Hypoglycemic Effects of Pills Made of Mulberry Leaves and Silkworm Powder in Streptozotocin-Induced Diabetic Rats (뽕잎과 누에가루 혼합환의 Streptozotocin유발 당뇨쥐에서의 혈당강하 효과)

  • This study was carried out to examine the effects of pills made of mulberry leaves and silkworm powder on lowering blood glucose level. Experimental animals were Sprague-Dawley male rat weighing 100$\pm$10 g and pills were supplemented with 0.4% (4 g/kg) diet. Experimental groups were assigned to diabetic group (DM group) and pill supplemented groups. Pill supplemented groups were classified 100% mulberry leaves (M group), mixing 25% silkworm powder to mulberry leaves (25SM group), mixing 50% silkworm powder to mulberry leaves (50SM group), mixing 70% silkworm powder to mulberry leaves (75SM group) and 100% silkworm powder (100S group). Experimental diets and water fed ad libitum, and streptozotocin was injected to induce diabetic state after 3rd weeks and sacrificed on the 9th day. The contents of 1-deoxynojirimycin(DNJ) were increased with adding the silkworm powder. The contents of GABA and rutin were increased with adding the mulberry leaves. In vitro, intestinal mucosa $\alpha$-glucosidase inhibitory activities were significantly increased in pills which mixed with silkworm powder by 50%. Blood glucose levels were high in groups which mixed with silkworm powder by 50% compared to DM group. Intestinal mucosa maltase activity in proximal part was significantly reduced in pill supplemented group compared to DM group and pill supplemented groups were no significant difference. Enzyme activity in middle part was no significant difference in experimental groups. Enzyme activity in distal part was decreased in pill supplemented groups, especially in 50SM, 75SM and 100S groups were significantly reduced compared to DM group. Sucrase and lactase activities in pill supplemented groups were significantly reduced at proximal part, and there was no significant difference in middle and distal parts. In conclusion, pills made of mulberry leaves and silkworm powder increased the $\alpha$-glucosidase inhibitory activity in vitro and reduced the blood glucose levels by controlling the disaccharidase activities of intestinal proximal part in STZ-induced diabetic rat. The synergistic effect was the highest when mulberry leaves was mixed with silkworm powder by the ratio of 50 : 50.

Effects of Silkworm Extract Powder on Plasma Lipids and Glucose in Rats (누에 추출 분말이 흰쥐의 혈장지질 및 혈당농도에 미치는 영향)

  • Changes of plasma lipids, total cholesterol and phospholipids content according to diet of silkworm extract powder were as follows. Decrease ratios of plasma lipids at 3, $5\%(w/w)$ treating groups were plasma lipids 5.16, $9.15\%$, those of total cholesterols were 1.63, $2.76\%$ and those of phospholipids were 1.37, $2.00\%$, respectively. Decreasing effects of plasma triglyceride content according to diet of silkworm extract powder was higher than those of total cholesterols and phospholipids contents. Decrease of plasma glucose concentrations were $4.62\%\;and\;6.46\%\;at\;3\%,\;5\%(w/w)$ treating group than control group, respectively. These results were not proportional decrease according to treating amount. In the other hand, changes of insulin concentration of plasma, pancreas and femur were appeared the similar tendency with plasma glucose concentration. Relationship between treating amount of silkworm extract powder and insulin concentration had not positive relativity, In conclusion, treatment of silkworm extract powder about $3\%(w/w)$ could be expected to prevention and/or medical treatment effect of diabetic patients.

