Effect of Prolonged Heat Exposure on Serum Glutamic Oxaloacetic and Glutamic Pyruvic Transaminase Activities of Rats (連續的 溫熱曝露가 흰쥐의 血淸 Glutamic Oxaloacetic Transaminase 및 Glutamic Pyruvic Transaminase의 活性에 미치는 影響)

  • Sera from male Spague-Dawley rats, exposed to $30\\pm 0.5^\\circ C$ for 240 hours or $33\\pm 0.5^\\circ C$ for 64 hours, were assayed for the activities of serum glutamic oxaloacetic transaminase(SGOT) and serum glutamic pyruvic transaminase(SGPT) at various time during the heat exposure. 1. When compared to control animals maintained at $23\\pm 1^\\circ C$, the animals exposed to $30\\pm 0.5^\\circ C$ ro $33\\pm 0.5^\\circ C$ showed a significant increase in SGOT and SGPT activities, 2. The SGOT activity incressed at 16 and 72 hours after the exposure to $30^\\circ C$, and at 30 and 64 hours after the exposure to $33^\\circ C$. After 72 hours, the activity returned to the initial value in case of $30^\\circ C$ exposure. 3. The SGPT activity increased significantly as early as 4 hours after the exposure to $30^\\circ C$ or $33^\\circ C$. It was also high at 16 hours after the exposure. The activity was also high at 72 hours and at 64 hours after the exposure to $30^\\circ C$ and $33^\\circ C$ respectively. After 144 hours, SGPT level increased slightly in the case of $30^\\circ C$ exposure. 4. The activities of SGOT and SGPT were significantly higher in rats exposed to $33^\\circ C$ at 16, 30, and 64 hours than those exposed to $30^\\circ C$. 5. It may be inferred from above data that the prolonged heat exposed rat has the abnormal metabolism of transamination.

Hepatoprotective and free radical scavenging activities of Lagerstroemia speciosa Linn. leaf extract

  • The present study deals with the amelioration by Lagerstroemia speciosa Linn. leaf extract against hepatotoxicity induced by carbon tetrachloride ($CCl_4$), which was evaluated in terms of serum marker enzymes like serum glutamate pyruvate transaminase, serum glutamate oxaloacetate transaminase, alkaline phosphatase, serum total bilirubin, total protein levels along with concomitant hepatic and antioxidants like superoxide dismutase, catalase, glutathione, glutathione peroxidase and lipid peroxidation enzymes were monitored. These biochemical parameters altered by the single dose level of $CCl_4$ (0.75 ml/kg body weight, i.p). Pre treatment with L. speciosa prior to the administration of $CCl_4$, at the doses of 50 and 250 mg/kg. body weight/day, p.o. for 7 days, significantly restored all the serum and liver tissue parameters near to the normal levels, respectively. Silymarin was used as a reference standard, prior to the administration of $CCl_4$ to rats. These findings indicate the protective potential of L. speciosa against hepato toxicity which possibly involve mechanism related to its ability of selective inhibitors of (reactive oxygen species like antioxidants brought about significant inhibition of TBARS suggesting possible involvement of $O_2{\cdot}-$, $HO_2{\cdot}$, and ${\cdot}OH$. In conclusion, the amelioration may be attributed to the synergistic effects of its constituents rather than to any single factor as the leaves are rich in tannins, sterols, flavonoids, saponins etc.

Prevalence of Anti-HCV among the Health-checkup Adults in Jeonbuk Province (전북 지역 건강 검진자들의 Anti-HCV 양성률 조사)

  • The author was performed to investigation of current status of prevalence for anti-hepatitis C virus (HCV) among the health-checkup adults in Jeonbuk province. A toal of 1,553 (male 1,046, female 507) serum samples were diagnosed by 3rd generation enzyme immunoassay (EIA) for anti-HCV. Total prevalence of anti-HCV was 0.9%, and prevalence of male and female were 0.8% and 1.2%, respectively. The prevalence of female was higher than male. According to ages group, prevalence of anti-HCV was highest in 60 age group, but it was not found in 20 age group. 14 samples with anti-HCV positive were diagnosed by EIA for hepatitis B virus surface antigen (HBs Ag), by chemiluminescence immunoassay (CLIA) for serum albumin, alanine transaminase (ALT) and asparagine transaminase (AST). Positive for HBs Ag was not found. The mean of serum albumin levels was 4.5 g/dL, and mean of ALT and AST were 34.3 IU and 31.9 IU, respectively. Through this study, I know that the prevalence of anti-HCV among adults in Jeonbuk, and suggest that the positive of anti-HCV persons who have lower serum albumin, normal to mild elevations in serum enzymes are chronic hepatitis.

  • Alcoholic extract of Terminalia arjuna [TA] was evaluated far its hepatoprotective activity against carbon tetrachloride (CCl$_4$)-induced hepatic damage in rats. The hepatoprotective activity of TA was evaluated by measuring levels of serum marker enzymes like serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), serum alkaline phosphatase (SALP). The serum levels of total proteins(TP), total albumins (TAL) and bilirubin (BILN) were also estimated. The histological studies were also carried out to support the above parameters. Silymarin (SM) was used as standard drug. Administration of TA (250 and 500 mg/kg/po) markedly prevented CCl$_4$-induced elevation of levels of SGOT, SGPT, SALP, TP, TAL and BILN. These biochemical observations were supplemented by histopathological examination of liver sections. Alcoholic extract of TA also shown significant in-vitro free radical scavenging activity against 1, 1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide (NO) radicals. Thus, the present study provides a scientific rationale for the traditional use of this plant in the management of liver diseases.

