할미꽃의 정단 분열조직배양에 의한 기내 효율적 미세번식에 미치는 식물생장조절물질의 효과를 조사하기 위해 2,4-D, NAA, kinetin, TDZ와 BA를 혼용처리한 MS배지에 정단분열조직을 배양하였다. 배지내 2,4-D와 kinetin, 2,4-D와 TDZ, NAA와 TDZ 혼용처리는 캘러스 유도에 효과적이지 못하였으나 NAA와 BA가 혼용 처리되었을 때 배발생적 또는 기관분화성 캘러스가 유도되었다. 특히 0.5mg/L NAA와 1.0mg/L BA 혼용처리된 MS 배지에서는 캘러스로부터 고빈도로(62%) 신초가 형성되어 캘러스 유도 및 신초형성에 적합하였다. 적합배지에 10% coconut water를 첨가하여 30일마다 계대배양 하므로 캘러스 증식률, 신초 분화율 및 신초 신장이 현저히 증가되었는데, 그 결과 절편체 당 평균 12.9개의 shoot bud를 분리할 수 있었다. 할미꽃 기내 유도된 식물체에서 뿌리는 쉽게 분화되지 못하였으나 9.0mg/L NAA 단용 또는 0.5mg/L NAA와 1.0mg/L BA 혼용처리된 MS배지에서 캘러스를 통해서만 분화되었다. 이들 뿌리는 동일 배지에서 캘러스유도 및 식물체 분화를 반복적으로 실시하는 시원체가 되기도 하였다.
A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.
Cleaning up the landfill soil by phytoremediation in association with biomass production and utilization of biosolid as a soil amendment will be an attractive green technology. In order to examine this integrated green technology, in the current study of pot trial, heavy metal removal rate and biomass production were determined following cultivation of three different plant species in the landfill soil incorporated with biosolid at two different levels (25 ton $ha^{-1}$ and 50 ton $ha^{-1}$). Among the three plant species including Indian mustard (Brassica juncea), giant sunflower (Helianthus giganteus. L), and giant cane (Arundo donax. L), sunflower appeared to produce the largest biomass yield (19.2 ton $ha^{-1}$) and the produced amounts were magnificently increased with biosolid treatment compared to the control (no biosoild treatment). The increased production associated with biosolid treatment was common for other plant species and this was attributed to the biosolid originated nutrients as well as the improved soil physical properties due to the organic matter from biosolid. The elevated heavy metals in soil which was originated from the incorporated biosolid were Cu and Zn. Based on the phytoavailable amount of heavy metals from biosolid, the removed amount by plant shoots were 95% and 165% for Cu and Zn, respectively, when sunflower was grown. This indicated that mitigation of heavy metal accumulation in soils achieved by the removal of metal through sunflower cultivation enables the successive treatment of biosolid to soils. Moreover, sunflower showed heavy metal stabilization ability in the rhizosphere resulting in alleviation of metal release to ground water.
The objective of this research was to produce sod by box seeding for zoysiagrass or by vegetative propagation for zoysiagrass and manilagrass.1 Various ratio of peatmoss to sand(v /v) were prepared to find idea[ medium for fast and light weight sod production. Then, the days required for sod formation, the effect of growth regulators on the growth of turfgrass, and the various storage methods for winter keeping of sods were also investigated. 1.The mixed medium of sand and peatmoss(v /v, 1 : 2) showed more biomass production than that of sand. 2.In comparison of seeding rate of zoysiagrass, the amount of log /$m^2$ was most effective in the fast and dense sod formation. The amount of 20g /$m^2$ also showed fast sod formation. But, it resulted in weak plant and less tillering. During April to June, about 100 days were required to form sod with seeding rate of 5g /$m^2$ regardless of seeding time. Whereas 80 days were required to form sod in the rate log /$m^2$, which was 20 days shorter than that of 5g /$m^2$. 3.More than 85% of shoots in sod stored in field or plastic house during the winter time resumed the growth in good appearance after transplanting. The whole covering of ground with sod resulted in less weeds and faster formation of lawn. 4.Vegetative propagation of manilagrass showed about 7 to 15 days faster formation of sod than that of zoysiagrass. Application of GA increased shoot growth and BA increased the total number of tillering. However, the effects of the combined application of GA and BA were negligable.