Effects of Silkworm Powder on Blood Glucose and Lipid Levels in NIDDM(Type II) Patients (인슐린 비의존형(Type II) 당뇨환자에서 누에분말 섭취가 혈당 및 혈중지질농도에 미치는 영향)

  • The purpose of this study is to investigate the effects of the silkworm powder on blood levels of glucose, Hb Alc, insulin, and lipids. Thirty-one NIDDM patients from Kyung Hee Medical Center were divided into two groups : patients with silkworm powder supplements and drug treatments(Drug diabetes) and patients with silkworm powder supplement only(Non-drug diabetes). For the control group, age-matched subjects were recruited. During the 4 weeks of the experimental period, silkworm powder(500mg/mea1) was given to the subjects right after each meal. Nutritional assessments and dietary education were carried out periodically, and body weight and blood pressure were measured when patients visited the hospital. Overnight fasting and 2-h postprandial blood glucose levels were measured at 2 week intervals. The blood levels of insulin, Hb Alc, and lipids were measured before and after the supplements. The mean ages of the three groups were 56.7-59.6 years old. The height, weight, and BMI did not differ among the groups. The fasting blood glucose levels were 138.1$\pm$22.0mg/dl for the Drug treated diabetes group, 175.0$\pm$32.0mg/dl for the Non-drug diabetes group, and 108. 3$\pm$16.gmg/dl for the control group at the begining of the supplement. After 4-wks of supplements, the blood levels of glucose tended to decrease in all three experimental groups. Before the supplements, the 2-h postprandial blood glucose levels of the Drug diabetes, Non drug diabetes, and control groups were 244.7$\pm$62.6mg/dl, 272.4$\pm$40.1mg/dl, and 147.7$\pm$28.0mg/dl, respectively. After the supplement, the levels were 197.2$\pm$30.gmg/dl, 208.6$\pm$ 56.6mg/dl, and 151.3$\pm$30.3mg/dl, respectively. This shows that silkworm powder tended to lower blood levels by 19.4% and 23.4% in NIDDM patient groups. However, the changes in the blood levels of insulin, Hb Alc, ind lipids were not observed after the supplement. In conclusion, the present study has demonstrated that silkworm powder has a tendency to decrease 2-h postprandial blood glucose levels, but it should be used with caution in controlling the diabetes. Effects of Silkworm Rearing Technique on the Cocoon Crops of Sericultural Farmers in Korea (육잠기술이 잠작에 미치는 영향)

  • This investigation was carried out to find the effects of silkworm rearing technique on the harvesting amounts. In spring and autumn of 1971, 14 silkworm rearing farmers at Botong Ri and Youngsoo Ri, Chongnam Myun, Hwaseung Kun, Kyunggi Do in Korea were selected as sample farmers. They were devided into two groups, high productivity group and low productivity group according to their harvesting amounts per box of silkworm eggs in 1970. The high productivity group represents the farmer who produced above average harvesting amounts per box, and the low group represents below average group. In this investigation, all farmers were adjusted to share uniform rearing condition with even mulberry leaves. Under this uniform condition, the effects of rearing techniques on the harvesting amounts per box of silkworm eggs and the effects of rearing condition on the harvesting amounts were inferred. Results obtained are as fellows; (1) Measuring from the 14 sample farmers, the effect of rearing technique on the harvesting amounts per box covers 15% of farmers in spring rearing season but 33% in autumn season. (2) Measuring from these sample farmers, the effects of the nutrition condition is less than 56% in spring rearing season but less than 69% in autumn, and it can be concluded that these effects include the effect of climate factor. (3) In the low productivity group. the most important facto. affecting the low productivity is non-disinfection of rearing environment.

ACE Inhibitory and Antioxidative Activities of Silkworm Larvae (Bombyx mori) Hydrolysate (번데기 가수분해물의 ACE 저해활성과 항산화활성)

  • In order to utilize the silkworm larvae (Bombyx mori) protein, defatted silkworm protein was hydrolysed by four enzymes (pepsin, trypsin, neutrase and alcalase) at various hydrolysis times (6, 12, 18, 24 and 30 hr) and suspension concentrations (2, 5, 10, 15 and 20%). Protein solubility index, ACE (angiotensin converting enzyme) inhibitory activity and antioxidative activity of silkworm protein hydrolysates were investigated. The optimum condition of hydrolysis was 10% suspension concentration and 18 hr. Protein solubility index of trypsin treatment was higher than other enzyme treatments. ACE inhibitory activity and $IC_{50}$ value of antioxidative activity of neutrase treatment were 86.16% at $100\;{\mu}g/mL$ and $352.75\;{\mu}g/mL$, respectively; also, these values were higher than other enzyme treatments.

