, hot water extract of Buja, and hot water extract of Sambu-tang) and the single oral dose toxicity of Sambu-tang-R in six-week-old Sprague-Dawley rats in order to compare the toxicity of Buja extracts.Methods: Aconitine content analysis is that Buja extracts were hold purity test. To detect single oral dose toxicity, six-week-old Sprague-Dawley rats were divided into two groups, a normal control group and a sambutang-R (2,000 mg/kg) group. For 14 days of treatment, clinical signs, body weight, clinical chemistry, necropsy, and histopathology were examined.Results: The aconitine contents of the Buja extracts were Buja-RH (0.1738%), Buja-RD (0.1746%), and Sambu-tang-R (0.0961%). There were no cases of death in either the control group or the experimental group. Nor was there any disorder to the clinical signs or any significant change in body weight in either group. There was no significant change of clinical chemistry or disorder of necropsy findings in either the control or the experimental group. And there was no difference in histopathological findings in comparing the control group with the experimental group.Conclusions: These results suggest that the aconitine content of the hot water extract of Buja was similar to the raw material of Buja, but the hot water extract of Sambu-tang had greatly decreased aconitine content. These results also suggest that a single oral lethal dose of Sambu-tang-R for Sprague-Dawley rats exceeds 2,000 mg/kg for both female and male rats.

Aldose Reductase Inhibitory Alkaloids from Corydalis ternata

  • A methanolic extract of Corydalis ternata having aldose reductase inhibitory activity was examined as a possible aldose reductase (ALR2) inhibitor, a key enzyme involved in diabetic complications. Seven alkaloids, tetrahydrocoptisine (1), corydaline (2), tetrahydropalmatine (3), isocorybulbine (4), corybulbine (5), dehydrocorydaline (6), and N-methyltetrahydroberbinium (7) were isolated from $CHCl_3$ fraction of C. ternata methanol extract. Among them, compounds 1, 5, and 7 exhibited $5.04{\pm}1.97%$, $5.00{\pm}1.26%$, and $1.80{\pm}2.33%$ inhibitions, respectively at $40{\mu}M$. The activities of the single compounds were not comparable to that of the whole extract, suggesting that the whole combination of each single compound was responsible for the activity of the extract as shown in many cases of natural medicines. Even though this is the second report on aldose reductase inhibition activity of C. ternata, recombinant human aldose reductase was employed in this study unlike in the previous report. Furthermore, the aldose reductase inhibitory activities of isocorybulbine, corybulbine, and N-methyltetrahydroberbinium, to the best of our knowledge, were evaluated for the first time in this study. These results suggest a use of the extract of C. ternata for ameliorating diabetic complications.

Benzo(a)pyrene 유도 DNA 손상에 대한 Genistein과 청국장추출물의 보호효과 (Protective Effect of Genistein and Korean Fermented Soybean (Chungkookjang) Extract against Benzo(a)pyrene Induced DNA Damage in HepG2 Cells)

  • Chungkookjang (CKJ) is a fermented soybean product and one of favorite traditional foods in Korea. In this study, the alcoholic extract from Korean fermented soybean (CKJ) and its one of major flavonoids, genistein were evaluated for their protective effect against B(a)P induced cytotoxicity and DNA damage in HepG2 cells. CKJ extract and genistein decreased B(a)P-induced cell cytotoxicity. CKJ extract inhibited DNA single strand breaks evaluated by single cell gel electrophoresis. From RT-PCR study, it was revealed that CKJ extract decrease DNA damage induced in HepG2 cells expressing CYP1A1 and 1A2 by B(a)P. The metabolizing activities of CYP1A1 and CYP1A2, as measured by the 7-alkoxy resorufin O-deethylation (AROD) assay, showed that CKJ extract and genistein inhibited CYP1A1 and CYP1A2 activities. Genistein may contribute to these biological effects of CKJ extract at least in part. All these results indicate that CKJ extract and genistein may be useful for protection against B(a)P-induced cytotoxicity and DNA damage. Therefore, the alcoholic extract of Korean fermented soybean (CKJ) is suggested to be promising functional food which can prevent the cellular genotoxicity of dietary and lifestyle related carcinogens.

