Recent Advances in Anti-inflammatory Flavonoid Research since 2004

  • Certain flavonoids possess anti-inflammatory activity. Besides their antioxidative property, the cellular action mechanisms of flavonoids include an inhibition of arachidonate metabolizing enzymes such as cyclooxygenases and lipoxygenases, and a down-regulation of proinflammatory gene expression such as cyclooxygenase-2, inducible nitric oxide synthase and tumor necrosis factor-$\alpha$. In this review, the recent findings of anti-inflammatory flavonoid research since 2004 were summarized. And the cellular mechanisms on signal transduction pathways were also discussed.

  • Plasma membrane Na$^{+}$-K$^{+}$ ATPase (pump) is an essential component to maintain asymmetrical ion distribution across cell membrane. The Na$^{+}$-K$^{+}$ ATPase was discovered by Jens C. Skou in 1957 and since then physiological and biochemical properties of the enzyme have been extensively studied. Jens C. Skou was awarded the 1997 Nobel Prize in chemistry for his discovery of the Na $^{+}$ - $K^{+}$ ATPase.(omitted)

가상세포를 활용한 항염증 효능 응용모델 연구 (Application model research on visualizing anti-inflammation effects by using the virtual cell)

  • Objective : The purpose of this study was to develop the simulator which can analyze the anti-inflammatory effects of herbs based on e-cell, or the virtual cell. Method : We have ensured the medical herbs and its active compounds by investigating the oriental medicine records and NBCI(Biomedicine database). Also we have developed the web-based search system for confirming database related to anti-inflammation. We have researched the cell signal pathway related with inflammatory response control and established the mathematical model of herb interaction on selected signal pathway in e-cell. Finally we have developed the prototype which can confirm the result of this model visibly. Results : We constructed the database of 62 cases of anti-inflammatory active compounds in 61 cases of medical herbs which have been known anti-inflammation effects in the paper, 16 cases of inflammatory factors, 10 cases of signal pathways related with inflammatory response and 6,834 cases of URL(Uniform Resource Locator) of referenced papers. And we embodied the web-based research system, which can research this database. User can search basic and detailed information of medical plants related with anti-inflammatory by using information system. And user can acquire information on an active compounds, a signal pathway and a link URL of related paper. Among investigated ten pathways, we selected NF-${\kappa}B$, which plays important role in activation of immune system, and we searched the mechanisms of actions of proteins which could be components of this pathway. We reduced total network into IKK-$I{\kappa}B$ - NF-${\kappa}B$, and completed mathematic modeling by using ordinary differential equations and response variables of $I{\kappa}B-NF-{\kappa}B$ signaling model network which is suggested by Baltimore Group. We designed OED(Ordinary Differential Equation) for response of IKK, $I{\kappa}B$, $NF-{\kappa}B$ in e-cell's cytoplasm and nucleus, and measured whether an active compound of medicinal plants which is inputted by an user would have a anti-inflammation effects in obedience to change in concentration over time. The proposed model was verified by using experimental results of the papers which are listed on NCBI.

Light Effects on the Membrane Potential in Oat Cells

  • One of the reaction pathways in light-invoked signal transduction can be initiated through ion fluxes across the plasma membrane in higher plants. We isolated protoplasts from oat coleoptile and examined the effects of light on the membrane potential using a membrane potential-sensitive fluorescent probe (bisoxonol). Both red and far-red light initially induced a hyperpolarization in oat cells. Red light-induced hyperpolarization was effectively dissipated by 100 mM $K^+$, but the hyperpolarization induced by far-red light was not depolarized by any of the cations ($K^+$, $Ca^{2+}$, $Li^+$, $Na^+$) tested. The depolarization induced by red light and $K^+$ was inhibited by 200 mM TEA, which is a $K^+$ channel blocker. These results suggest that $K^+$ influx through the inward $K^+$ channel may be a depolarization path in the phytochrome-mediated signal transduction.

Expression and Role of ICAM-1 in the Occurrence and Development of Hepatocellular Carcinoma

  • Intercellular adhesion molecule-1 (ICAM-1) is a member of the immunoglobulin superfamily, its main function being to participate in recognition and adhesion between cells. ICAM-1 is considered closely related to occurrence, development, metastasis and invasion process of hepatocellular carcinoma (HCC). A variety of inflammatory cytokines and stimulus affect its expression through the nuclear factor-kappa B (NF-${\kappa}B$) signal transduction pathway. In the initial stage of inflammation, hepatocirrhosis and tumor development, ICAM-1 is expressed differently, and has varied effects on different cells to promote occurrence of malignancy and metastasis. ICAM-1 has diagnostic significance for AFP-negative or suspected HCC, and may be a prognositic significance. It is thus widely used in studies as a biomarker which reflects cancer cells metastasis as well as curative effect of drugs. Many new treatments of HCC may be based on the effects of ICAM-1 on different levels of function.

Dual roles of estrogen metabolism in mammary carcinogenesis

  • A female hormone, estrogen, is linked to breast cancer incidence. Estrogens undergo phase I and II metabolism by which they are biotransformed into genotoxic catechol estrogen metabolites and conjugate metabolites are produced for excretion or accumulation. The molecular mechanisms underlying estrogen-mediated mammary carcinogenesis remain unclear. Cell proliferation through activation of estrogen receptor (ER) by its agonist ligands and is clearly considered as one of carcinogenic mechanisms. Recent studies have proposed that reactive oxygen species generated from estrogen or estrogen metabolites are attributed to genotoxic effects and signal transduction through influencing redox sensitive transcription factors resulting in cell transformation, cell cycle, migration, and invasion of the breast cancer. Conjuguation metabolic pathway is thought to protect cells from genotoxic and cytotoxic effects by catechol estrogen metabolites. However, methoxylated catechol estrogens have been shown to induce ER-mediated signaling pathways, implying that conjugation is not a simply detoxification pathway. Dual action of catechol estrogen metabolites in mammary carcinogenesis as the ER-signaling molecules and chemical carcinogen will be discussed in this review.

  • The contamination of the environment with pollutants is one of the main problems of modern life, and the levels pollution in industrialized regions are giving raise to increased public concern. The mitogen-activated protein kinase (MAP kinase) are playa pivotal role in the regulation of important cellular functions by activation of specific signal transduction pathways from cell the surface to the nuclei. Three major subgroups of MAP kinases have been identified, and these comprise the extracellular signal-regulated kinase (ERK), the c-Jun amino-terminal kinase (JNK), and the p38 MAP kinases [1-3]. Herein, we investigated the effect of regulation of phospho-JNK (p-JNK), phospho-p38 (p-p38) and phospho-ERK (p-ERK) by TCDD. (omitted)

The Suppressing of MR Image Artifacts using Phases Cycling in Fast SE Sequence

  • The correction of image artifacts due to misadjustment in tuning of RF coils (tip angle) and in the RF single sideband spectrometer was investigated using phase cycling of the $\pi$/2 and $\pi$ pulses in spin-echo sequences. A general procedure was developed for the systematic design of phase cycles that select desirable coherence transfer pathways. To analyze a phase cycling sequence, changes in the coherence level and phase factor for each RF pulse in the spin-echo cycle must be determined. Four different phase cycling schemes (FIXED, ALTERNATE, FORWARD, REVERSED) to suppress unwanted signal components such as mirror and ghost images were evaluated using two signal acquisitions. When the receiver phase factor is cycled counter-clockwise (REVERSED), these artifacts are completely removed.
