Chromium is one of the representative toxic substance by occupational exposure which damage the mucosa of respiratory tract including nasal septal perforation. The aim of this study is to evaluate the effect of chromium exposure on olfactory function and to obtain the fundamental information about chromium exposure. The authors performed olfactory function test, laboratory tests and questionnaire interview on the subject of three groups, that is, two exposed groups and one nonexposed group from May 1 to June 30, 1994. Exposed group 1 was 15 male workers without nasal septal perforation, exposed group 2 was 15 male workers with nasal septal perforation among 103 workers in 22 chromium plating factories, and nonexposed group was 15 male medical students. The gathered informations were histories of chromium exposure, habits of smoking and alcohol drinking, the concentrations of chromium in serum and urine, and asparate aminotransferase(AST), alanine aminotransferase(ALT), gamma-glutamyl transferase, etc. Olfactory function was checked by T and T olfectometer using phenyl ethyl alcohol(material A), methyl cyclopentenolone(material B), iso-valeric acid(material C), $\gamma$-undecalactone(material D), skatole(material E) and the results were expressed by detection threshold(DT) and recognition threshold(RT). There was a significant difference between exposed groups and nonexposed group in A, B, C, D, E substances by DT and in A, B, C, D substances by RT(P<0.01). The degree of olfactory dysfunction was highest in the exposed group 2 and lowest in the nonexposed group in all five substances by DT and it was same in A B, D substances RT and the difference of RT and DT. As summary, olfactory dysfunction by chromium exposure was recognized and the degree of olfactory dysfunction was higher in the exposed group with nasal septal perforation. Therefore, it would be helpful to apply olfactory function test for the early detection of olfactory dysfunction, and this test would be considered as the basic tool within workers' compensation system.
타우린은 심혈관 질환을 예방하고 간 기능 개선 및 당뇨병 및 혈소판 기능을 향상시키는 여러 작용을 보고하고 있다. 하지만, 타우린이 인체에 미치는 영향에 대하여 우리나라에서 연구된 결과가 많지 않다. 이에 기본 용량을 복용 후 혈당질환과 간질환, 지질 질환에 대한 영향을 알아보고자 하였다. 대상자는 타우린 복용군 15명과 대조군 15명을 대상으로 시행하였다. 타우린은 기본 용량인 1,000 mg을 식후에 2주 동안 복용 후 변화를 확인하였다. 대상자 모두 기숙사에서 제공하는 식사 외에 약이나 기타 음식을 절제하도록 하였다. 그 결과 타우린 복용군에서 간 기능 검사인 GGT는 섭취 전 $23.53{\pm}25.73IU/L$, 섭취 후 $15.15{\pm}4.91IU/L$로 감소하였다(P=0.186). 지질 대사인 TG는 섭취 전 $100.4{\pm}28.33mg/dL$, 섭취 후 $80.22{\pm}17.08mg/dL$로 유의한 결과를 보였다(P<0.05). T-cho, LDL-C이 감소를 보였고, HDL-C이 약간 증가를 보였다. 결과적으로 간 기능과 지질대사 개선에 도움을 주는 것으로 나타났다. 혈액학적 검사에서는 segmented neutrophil 백분율이 감소하고, lymphocyte 백분율이 증가를 보였다. 결과적으로 면역학적 기능과 관계가 있으리라 사료된다.

