스테로이드 성호르몬이 돼지 지방전구세포의 분화에 미치는 영향 (Effects of Sex Steroid Hormones on Differentiation of Pig Preadipocytes)

  • 본 연구는 스테로이드 성호르몬 즉 에스트로겐, 테스토스테론 및 노르테스토스테론이 돼지 지방전구세포의 분화와 증식에 미치는 영향을 구명하기 위해서 수행하였다. 지방전구세포는 갓난 암퇘지의 등지방조직을 떼어내어 coll- agenase를 처리해서 분리해서 배양했다. 세포 배양중에 10-8M과 10-7M의 스테로이드 성호르몬을 처리했다. 세포배양 전기에 스테로이드 성호르몬을 처리했을 때 지방전구세포의 분화나 증식에 아무런 영향을 미치지 않았고, 세포배양 후기에 처리했을 때는 테스토스테론과 노르테스토스테론이 세포분화를 촉진시켰다.

  • 본 연구는 스테로이드 성호르몬인 에스트로겐(estrogen), 테스토스테론(testosterone) 및 노르테스토스테론(19-nortestosterone)이 암, 수 돼지 지방전구세포의 증식과 분화에 미치는 영향을 구명하기 위해서 수행하였다. 지방전구세포는 암, 수 갓 난 돼지의 등지방 조직을 떼어 내어 collagenase를 처리한 후 분리해서 $CO_2$ 배양기에서 배양했다. 세포 배양 중에 $10^{-7}M$$10^{-6}M$의 스테로이드 성호르몬을 처리했다. 먼저 지방전구세포의 증식에 미치는 영향을 보면, 암퇘지에서 분리한 지방전구세포의 증식을 높은 농도의 스테로이드 성호르몬 모두가 촉진했다. 수퇘지에서 분리한 지방전구세포의 증식은 에스트로겐과 테스토스테론만이 촉진했다. 지방세포의 분화에 미치는 작용을 보면, 세 호르몬 모두, 농도에 관계없이 암수 성별에 관계없이 지방전구세포의 분화를 촉진했다. 촉진 정도는 증식보다 분화에 더 크게 나타났다.

  • 어류의 체내에서 성분화를 유도하는 물질이 성스테로이드호르몬(sex steroid hormone)이라는 사실이 잘 밝혀져 있으며, 성스테로이드 생합성 효소의 하나인 aromatase도 성분화에 직접적인 역할을 하는 것으로 알려져 있다. 본 연구에서는 유전적으로 암컷인 틸라피아 자어(larvae) 집단을 aromatase 저해제(aromatase inhibitor, AI)인 Fadrozole로 침지 처리하여 초기 발생단계 중 어느 시기에 aromatase가 성분화 유도 작용을 하는 지를 조사하였다. Fadrozole 처리 유무 및 처리 농도의 차이는 부화 자어의 생존율에 유의한 차이를 유발하지 않았다. 하지만, 부화후 11일과 13일째에 고농도의 Fadrozole로 처리한 실험군의 자어는 유전적인 성이 암컷임에도 불구하고 유의하게 높은 비율의 자어가 수컷으로 분화하였다. 이 결과는 틸라피아 부화 자어가 스테로이드 생합성 효소의 저해에 아주 민감하게 반응하며, 이 종에서 aromatase의 주된 작용시기가 예상보다 훨씬 빠른 부화 후 11일 전후라는 사실을 보여준다. 또한 이상의 결과는 단 3시간의 AI 침지 처리가 유전적으로 설정되어 있는 성과 반대방향으로의 성분화를 유도하기에 충분할 정도로 강력함을 의미한다.

