Effect of golden needle mushroom (Flammulina velutipes) stem waste on laying performance, calcium utilization, immune response and serum immunity at early phase of production

  • Objective: This experiment was conducted to evaluate the effects of golden needle mushroom (Flammulina velutipes) stem waste (FVW), on organic eggs production, calcium utilization, antibody response, serum immunoglobulin, and serum cytokine concentration at early phase of production in laying hens. Methods: A total 210, 19 weeks old aged ISA Brown layers were randomly assigned into 5 equal treatment groups, with 7 replications of 6 hens each. Dietary treatment included a standard basal diet as control; antibiotic (0.05% flavomycin); 2% FVW; 4% FVW; and 6% FVW. The experimental duration was 10 weeks. Results: There was no significant differences (p>0.05) on hen day egg production, egg weight, egg mass, feed intake, and feed conversion ratio (FCR) among experimental groups. Unmarketable eggs were significantly lower (p<0.05) both in 4% FVW and 6% FVW fed groups than control group. The calcium retention and calcium in egg shell deposition were significantly higher (p<0.05) in FVW inclusion groups than control and antibiotic groups. Antibody titers against Newcastle diseases were significantly higher (p<0.05) in 6% FVW fed group (except combined with 4% FVW at day 147) and infectious bronchitis were significantly higher (p<0.05) in FVW fed groups (except 2% FVW and 4% FVW at day 161) than control and antibiotic groups. Serum immunoglobulin sIgA was significantly higher (p<0.05) in all levels of FVW and IgG was significantly higher (p<0.05) in 4% FVW than control and antibiotic groups. Serum cytokine concentration interleukin-2 (IL-2) was significantly higher (p<0.05) in 6% FVW; IL-6 and tumor necrotic $factor-{\alpha}$ were significantly higher (p<0.05) both in 4% FVW and 6% FVW than control and antibiotic groups; IL-4 was significantly higher (p<0.05) in antibiotic, 2% FVW and 4% FVW fed groups than control. Conclusion: F. velutipes mushroom waste can be used as a novel substitute for antibiotic for organic egg production and sound health status in laying hens.

Effects of Dietary Protein and Calcium levels on Ca, Fe, Cu, Zn and Mg Level of the Tissues of the Pb-administered Rats (단백질과 칼슘의 급여 수준이 납중독된 흰쥐의 체내 무기질함량에 미치는 영향)

  • This study was designed to investigate the effects of dietary protein and calcium levels on Ca, Fe, Cu, Zn and Mg accumulation of the tissue of the Pb-administered rats. Male Sprague-Dawley rats were assigned to a 3$\times$3$\times$2 factorial design with 3 levels of protein (40%, 15%, 6%), 3 levels of Ca (1.2%, 0.6%, 0.12%) and 2 feeding periods (3 and 7 weeks). The control group was included separately. The rats were exposed to the drinking water containing 2,000ppm of lead. Calcium contents in serum, liver and femur were decreased with Pb administration. Calcium contents in serum and femur were reduced with dietary protein and Ca levels. Level of serum Fe showed no significant difference with Pb administration in the rats fed the high Ca diet. Iron content in liver was not affected by the lead when the rats fed the diet containing high protein and Ca. Level of serum Cu was lower in the Pb added groups than in the control group and tended to be reduced with decreasing dietary protein and Ca levels. Copper content in liver was not affected by the lead when the rats fed the high Ca diet. Level of serum Zn was decreased in the low protein-low Ca group. Magnesium content in serum was decreased with Pb administration when the rats fed the diet containing low protein and Ca. However, magnesium content in liver was reduced with Pb administration and affected only by dietary protein level.

  • Milk-alkali syndrome (MAS), a triad of hypercalcemia, metabolic alkalosis, and renal failure, is associated with ingestion of large amounts of calcium and absorbable alkali. MAS is the third most common cause of hypercalcemia in hospital, after primary hyperparathyroidism and malignant neoplasm. MAS is not often reported in the Korean literature. We describe MAS secondary to intake of calcium citrate for the treatment of osteoporosis with thoracic spine compression fracture. A 70-year-old man presented to our hospital with a 1-week history of general weakness and lethargy. He was found with acute kidney injury (serum creatinine, 4.6 mg/dL), hypercalcemia (total calcium, 14.8 mg/dL), and alkalosis. Laboratory evaluation excluded both hyperparathyroidism and malignancy. Mental status and serum calcium level was normalized within a week after proper hydration and intravenous administration of furosemide. However, he developed aspiration pneumonia, pseudomembranous colitis, and sepsis with multi-organ failure. Despite intensive treatment including inotropics, mechanical ventilation, and renal replacement therapy, he expired with no signs of renal recovery on the 28th hospital day.