  • Biotechnological researches in Central Asian Research Institute of Sericulture were developed since 1981 when laboratory of artificial diets for mulberry silk­worm was created. The researches were directed not only to creation of accessible artificial diets (AD) for off-season mass rearing of economical beneficial insect and for industrialization of sericulture but also to obtaining of easy reproducible source of animal proteins, carbohydrates, lipids, enzymes, enzymes inhibitors and other metabolites - bioresource for manufacture of oriental medicine preparations. Space experiments with silkworms became possible with use of AD. These experiments used mulberry silkworm as high effective board test organism with outstanding developmental, reproductive and transgenic properties having impor­tant significance in future of cosmonautics. Introduction to practice of bioprotective and ecologically pure method of drying and sterilizing of green cocoons and by-products of silk manufacture allows to increase efficiency of cocoons reeling technology as well as to preserve nativity of biological active substances in mulberry silkworm pupae that is unachievable by traditional methods of raw material processing. New spheres of application of two important silk proteins - fibroin and sericin in medicine, cosmetics, for bio- and nanotechnology are opening with use of biotechnological approaches. At present the achievements and experience of R & D in these fields are used in creation of modern pests biocontrol agents. Factors Affecting the Extractability of Protein of Silkworm Larvae (번데기 단백질(蛋白質)의 추출성(抽出性)에 영향을 미치는 요인(要因))

  • This experiment was designed to investigate the optimum conditions for the extraction of protein from the silkworm larvae by using the oven-dried and freeze-dried samples. The extractability was examined on the basis of kinds of extractants, ratio of sample to extractants, extraction time, temperature, and the effect of pH. The following results were obtained: (1) About 56% of proteins were extracted when 0.05M NaOH was used and the highest extractability was obtained at the ratio of 1 : 40 (sample: extractant). (2) The amount of nitrogen extracted was gradually increased when the extraction temperature was increased from $10^{\circ}C$ to $50^{\circ}C$ in both oven-dried and freeze-dried silkworm larvae. The highest amount of nitrogen was extracted at $50^{\circ}C$ in freeze-dried sample and at $60^{\circ}C$ in oven-dried sample. In addition, sixty min. of extraction time showed the highest extractability of nitrogen, however, not much difference in extractability was found after 10 min of extraction time. (3) More than 60% of nitrogens were extracted at pH 13.0, but less than 20% were extracted from pH 1.0 to 11.0. There was only slight difference in extractability of nitrogen between freeze-dried and oven-dried samples.

Study on the effect of different temperatures on the main productive characters of the silkworm larvae, Bombyx mori under Egyptian agro-climatic conditions (에집트 기후조건 특히 온도의 변화가 원원잠품종의 주요 유전형질에 미치는 영향)

  • Eleven inbred lines of silkworm, Bombyx mori L. were exposed to two main different temperature (23C and 25C) during the larval period in order to elucidate the effect of the temperature differences on their main characters under the, prevailing in Egyptial agro-climactic conditions. The results show that the temperature differences did not affect significantly the pupation ratio and cocoon shell ratio. However, they affected their larval duration to become shorter with the high temperature treatment ($27^{\circ}C$) and their cocoon shell weight and cocoon yield per box of silkworm eggs to be higher in the lower temperature($23^{\circ}C$). It was recommended that some inbred lines should be bred at the 23C throughout the whole larval period, particularly in being reared with high nutritional leaves in order to maximize the hybridization of cocoon yields.

Inhibitory Effect of Cordyceps Militaris Water Extracts on Sarcoma-180 Cell-Induced Ascities Tumor in ICR Mice (누에번데기 및 누에애벌레 밀리타리스동충하초 (Cordyceps Militaris) 열수추출물이 Sarcoma-180 세포로 유발한 마우스 복수암에 미치는 영향)

