Aspergillus niger를 이용한 유청으로부터 구연산의 생산에 있어서 온도와 pH의 영향 (Effects of Temperature and pH on the Production of Citric Acid from Cheese Whey by Aspergillus niger)

  • Cheese 제조시 부산물로 생성되는 whey를 배지로 사용하여 Aspergillus niger를 이용하여 citric acid를 생산하는데 영향을 미치는 여러 가지 요인 중 중요한 요인인 온도와 pH의 영향에 대하여 고찰하였다 15일간 27, 30, 33, $36^{\circ}C$와 pH 2, 3, 4, 5에서 각각 배양하면서 소비된 lactose의 양과 생산된 citric acid의 양을 측정하였다. 생산된 citric acid의 최대 농도는 33.9 g/l(구연산 생산에 쓰여진 유당을 기준으로 할 때 68.26%)이었으며, shaking speed는 citric acid 생산에 직접 영향을 주기보다는 pellet 형성시 그 형태에 영향을 미치는 것으로 나타났다. 배양 온도가 $33^{\circ}C$, pH는 3일때 가장 많은 양의 citric acid가 생산되었다.

Optimizing Fermentation Medium Composition for Bacterial Cellulose Production by a Newly Isolated Gluconacetobacter sp. RKY5

  • In this study, we investigated the optimal medium composition for bacterial cellulose (BC) production by Gluconacetobacter sp. RKY5. Among the various kinds of carbon sources, glycerol was the most efficient as a sole carbon source and its optimal concentration for BC production was 15 g/L. The optimal concentration of yeast extract as a nitrogen source for BC production was found to be 8 g/L. $K_{2}HPO_{4}$ and acetic acid were selected respectively as a phosphate source and a secondary substrate, and both optimal concentrations were 3 g/L. The amount of produced BC was 4.59 g/L in a static culture and 6.5 g/L in a shaking culture condition with 150 rpm. These values were 2.1 and 2.7 times higher than those in a static (2.16 g/L) and a shaking (2.41 g/L) cultures using HS medium generally used for BC production.

  • 비교적 연구가 미비한 Fe-SOD의 효소화학적 특성 및 그 생리적 기능을 검토하기 위해 여러 종의 세균을 대상으로하여 Fe-SOD의 고생산균주를 screening하였다. 그 결과 Fe-SOD를 대량 세포내에 생성하는 Pseudomonas polycolor를 선발하여, 이 균주의 효소생산 최적 배양조건을 설정하였다. 본 균주가 생산하는 효소는 특이적인 저해제의 작용양식에 의해 Fe을 cofactor로 요구하는 Fe-SOD임이 밝혀졌다. SOD 생성을 위한 최적배지조성은 glycerin 3%, polypeptone 1%, meat extract 0.5%, KCI 0.2%이었고, 최적 초발 pH는 9.0이었으며, 이 조건에서 500ml용 shaking flask에 배지 100ml를 넣어 15시간 전 후 배양했을 경우가 효소생산량은 최대가 되었다.

잿빛공팜이에 대한 길항균 Bacillus Iicheniformis N1의 배양적 특성 (Cultural Characteristics of Antagonistic Bacterium, Bacillus licheniformis N1 against Botrytis cinerea)

  • This study was conducted to estimate the cultural characteristics, the production of antibiotic, and the selection of optimal media for mass culture of Bacillus licheniformis N1 isolate which was previously reported as an antagonistic bacterium to Botrytis cinerea. We investigated initial pH, temperatures and shaking speed for good cultural conditions and antibiotics production by N1 isolate. According to the results, the optimal conditions of initial pH, temperatures, and shaking speed were determined to be pH 5.0~5.5, 30~35$^{\circ}C$ and 250 rpm, respectively. Also, the optimal conditions for the antagonism by N1 isolate highly appeared in the initial pH as 5.0, and the mycelial growth inhibition was high when the substances used such as glucose or corn starch as carbon sources, and biji(soybean curd residue) flour as a nitrogen source. Furthermore, inhibitory area was significantly expanded, when 3% or 5% of corn starch was added into 5% of Biji flour as nitrogen source, were respectivley selected for mass culture of N1 isolate. Among them, 5% Biji flour medium showed higher cell density more than 10 times that in NB medium after 48 hour incubation. Therefore, the optimal medium was determined as 5% biji flour added 3~5% of corn starch for high density of cells.

Effect of Culture Conditions on Cathepsin B Inhibitor Production by a Marine Bacterium, Pseudomonas sp. Strain PB01

  • A novel cathepsin B inhibitor-producing bacterium was isolated from marine sediments and identified based on its 16S rDNA sequence as Pseudomonas sp. strain PB01 (Accession No. EU126129). The growth and enzyme inhibitor production were investigated under various culture conditions. A mixture of organic nitrogen source was required for the optimal production, whereas both glucose and maltose proved to be the effective carbon sources for cathepsin B inhibitor production. Other optimal culture conditions included temperature range between 25 and $28^{\circ}C$, initial medium pH of 6.6, and shaking speed of 200 rpm. Under these optimal conditions, the maximum inhibitory activity from culture broth was approximately 50% after 30 h of cultivation. Additionally, kinetic study revealed that inhibitor production paralleled with cell growth, which suggested that the inhibitor may be a primary metabolite of that bacterium.