ICR 마우스를 이용한 귀비탕 열수 추출물의 단회투여 독성 실험 (Single Dose Oral Toxicity Study of the Gwibi-tang Extract in ICR Mice)

  • Purpose: This study was performed to evaluate the single dose oral toxicity of Gwibi-tang extract in ICR mice. Methods: 0(control group), 1250, 2500 and 5000 mg/kg of Gwibi-tang extracts were orally administered to 20 male and 20 female ICR mice. After single oral administration of Gwibi-tang extract to ICR mice, we observed number of the death, clinical signs, changes of body weights for 14 days. After 14 day of Gwibitang extract administration, all mice were sacrificed and major organs were observed. Results: Compared with the control group, we could not find any toxic signs in the mortalities, clinical signs, body weight changes, necropsy findings and hematological values in all treated groups(1250, 2500 and 5000 mg/kg). Conclusions: $LD_{50}$ value of Gwibi-tang extracts may be over 5000 mg/kg and it may have no side toxic effect to ICR mice.

총 디젤분진의 DNA 손상작용과 야채 및 과일추출물의 보호효과 (Chemopreventive Effect of Vegetable or Fruit Extract Against Total Diesel Exhaust Particle Extract in NIH/3T3 Cells Using Alkaline Single Cell Gel Electrophoresis)

  • In urban areas, diesel exhaust particles (DEP) are probably a major component of particulate matters, especially in Korea where drive many diesel vehicles. The aim of this study was to investigate genotoxic effects of DEP using single ceil gel electrophoresis. In order to evaluate the mechanisms of DEP genotoxicity, the rat microsome mediated and DNA repair enzyme treated comet assays together with conventional comet assay were performed. Total diesel particles (DEPT) was collected without site fractionation from diesel engine bus and dichloromethane extract was obtained. The organic extract of DEPT revealed DNA damage itself in NIH/3T3 cells. The level of DNA breaks plus oxidative DNA lesions and microsome mediated DNA damage was assessed by modified single cell gel eletrophoresis. DEPT was able to induce oxidative DNA damage as well as microsome mediated DNA damage. Vitamin C as an model antioxidant reduced DNA damage in endonuclase III treated comet assay. One of flavonoid, galangin as a CYP1A1 inhibitor. reduced DNA damage in the presence of S-9 mixture. $DEP_T$ is the sources of oxidative stress, but antioxidants can significantly reduce oxidative DNA dmage. And $DEP_T$ may contain indirect mutagens which can be inhibited by CYP1A1 inhibitors. The ethanol extracts of the mixed vegetables (BV) or the mixed fruits (BF) were evaluated for their in vitro antigenotoxic effects. BV and BF showed potent Inhibitory effects against DEPT induced DNA damage with oxidative DNA lesions and in the prescence of S-9 mixture. These results indicate that BV and BF could prevent cellular DNA damage by inhibiting oxidative stress and suppressing cytochrome P4501A1 in cell culture.

  • Panax ginseng C.A. Meyer has been extensively used in the traditional oriental medicine as a restorative, tonic and prophylatic agent. This study was devised to develop a chemopreventive agent from panax ginseng to be able to suppress the genotoxicity and oxidative damage by ractive oxygen species, which are involved with cancer or aging. Ginseng petroleum ether extract (GPE) and one of its fraction, P2, showed an antioxidative effect on the lipid peroxidiphenyl-2-picryl hydrazil (DppH) radical generation. They also showed the suppressive effect of H2O2 or KO2 induced DNA damage by single cell gel electrophoresis (SCGE). Results from our study indicate that GPE and P2 are capable of protecting lipid peroxidation, and oxidative DNA damage. Therefore, GPE and P2 may be useful chempreventive agents which are involved with cancer and aging.

Production of Platinum Nanoparticles and Nanoaggregates Using Neurospora crassa

  • Fungal biomass and fungal extract of the nonpathogenic fungus Neurospora crassa were successfully used as reducing agents for the biosynthesis of platinum nanoparticles (PtNPs). The experiment was carried out by exposing the fungal biomass or the fungal extract to a 0.001 M precursor solution of hexachloroplatinic(IV) acid ($H_2PtCl_6$). A change of color of the biomass from pale yellow to dark brown was the first indication of possible formation of PtNPs by the fungus. Subsequent analyses confirmed the intracellular biosynthesis of single PtNPs (4-35 nm in diameter) and spherical nanoaggregates (20-110 nm in diameter). Using the fungal extract, similar results were obtained, producing rounded nanoaggregates of Pt single crystals in the range of 17-76 nm.