  • The role of estrogen in calcium metabolism has not been well documented. To further investigate the effects of endogenous estrogen on the calcium regulating hormones in women, we correlated the changes of estrogen level with those of calcium regulating hormones during ovulation induction in 12 hypogonadal and 8 normally menstruating women. During ovulation induction, the serum leveles of estradiol rose from 40.0 to 831.0pg/ml. There were no significant changes in the serum leveles of total calcium, inorganic phosphorus, parathyroid hormone, calcitonin. However, 1, $25-(OH)_2D_3$ rose significantly from 31.0 to 47.2pg.ml as the endogenous $E_2$ increaed (p<0.005).

  • This study was designed to investigate the effects of dietary protein and caffeine consumption levels on Ca, P, Na and K metabolism. One hundred twenty rats were divided into twelve groups according to age, protein level and caffeine consumption such as group(120-130g young rat, 250-300g adult rat) , dietary protein group (20% normal protein , 85 low protein) , caffeine consumption group(0, 3.5mg, 7.0mg). Low protein diet containing high caffeine levels caffeine levels increased calcium, phosphorus, sodium and potassium contents of urine and fecal in rats. Young rat has higher level of calcium, phosphorus , sodium and potassium than adult rat. In the serum, calcium contents were not affected by age, dietary protein levels and caffeine consumption . However, phosphorus, sodium and potassium contents of serum in adult rat were higher than young rat. In the liver, potassium decreased with decreasing dietary protein levels. In the kidney, calcium , phosphorus and potassium contents were not different by age group, dietary protein levels and caffeine consumption , but sodium content was significantly reduced in the adult rat.

Effects of Chitosan on Mineral Metabolism in Rats Exposed to Cadmium (카드뮴 투여 흰쥐에서 키토산 섭취가 무기질 대사에 미치는 효과)

  • This study investigated the effects of chitosan on mineral metabolism in rats exposed to cadmium by oral administration. Six week-old Sprague-Dawley rats were divided into eight groups. Four groups were fed AIN-93G based 3% ${\alpha}$-cellulose diets and the other four groups were fed 3% chitosan diets for four weeks with the oral administration of 0, 0.5, 1.0, 2.0 mg Cd/2ml distilled water three times per week, respectively. The essential mineral contents of serum, liver, kidney and bone (femur and lumbar), and the excretion of calcium in feces and urine were determined. There was no significant difference in weight gain and food intake among groups. The cadmium administration significantly decreased calcium in serum, iron in blood, calcium and iron in liver and iron contents in kidney. In contrast, calcium and zinc contents in kidney increased by the administration of cadmium. The weight, length and breaking forces of the femur and lumbar were not significantly different due to cadmium administration and chitosan among the groups. The fecal excretion of calcium was increased by the administration of cadmium. On the other hand, calcium absorption and the absorption rate were decreased by Cd administration. In the groups without Cd administered (N, N-Chi groups), chitosan significantly decreased the absorption rate of calcium by increasing the excretion in feces (p<0.05). These results suggest that cadmium administration may facilitate the decline of essential minerals in rats and also, chitosan may have a conflicting effect between cadmium and the essential minerals of tissues.

Effect of Calcium Channel Blocker on Acetaminophen-induced Hepatotoxicity in Rats and Histopathologic Examination (Acetaminophen에 의해 유도된 흰주의 간손상에 미치는 Calcium channel Blocker의 효과 및 조직학적 소견)

  • The acetaminophen (AP AP), an antipyretic and analgesic agent, induces the hepatotoxicity by increasing influx of calcium and destabilizing the cellular membrane which can be caused by N-acetyl-p-benzoquinoneimine generated by cytochrome P-450 (CYF-450) when it is overdosed. Diltiazem (DIL), a calcium channel blocking agent, has been known to suppress the CYF-450 activities. To study the effect of DIL in APAP treated rats, the serum biotransformational enzyme analyses and the liver histopathologic examination were conducted on the rats which had been administered DIL at 3, 6, 9 and 12 hours after the 3,000 mg/kg of APAP administration. Following a single dose of DIL administered 12 hours after AP AP administration, serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities, malondialdehyde and calcium contents of liver and microsome were significantly reduced. Glutathione S-transferase (GST) activity was significantly increased. Histopathologic studies showed that DIL had prevented the development of centrilobular necrosis induced by AP AP in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radical and the influx of calcium and could increase GST activity. Therefore, diltiazem can be administered at the time of 12 hours after overdosed AP AP to diminish the liver damage.