성인초기 미혼남녀의 자아분화와 자기개방이 이성과의 친밀감에 미치는 영향 (The Impact of the Self-Differentiation and Self-Disclosure of Unmarried Persons in Early Adulthood on Their Intimacy with the Opposite Sex)

  • The purpose of this study was to examine how the family of origin and self-disclosure of unmarried persons in early adulthood affected their own intimacy with the opposite sex, which was one of the major developmental tasks for them. The findings of the study are as follows: First, the self-differentiation, self-disclosure, and intimacy with the opposite sex of the single male and female adults investigated were generally at a moderate level. Second, for self-differentiation, the background variables of the male and female adults made no statistically significant difference. For self-disclosure, the background variables of the female adults made no statistically significant difference to that, but the male adults differed significantly from one another in that regard according to their family makeup and birth of order. As for connections between the background variables of the single male and female adults and their intimacy with the opposite sex, the male adults varied in that aspect with their birth order, and whether they were dating and considered getting married made a difference to that as well. In the event of the female adults, whether they considered getting married and how long they were dating made a difference in their intimacy with the opposite sex. Third, concerning the relative influence of the self-differentiation and self-disclosure of the unmarried male and female adults on their intimacy with the opposite sex, their intimacy with the opposite sex improved as division occurred from their original families, when they opened themselves up to more people, when they thought of marriage more and when they were dating for a longer period of time.

성별을 고려한 중풍 변증진단 판별모형개발(V) (Discriminant Model V for Syndrome Differentiation Diagnosis based on Sex in Stroke Patients)

  • In spite of abundant clinical resources of stroke patients, the objective and logical data analyses or diagnostic systems were not established in oriental medicine. As a part of researches for standardization and objectification of differentiation of syndromes for stroke, in this present study, we tried to develop the statistical diagnostic tool discriminating the 4 subtypes of syndrome differentiation using the essential indices considering the sex. Discriminant analysis was carried out using clinical data collected from 1,448 stroke patients who was identically diagnosed for the syndrome differentiation subtypes diagnosed by two clinical experts with more than 3 year experiences. Empirical discriminant model(V) for different sex was constructed using 61 significant symptoms and sign indices selected by stepwise selection. We comparison. We make comparison a between discriminant model(V) and discriminant model(IV) using 33 significant symptoms and sign indices selected by stepwise selection. Development of statistical diagnostic tool discriminating 4 subtypes by sex : The discriminant model with the 24 significant indices in women and the 19 significant indices in men was developed for discriminating the 4 subtypes of syndrome differentiation including phlegm-dampness, qi-deficiency, yin-deficiency and fire-heat. Diagnostic accuracy and prediction rate of syndrome differentiation by sex : The overall diagnostic accuracy and prediction rate of 4 syndrome differentiation subtypes using 24 symptom and sign indices was 74.63%(403/540) and 68.46%(89/130) in women, 19 symptom and sign indices was 72.05%(446/619) and 70.44%(112/159) in men. These results are almost same as those of that the overall diagnostic accuracy(73.68%) and prediction rate(70.59%) are analyzed by the discriminant model(IV) using 33 symptom and sign indices selected by stepwise selection. Considering sex, the statistical discriminant model(V) with significant 24 symptom and sign indices in women and 19 symptom and sign indices in men, instead of 33 indices would be used in the field of oriental medicine contributing to the objectification of syndrome differentiation with parsimony rule.

Sex Differentiation and Early Gonadogenesis in Sebastes inermis Cuvier

  • Early gonadal development and sexual differentiation of dark-banded rockfish (Sebastes inermis Cuvier) were followed from parturition to 400 days post parturition (dpp). During this period, average total length (TL) increased from 0.57 to 13.18 cm. Primordial germ cells (PGCs) were first detected at 0.68 cm TL (10 dpp). When fish reached 1.52 cm TL (50 dpp), initial stages of ovarian differentiation were identified by the presence of PGCs containing condensed chromatin and their transformation into meiotic oocytes. At 10.23 cm TL (300 dpp), the ovaries gradually developed into oocytes in the primary yolk stages. Ovary growth was rapid after sex differentiation, but testis tissue continued to multiply without growing until fish reached 6.97 cm TL (200 dpp), after which the production of spermatocytes, spermatogonia, and cyst cells was apparent. Histological analysis of gonadal structure suggested a gonochoristic sexual development pathway. Our analysis of the sex ratio at 400 dpp showed a significantly higher proportion of males.