아열대 채소인 모로헤이야의 남부지역 무가온 하우스 재배시 신초 다수확을 위한 적정 파종시기와 적심높이를 설정하기 위해 본 시험을 수행하였다. 모로헤이야 종자는 $10^{\circ}C$에서는 전혀 발아되지 않았으나 $18{\sim}30^{\circ}C$에서는 95.5~98.5%가 발아되었으며, $15^{\circ}C$에서는 75.0%의 발아율을 보였으나 발아세가 아주 낮았다. 또한, 4월 28일 이전 파종시 8월 하순 이후 꼬투리가 성숙되었으나, 5월 7일 이후 파종시에는 꼬투리 성숙이 이루어지지 않았다. 모로헤이야 신초의 수확량과 수확횟수는 4월 14일 파종한 경우에 가장 많았고, 상품수량도 다른 시기에 비해 14~42% 증가하였다. 한편, 지제부로부터 100cm 높이에서 적심한 후 연속 수확한 처리에서 분지수가 가장 많이 증가하였으며, 이에 따라 신초수량도 다른 높이의 적심처리에 비해 많았다. 따라서 모로헤이야를 남부지역에서 무가온 하우스 재배시 4월 중순에 파종하고 100cm 높이에서 적심하는 것이 지속적인 신초수확에 유리한 것으로 나타났다.

Because seagrass production significantly contributes to the biodiversity and production of coastal and estuarine ecosystems, accurate estimation of seagrass productivity is a critical step toward understanding the ecological roles of seagrass in these ecosystems. To develop an accurate and effective method of measuring seagrass productivity, we estimated leaf productivity of eelgrass (Zostera marina) on the southern coast of Korea using three methods, the conventional leaf marking method, the elongation-mass method (Short '87 method), and the plastochrone method. In each season, shoots were pierced through the bundle sheath using a hypodermic needle and were collected after 2-4 weeks had elapsed to estimate their productivity. The leaf elongation and the leaf plastochrone intervals varied significantly among seasons. On an annual basis, the conventional leaf marking method showed the lowest leaf productivity estimates compared to the elongation-mass method and the plastochrone method, suggesting that the conventional leaf marking method underestimated leaf productivity as it ignored leaf maturation processes and new leaf growth within the sheath. Since the elongation-mass method considered leaf maturation processes, this method produced higher leaf productivity estimates than the conventional leaf marking method. On an annual basis, the plastochrone method produced the highest leaf productivity estimates. Below-ground productivity, which can be easily estimated using the plastochrone method, ranged between 3.29 and 5.73 (mg dry weight $shoot^{-1}\;day^{-1}$) and accounted for about 17.8% to 30.3% of total productivity. Because of the high contributions of below-ground productivity to total seagrass production, we suggest that the plastochrone method is an effective and simple technique for assessing both above- and below-ground productivities.
다중 공간에서의 촬영을 필요로 하는 영상콘텐츠 제작은 많은 인력과 장비를 요하는 작업이다. 특히 소규모 방송사들의 입장에서는 감당하기 힘든 작업이지만, 다양한 형태의 영상콘텐츠를 얻기 위해서는 필요한 작업이다. 본 논문에서는 다중 공간 촬영 시 요구되는 인력과 장비의 절감을 위해, IP 카메라 기반의 실시간 웹캐스팅 시스템을 이용한 다중 공간 동시 촬영과 그를 바탕으로 한 1인 영상콘텐츠 제작 방법이 제안된다. 제안된 방법은 다중 공간에 각각 설치된 IP 카메라가 담는 화면을 수집하여 그들을 인터넷으로 송출하고, 인터넷을 통해 들어온 그들을 동시 수신하며, 화면의 저장과 영상 녹화 및 편집 기능들을 기반으로 하고 있다. 본 제작 방법을 사용함으로써 과거에 어려웠던 1-인/다중-카메라 원격 제어가 가능하게 됨에 따라, 기존의 1-인/1-카메라 사용의 한계에서 벗어나 다중 공간 동시 촬영이 가능하게 되어, 다양한 영상콘텐츠를 적은 인력과 시간을 확보할 수 있게 되었다. 따라서 소규모 웹캐스팅사의 경우, 짧은 시간에 다중 공간에서 촬영된 영상을 기반으로 한 영상콘텐츠 1인 제작이 가능해지고 적은 비용으로 고급 영상콘텐츠의 제작을 기대할 수 있다.
