Effects of Phytohormone and Activated Carbon on the Growth and Rooting of Teratoma Shoot Induced from Crown Gall Callus in Nicotiana tabacum cv. NC 2326 (연초 Crown Gall Callus유래 Teratoma Shoot의 생육과 발근에 미치는 식물호르몬과 활성탄의 영향)

  • These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

  • This experiment was carried out to find the proper way of shoot induction and elongation of shoots in Rehmannia glutinosa. Shoot induction and multiplication rate from stem explants were significantly improved in the induction media with 1.0mg/L thidiazuron compared to BA and NAA combinations. Shoot elongation was better in the media with thidiazuron 1.0mg/L alone than in the presence of paclobutrazol.

  • The present study was conducted to obtain some basic information on the shoot formation from crown gall callus and the characteristics of teratoma shoot derived from crown gall induced by inoculation of Agrobacterium tumefaciens C58. Crown gall callus could be continuously cultured on the phyohormone free basic medium. The growth of crown gall callus was inhibited when BA were added to the cultural media. Shoot formation from crown gall callus fail to be initiated except teratoma shoot which induced on the phytohormone free medium after several subculture on rare occasions. Teratoma shoot could not form root and grow as normal shoot. Addition of BA to cultural media was not effective for shoot elongation, reduction in multiple shoot formation, but IBA was somewhat effective for shoot elongation of teratoma shoot, never for root formation.

Effects of Restricted Oxygen, Nitric oxide, and Mercuric Chloride on the Seed Germination and Early Elongation Growth of Rice

  • Germination and early elongation of rice after germination were investigated in anoxic air treatment, nitric oxide gas treatment, and six concentrations of mercuric chloride solutions to determine the effects of limited oxygen environment, nitric oxide, and inhibited water flux through cell membrane in $17^{\circ}C$. Anoxic air treatment affected germination of tested six varieties very little. However root elongation rates were severely inhibited while shoot growth was affected less. Reductions in shoot and root elongations demonstrated genotypic variations. Nitric oxide delayed the germination of rice even though it didn't affect the final percent germination. Elongations of root and shoot were inhibited in nitric oxide treatment. The inhibitor effect of nitric oxide on the shoot elongation of rice was less severe, while nitric oxide completely inhibited the root emergence of rice. Concentrations of $HgCl_2$ greater than $300{\mu}M$ dramatically reduced the rate and percentage of germination when compared to distilled water treatment. The reduced percent germination showed the greatest variation among rice varieties in $500{\mu}M$ solution of mercuric chloride. Ansanbyeo, Jinheung, and Odaebyeo were affected less by $HgCl_2$, Nonganbyeo and Sangmibyeo were intermediate, and the germination of Andabyeo was greatly reduced by $HgCl_2$. Root elongation of germinated rice seedlings was more sensitive to oxygen deficits, nitric oxide, and $HgCl_2$ treatments than germination and shoot elongation. In conclusion, poor seedling establishment of rice sown in flooded paddy soils, in which the oxygen supply to the seeds is restricted, appears to the result of limited root elongation rate.

Effect of Carbohydrates on in vitro Shoot Growth of Various Prunus Species

  • Carbohydrate sources are one of important factors associated with macro- and micro nutrients and phytohormones in vitro culture medium for shoot growth. The optimal carbohydrates for eight species of the genus Prunus which are economically important fruit crop was evaluated at the initiation and elongation stages. All carbohydrate seemed utilized for the bud break and leaf growth at the early stage of culture. However, shoot elongation and fresh weight of species tested were superior in the medium containing 90 mM of fructose or glucose rather than sucrose. There was no difference between glucose and fructose. Adventitious shoots from the axillary buds were induced in most species but no significant differences were observed except for two species (P. salicina ‘Shiro’ and P. tomentosa). These result demonstrated that glucose and fructose were suitable carbohydrate sources for diverse Prunus species than sucrose, although the response to the carbohydrates in the medium were slightly different in the species.

Promotion of in vitro growth and rooting of micropropagated shoots in Spathiphyllum floribundum by the addition of liquid medium (액체배지 첨가에 의한 Spathiphyllum floribundum 신초의 기내생육 및 발근 촉진)

  • This experiments were carrid out to examine the effects of liquid medium addition in same vessels on shoot elongation and rooting, and soil survival of plantlets after the shoot cluster sections of Spathiphyllum floribundum 'Cupid' were pre-cultured. The shoot clusters with 3 to 4 small shoots were proliferated on LS medium supplemented with 2.0 mg/L BA for 8 weeks, and then 15 mL of various kinds of liquid medium was added in the same vessels. The addition of 15 mL liquid medium containing l/2 MS macro elements, 50 g/L sucrose and 5.0∼10.0 g/L activated charcoal was significantly stimulated the elongation and rooting of proliferated shoots. The medium addition was resulted in the enhanced soil survival of plantlets.