Effects of Caffeine Intake on Calcium Utilization in Rats of Different Age and Sex (카페인 섭취수준이 연령이 다른 흰쥐의 체내 칼슘 이용에 미치는 영향)

  • The purpose of this research was to investigate the effect of caffeine levels on calcium utilization in rats of different age and sex. Calcium utilization was compared in rats of different age(4 weeks and 10 months) and sex that were fed various levels of caffeine (0, 3.5, and 7mg/100g body weight) for 3 weeks. There was no significant difference in feed intake, serum calcium level, and ash content in tibia among the groups. Fecal calcium excretion was lower in young rats than in adults, Urinary calcium excretion significantly higher in the caffeine groups than that in the no-caffeine group. Daily retention and apparent absorbability of calcium in young rats were higher than those in adult rats. However, there was no significant difference among groups of different sex and caffeine levels. The results of this study suggest that caffeine consumption promotes urinary calcium excretion.

Comparison of Nutrient Intakes, Serum Minerals and Lipids between Physical Education Major and Non-major Students (체육전공과 비전공 남학생의 영양섭취, 혈청 무기질과 지질 함량 비교)

  • The purpose of this study was to investigate daily nutrient intakes, serum minerals and lipids in university students majoring in physical education. Anthropometric measurement, dietary survey, and blood analysis were conducted and compared between physical education major (PM) and other major (OM) students. Average age was 21.3 years for PM students and 20.8 years for OM students. Body weight and BMI of PM students were significantly higher than those of OM students (p<0.05). Systolic blood pressure of PM students was significantly lower than that of OM students (p<0.05). Daily energy intake was 2,047.4 kcal for PM students and 2,393.9 kcal for OM students, indicating a significant difference. Carbohydrate and vitamin B1 intakes of PM students were significantly lower than those of OM students. Serum calcium was significantly higher in PM students while serum phosphorus and magnesium levels were higher in OM students. Serum total cholesterol and LDL-cholesterol were significantly lower in PM students while HDL-cholesterol was lower in OM students. In conclusion, university students who major in physical education showed higher serum calcium, lower serum phosphorus and magnesium, and lower systolic blood pressure and serum cholesterol levels than other major students. These results show that physical activity as a major course of study is associated with decreased blood pressure and blood lipids as well as altered serum mineral levels.

Relationship Among Nutritional Intake, Duration of Outdoor Activities, Vitamin D Status and Bone Health in High School Girls

  • This study investigated the interactions of bone health with several variables such as outdoor activity hours, nutritional status including habitual intake of calcium andvitamin D status in 72 high school girls aged 16-17 yearsattending day classes or night classes. The subjects consisted of 39 day-class students and 33 night-class students. Dietaty nutrient intakes were estimated using the 24-hour recall method. The daily activities of each subject were assessed using an activity questionnaire. Urinary calcium and creatinine excretion were assayed from subjects' 24-hoururine, while 25-OH-vitamin D[25-(OH)-D] and osteocalcin were measured from the subjects' fasting blood. Intake of energy, iron, vitamin A and vitamin C were worse in the night-class students. There was no significant difference in dietary calcium between the subjects in the different class types. Time spent on outdoor activities was significantly less in subjects attending night classes. Urinary calcium excretion of the night-class subjects was significantly higher than that of the day-class subjects (p<0.05). There was no significant difference in serum 25-(OH)-D level according to class type. Serum osteocalcin for night-class subjects was significantly higher than that for day-class subjects (p<0.01). It appeared that the night-class students had poorer dietary habits as well as fewer outdoor activities. Even though the estimated bone health of both groups of subjects appeared to be normal, the overall nutritional intake and duration of outdoor activities appeared to be important for maintaining bone health and lowering the future risk of osteoporosis.