Acetolactate synthase (ALS, EC 4.1.3.18) is the first common enzyme in the biosynthesis of leucine, isoleucine, and valine. It is the target enzyme for several classes of herbicides, including the sulfonylureas, the imidazolinones, the mazolopyrimidines, the pyrimidyl-oxy-benzoates, the pyrimidyl-thio-benzens, and the 4,6-dimethoxypyrimidines. An amino-terminal fragment of the sulfonylurea-resistant ALS gene (SurB) from Nicotiana tabaccum was cloned into the bacterial expression vector pGEX-2T. The resulting recombinant plasmid pGEX-ALS1 was used to transform Escherichia coli strain BL21, and the tobacco ALS was expressed in the bacteria as a protein fused with glutathione S-transferase (GST). Polyclonal antibodies against the fusion product (GST-ALS) were produced, and the sensitivity of GST-ALS with the rabbit anti-GST-ALS IgG was up to 50 ng. This antibody was used for Western blot analysis of the partially purified ALS from barley shoots. The results suggest that the polyclonal antibody produced in this study can be used to detect plant ALS.
Allium hookeri L. (Alliaceae family) is an important ethnomedicinal plant native to the Himalayan region of Asia. The aim of this research was to establish a high-frequency plant regeneration system for in vitro propagation of A. hookeri. Among the tissue types examined, receptacle explants derived from immature flower buds showed the highest regeneration rate of shoots ($93.33{\pm}4.63%$), roots ($76.67{\pm}7.85%$), and calli ($80.00{\pm}7.43%$) when cultured on Gamborg B5 (B5) medium containing $10{\mu}M$ 6-benzylaminopurine (BA) + $1{\mu}M$ naphthalene acetic acid (NAA), $0.5{\mu}M$ BA + $5{\mu}M$ NAA, and $1{\mu}M$ BA + $10{\mu}M$ NAA, respectively. Shoot multiplication was superior when cultured in liquid rather than on solid medium and relatively high concentrations of BA, ranging from 5 to $10{\mu}M$. Efficient bulblet formation following root induction from shoot clumps was achieved with culture in liquid B5 medium containing 7% (w/v) sucrose. Regenerated bulblets were successfully acclimatized to ex vitro conditions with a greater than 95% survival rate. By this method, a maximum of 62 plantlets per receptacle could be propagated within 9 weeks of initial culture. The in vitro propagation system established in this study will promote A. hookeri biotechnology, including large-scale production of healthy and aseptic clones, preserving parental genotypes with desirable traits, and genetic manipulation to enhance medicinal value.
Ginger is an important monocotyledonous plant belonging to the family Zingiberaceae. The objective of this study was to investigate the regeneration potential of ginger using leaf base explants. Auxins such as 2, 4-D and NAA in combination with BA were used for initiation of callus. Different combinations of both ammonium ($NH^{4+}$) and nitrate ($NO^{3-}$) were also studied for efficient callus production. High frequency of white friable calli was observed on modified Murashige and Skoog (MS) medium supplemented with 2.0 mg/L 2, 4-D, 0.5 mg/L NAA and 0.5 mg/L BA. The highest shoot induction (92.33%), shootlets number ($7.33{\pm}0.33$) and length ($88.33{\pm}4.40$) mm were achieved on MS media containing 0.5 mg/L BA. Regenerated shoots were transferred to in vitro rooting media containing 1.0 mg/L IBA. Afterwards, plantlets with well-developed root and shoot system were subjected to a twostep hardening process. 71% of plantlets survived after secondary hardening without any abnormal morphology.
