Spatial and Temporal Occurrence of Edwardsiella tarda at Flounder Farms in Jeju (제주도 넙치 양어장의 Edwardsiella tarda균의 분포에 관한 연구)

  • To find the appearance period and distribution of Edwardsiella tarda which causes severe damage to flounder (Paralichthys olivaceus) farms in Jeju-do, the rearing seawater (inflow, rearing water, outflow) and internal organs of flounders from 5 flounder farms were examined from June, 1997 to May, 1998. The number of bacteria in seawater was counted by plating the seawater on DSSS (Double Strength Salmonella-Shigella) agar plates or by plating after cultivation of bacteria. Bacteria in internal organs were counted by plating series of dilution of homogenized organ. The results are summarized as follows. E. tarda was detected in inflow seawater of five flounder farms in July, September and November, 1997 and February, March, April of 1998. In the rearing water and outflow water, the bacterium was detected throughout the year and the number of bacteria was much higher in summer than any other seasons. A large number of E. tarda in the internal organs were detected at farm B where a track-shaped tank was used, which has the characteristics of low circulation rate and bad discharge of excrement and residuals. In contast, none of E. tarda was detected at farm A where high circulation rate and good discharge of organic materials were applied. A few number of E. tarda at farm E were detected at the same condition as the farm A. A large number of E. tarda was observed in liver and intestines among the internal organs, and the number was higher from June to September in summer.

A Study of Antimicrobial & Antiviral Effect of Natural Product (천연물을 이용한 살균 및 살바이러스 효과에 관한 연구)

  • Bactericidal effect of $Green-Zone^{(TM)}$ was observed, when Staphylococcus aureus, E. coli O157:H7, Salmonella typhimurium, S. enteritidis, Listeria monocytogenes, the causative bacteria of food poisoning, Vibrio parahaemolyticus, and Shigella sonnei were treated with the diluted solution of $Green-Zone^{(TM)}$(33.3%~4.1%) for 30min at $20^{\circ}C$. All the bacteria were killed in 30 sec, when 33.3%-diluted $Green-Zone^{(TM)}$ was applied, except for S. aureus. Coronavirus, the same virus with SARS virus taxonomically, was also lilled with the 20%-diluted $Green-Zone^{(TM)}$. Canine parvovirus and Canine distermper virus were also killed even in the organic matter and hard water when treated with $Green-Zone^{(TM)}$. When applied to food such as raw fish, chilled meat, vegetables, $Green-Zone^{(TM)}$ could also decrease the number of microorganism, expecially for E. Coli. From these results, $Green-Zone^{(TM)}$ is thought to be effective for killing virus and bacteria, and also was proved to be safe when applied directrly to food.

Analysis of Genes Involved in the Pathogenesis of Intracellularly Survival Bacteria (세포내 기생세균의 병원성 관련 유전자의 분석에 관하여)

  • Eight bacterial strains were examined whether they have phoP/phoQ genes which were known to be involved in the intracellular survival of Salmonella typhimurium. The phoP/phoQ operon were known to sense the stimuli of the genes involved in the adaptation of the environment. Using 514-basepairs EcoRV DNA fragment of phoP region of Salmonella typhimurium as a probe, dot blot hybridization were performed. Chromosomal DNAs of Klebsiella pneumoniae, Pseudomonas aeruginosa, Serratia marscescens, Enterobacter cloacae, Salmonella typhimurium, Escherichia coli, Shigella dysenteriae, and Listeria monocytogenes were examined by DNA hybridization assay. Against our expectation, intracellular pathogen, L. monocytogenes, did not have similar DNA sequences to phoP/phoQ of S. typhimurium, while E. coli, S. dysenteriae, and E. cloacae showed the positive signal even though they were not intracellular pathogens. This result suggested that the phoP/PhoQ operon was absent in intracellular pathogenic bacterias other than S. typhimurium. Rather it was found in phylogenetically closer bacterias to S. typhimurium, which were not able to survive in intracellular environment. Some different mechanism, which is not dependent on phoP/PhoQ operon, could be involved in the intracelluar survival of L. monocytogenes.

