Effect of Working on Physiological, Biochemical and Haematological Parameters in Hariana Bullocks

  • An experiment was conducted on eight bullocks of Hariana cattle study draft efficiency using different appliances, changes in their physiological, biochemical and haematological parameters, and how the body tries to maintain these within normal physiological range. Blood collection was done at pre-exercise, three hours after exercise and two hours post exercise in summer and winter seasons of the year 1990-91. Average pulse rate and respiration rate per minute were found to be $63.09{\pm}0.78,\;25.60{\pm}0.54,\;97.21{\pm}2.51,\;63.37{\pm}2.70,\;64.05{\pm}1.90,\;25.33{\pm}1.20$, respectively, at pre-exercise, three hours exercise and two hours post exercise rest. The seasonal differences were found to significant and the values were higher in summer than in winter. After two hours post exercise rest Hariana bullocks regained their normal values for all the parameters, except that pulse rate, respiration rate and serum sodium during summer did not reach the pre - exercise levels.

  • Hematological and serum biochemical analyses are routinely used to screen dogs for disease and monitor their clinical progression. These values in dogs may be affected by internal factors, such as breed and age, and external factors like environment, diet, and lifestyle. Apparently, subclinical breed-related variations in hematologic and serum biochemical results have been characterized for Bernese Mountain dogs, Alaskan Malamutes, English Setters, and Golden Retrievers. It is possible that some breed-related differences in clinical pathology analyses may reflect evidence of underlying disease and impact clinical decision plan. In this study, we estimated hematologic and serum biochemical parameters in small breed dogs and compared among four different breed dogs. Blood samples were collected from 38 domestic dogs that were 10 Beagles, 9 Malteses, 10 Poodles, and 9 Miniature Schnauzers. In terms of RBC count, Beagle ($7.2{\pm}0.24{\times}10^3cells/{\mu}L$) showed significantly (P<0.05) higher count than other breeds (Maltese, $6.51{\pm}0.25{\times}10^3cells/{\mu}L$; Poodle, $6.35{\pm}0.24{\times}10^3cells/{\mu}L$; and Miniature Schnauzer, $6.35{\pm}0.25{\times}10^3cells/{\mu}L$). For MCV, Miniature Schnauzer ($67.05{\pm}0.84fl$) results were significantly (P<0.05) higher than Beagle with $64.33{\pm}0.8fl$. For cholesterol value, Beagle ($163.3{\pm}9.15mg/dl$) had significantly (P<0.05) higher values than Miniature Schnauzer with $119.12{\pm}9.64mg/dl$. There was no statistically difference among breeds in terms of enzyme markers of liver diseases. In conclusion, data obtained from this study may be valuable as breed-related variability for interpretation of the results in hematologic and serum biochemical analysis among four small breed dogs.

Cardiocirculatory, biochemical and hemostatic evaluation of dogs with hyperadrenocorticism at diagnosis and after treatment

  • Hyperadrenocorticism (HAC) is a common endocrinopathy among dogs that causes multisystemic signs. This study was conducted to evaluate cardiocirculatory, biochemical, and hemostatic parameters in dogs with HAC at diagnosis, in addition to verifying whether abnormal parameters could be controlled by initial treatment with trilostane. Fifteen dogs with HAC were assessed by systolic blood pressure measurement, electrocardiography, Doppler echocardiography, serum concentration of troponin I, and biochemical and hemostatic profile at diagnosis and after trilostane therapy. Unlike biochemical parameters, hemostatic and cardiocirculatory parameters were not significantly influenced by the onset of treatment. The authors believe that clinical treatment with trilostane for 3 to 4 months might not be sufficient for the stabilization of cardiocirculatory abnormalities such as hypertension. Therefore, dogs with HAC must receive cardiocirculatory monitoring at diagnosis and during drug treatment.

