Effect of Physical Pre-treatment of Mature Seed in Callus Formation and Plant Regeneration of Zoysiagrass (잔디종자의 물리적 전처리가 캘러스 형성과 식물체 재분화에 미치는 효과)

  • In order to improve the transformation efficiency of zoysiagrass by increasing the frequency of callus formation from mature seeds and plant regeneration, the effect of pre-treatment with sea sand was examined. Mature zoysiagrass seeds were given 10 min of swelling time before sea sand treatment using a sea sand and seed ratio of 1 : 1 and a vortex shaking speed of 6 (1,000 rpm) for 10 min. The seeds showed increased callus formation that was more than 2 times the rate in the control. In addition, plant regeneration efficiency was also increased when embryogenic callus induced from sea sand-treated mature seeds was cultured in regeneration medium. These results will be very helpful for improving the genetic transformation frequency of zoysiagrass, a recalcitrant monocot grass.

  • Shaking cultivation was carried out in 250 mL Erlenmeyel' flasks containing 60 mL of culture solution for 7 days. Artificial seawater was used as a basal culture medium. The cultivation was performed under the conditions that the temperature ranged from $4^{\circ}C$ to $32^{\circ}C$, the agitation speed 50 rpm to 200 rpm, and the sugar concentration 5 g/L to 35 g/L. The medium condition was cultured that sugar and nitrogen ratio 1.25 to 10. The biomass. the lipid content in biomass, and the DBA content of lipid were investigated according to the cultivation conditions. The lipid in biomass was distributed between 10 and 349'0, the DHA in lipid 34% to 43% of the lipid. and the biomass concentration 0.425 g/L to 4.50 g/L.

  • This paper proposes a hybrid image stabilizing system which uses both optical image stabilizing system based on EKF (Extended Kalman Filter) and digital image stabilization based on SURF (Speeded Up Robust Feature). Though image information is one of the most efficient data for object recognition, it is susceptible to noise which results from internal vibration as well as external factors. The blurred image obtained by the camera mounted on a robot makes it difficult for the robot to recognize its environment. The proposed system estimates shaking angle through EKF based on the information from inclinometer and gyro sensor to stabilize the image. In addition, extracting the feature points around rotation axis using SURF which is robust to change in scale or rotation enhances processing speed by removing unnecessary operations using Hessian matrix. The experimental results using the proposed hybrid system shows its effectiveness in extended frequency range.

Amylase Production from Haloarcular sp. EH-1 (고호염성 Haloarcular sp. EH-1으로 부터 amylase 생산)

  • The extremely halophilic archaebacterium Haloarcular sp. EH-1 was isolated from solar salts. Amylae production from Halonrcular sp. EH-1 have been studied. The results obtained were as follows. The optimal medium composition for the production of amylase from Haloarcular sp. EH-1 were soluble starch 1.5%, yeast extract 1.0%, MgSO$_4$ 7h$_2$O 2.0%, KCI 0.1%, NaCl 25% (pH 7.5). The incubation temperature, aeration rate and agitation speed were 4$0^{\circ}C$, 100 $m\ell$ medium / 500 $m\ell$ shaking flask, and 110 rpm. The cell growth and enzymatic activity was highest at 9 days of incubation. So amylase production appeared to be a growth-related phenomenon.

  • Most of outdoor mobile robots have a suspension on each wheel in order to relieve the shock by ground obstacles and to improve the driving stability. Typically, in the actual operations, the suspensions have been used under a given set of conditions as all the damping and spring coefficients of the suspensions are fixed. However, it is necessary to readjust the coefficients of the suspensions according to surface conditions that may cause the unstable shaking of a robot body at high speed driving. Therefore, this paper is focused on the mobility analysis of an outdoor robot when the coefficients of suspensions (in particular, damping coefficients) are changed while driving on an uneven road surface. In this paper, a semi-active suspension with twelve damping coefficient levels was used and a small sized vehicle with the suspensions was employed to analyze the mobility dependent on a change of the damping coefficient. And the mobility was evaluated through driving experiments on a bumped slope.

  • The optimum cultural conditions for production of red pigment from Monascus rubber KCTC 6122 is liquid culture were studied. Monascus robber KCTC 6122 was shown to give the maximum production of red pigment in the medium containing 4% rice powder, 0.2% NaNO$_3$, 0.3% Na$_2$HP0$_4$ and 0.15% MgSO$_4$. The optimum culture conditions, temperature, initial pH and shaking speed were 30$^{\circ}C$, 6.5 and 150 rpm, respectively. The red pigment production reached a maximum level at 8days of cultivation.