An experiment was conducted to determine the effects of different mycotoxin adsorbents including esterified glucomannan (EGM), hydrated sodium calcium aluminosilicate (HSCAS) and compound mycotoxin adsorbent (CMA) on performance, blood parameters, and liver pathological changes in broilers fed mold-contaminated feed. Two hundred and forty 10-day-old broilers were randomly assigned to one of the five dietary treatments including: i) control diet; ii) mold-contaminated diet; iii) moldcontaminated diet+0.05% EGM; iv) mold-contaminated diet+0.2% HSCAS; v) mold-contaminated diet+0.1% CMA. At 35-days-old, blood and liver tissue samples were collected for analysis. 0.1% CMA improved ADG and ADFI during 10-42 d compared to the moldcontaminated group (p<0.05). The mold-contaminated diet increased total white blood cell (WBC) number, haemoglobin (Hgb) concentration, hematocrit (Hct) level, serum aspartate aminotransferase (AST) and ${\gamma}$-glutamyl transferase (GGT) activities, and decreased red blood cell (RBC) number and serum globulin (GLB) and urea nitrogen (BUN) concentrations (p<0.05). The three mycotoxin adsorbents alleviated the alteration of RBC, WBC, Hgb and AST caused by the mold-contaminated diet. Furthermore, 0.1% CMA increased GLB concentration and decreased Hct level and GGT activity (p<0.05). Liver superoxide dismutase (SOD) activity was reduced, and myeloperoxidase (MPO) activity was increased by the mold-contaminated diet (p<0.05). Both EGM and HSCAS prevented the increase of MPO activity (p<0.05). Liver lesion, including severe vacuolar degeneration of hepatocytes, was observed in chicks fed the mold-contaminated diet. 0.05% EGM prevented these effects except for biliary hyperplasia and mild vacuolar degeneration. 0.2% HSCAS showed medium vacuolar degeneration of hepatocytes. Liver of broilers fed 0.1% CMA revealed a mild vacuolar degeneration. These results indicate that a mold-contaminated diet results in adverse effects on blood parameters and liver morphology. 0.05% EGM and 0.2% HSCAS partially alleviated the adverse effects. However, 0.1% CMA almost completely ameliorated the adverse effects.
본 연구에서는 다양한 약리 활성 및 항산화 활성이 높다고 보고된 신선초 추출물을 대상으로 in vitro 및 in vivo계에서 알코올성 간세포 손상을 유도하여 그 효과를 검토하였다. 알코올 산화 효소인 cytochrome P4502E1(CYP2E1)이 과발현된 HepG2 세포에서 200 mM의 알코올과 신선초 추출물을 투여하였을 때 농도 의존적으로 세포생존률 및 catalase(CAT) 활성이 증가하였다. In vitro에서 신선초 추출물의 보호 효과를 확인한 후 7주령의 C57BL/6J 마우스에 알코올과 신선초 추출물 20, 100 mg/kg BW/d를 급여한 결과 혈중 ALT, AST, GGT의 농도는 대조군보다 유의적으로 증가한 알코올군에 비해 신선초 추출물을 급여한 군에서 유의적 감소했음을 확인할 수 있었다. 그뿐만 아니라 알코올 투여로 세포변성과 지방구가 보이는 간 조직의 변화가 신선초 투여로 의해 대조군과 유사하게 관찰되었다. 항산화 효소의 변화와 지질과산화 수준은 대조군보다 알코올군이 유의적 증가했으며, 신선초 추출물 급여군에서 감소하는 결과를 보였고, 특히 신선초 20 mg/kg BW/d로 급여한 군에서 CAT, glutathione peroxidase, glutathione reductase, malondialdehyde(MDA) 등의 유의적 감소를 보였다. 이 같은 변화를 매개했다고 생각되는 CYP2E1의 발현과 활성은 대조군보다 알코올 투여군에서 유의적으로 상승하였으며, 특히 항산화 효소와 MDA 함량에서 유의적 감소했던 20 mg/kg BW/d의 신선초 투여군에서 유의적 감소하는 것으로 나타났다. 그러므로 이는 신선초 추출물에 함유된 luteolin, quercetin, chalcone 화합물 등의 성분에 의해 알코올 유도 산화적 스트레스가 감소하였다고 생각되며, 신선초 추출물은 알코올 대사과정에서 산화적 스트레스를 억제하여 간 보호 효과가 있을 것으로 생각한다.