Effects of Dietary Synbiotics from Anaerobic Microflora on Growth Performance, Noxious Gas Emission and Fecal Pathogenic Bacteria Population in Weaning Pigs

  • Synbiotics is the term used for a mixture of probiotics (live microbial feed additives that beneficially affects the host animal) and prebiotics (non-digestible food ingredients that beneficially affect the organism). This study investigated the effect of probiotics from anaerobic microflora with prebiotics on growth performance, nutrient digestibility, noxious gas emission and fecal microbial population in weaning pigs. 150 pigs with an initial BW of 6.80${\pm}$0.32 kg (20 d of age) were randomly assigned to 5 dietary treatments as follows: i) US, basal diet+0.15% antibiotics (0.05% oxytetracycline 200 and 0.10% tiamulin 38 g), ii) BS, basal diet+0.2% synbiotics (probiotics from bacteria), iii) YS, basal diet+0.2% synbiotics (probiotics from yeast), iv) MS, basal diet+0.2% synbiotics (probiotics from mold), v) CS, basal diet+0.2% synbiotics (from compounds of bacteria, yeast and mold). The probiotics were contained in $10^{9}$ cfu/ml, $10^{5}$ cfu/ml and $10^{3}$ tfu/ml of bacteria, yeast and molds, respectively. The same prebiotics (mannan oligosaccharide, lactose, sodium acetate and ammonium citrate) was used for all the synbiotics. Pigs were housed individually for a 16-day experimental period. Growth performance showed no significant difference between antibiotic treatments and synbiotics-added treatments. The BS treatment showed higher (p<0.05) dry matter (DM) and nitrogen digestibility while ether extract and crude fiber digestibility were not affected by the dietary treatment. Also, the BS treatment decreased (p<0.05) fecal ammonia and amine gas emissions. Hydrogen sulfide concentration was also decreased (p<0.05) in BS, YS and MS treatments compared to other treatments. Moreover, all the synbioticsadded treatments increased fecal acetic acid concentration while the CS treatment had lower propionic acid concentration than the US treatment (p<0.05) gas emissions but decreased in fecal propionate gas emissions. Total fecal bacteria and Escherichia coli populations did not differ significantly among the treatments, while the Shigella counts were decreased (p<0.05) in synbiotics-included treatment. Fecal bacteria population was higher in the YS treatment than other treatments (p<0.05). The BS treatment had higher yeast concentration than YS, MS and CS treatments, while US treatment had higher mold concentrations than MS treatment (p<0.05). Therefore, the results of the present study suggest that synbiotics are as effective as antibiotics on growth performance, nutrient digestibility and fecal microflora composition in weaning pigs. Additionally, synbiotics from anaerobic microflora can decrease fecal noxious gas emission and synbiotics can substitute for antibiotics in weaning pigs.

Long-Term Monitoring of Noxious Bacteria for Construction of Assurance Management System of Water Resources in Natural Status of the Republic of Korea

  • Climate change is expected to affect not only availability and quality of water, the valuable resource of human life on Earth, but also ultimately public health issue. A six-year monitoring (total 20 times) of Escherichia coli O157, Salmonella enterica, Legionella pneumophila, Shigella sonnei, Campylobacter jejuni, and Vibrio cholerae was conducted at five raw water sampling sites including two lakes, Hyundo region (Geum River) and two locations near Water Intake Plants of Han River (Guui region) and Nakdong River (Moolgeum region). A total 100 samples of 40 L water were tested. Most of the targeted bacteria were found in 77% of the samples and at least one of the target bacteria was detected (65%). Among all the detected bacteria, E. coli O157 were the most prevalent with a detection frequency of 22%, while S. sonnei was the least prevalent with a detection frequency of 2%. Nearly all the bacteria (except for S. sonnei) were present in samples from Lake Soyang, Lake Juam, and the Moolgeum region in Nakdong River, while C. jejuni was detected in those from the Guui region in Han River. During the six-year sampling period, individual targeted noxious bacteria in water samples exhibited seasonal patterns in their occurrence that were different from the indicator bacteria levels in the water samples. The fact that they were detected in the five Korea's representative water environments make it necessary to establish the chemical and biological analysis for noxious bacteria and sophisticated management systems in response to climate change.

  • This study was conducted to detect and identify Staphylococcus aureus, Vibrio parahaemolyticus, and Salmonella enterica subsp. using simultaneous multiplex polymerase chain reaction (multiplex PCR) assay. 23S rRNA partial gene (S. aureus), tox R gene (V. parahaemolyticus), and inv A gene (S. enterica subsp.) as diagnostic marker gene were suggested, and their amplicon sizes were 482 bp, 368 bp, and 284 bp, respectively. Non specific amplicons by STA-5F/STA-5R primer, ToxR-F/ToxR-R primer, and 139/141 primer were not observed in genomic DNA of pathogen bacteria as Bacillus cereus, Listeria monocytogenes, Escherichia coli, Proteus vulgaris, Streptococcus pyogenes, Candida albicans, and Shigella sonnei. The extracted crude DNA of targeted bacteria was detected as PCR template successfully. The detection limits were $10^5\sim10^4$ CFU/mL and 10 pg of purified genomic DNA of S. aureus, V. parahaemolyticus, and S. enterica subsp. by using simultaneous multiplex PCR.

