Comparison of Photocyclization Reactions of Fluoro- vs Nonfluoro-Substituted Polymethyleneoxy Donor Linked Phthalimides

  • Photochemical reactions of fluoro- vs. nonfluoro-substituted polymethylenoxy chain linked phthalimide were carried out to explore how electronegative fluorine atoms inside the donor chain influence photocyclization reaction efficiencies and to briefly determine the alkali metal binding properties of the photoproducts. The results of this study show that the fluorine-substituted donor chain linked phthalimide undergoes inefficient photocyclization via single electron transfer (SET)-induced excited state pathways to generate 14-membered cyclic amidol compared to nonfluoro-analog due to low electron donor ability of the terminal oxygen donor site. These results show that photoinduced intramolecular SET processes arising from ${\alpha}$-silyl ether electron donors to phthalimides are largely dependent on the kinds of substituents inside donor chain. Finally, a preliminary study with the cyclic amidols generated in this effort showed that they have weak alkali metal cation binding properties regardless of absence/presence of fluoro-substituents.

  • In order to prepare a novel human insulin analogue suhbstituted with homoserine at B$^{30}$ / position, (B$^{30}$ /-homoserine) human insulin, a synthetic gene was designed by linking directly a gene for B chain with that for A chain. This gene was constructed by enzymatic joining of 10 different synthetic oligonucleotides, and then inserted at the polylinker region of pUC19 plasmid. To achieve a high level of gene expression, the gene fusion technique region of pUC19 plasmid. To achieve a high level of gene expression, the gene fusion technique was employed using amino terminal regions of lacZ gene up to Clal or hpal, and either of them has been located under tac promoter. The chemical induction of these fused genes by isopropyl-.betha.-D-thiogalactopyranoside (IPTG) gave a satisfactory level of expression in Escherichia coli harboring the ocnstructed plasmids. It was observed that the fused gene product as a single chain insulin precusor was produced more than 30% of total cell protein of E. coli as a form of inclusion body.

Temperature-dependent Conformational changes of Single Polymer Chains

  • Atomic force microscope (AFM) enables us to stretch a single polymer chain by picking it at its two modified termini. Using this method called "nanofishing," we have studied statistical properties of single polymer chains. A force-extention curve obtained for a polystyrene with thiol termini in a cyclohexane showed a good agreement with a worm-like chain (WLC) model, and thus gave microscopic information about entropic elasticity. In this report, the experiments were performed at wide-range temperatures, resulting in wide-range solvent qualities from poor to good solvent condition. The temperature dependence of statistical properties of polystyrene was examined. The coil-strand transformation realized in a poor solvent was also discussed.

Highly flexible dielectric composite based on passivated single-wall carbon nanotubes (SWNTs)

  • Single-walled carbon nanotubes (SWNTs) was modified with various length of linear alkyl chains and passivated to form dielectric filler. The modified SWNTs embedded into epoxy matrix to fabricate a flexible composite with high dielectric constant. The dielectric behavior of the composite was significantly changed with various alkyl chain length(n) of pyrene. The dielectric constant of the epoxy/SWNTs composite significantly increased with respect to increase in length of alkyl chain at the frequency range from 10 to 105Hz (n=12and18).We also found that the passivated epoxy/SWNTs composite with high dielectric constant presented low dielectric loss. The resulted dielectric performances corresponded to de-bundling of nanotubes and their distribution behavior in the matrix in terms of tail length of alkyl pyrene in the passivation layer.

  • Sex determination in genomic DNA from human blood leucocytes was performed by amplification of human Y chromosome-specific DNA sequences using PCR technique. A clear DNA fragment(154 nucleotides long) was appeared only in the male genomic DNA, but no specific band was observed in the case of female genomic DNA and negative control. To know the sensitivity of this method, the amplification reaction was performed in genomic DNA diluted to 2pg equivalent to the amonut present in the single human cell, and clear band also observed. The PCR amplification was so succesfully performed in the single leucocyte separated from human blood using micromanipulator that this techniqe is assumed to be applied to single blstomere before embryo transfer.