This study was conducted to investigate the effects of dietary Allium hookeri on growth performance, bone strength, and blood biochemical profiles in growing broiler chickens. Twelve hundreds of one-day old Arbor Acres male broilers were divided into 6 treatments with 4 replicates and 50 birds per replicate (n=200 chicks/treatment). Chickens fed basal diet (Control), basal diet with commercial X (Positive control) at 0.05% of diet, or each one of the experimental diets (L3, L5, R3, R5) supplemented with the powder of A. hookeri leaf or root at 0.3 and 0.5% of diet respectively for 5 weeks. At the 5th week of feeding the diets, body weight, tibia strength, and blood biochemical profiles including antibody titers were measured. Dietary A. hookeri (L3, L5, R3, R5) significantly increased final body weight than the control group. And the dietary leaf of A. hookeri effectively increased the growth performance than dietary root of A. hookeri. Interestingly dietary leaf of A. hookeri improved tibia strength than the control group and L3 showed the highest value. The antibody titers against infectious bursal disease (IBD) increased with the addition of dietary leaf of A. hookeri compared with positive control, R3, and R5 groups. But there was no significant difference in serum biochemical parameters such as albumin, globulin, glucose, cholesterol, Ca, P, total protein, total bilirubin, alanine aminotransferase, alkaline phosphatase, and gamma-glutamyl transferase. These results suggest that A. hookeri can be used as a good supplement to improve growth performance and health by increasing bone strength and antibody titer against IBD without any anti-nutritional or toxic effects in growing broilers.
Metabolic profile test is used to evaluate nutritional imbalance and metabolic disease in dairy cows. The reference intervals of metabolic parameters may change according to nation, region, decades, and maintenance system. Despite the need to be periodically updated for the reference intervals of metabolic parameters, it has rarely been investigated in Korea. Therefore, this aim of study was to provide the reference intervals of metabolic parameters using dairy cows surveyed in Korea during recent years. A metabolic profile test was conducted for 2,976 clinically healthy dairy cows in Korea. Blood samples were collected for the analysis of serum metabolites. This study provided reference intervals of thirteen metabolic parameters (${\beta}$-hydroxybutyrate [${\beta}-HB$], non-esterified fatty acids [NEFA], glucose, total cholesterol [T-COL], total protein, albumin, globulin, blood urea nitrogen [BUN], aspartate aminotransferase [AST], gamma-glutamyl transferase [GGT], calcium, phosphorus, and magnesium). BUN and AST values of the current study were higher than those of previous studies. In the present study, the other metabolic parameters showed low or similar value compared to previous results. Moreover, ${\beta}-HB$, NEFA, T-COL, ALB, BUN, AST, and GGT values were affected by lactation period. This study provided information on the reference intervals of metabolites in healthy dairy cows in Korea. The reference intervals from the present study would be useful in managing and diagnosing disease of dairy cows. However, careful attention should be given in interpreting disease condition for metabolites affected by lactation.