Pathogenesis of Inflammation in H. pylori Infection

  • 위의 parietal cell 혹은 대식세포와 유사한 세포 내부에서 H. pylori가 발견된다는 보고가 있기는 하나 일반적으로 H. pylori는 Shigella와 같은 침습성 세균은 아닌 것으로 알려져 있다. 그럼에도 불구하고 H. pylori에 감염된 위점막에는 많은 수의 호중구를 위시한 염증세포의 침윤이 관찰되는데 H. pylori가 위상피세포에 부착 할 경우 위상피세포를 자극하여 interleukin-8을 위시한 cytokine 을 발현케하고 이에 의하여 호중구 등의 염증세포가 몰려들게 된다. 한편 고유층에 몰려든 호중구에서는 다시 interleukin-8을 위시한 일련의 호중구 활성화 chemokine을 분비하여 염증반응을 증폭해 나갈 것이다. 호중구에서 발현되는 myeloperoxidase나 활성 산소 등도 위점막의 조직 손상에 기여할 것이다. 위상피세포를 덮고 있는 점액층은 위상피세포를 보호한다고 알려져 있으나 H. pylori 감염의 경우 점액층에 의하여 H. pylori의 운동성이 증가하고 이것이 위상피세포로부터의 cytokine 발현을 자극하여 염증반응을 증폭하는데 관여할 가능성도 있다. H. pylori는 위상피세포에 대하여 apoptosis를 유도함과 동시에 고유층에 몰려든 호중구에 대하여는 apoptosis를 억제케하여 궁극적으로 염증반응을 증폭 및 지속시켜 나가는 쪽으로 작용한다. 한편 H. pylori는 위상피세포로부터 COX-2의 발현을 증가시키는데 이는 위상피세포의 apoptosis를 억제하는 방향으로 작용한다. 이외에 H. pylori의 urease에 의하여 발생한 암모니아나 H. pylori 자신이 분비하는 세포독소가 세포 손상을 유발할 가능성도 있다. 상술한 여러 독성 인자들 중 어느 하나가 단독으로 작용하기보다는 여러 인자가 같이 동시에 또는 시차를 두고 작용할 가능성이 많다고 생각된다.(\gamma-FeOOH)$, 침철광$(\alpha-FeOOH)$, 적금광$(\beta-FeOOH)$, 그리고 자철광$(Fe_3O_4)$이다. 인위적 부식에서는 전부 인철광의 부식물이 생성되었고 자연적 부식에서는 모두 침철광의 부식물이 생성되었다. 특히 철제 표면에 자연적으로 생성된 공식 녹을 XRD 분석한 결과 적금광으로 동정되었다. 이런 모든 시편들을 각 탈염방법에 따라 탈염처리한 후 XRD와 SEM-EDS으로 분석한 결과 인철광과 침철광은 어떠한 변화도 보이지 않았고, 다만 적금광으로 동정된 시편만이 잔존하지 않았다. 철기 제작별 $Cl^-$ 이온 추출량과 탈염효과에 대한 비교 실험은 이온 크로마토그래피 분석 결과와 마찬가지로 단조 철제유물이 주조 철제보다 $Cl^-$ 이온을 많이 가지고 있었으며, 탈염 처리 후에는 $Cl^-$ 이온은 전혀 발견되지 않았다. 이상의 결과 $K_2CO_3$와 Sodium 용액은 탈염처리에서 가장 적합한 탈염처리 용액으로 알수가 있었으며 특히 어떠한 탈염 용액으로 유물을 처리한다 해도 철제유물에 생성된 부식물은 제거되지 않는다는 것을 알게 되었다. 따라서 보존처리자는 유물 표면의 부식 상태만을 보고 처리하기 보다는 철기제작물로 고려하여 처리하는 것이 필요하다. 또한 금속에 부식을 야기시키는 $Cl^-$ 이온과 부식물을 완전하게 제거하여 탈염처리를 하는 것이 유물 부식을 최대한 지연시킬 수 있는 것이라 생각된다.TEX>$88\%$)였다.(P=0.063). 결론: 본 연구에서는 MTHFR C/T & T/T 유전자 다형성이 위암의 발생과 그 위치에 대해 관련이 있는 것으로 여겨지고, 흡연력, 음주력과는 관련이 없는 것으로 여겨진다.험이

Probiotic Properties and Immunomodulator Evaluation of the Potential Feed Additive Pediococcus acidilactici SRCM102607 (잠재적 사료첨가제로서 Pediococcus acidilactici SRCM102607의 생균제 특성 및 면역활성 효과)

