A Novel Marker for the Species-Specific Detection and Quantitation of Shigella sonnei by Targeting a Methylase Gene

  • Shigella sonnei is a causal agent of fever, nausea, stomach cramps, vomiting, and diarrheal disease. The present study describes a quantitative polymerase chain reaction (qPCR) assay for the specific detection of S. sonnei using a primer pair based on the methylase gene for the amplification of a 325 bp DNA fragment. The qPCR primer set for the accurate diagnosis of Shigella sonnei was developed from publically available genome sequences. This quantitative PCR-based method will potentially simplify and facilitate the diagnosis of this pathogen and guide disease management.

  • This study is to be surveyed the rural environmental conditions such as housing, water works, sewerage, road and farm road, land utilization, natural conservation and sightseeing, public damage and disaster, and rural water requirement through the 50 villages (10995 home unit) The brief results summarized in this study are as follows. 1.Modernization of rural housing, toilet room and arrangement of dust materials are getting better than before l0yrs. 2.Water works are highly improved but sewerage problem is still in the serious problems. 3.Rood achievement is quite good but farm road is in still insufficiency 4.Utilization of land resources should be maximaized but land conservation is still insufficient condition and farm land is getting polluted. 5.Natural environmental conservation is much more improved but landscape is still insufficient. 6.Public damages such as water pollution, air pollution, and others are getting greater and greater but meteorological damage should be decreased. 7.Watershed condition is getting better and better but because of the large requiriment of the water use for the crops, agricultural water use should be needed much more than before l0yrs.

  • Pollution load allocation can likely be much controversial, which is essential to formulate the total water pollution load management plan. Existing rules(or guidances) in Korea, can provide no specific criteria for load allocation. Therefore, this paper studied(comprehensively) possible standards how or why to choose any particular allocation method, which was applied in the Gwangju City's load allocation for the satisfaction of set water quality goal. This load allocation is basically focused on the load reduction of domestic wastewater rather than industrial wastewater, because the land-use is strictly regulated and larger sources of pollution are few in the Gwangju City. This paper recommends the city to increase the capacity of sewage treatment plants, promote sewerage maintenance, and set higher effluent standards.

  • New type of the sewer drainage has been developed to overcome present sewer drainage's shortcoming. This system has the function of drainage, sewerage, reverse flow prevention, and harmful insect and smell prevention. Numerical simulation has been carried out in order to minimize the troubles that can be happened in the process of manufacture and installation process. This sewer drainage system for harmful insect and smell prevention intercepts pollution source, and then it prohibit second pollution. Harmful insect cannot go in and out in this system. Also, this system can reduce the damage of flooded districts due to heavy rain because it is impossible to flow backward from sewer drainage.

  • In this presentation I would like to introduce the Rural Sewerage Project, subsidized by the Ministry of Agriculture, Forestry and Fisheries $\alpha$UFF), for the purpose of water quality conservation and improvement onife in rural areas of Japan. Specifically, it will cover background information on the inauguration ofthe Project, its strong points, wastewater treatment technology and some of its problems, systems of t the Project, etc.