The present study evaluated the effect of controlled ryegrass-white clover herbage availability from day 128 until day 142 of pregnancy in comparison to unrestricted availability, on the performance of twin-bearing ewes of varying body condition score (BCS; 2.0, 2.5, or 3.0) and their lambs. It was hypothesised that under conditions of controlled herbage availability, the performance of lambs born to ewes with a greater BCS would be greater than those born to ewes with a lower BCS. During the period that the nutritional regimens were imposed, the pre- and post-grazing herbage masses of the Control regimen ($1,070{\pm}69$ and $801{\pm}30$ kg dry matter [DM]/ha) were lower than the ad libitum regimen ($1,784{\pm}69$ and $1,333{\pm}33kg\;DM/ha$; p<0.05). The average herbage masses during lactation were $1,410{\pm}31kg\;DM/ha$. Nutritional regimen had no effect on ewe live weight, BCS and back fat depth or on lamb live weight, indices of colostrum uptake, maximal heat production, total litter weight weaned or survival to weaning (p>0.05). The difference in ewe BCSs and back fats observed among body condition groups was maintained throughout pregnancy (p<0.05). At weaning, ewes from the BCS2.0 group had lower BCS and live weight ($2.4{\pm}0.2$, $74.3{\pm}2.6kg$) than both the BCS2.5 ($2.6{\pm}0.2$, $78.6{\pm}2.4kg$) and BCS3.0 ewes ($2.7{\pm}0.2$, $79.0{\pm}2.6kg$; p<0.05), which did not differ (p>0.05). Ewe BCS group had no effect on lamb live weight at birth or weaning or on maximal heat production (p>0.05). Serum gamma glutamyl transferase concentrations of lambs born to BCS3.0 ewes were higher within 36 hours of birth than lambs born to BCS2.0 ewes and BCS2.5 ewes ($51.8{\pm}1.9$ vs $46.5{\pm}1.9$ and $45.6{\pm}1.9IU/mL$, respectively [p<0.05]). There was, however, no effect of ewe body condition on lamb plasma glucose concentration (p>0.05). Lamb survival was the only lamb parameter that showed an interaction between ewe nutritional regimen and ewe BCS whereby survival of lambs born to BCS2.5 and BCS3.0 ewes differed but only within the Control nutritional regimen ewes (p<0.05). These results indicate farmers can provide twin-bearing ewes with pre- and post-grazing ryegrass-white clover herbage covers of approximately 1,100 and 800 kg DM/ha in late pregnancy, provided that herbage covers are 1400 in lactation, without affecting lamb performance to weaning. The present results also indicate that under these grazing conditions, there is little difference in ewe performance within the BCS range of 2.0 to 3.0 and therefore they do not need to be managed separately.
본 연구는 산채류 중 널리 섭취되고 있는 곰취의 활용가치를 높이고 건강기능식품 소재로써의 가능성을 확인고자 DGalN 동물모델을 통해 곰취 열수 추출 발효물의 간 기능 개선 효과를 연구하였다. 실험기간 동안 실험동물의 체중 증가량은 처리군 간 유의적인 차이가 없었으며(P>0.05), 간 무게는 곰취 열수 추출물 투여군에서 유의하게 증가하는 것으로 나타났다(P<0.05). 혈청 중 GGT 분석 결과 곰취 열수 추출물과 발효물 투여군에서 대조군과 비교하여 유의하게 감소하였다(P<0.05). 특히 발효물 투여군의 경우 GGT, AST, ALT 및 LDH의 효소 활성이 대조군과 비교하여 40% 이상 감소하였다(P<0.05). 총 콜레스테롤의 함량을 측정한 결과 정상식이군과 비교하여 모든 투여군에서 유의하게 감소하였으며(P<0.05), HDL-콜레스테롤의 경우 발효물 투여군에서 대조군과 비교하여 유의하게 증가하였다(P<0.05). LDL-콜레스테롤은 농도 의존적으로 유의하게 감소하였다(P<0.05). 중성지방의 함량은 발효물 투여군에서 대조군과 비교하여 유의하게 감소하였으며(P<0.05), 총 콜레스테롤 함량 중 HDL-콜레스테롤 함량의 비율은 발효물 투여군에서 대조군과 비교하여 유의하게 증가하는 것으로 나타났고(P<0.05), 동맥경화지수의 경우 발효물 투여군에서 대조군과 비교하여 유의하게 감소하였다(P<0.05). 발효물의 경우 SOD 활성이 유의하게 증가하였으며(P<0.05), 간 조직 내 과산화지질의 분해산물인 MDA 함량은 곰취 열수 추출물 및 발효물 투여군에서 대조군과 비교하여 MDA 함량이 유의하게 감소하였다(P<0.05). 간 조직 병리학적 분석 결과 곰취 열수 추출물 및 발효물 투여군에서 대조군과 비교하여 세포사멸사 지수, 문맥엽 염증 지수 등이 개선되는 경향을 보였으나 유의적인 차이는 없는 것으로 나타났다(P>0.05). 이상의 결과로 보아 곰취 열수 추출 발효물이 간 기능 효소의 활성 억제, 혈청지질 대사 개선, 간 조직 내 SOD 활성증가 및 과산화지질을 감소시켜 D-GalN으로 인한 간 손상을 완화시키는 데 효과적일 것으로 사료되며, 향후 건강기능 식품 소재로 이용 가능할 것으로 판단된다.