  • Serum glutamic oxaloacetic transaminase (GOT), latic dehydrgenase (LDH), and alkaline phosphatase (ALP) activities were determined in 5 Korean native cattle to obtain the base line of blood enzyme activities in relation to the stage of gestation. Serum GOT activities showed an increasing tendency with the course of gestation period, serum LDH levels were within normal range through all the stage of pregnancy, and the slight increase in serum ALP activities was acknowledged at the 2nd and 3rd stage of gestation.

Protective Effects of Extracts of Mori Cortex Radicis on Carbon Tetrachloride-induced Hepatotoxicity (사염화탄소에 의해 유발된 간독성에 대한 상백피 추출물의 간보호효과)

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride-treatment were markedly increased than normal control but those levels were significantly decreased by the treatment of butanol fraction of Mori cortex radicis mathanol extract. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

Effect of G009 on Lipid Peroxidation Induced by Peroxidizer in Rats

  • In order to elucidate the correlation between the lipid peroxidation and hepatotoxicity, the formation of malondialdehyde (MDA) in liver homogenate and serum, and the transaminase activities were determined in intoxicated by ascorbic acid-Fe$\^$2+/-ADP in rat. In a model of ascorbic acid-Fe$\^$2+/-ADP hepatotoxicity, G009, which was obtained from Ganoderma lucidum IY009, exhibited anti-lipid peroxidative effect in rat liver homogenate, and that MDA values of the liver homogenate decreased from 48.1% to 74.8% in comparision to controls (p<0.01) Also, the MDA formation in serum inhibited 66.5% at 100 mg/kg of G009. Serum levels of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) in peroxidizer-induced rats treated with G009 was decreased compared with control. Especially, The formation of lipid peroxides in serum was related to GPT levels. These results that G009 has a protective effect on ascorbic acid-Fe$\^$2+/-ADP-induced hepatic injury through an inhibition of lipid peroxidation in liver.

Effect of Soybean Powder on Lipid Metabolism and Enzyme Activities in Induced Hyperlipidemic Rats (대두분 급여가 고지혈증 흰쥐의 지질대사 및 효소 활성도에 미치는 영향)

  • To investigate the effects of yellow soybean powder on serum lipid level and enzyme activities, male Sprague-Dawley(SD) rats were given hyperlipidemic diets for 6 weeks. Rats(n=8) were divided into three groups consisting the normal group(N), hyperlipidemic diets group(C) and hyperlipidemic diets and yellow soybean powder group(YSP). YSP group lowered triglyceride, total lipid, phospholpid, ester-cholesterol and free-cholesterl than control group. YSP group lowered total-cholesterol, VLDL-cholesterol and LDL-cholesterol but increased HDL-cholesterol of the serum compared to the control group. Athergenic Index(AI) and LHR(LDL-cholesterol/HDL-cholesterol ratio) of YSP group was decreased by control group (p<0.05). On the other hand, glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GTP), lactate dehydrogenase(LDHase) and alkaline phosphatase(ALP) enzymes activities were decreased by hyperlipidemic diets group (C). These results suggest that yellow soybean powder fed a lipid lowering effect in serum of rats.

Hepatoprotective and antioxidant activity of Leea asiatica leaves against acetaminophen-induced hepatotoxicity in rats

  • Leea asiatica (L.) Ridsdale, a folk medicinal plant is used by the ethnic people of North East India for the treatment of hepatic disorder. In this study, we have investigated the hepatoprotective and antioxidant activity of L. asiatica leaves against acetaminophen induced hepatotoxicity. Methanol extract of L. asiatica (150 and 300 mg/kg/day, p.o.) were administered to rats for three consecutive days followed by single acetaminophen (3000 mg/kg, p.o.) administration on $3^{rd}$ day. After 48 h of acetaminophen administration animals were sacrificed and biochemical estimation of serum, in vivo antioxidant activity using liver tissue were carried out. High levels of serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, serum alkaline phosphatase, total bilirubin, direct bilirubin, total cholesterol and triglycerides were observed in disease control group, which found near to normal in extract treated groups. Higher dose exhibited significant hepatoprotective activity against acetaminophen induced toxicity. Level of superoxide dismutase, catalase, glutathione peroxidase in liver tissue, and reduced glutathione in liver and blood were also significantly increased in extract (300 mg/kg) treated animals compare to disease control group. In this study we found that leaves of L. asiatica exhibited potent hepatoprotective activity against acetaminophen induced hepatic damage in experimental animals which justify the folklore claim, and the possible mechanism of this activity may be due to strong antioxidant activities of extract.

Effects of Dietary Dae-Chu (Rhamnace ziziphus), Onion (Allium cepa L.), Mixture Extract on Serum Composition in Rats (대추, 양파, 혼합추출물이 흰쥐의 혈청조성에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary dae-chu(Rhamnace ziziphus, A), onion(Allium cepa L., O), mixture extracts (mulberry leaf, licorice root, pine needle, angelica gigas, jujube, onion, M) on serum glucose, lipid, enzyme, phosphorus levels in rats (Sprague-Dawley male rats, $357.03{\pm}7.08g$). Serum calcium of onion group was significantly decreased (p<0.05), but mixture extracts group of Cl (p<0.05) and TBIL (total bilirubin, p<0.05) were significantly increased. Serum glucose, total cholesterol, HDL-cholesterol and triglyceride were increased experimental rats than those of the normal rats. Mixture extracts was better than other groups for lipid metabolism. Also, GPT(glutamic pyruvic transaminase) and GOT(glutamic oxaloacetate transaminase) of onion extracts were protected to liver. So mixture and onion extracts were good drink for health.