Enhancement of eurycomanone biosynthesis in cell culture of longjack (Eurycoma longifolia) by elicitor treatment

  • In this study, the effect of elicitors such as yeast extract (YE), methyl jasmonate (MeJA) and salicylic acid (SA) on the accumulation of eurycomanone in Eurycoma longifolia cell cultures were investigated. Suspension cells of E. longifolia was cultured in Murashige and Skoog (MS) medium supplemented with 30 g/L sucrose, 1.25 mg/L naphthaleneacetic acid (NAA) and 1 mg/L kinetin at a shaking speed of 120 rpm. Elicitors were added in the culture at different concentrations and times to stimulate eurycomanone accumulation in the Eurycoma longifolia cells. Eurycomanone content was determined by HPLC with a C18 column, flow rate of 0.8 mL/min, run time of 17.5 min, and a detector wavelength of 254 nm. The stationary phase was silica gel and the mobile phase was acetonitrile: $H_2O$. Non-elicited cells were used as the control. The study showed the effect of different elicitor concentrations, YE at 200 mg/L, MeJA at $20{\mu}M$ and SA at $20{\mu}M$ stimulated high production of eurycomanone. In which, treatment of $20{\mu}M$ MeJA after 4 days of culture resulted in the highest accumulation of this compound (17.36 mg/g dry weight), approximately 10-fold higher than that of untreated cells (1.70 mg/g dry weight).

Optimal Conditions for Chitinase Production by Serratia marcescens

  • A chitinase-producing bacterium was isolated from seashore mud around Beobseongpo in Chunmam province through the use of a selective enrichment culture. The best chitinase producing strain was isolated and identified as Serratia marcescens KY from its characteristics. For effective production of chitinase, optimum pH, temperature, and agitation speed were investigated in flask cultures. The optimum pH using Serratia marcescens KY was between pH 6 and 7 and the chitinase produced was 37.9 unit/mL. On the other hand, the optimal pH of the Serratia marcescens ATCC 27117 was 7.5, and the produced amount of chitinase was 35.2 unit/mL. The optimal temperature for chitinase production for Serratia marcescens KY and Serratia marcescens ATCC 27117 was $30^{\circ}$. The cell growth pattern at different temperature was almost identical to the chitinase production. To investigate the optimal shaking speed under optimal culture, speeds were varied in the range of 0∼300 rpm. The maximum production of chitinase was carried at 200 rpm although the cell growth was the highest at 150 rpm. It indicates that oxygen adjustment is required for the maximum chitinase production. Using optimal conditions, batch cultures for comparing Serratia marcescens KY and Serratia marcescens ATCC 27117 were carried out in a 5 L fermentor. The oxygen consumption was increased with the increase of culture. Especially, at 120 h of culture Serratia marcescens KY and Serratia marcescens ATCC 27117 produced 38.3 unit/mL, and 33.5 unit/mL, respectively.

Medium Composition Affecting Production of Bacterial Cellulose by Gluconacetobacter hansenii PJK in an Agitated Culture (배지조성이 Gluconacetobacter hansenii PJK의 Bacterial Cellulose의 교반 생산에 미치는 영향)

  • The effects of variation in composition of the medium on the conversion of Gluconacetobacter hanseii PJK cells producing cellulose ($Cel^+$) to non-cellulose producing ($Cel^-$) mutants and the production of bacterial cellulose (BC) in an agitated culture were investigated. The impeller speed greater than 500 rpm was required to decrease the population of $Cel^-$ mutants to minimum in a basal medium containing $1.5\%$ ethanol because the optimum impeller speed to minimize the population of $Cel^-$ mutants increased with the concentration of ethanol added to a basal medium. Ethanol fed-batch culture could not increase the BC production in an agitated culture unlike that of a shaking culture. The amount of BC produced in a basal medium containing $1\%$ ethanol was $39\%$ more than that of the same medium with $0.27\%\;Na_{2}HPO_4$. Increase in the concentration of acetic acid in a basal medium decreased the BC production. The pH control of the culture broth increased the cell mass in the batch culture and improved the production yield of water-soluble polysaccharide (WSPS), but did not affect the production of BC.

Influence of Vehicle Vibration on Track Geometry Measurement (차량 진동이 궤도 선형 측정에 미치는 영향)

  • Track maintenance works based on track geometry recordings are essential to enhance the safety and comfort of railway transportation. Usually, the track irregularity has been measured by a special inspection trains which all were imported from abroad. Because the inspection train speed is limited under 160 km/h, it takes a long time to inspect railways and there is difficulty in daytime operation. To solve this problem, we started to develop a track geometry measuring system (TGMS) with measurement speed up to 300 km/h which can be installed in commercial vehicles such as HEMU-430X. In this paper, we introduce a newly developed inertial TGMS and propose two inertial navigation system (INS) algorithms (method A, B) for measuring track geometry. In order to investigate advantage and disadvantage of each algorithm, we performed vibration test of the TGMS, which was simulated by 6-axis shaking table. Through the vibration test, we analyzed the influence of vehicle vibration on the TGMS which will be installed on bogie frame. To the vibration test, two methods satisfied the required accuracy of track geometry measurement under the level of the actual vehicle vibration of HEMU-430X train. Theoretically, method A is sensitive to vehicle vibration than method B. However, HEMU-430X's bogie vibration frequency range is out of interest range of measurement system. Therefore, method A can also apply the HEMU-430X train.
