Isolation and characterization of sigH from Corynebacterium glutamicum (Corynebacterium glutamicum의 sigH 유전자의 분리 및 기능분석)

  • Corynebacterial clones which exert regulatory effects on the expression of the glyoxylate bypass genes were isolated using a reporter plasmid carrying the enteric lacZ fused to the aceB promoter of Corynebacterium glutamicum. Some clones carried common fragments as turned out by DNA mapping technique. Subcloning analysis followed by the measurement of $\beta-galactosidase$ activity in Escherichia coli identified the region responsible for the aceB-repressing activity. Sequence analysis of the DNA fragment identified two independent ORFs of ORF1 and ORF2. Among them, ORF2 was turned out to be responsible for the aceB-repressing activity. ORF1 encoded a 23,216 Da protein composed of 206 amino acids. Sequence similarity search indicated that the ORF may encode a ECF-type $\sigma$ factor and designated sigH. To identify the function of sigH, C. glutamicum sigH mutant was constructed by gene disruption technique and the sigH mutant showed growth retardation as compared to the wild type strain. In addition, the mutant strain showed sensitivity to oxidative-stress generating agent plumbagin. This result imply that sigH is probably involved in the stress response occurring during normal cell growth.

  • This study aims to find out whether political parties apply financial reports under PSAK (Statement of Financial Accounting Standards) No. 45. The Stewardship Theory provides the analytical framework. The objects in this study were 16 Yogyakarta City Political Parties in the Special Region of Yogyakarta, with the total number of 64 respondents. The application for processing data in this study used SPSS 22 version. Based on the results of analyzing data, H1, which had a sig (2-tailed) value of 0.026 <0.05, then H0, stating that there was an influence of the statement of financial position on the implementation of PSAK, was rejected. H2 had a sig (2-tailed) value of 0.116> 0.05, so that H0, stating that there was no significant effect of the activity report on the implementation of PSAK, was accepted. H3 had a sig (2-tailed) value of 0.027 <0.05, meaning that it rejected H0, which stated that there was a significant effect of the cash flow statement on the implementation of PSAK. Furthermore, H4 had a sig (2-tailed) value of 0.744> 0.05, indicating that H0 was accepted, stating that there was a significant effect of notes to the financial statements on the implementation of PSAK.

Transcriptional Features of the Chromosomal Region Located between the sigH and rplA Genes of Bacillus subtilis

  • In this study. the transcriptional features of a 2.8 kb region spanning the sigH and rplA genes of Bacillus subtilis were clarified using synthetic oligonucleotides complementary to the transcripts of the rpmG, secE, nusG, and rplK genes. The 5' ends of three transcripts corresponding to this region were located and mapped on the chromosome via primer extension analysis. Three regions, designated Prg, Pn, and Prk, which partially share the consensus sequence recognized by ${\sigma}^A$ RNA polymerase, were theorized to function as promoter elements. The rpmG and secE genes of B. subtilis were cotranscribed from the designated prg promoter, whereas the nusG and rplK genes were transcribed separately from the Pn and Prk promoters, respectively. Accordingly, the transcriptional features, as well as the gene organization, of the region encompassing the sigH and rplA genes of B. subtilis, including the rpmG-secE-nusG-rplK genes, were determined to be distinct from those of Escherichia coli. Divergences in terms of gene organization and transcriptional features within the relevant region would serve as excellent criteria for the delineation of phylogenetic relationships among bacteria.

Influence of hydraulic retention time on biogas production during leachate treatment

  • The main objective of this study is to investigate the influence of hydraulic retention time (HRT) on biogas production during leachate treatment using an anaerobic reactor type Upflow Anaerobic Sludge Blanket. For this purpose, four HRTs ranging from 12 to 48 h were experienced. The obtained results showed that higher amount of biogas could be produced during longer HRTs. However, HRTs longer than 48 h could not affect clearly the biogas generation and considered as unnecessary given the small additional amount of biogas produced during the degradation process. A volume of $0.434L/L_{leachate}/d$ was achieved during the treatment with a HRT of 48 h. The higher biogas production, the smaller chemical oxygen demand (COD) values achieved. Besides, COD removal and biogas production positively correlate, showing that the active biomass has degraded effectively the organic load to produce biogas. Moreover, all the analyzed physicochemical parameters have experienced a decrease after 48 h except for the pH, which increased to approximately neutral value. More precisely, a decrease of 93.8%, 89.7%, 95%, 70%, 77%, and 84.4% was recorded for COD, electrical conductivity, total suspended solid, turbidity, $NH_4{^+}-N$, and $NO_3{^-}-N$, respectively.

Implementation of the robust $H^{\infty}$ speed controller by auto-tuning of the weighting function (하중함수의 오토 튜닝에 의한 강인한 $H^{\infty}$ 속도제어기의 구현)

  • In this paper, we are applied the Genetic Algorithm(GA) to design of the robust $H^{\infty}$ speed controller by auto-tuning of the weighting function. GA is used to design of the weighting functions in the robust $H^{\infty}$ controller. To evaluate the performances of the proposed robust $H^{\infty}$ controller, we make an experiment on $H^{\infty}$ speed controller of an actual DC servo- motor system with nonlinear characteristics. Experimental results show that proposed controller have better performance than those of PD controller.