Effects of Medium Compositions and Plant Growth Regulators on in vitro Organogenesis in Cultured Explants of Platycodon grandiflorum Species

  • Platycodon grandiflorum (Bell flower) is an important plant that has traditionally been used as herbal medicine for the treatment of cough, phlegm, sore throats, lung abscesses, chest pains, dysuria, and dysentery. The present study was initiated to investigate the feasibility of inducing shoot and root organogenesis in cultured explants of P. grandiflorum in a range of culture media and through use of various plant growth regulators (PGRs). The plantlets (Stem containing one node) were isolated and cultured on different concentrations of Murashige and Skoog (MS) medium supplemented with PGRs. We found that proliferation and elongation of shoots and roots could be achieved on 1/4 MS for P. grandiflorum with wild and green petals and on 1/8 MS for P. grandiflorum with double petals. The highest levels of development and elongation of adventitious shoots and roots were observed when petal explants were cultured on 1/4 MS (pH 3.8) supplemented with 5% sucrose. Increasing the agar concentration reduced shoot growth and rooting potential; nevertheless, the highest number of shoots and roots was observed on 0.6% agar. In the case of growth regulators, 1/4 MS supplemented with $1mg\;L^{-1}$ 6-benzylaminopurine (BA) was found to be best for shooting, although higher concentrations of BA tended to reduce shoot and root elongation. The highest number of shoots was achieved on $0.5mg{\cdot}L^{-1}$ thidiazuron (TDZ) from double petal explants grown on 1/8 MS. However, root and shoot elongation were found to decrease when TDZ concentrations were increased. Low concentrations of kinetin, naphthalene acetic acid, indole acetic acid, and 3-indole butyric acid induced shoot and root proliferation and elongation. Taken together, our study showed that low concentrations of PGRs induced the greatest root formation and elongation, showing that the optimal concentration of PGRs for shoot proliferation was species-dependent.

  • The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

Factors influencing Adventitious Shoot induction from Leaves of Populus deltoides Bartr (미류나무 잎절편체 조직배양을 통한 식물체 재분화에 미치는 요인)

  • We have investigated several factors influencing adventitious shoot induction from the Populus deltodies bartr. leaf segments. To obtain in vitro materials, at first, stem segments from ex vivro were tested for axillary bud breaking on the five macronutrients levels of two differenr media. WPM was better both in bud breaking and leaf expansion than MS medium. The amount of NH$_{4}$NO$_{3}$ added in the medium did not seem to affect axillary bud breaking, significantly and subsequent shoot elongation from the stem segments of P. deltodies. However, other components in WPM might have played important roles in axillary bud breaking and shoot elongation. Regenerability from three sections (the distal, the middle and the base) of leaves cultured on WPM supplemented with TDZ and NAA combinations appeared to be different; middle and basal sections of leaves produced more organogenic sites than those of top section on WPM and those sesctions produced the highest ogranogenic sites on the same medium supplemented with 0.01 mg/l TDZ and 0.02mg.l NAA. Among 3 carbon sources tested for adventitious shoot elongation, fructose seemed to be stimulating the elongation of adventitious shoots. Sucrose and glucose added in the medium resulted in the necrosis which caused dying of adventitious shoots, eventually.

Growth regulation and Nitrogen Uptake Inhibition of Zoysia matrella Applying Prohexadione Calcium (Prohexadione-calcium 처리에 따른 금잔디의 생육 조절 및 질소 흡수 억제)

  • BACKGROUND: Prohexadione-calcium (PC) is a plant growth regulator of cyclohexanedione family acting on inhibiting gibberellin biosynthesis and has been used to regulate shoot elongation of turfgrass in the golf course during the summer season. This study was conducted to investigate the effects of PC on the inhibition of shoot elongation, growth, and nitrogen uptake in Zoysia matrella. METHODS AND RESULTS: Treatments were as follows; non-treatment, control (TE 0.01 a.i. g/m2/100 mL), 0.0025PC (PC 0.0025 a.i. g/m2/100 mL), 0.005PC (PC 0.005 a.i. g/m2/100 mL), 0.01PC (PC 0.01 a.i. g/m2/100 mL) and 0.02PC (PC 0.02 a.i. g/m2/100 mL). Visual quality and chlorophyll content were not significantly different. As compared to non-treatment, shoot length and clipping yield in 0.005PC, 0.01PC and 0.02PC treatments were decreased by 29-36% and 65-82%, respectively, and those of 0.0025PC were not significantly different. N uptake of Z. matrella after applying PC was decreased by 28.9-77.8% than that of non-treatment. Inhibition effects of PC treatment in Z. matrella were not significantly different from those of control. CONCLUSION(S): These results indicated that the application of prohexadione calcium inhibited shoot elongation, clipping yield and N uptake amount in Z. matrella, and trinexapac-ethyl could be replaced with prohexadione-calcium in turfgrass management.
