Micropropagation by Apical Meristem Culture of Wasabia japonica Matsum (고추냉이의 頂端分裂組織培養에 의한 微細增殖)

  • Apical meristems of Wasabia japonica were cultured on Murashige and Skoog's medium supplemented with cytokinins alone or together with 1.0 mg/L IAA. Shoot initials could be induced from leaf primordia on apical meristems. Calli and roots were formed on the medium containing cytokinins and 1.0 mg/L IAA in combination after 30 days of culture, but there were no callus proliferation. Shoot organogenesis began after 60 days of culture and these small shoots elongated when transferred to a medium containing 1.0 mg/L BA or kinetin. Shoots were formed directly without callus induction from apical meristems all the explants on the medium containing cytokinins variously, and most of the shoots proliferated multiple shoots which could be divided to obtain plantlets. Shoot multiplication rate in response to cytokinins was best on the medium containing 1.0 mg/L BA or 2.0 mg/L zeatin. Divided plantlets rooted well on MS medium containing 0.01 mg/L IBA after 15~30 days of subculture and the rooted plantlets developed into whole plants with multiple shoots. After rooting, the regenerated plants were washed and transferred to the pots containing sterilized soil.

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants.   • The effects of explant sources, plant growth regulators on callus induction and plantlet differentiation from leaf blade, petiole, and meristem tissue of Pelalgonium citrosa were investigated under illumination or in dark condition Leaf blade explants cultured on Murashige and Skoog's medium containing 2,4-D and kinetin did not form callus or organ. But those cultured on medium with NAA and BA produced callcus and shoots. Dark condition was more effective than light condition to callus induction and showed that some of shoot were differentiated directly from leaf blade explane. Callus proliferated vigorously on meristem tissue after 7 days of culture, and multiple shoots were obtained Sum callus on medium with 0.5 mg/L NAA and BA. Roots formed readily from about 80% of the shoots cultured on medium with 1.0 mg/L NAA. Regenerated plantlets regenerated had phenotypically normal leaves and roots.

In vitro Shoot Propagation Derived from Stem and Shoot Tip in Hovenia dulcis var. koreana Nakai by Plnat Growth Regulators and Light Resources (식물생장조절제 및 광원처리에 따른 헛개나무 줄기와 경정유래 신초의 기내증식)

  • This study was conducted to examine effects of plant growth regulators and light resources on the formation of multiple shoot and plant regeneration of Hovenia dulcis var. koreana Nakai. Stem and shoot tip were cultured on MS medium or WPM supplemented with various plant growth regulators. At the single treatment, the highest shoot formation was obtained when stem explants were cultured on WPM supplemented with kinetin $1.0mg{\cdot}L^{-1}$. MS medium containing NAA 0.1 and TDZ $0.1mg{\cdot}L^{-1}$ gave the best results for shoot induction rate and shoot growth in combination treatments. Of the BAP and kinetin tested, BAP $0.5mg{\cdot}L^{-1}$ on WPM was found to be more effective for shoot growth from shoot tip. Under white fluorescent light treatment, shoot growth was much higher than blue, red LED treatments. Root induction from in vitro growth of plantlet was the best on WPM supplemented with $1.0mg{\cdot}L^{-1}$ IBA. The results suggest that selection of plant growth regulators and light resources could be important factor to achieve an efficient in vitro growth.

  • Spathiphyllum floribundum cv. Cupid was micropropagated on LS medium containing various growth regulators. Shoot tips were cultured on media with different concentrations of thidiazuron (TDZ) or banzyladenine (BA). The medium containing 2.0 mg/L BA was most effective on the shoot multiplication and the formation of adventitious multi-shoot cluster from shoot tips among the different concentrations of TDZ and BA. The combined treatments of BA and paclobutrazol (PBZ) were stimulated the shoot multiplication and the formation of adventitious mu]ti-shoot clusters from shoot tips compared to the ones of BA alone. The adventitious multi-shoot clusters were culled with 5∼7 mm in size and cultured on media with BA or thidiazuron (TDZ). Shoot multiplication and shoot cluster proliferation from the shoot clusters were very favorable on media supplemented with 2.0∼3.0 mg/L BA. The shoots grew and rooted out vigorously on media with 2.0∼3.0 mg/L IBA. The rooted plantlets were acclimatized in the soil mixed with a rate of vermiculite 1 and perlite 1, and survived more than 95% in greenhouse.

Mechanization of Pine Cone Harvest(II) -Shearing Characteristics of Shoots of Korean Pine Trees- (잣 수확의 기계화 연구(II) -잣나무 가지의 전단 특성-)

  • This reasearch was performed to provide the fundamental intonation for the mechanization of Korean pine cone harvest when the shoot shearing method is adopted. Shear force and stress of pine cone shoots were measured and analyzed for this purpose. Samples are selected along their harvesting time and tested in 17 levels of shoot diameter from 10 to 26mm with 1mm increment. 1) Shear force-deformation characteristics showed that shoot reached its rupture point after 2 to 4 of bio-yield points. It was supposed that these multiple bio-yield points were caused by the discrete compression of wood parts which are composed of water, nutrient, resin, etc. 2) Required shear force to shear shoot was proportional to the square of shoot diamter, however, shear force for shoots of early harvesting time(Aug. 31) was proportional to the shoot diameter. Variance of shear force was increased as the harvesting time was delayed. Shear forces were distributed from 468N(Aug. 31, 12mm dia) to 4153N(Aug. 31, 26mm dia) disregarding the sampling date. 3) The average shear stresses by sampling dates were 744,822, and 883N/m2, respectively, and for the earlier shoot samples shear stress was quite smaller than the others. Shear stress was proportional to shoot diameter squared, and the effect of shoot diameter on the shear stress was decreased as harvesting time was delayed.