How Quick Response affects the Supply Chain Performance

  • Purpose - The goal of this research is to examine the influence of Quick Response on the supply chain performance. Furthermore, this study investigates the potential of Quick Response to be a more advanced form of supply chain collaboration program with extensive information sharing activities. Research design, data, and methodology - The mathematical model is developed to represent the two stage supply chain system with a single manufacturer and one retailer. In the numerical study with the proposed mathematical models, three supply chain systems including the traditional system, Quick Response, and the fully shared information system are compared in terms of their profits. Results - The numerical analysis shows both manufacturer and retailer obtain greater profits under Quick Response than in the traditional system. While the fully shared information outperforms Quick Response as well as the traditional system, it results in lower manufacturer's profit compared with Quick Response. Conclusions - According to the numerical examples, Quick Response is the effective supply chain collaboration program that is beneficial to every supply chain member. The fully shared information system, as a more advanced form of collaboration than Quick Response can bring more benefits to the whole supply chain system, but it is necessary to prepare the proper incentive program that enables every member to share its benefits equally.

Molecular Dynamics Study of the Self-Diffusion Coefficient and Velocity Autocorrelation Function of a Polymer Molecule in Solution

  • A molecular dynamic computer experiment was performed on a system of 108 particles composed of a single polymer chain and solvent molecules. The state considered was in the immediate neighborhood of the triple point of the system. The polymer itself is an analog of a freely jointed chain. The Lennard-Jones potential was used to represent the interactions between all particles except for that between the chain elements forming a bond in the polymer chain, for which the interaction was expressed by a harmonic potential. The self-diffusion coefficient and velocity autocorrelation function (VACF) of a polymer were calculated at various chain lengths $N_p$, and various interaction strengths between solvent molecules and a polymer chain element. For self-diffusion coefficients D, the Einstein relation holds good; as chain length $N_p$ increases the D value decreases, and D also decreases as ${\varepsilon}_{cs}$ (the interaction parameter between the chain element and solvent molecules) increases. The relaxation time of velocity autocorrelation decreases as ${\varepsilon}_{cs}$ increases, and it is constant for various chain lengths. The diffusion coefficients in various conditions reveal that our systems are in a free draining limit as is well known from the behavior of low molecular weight polymers, this also agrees with the Kirkwood-Riesman theory.

An Inventory Rationing Method in a M-Store Regional Supply Chain Operating under the Order-up-to Level System

  • This paper addresses the inventory rationing issue embedded in the regional supply chain inventory replenishment problem (RSIRP). The concerned supply chain, which was fed by the national supply chain, consisted of a single warehouse distributing a single product to multiple stores (M-stores) with independent and normally distributed customer demand. It was assumed that the supply chain operated under the order-up-to level inventory replenishment system and had only one truck at the regional warehouse. The truck could make one replenishment trip to one store per period (a round trip per period). Based on current inventories and the vehicle constraint, the warehouse must make two decisions in each period: which store in the region to replenish and what was the replenishment quantity? The objective was to position inventories so as to minimize lost sales in the region. The warehouse inventory was replenished in every fixed-interval from a source outside the region, but the store inventory could be replenished daily. The truck destination (store) in each period was selected based on its maximum expected shortage. The replenishment quantity was then determined based on the predetermined order-up-to level system. In case of insufficient warehouse inventories to fulfill all projected store demands, an inventory rationing rule must be applied. In this paper, a new inventory rationing rule named Expected Cost Minimization (ECM) was proposed based on the practical purpose. The numerical results based on real data from a selective industry show that its performance was better and more robust than the current practice and other sharing rules in the existing literature.

Sensitivity of Conditions for Lumping Finite Markov Chains

  • Markov chains with large transition probability matrices occur in many applications such as manpowr models. Under certain conditions the state space of a stationary discrete parameter finite Markov chain may be partitioned into subsets, each of which may be treated as a single state of a smaller chain that retains the Markov property. Such a chain is said to be 'lumpable' and the resulting lumped chain is a special case of more general functions of Markov chains. There are several reasons why one might wish to lump. First, there may be analytical benefits, including relative simplicity of the reduced model and development of a new model which inherits known or assumed strong properties of the original model (the Markov property). Second, there may be statistical benefits, such as increased robustness of the smaller chain as well as improved estimates of transition probabilities. Finally, the identification of lumps may provide new insights about the process under investigation.

Joint Optimization of the Number of Suppliers and the Order Quantities Considering Compensation Orders under Supply Chain Disruptions (공급사슬 중단에 대비한 공급업체의 수와 주문량 및 보완주문 최적화 방안에 관한 연구)

  • In this study, we develop an optimal sourcing strategy considering compensation orders to mitigate the supply chain disruption risks. We considered two-echelon supply chain consisting of a single buyer and multiple suppliers who have fixed transaction cost and probabilistic disruption risks. Under this setting, we provide the joint optimization method to determine the number of suppliers and the order quantities. Through numerical examples, we provide managerial implications on the sourcing strategy by investigating changes in the order quantities and the number of suppliers due to the degree of supply chain disruption risks.