  • Red spotted grouper, Epinephelus akaara, is a popular aquaculture species and a protogynous hermaphrodite. Induction of artificial sex change at the time of primary sex differentiation is of interest but has not been successful due to the lack of necessary basic information. To find out the potential neuroendocrine influence on the primary sex differentiation, the expression of key genes in the brain was investigated during the formation of ovarian cavity. Expression of cyp19a1b, esr1, gnrhr1, fsh, lh and cga in the brain was positively associated with the formation of ovarian cavity, showing gradual increase as the formation proceeds. However, the expression of gnrh1 was suppressed during the early part of the ovarian cavity formation, signifying potential hypothalamic influence on the primary sex differentiation in this species.

조피볼락, Sebastes schlegeli의 성분화 기간 중 Cytochrome P450 Aromatase 유전자의 발현 (Expression of Cytochrome P450 Aromatase Genes during Sex Differentiation in Korean Rockfish, Sebastes schlegeli)

  • 자웅이체형 어류의 성결정 및 성분화는 일반적으로 각 개체의 유전형을 따른다. 자연환경에서는 자신이 가진 유전정보의 조절에 따라 성분화 시기에 아로마테이즈 유전자의 발현이 증가하거나 감소하고, 그 결과 스테로이드 호르몬들의 조성이 결정되어 각기 다른 방향으로 성분화하는 것으로 알려져 있다. 본 연구에서는 해산 태생 어류인 조피볼락(Sebasts schlegeli)의 ovarian type aromatase (P450aromA)와 brain type aromatase (P450aromB)의 유전자를 부분적으로 클로닝하여, 이들의 염기서열을 밝혔으며, 각 유전자에 대한 primer를 제작한 후 성분화 기간 중 이 유전자들의 발현을 조사하였다. 조피볼락 아로마테이즈 유전자들은 조사를 시작한 출산 후 35일째에 여러 개체의 머리와 몸에서 각각 발현되었으나, 52일째에는 아로마테이즈 유전자들의 발현 개체수가 현저히 감소하였다. 그리고 출산 59일째에는 발현 개체 수가 다시 증가하였다. P450aromA과 P450aromB의 발현 양상은 전반적으로 유사하였으나, 45일째에는 P450aromB의 발현 개체수가 P450aromA 발현 개체 수보다 훨씬 많았다. 조직학적 분석을 통해 제시된 이 종의 성분화 시기는 지금까지 출산 $50{\sim}65$일 전후로 알려져 왔으나, 본 연구에서 나타난 아로마테이즈 유전자들의 발현 결과를 볼 때, 이 종의 실질적인 성분화는 알려진 것보다 최소한 $1{\sim}2$주 정도 빨리 진행될 가능성이 있다. 본 연구의 결과는 또한 태생어류의 성분화도 난생어류의 성분화에서 보고된 것과 마찬가지로 아로마테이즈의 작용과 밀접하게 연관되어 있다는 사실을 시사한다.

  • Uterine stromal tumors with features of ovarian sex-cord differentiation are relatively rare. The neoplasms composed of sex cord-like components in more than 50% of the tumor are classified as group II. We report the cytologic findings of a case of uterine tumor resembling ovarian sex-cord tumor. The cervical smears of a 62-year-old woman with submucosal tumor showed loose aggregates of spindle cells as well as glandular or tubular structures of round cells with a distinct ceil membrane and a prominent small nucleolus. Because uterine stromal tumor can have sex cord differentiation, its possibility should be considered in the interpretation of cervical smears.

Histological Analysis of Early Gonadal Development and Sex Differentiation in Chameleon Goby, Tridentiger trigonocephalus

  • This study describes the developmental process of gonads in chameleon goby, Tridentiger trigonocephalus from the stage of hatching to 100 days after hatching (DAH). Based on histological observation, the primordial germ cells were observed in mesentery between mesonephric duct and gut at 15 DAH (total length, TL: $6.8{\pm}0.2mm$). At 20 DAH (TL: $7.9{\pm}0.1mm$), the primordial gonad began to protrude into peritoneal cavity and developed between mesonephric duct and gut. Initial ovarian differentiation was identified by the presence of ovarian cavity and oogonia in the gonads at 55 DAH (TL: $21.1{\pm}1.3mm$). Testicular differentiation started at 65 DAH (TL: $23.7{\pm}0.9mm$) with appearance of spermatogonial cells in the gonads. These findings indicate that sex differentiation in T. trigonocephalus occurs earlier in females than males, suggesting that this species can be classified as an undifferentiated gonochorist.