  • This experiment was to study the relationship between shoot and root system in perennial ryegrass at different stages of growth. The results are summarized as follows; 1. The dry weight of shoot was significantly correlated with the dry weight of root at both stages of growth. 2. The yield components of shoot and root systems were changed by the stages of growth. Thus, the dry weights of shoot and root were positive correlated with the number of tillers and roots at 60 days after sowing. But, the dry weights of shoot and root were positive correlated with the dry weight of root, the dry weight of tiller, length of plant and length of root at 90 days after sowing. 3. Maprirna variety was obtained a higher dry weights of shoot and root at 60 days after sowing, but, Alta variety was obtained at 90 days after sowing, respectively. 4. The dry weight of shoot was positive correlated with leaf area at both stages of growth. 5. The dry weight of shoot was significantly negative correlated with specific leaf weight at 60 days after sowing, but positive correlated with specific leaf weight at 90 days afikr sowing. 6. The dry weight of root was smcantly positive correlated with leaf area at 60 days after sowing, but not recognized at 90 days after sowing.

  • E3 ligase BRUTUS (BTS), a putative iron sensor, is expressed in both root and shoot tissues in seedlings of Arabidopsis thaliana. The role of BTS in root tissues has been well established. However, its role in shoot tissues has been scarcely studied. Comparative transcriptome analysis with shoot and root tissues revealed that BTS is involved in regulating energy metabolism by modulating expression of mitochondrial and chloroplast genes in shoot tissues. Moreover, in shoot tissues of bts-1 plants, levels of ADP and ATP and the ratio of ADP/ATP were greatly increased with a concomitant decrease in levels of soluble sugar and starch. The decreased starch level in bts-1 shoot tissues was restored to the level of shoot tissues of wild-type plants upon vanadate treatment. Through this study, we expand the role of BTS to regulation of energy metabolism in the shoot in addition to its role of iron deficiency response in roots.

Factors for high frequency plant regeneration in tissue cultures of Indian mustard (Brassica juncea L.)

  • An efficient system for high frequency plant regeneration was established through investigating various factors such as plant growth regulator combinations, explant types and ages, and addition of $AgNO_3$ influenced on shoot regeneration in Brassica juncea L. cv. BARI sarisha-10. Murashige and Skoog (MS) medium supplemented with 0.1 mg/L NAA (1-naphthaleneacetic acid) and 1 mg/L BA (6-benzyladenine) showed the maximum shoot regeneration frequency (56.67%) among the different combinations of NAA and BA. Explant type, explant age, and addition of $AgNO_3$ also significantly affected shoot regeneration. Of the four type of explants (cotyledon, hypocotyl, root, and leaf explants)- cotyledon explants produced the highest shoot regeneration frequency and hypocotyls explants produced the highest number of shoots per explant, whereas root explants did not produce any shoot. The cotyledonary explants from Four-day-old seedlings showed the maximum shoot regeneration frequency and number of shoots per explant. Shoot regeneration frequency increased significantly by adding $AgNO_3$ to the medium. Two mg/L $AgNO_3$ appeared to be the best for shoot regeneration with the highest shoot regeneration frequency (86.67%) and number of shoots per explant (7.5 shoots). Considerable variation in shoot regeneration from cotyledonay explants was observed within the B. juncea L. genotypes. The shoot regeneration frequency ranged from 47.78% for cv. Shambol to 91.11% for cv. Rai-5. In terms of the number of shoots produced per explant, B. juncea L. cv. Daulot showed the maximum efficiency. MS medium supplemented with 0.1 mg/L NAA showed the highest frequency of rooting. The regenerated plantlets were transferred to pot soil and grown to maturity in the greenhouse. All plants were fertile and morphologically identical with the source plants.

Correlation between In vitro Plant Regeneration and Polarity with Boxthorn (Lycium chinense Mill.) Seedlings (구기자나무 유식물체를 이용한 식물체의 재분화와 극성과의 관계)

  • This experiment was conducted out to investigate the effect of plant growth regulators on callus and shoot formation. The callus formation was effective on 1/2 MS medium containing 2,4-D, while shoot formation was suppressed. Shoot formation and differentiation were the highest in combination 0.1 mg/L of IAA and 0.1 mg/L of BA. The polarity of explants was investigated from cotyledon, which excised 20% of each basal and terminal parts. Formation of shoot was induced from excised ends of the basal part. In excised ends of the basal part, callus was induced vigorously and shoots were produced lately. Root induction was easily achieved in 1/3 MS medium from the adventitious shoot and more than 90% of regenerated plantlets acclimatized successfully and flowered normally.