  • The purpose of this study was to investigate the probiotic characteristics and immune activities of selected lactic acid bacterial (LAB) strains as feed additives in livestock. 301 LAB strains isolated from traditional fermented foods were first assessed for their antibacterial activity potential. Of the 301 isolates, five showed antibacterial activity against five livestock pathogens (Esherichia coli KCCM11234, Listeria monocytogens KCTC3710, Salmonella Typhimurium KCTC1926, Staphylococcus aureus KCCM11593, and Shigella flexneri KCTC2517). The probiotic characteristics of the five selected strains were also investigated by antioxidative activity, hemolysis, bile salt hydrolase, acid resistance and bile tolerance. The SRCM102607 strain was found to have superior probiotic properties and was selected for further experimentation. 16S rRNA gene sequencing showed that SRCM102607 is Pediococcus acidilactici, which was labeled as P. acidilactici SRCM102607 (KCCM 12246P). The survival characteristics of P. acidilactici SRCM102607 in artificial gastrointestinal conditions were assessed under exposed acidic (pH 2.0) and bile (0.5% and 1.0%) conditions. P. acidilactici SRCM102607 was also confirmed to have resistance to various antibiotics, including amikacin, gentamicin, vancomycin, and etc. The TNF-α production by P. acidilactici SRCM102607 was 171.86±4.00 ng/ml. These results show that P. acidilactici RCM102607 has excellent potential for use as a probiotic livestock feed additive.

Intestinal Parasitic Infections and Environmental Water Contamination in a Rural Village of Northern Lao PDR

  • A field survey studying intestinal parasites in humans and microbial pathogen contamination at environment was performed in a Laotian rural village to identify potential risks for disease outbreaks. A parasitological investigation was conducted in Ban Lak Sip village, Luang Prabang, Lao PDR involving fecal samples from 305 inhabitants as well as water samples taken from 3 sites of the local stream. Water analysis indicated the presence of several enteric pathogens, i.e., Aeromonas spp., Vibrio spp., E. coli H7, E. coli O157: H7, verocytotoxin-producing E. coli (VTEC), Shigella spp., and enteric adenovirus. The level of microbial pathogens contamination was associated with human activity, with greater levels of contamination found at the downstream site compared to the site at the village and upstream, respectively. Regarding intestinal parasites, the prevalence of helminth and protozoan infections were 68.9% and 27.2%, respectively. Eight helminth taxa were identified in fecal samples, i.e., 2 tapeworm species (Taenia sp. and Hymenolepis diminuta), 1 trematode (Opisthorchis sp.), and 5 nematodes (Ascaris lumbricoides, Trichuris trichiura, Strongyloides stercoralis, trichostrongylids, and hookworms). Six species of intestinal protists were identified, i.e., Blastocystis hominis, Cyclospora spp., Endolimax nana, Entamoeba histolytica/E. dispar, Entamoeba coli, and Giardia lamblia. Questionnaires and interviews were also conducted to determine risk factors of infection. These analyses together with a prevailing infection level suggested that most of villagers were exposed to parasites in a similar degree due to limited socio-economic differences and sharing of similar practices. Limited access to effective public health facilities is also a significant contributing factor.

Change of Microbiological Quality according to Various Storage Conditions in the Drinking Process of Bottled Mineral Water (먹는 샘물의 개봉 후 음용과정에서의 보관 조건에 따른 미생물학적 수질 변화)

  • Objective: This study was conducted to investigate changes in microbiological quality according to various storage conditions in the drinking process of bottled mineral water. Methods: Heterotrophic plate counts ($21^{\circ}C$ and $36^{\circ}C$) and pathogenic indicators (total coliforms, fecal Streptococcus, Pseudomonas aeruginosa, Clostridium perfringens, Salmonella, and Shigella) were analyzed in commercial bottled mineral water stored under different conditions ($4^{\circ}C$, $20-25^{\circ}C$, $36^{\circ}C$) after injecting saliva. The heterotrophic plate counts were analyzed twice per day for the first week and once per day for the three weeks after. Pathogenic indicators were analyzed at the beginning and end (initial and final). Results: The results of the microbiological quality of the bottled mineral water in contact with saliva showed that heterotrophic plate counts ($21^{\circ}C$) had a tendency to be sustained or decrease slightly after 10 days. Heterotrophic plate counts ($36^{\circ}C$) had a high population in the initial samples and gradually decreased at $4^{\circ}C$ storage, but it remained constantly high in storage at $20-25^{\circ}C$ and $36^{\circ}C$. In the general drinking condition, the population was slightly higher than the control, but the overall trend was similar. Conclusions: As a result of the microbiological quality of mineral bottled water in contact with saliva during the process of drinking, heterotrophic plate counts ($21^{\circ}C$ and $36^{\circ}C$) showed a high population compared to the control, which was only opened and not in contact with saliva. In some samples, pathogenic indicators were also detected. Therefore, it is desirable to consume bottled mineral water as soon as possible after opening.