  • This study was undertaken to evaluate the antitumor activities of Cordyceps militaris of silkworm pupa (CMP) and silkworm larva (CML), as compared with the effect of cordycepin, an active compound found in Cordyceps militaris. Antiproliferation effect of the test materials were evaluated in the sarcoma-180 cells using the MTT test. For the in vivo study, ICR mice were inoculated i.p. with 1.0 ${\times}$ 10$^{6}$ sarcoma-180 cells/mouse on Day 0, and were again i.p. injected with one of the following substances from Day 1 to Day 10 : saline (control group), 50 mg/kg (CMP50, CML50) ,100 ma/kg (CMP100, CML100), or 200 mg/kg (CMP200, CML200) of Cordyceps militaris water extracts, or 1 mg/kg (C1), 2 mg/kg (C2), or 4 mg/kg (C4) of cordycepin. Pretreatment of the sarcoma-180 cells with 100 mg/ml, 500 mg/ml, and 1000 mg/ml of CML (60.1$\pm$2.5%, 49.8$\pm$3.7%, and 45.4$\pm$0.1% of the value for untreated control cells, respectively) or CMP (68.3$\pm$2.1%, 55.1$\pm$0.9%, and 51.4$\pm$3.5% of the value for control cells, respectively) for 48 hrs significantly decreased the survival rate (proliferation) of tumor cells (p<0.05). Body weight of the control mice bearing ascites tumor and injected with saline was 1.4 times of the value for normal animals at day 18. Mice bearing ascites tumor and injected with cordycepin (1, 2, or 4 mg/kg) exhibited a significantly lighter body weight compared with the control mice, while animals injected with CMP or CML (50, 100, or 200 mg/kg) showed a significantly lighter body weight compared with the mice injected with cordycepin. Mice injected with CMP50, CMP100, or CMP200 mg/kg (or CML50, CML100, or CML200 mg/kg) showed a 133% (or 90%), 80% (or 62%), and 68% (or 52%) longer mean survival time, and those treated with C1, C2, or C4 exhibited a 54%, 91% and 80% longer survival time compared to the value for control mice injected with saline. These results indicate that the hot-water extracts of Cordyceps militaris of both silkworm pupa and silkworm larva have an anti-proliferation effect of tumor cells as well as the life prolongation effect in mice bearing ascites tumor, which are superior to the activities of cordycepin.

A Study on the Sensory Evaluation and Nutritional Analysis of Functional Laver Added with Sericulture Powder (양잠산물분말을 첨가한 조미김의 영양성분분석에 관한 연구)

  • This study was carried out to make functional laver added with sericulture powder which have a effect of prevention adult diseases such as diabetes mellitus, hypertention, and obesity. In our lab. a lot of study have been performed about functional effect of sericulture(mulberry leaf, silkpeptide and silkworm) for examples decrease of serum cholesterol, triacylglyceride and control of serum glucose in rat. sericulture powder was prepared by pulverizing freeze-dried. Functional laver samples were prepared, and the sensory evaluation quality, physical characteristics and mineral contents of those were compared. In case of overal quality of sensory evaluation, silkpeptide powder laver was lower than other samples. The content of moisture of functional laver added with sericulture powder samples(control, sample 1, 2, 3) were 0.30, 0.98, 0.24 and 0.28%, respectively. The content of crude protein of functional laver added with sericulture powder samples were 40.50, 44.10, 56.75, and 62.50%, respectively. The content of crude fat of functional laver added with sericulture powder samples were 4.00, 4.10, 4.00 and 4.40%, respectively. The content of ash of functional Laver added with sericulture powder samples were 7.07, 7.53, 7.60 and 7.27%, respectively. The content of calcium of functional laver added with sericulture powder samples were 219, 253, 224 and 229mg/100g, respectively. The content of potassium of functional laver added with sericulture powder samples were 1879, 1919, 1890 and 1907mg/l00g, respectively. The content of iron of functional laver added with sericulture powder samples were 18.03, 18.23, 18.46 and 18.53mg/100g, respectively. The content of zinc of functional laver added with sericulture powder samples were 1.86, 4.47, 4.57 and 5.64mg/100g, respectively. The content of manganese of functional laver added with sericulture powder samples were 4.50, 8.50, 7.00 and 6.00 mg/100g, respectively. It was concluded that this functional laver added with sericulture powder is low calorie, high protein and high mineral health food which have a preventive effect of adult disease.