플라스크 배양에서 Bacillus subtilis BK-17의 혈전용해효소 생산에 대한 환경 및 영양 조건의 영향 (Effects of Environmental and Nutritional Conditions on Fibrinolytic enzyme Production from Bacillus subtilis BK-17 in Flask Culture)

  • The production of fibrinolytic enzyme from Bacillus subtilis BK-17 was studied in the shake flask cultures. The important medium components studied include nitrogen source, carbon source and inorganic salts. The environmental conditions include initial pH, temperature, shaking speed and working volume. Among various N-sources, C-sources and inorganic salts tested, soybean flour, D-glucose and Na2HPO4 gave the best results, and their optimal concentrations were 1.5%, 0.5% and 0.05%, respectively. The optimal pH and temperature were 9.0 and 37$^{\circ}C$. With decreasing working volume in the range of 25∼100ml in the 250ml flask or increasing shaking speed in the range of 100∼300rpm, the enzyme production was greatly enhanced. The enzyme activity under the optimal conditions was about 1400I.U./ml with urokinase as a standard.

Effects of Different Sizes of Glass Beads on the Release of Sporocysts from Eimeria tenella Oocysts

  • The oocyst wall is severed by means of mechanical injury or chemical agents. This study reports the percentage of in vitro sporocyst release following mechanical shaking in the presence of varying sizes of glass beads. Glass beads measured 0.5, 1, and 3 mm in diameter and were shaken with the oocysts for different times ranging from 5 sec to 5 min. Approximately 80% of sporocysts were released with 5 min of shaking in the presence of 3 mm glass beads, as well as 30 sec with 0.5 mm beads and 1 mm glass beads. The release of sporocysts of E. tenella was most efficient using 1 mm glass beads and treatment times of 30 sec to 1 min. Therefore, the use of 1 mm glass beads with 30 sec to 1 min of agitation is recommended in order to maximize sporocyst release and recovery and to improve the yield of viable sporozoites for use in biochemical, tissue culture, and immunological applications of coccidia.

미생물 셀룰로오스 생산을 위한 당밀의 전처리 및 생산된 셀룰로오스의 물리화학적 특성 (Pretreatment of Cane Molasses for Production of Bacterial Cellulose and Its Physico-Chemical Properties)

  • BC 합성능이 우수하며, 진탕배양에서도 BC를 생산할 수 있는 능력이 있음이 확인된 Aectobacter sp. V6로부터 BC 생산을 위한 당밀 전처리 조건을 검토하였으며, 생산된 BC의 물리화학적 특성을 검토하였다. 당밀을 배지성분으로 사용하기 위해 전처리를 실시한 결과, 1%의 TP가 BC 생산에 가장 효율적이었다. 전처리한 당밀배지를 이용하여 정치 및 진탕배양에서 생산된 BC의 물리화학적 특성을 조사한 결과, 모든 BC는 유화능이 있었으나 유화 안정능은 낮았다. 또한 모든 BC에서 높은 수분 보유능이 나타났으며, 특히 정치배양에서 생산된 BC는 $\alpha$-cellulose보다 14배 이상 높았다. BC의 점도는 모두 $\alpha$-cellulose보다 높았으며, 점도계 회전속도의 증가에 따라 급격히 감소하였다. FT-IR을 통한 조성 분석에서 BC는 식물성 셀룰로 오스와 차이가 없었으며, XRD를 통한 결정성 분석에서는 모든 BC가 결정성을 나타내었다. BC의 미세구조를 조사한 결과, 모든 BC가 미세망상구조로 이루어져 있었는데, 이로 인해 BC는 식물성 셀룰로오스와는 달리 독특한 물성을 가지는 것으로 판단되었다. 또한 정치배양에서 생산된 BC는 진탕배양에서 생산된 것보다 셀룰로오스 미세섬유가 조밀하게 얽혀있음을 알 수 있었다.

Thidiazuron이 무궁화 '홍화랑' 품종 액체 현탁 배양시 신초형성에 미치는 영향 (Thidiazuron-induced Shoot Formation of Hibiscus syriacus L. 'Honghwarang' by Suspension Culture)

  • 본 실험은 무궁화 '홍화랑' 품종의 기내 대량 생산을 위한 액체 현탁배양에서의 배양조건과 방법을 구체적으로 규명하고자 수행되었다. Mesh 종류별로 비교해 보았을 때 가장 굵은 10 mesh를 사용한 것이 callus형성에 효과적이었으며, 회전속도에 따른 callus의 형성은 80 rpm에서 가장 효과적이었다. TDZ 0.01 mg/L 첨가된 MS 고체배지에서 유기된 callus를 TDZ 0.001 mg/L가 첨가된 액체배지에 현탁배양했을 경우 callus 형성과 신초원기 형성이 가장 왕성하였다. 형성된 신초원기를 MS와 1/2 MS 배지에 생장조절제가 첨가되지 않은 조건에서 현탁배양시 신초 재분화율이 가장 높았다. 따라서, 이러한 액체 현탁배양 조건과 방법의 구체적인 규명은 무궁화 급속 대량 번식체계 확립을 위한 가능성을 보여 주었다.

Plant Regeneration from Mesophyll Protoplasts Culture of Solanum sisymbriifolium

  • The optimal culture conditions were studied for plant regeneration from mesophyll protoplasts of Solanum sisymbriifolium. Axenic seedlings of S. sisymbriifolium were used as a explant for protoplast culture. Many viable protoplasts were isolated by incubating leaf slices in an enzyme solution containing 0.25% Meicerase and 0.05% Macerozyme for 16 hr at $25^{\circ}C$ without shaking. Protoplast density of $5.0{\times}10^4\;ml^{-1}$ in Kao medium containing 5.0 mg/L NAA, 1.0 mg/L 2,4-D and 1.0 mg/L BA was optimal for colony formation. Most colonies were formed when protoplasts were cultured at $25^{\circ}C$ after initial culture at $30^{\circ}C$ for one week. On the MS agar medium with 1.0 mg/L zeatin, 38.4% of protoplast-derived calli differentiated shoots. These shoots rooted on 1/2MS medium with 5.0 g/L sucrose and 2.5 g/L gellan gum, and developed into whole plants.