Effects of supplemented culture media from solid-state fermented Isaria cicadae on performance, serum biochemical parameters, serum immune indexes, antioxidant capacity and meat quality of broiler chickens

  • Objective: The objective of this study was to investigate effects of supplementation of culture media from solid-state fermented Isaria cicadae (I. cicadae) on performance, serum biochemical parameters, serum immune indexes, antioxidant capacity and meat quality of broiler chickens. Methods: A total of 648 Arbor Acres male broiler chickens(1 d; average body weight, 42.93±0.47 g) were randomly assigned to 6 treatments, each with six replicates and 18 broiler chickens per replicate. Broiler chickens were fed phase I (d 1 to 21) and phase II (d 22 to 42) diets. The phase I diets were corn and soybean-meal based diets supplemented with 0%, 2%, 4%, 6%, 8%, or 10% culture media from solid-state fermented I. cicadae respectively. The phase II diets were corn and soybean-meal based diets supplemented with 0%, 1.33%, 2.67%, 4.00%, 5.32%, or 6.67% culture media from solid-state fermented I. cicadae respectively. Results: In phase I, the broiler chickens with the supplementation of culture media had increased body weight gain and feed intake (linear and quadratic, p<0.05) with increasing inclusion of culture media. The levels of serum total antioxidant capacity (T-AOC), glutathione (GSH) and superoxide dismutase (SOD) increased linearly (p<0.05). In phase II, levels of serum T-AOC and interleukin-1β increased linearly (p<0.05), and GSH increased (p<0.05). In the kidney, GSH and glutathione peroxidase (GSH-Px) concentrations increased (linear and quadratic, p<0.05) and SOD concentration increased linearly (p<0.05). Compared to the control, shear force and drip loss of breast muscle decreased (linear and quadratic, p<0.05). Drip loss of leg muscle decreased linearly and quadratically (p<0.05). Conclusion: Dietary supplementation of culture media from solid-state fermented I.cicadae which was enriched in both wheat and residual bioactive components of I. cicadae enhanced the growth performance of broiler chickens. It also improved body anti-oxidative status and contributed to improve broiler meat quality.

Removing Lipemia in Serum/Plasma Samples: A Multicenter Study

  • Background: Lipemia, a significant source of analytical errors in clinical laboratory settings, should be removed prior to measuring biochemical parameters. We investigated whether lipemia in serum/plasma samples can be removed using a method that is easier and more practicable than ultracentrifugation, the current reference method. Methods: Seven hospital laboratories in Spain participated in this study. We first compared the effectiveness of ultracentrifugation ($108,200{\times}g$) and high-speed centrifugation ($10,000{\times}g$ for 15 minutes) in removing lipemia. Second, we compared high-speed centrifugation with two liquid-liquid extraction methods-LipoClear (StatSpin, Norwood, USA), and 1,1,2-trichlorotrifluoroethane (Merck, Darmstadt, Germany). We assessed 14 biochemical parameters: serum/plasma concentrations of sodium ion, potassium ion, chloride ion, glucose, total protein, albumin, creatinine, urea, alkaline phosphatase, gamma-glutamyl transferase, alanine aminotransferase, aspartate-aminotransferase, calcium, and bilirubin. We analyzed whether the differences between lipemia removal methods exceeded the limit for clinically significant interference (LCSI). Results: When ultracentrifugation and high-speed centrifugation were compared, no parameter had a difference that exceeded the LCSI. When high-speed centrifugation was compared with the two liquid-liquid extraction methods, we found differences exceeding the LCSI in protein, calcium, and aspartate aminotransferase in the comparison with 1,1,2-trichlorotrifluoroethane, and in protein, albumin, and calcium in the comparison with LipoClear. Differences in other parameters did not exceed the LCSI. Conclusions: High-speed centrifugation ($10,000{\times}g$ for 15 minutes) can be used instead of ultracentrifugation to remove lipemia in serum/plasma samples. LipoClear and 1,1,2-trichlorotrifluoroethane are unsuitable as they interfere with the measurement of certain parameters.

Paraquat 독성에 미치는 Glycyrrhizin의 효과 (Effect of Glycyrrhizin on Paraquat Intoxication)

  • The herbicide, paraquat, has caused numerous poisonings in man and farm animals since its interoduction. Althought the lungs are the primary sites of toxicological effect, other organs are also damaged, such as liver, heart, and kidney. The biochemical mechanisms responsible for the toxicity of the herbicide are not clearly understood. The purpose of the present study was to determine the hepatotoxity of paraquat in rats and the effects of glycyrrhizin on the paraquat toxicity. The experimental results are the followings. 1. The values of serum AST, ALT, LDH, ALP, glucose, and cholesterol were significantly increased in the paraquat administered group. 2. The TBA values in serum and liver were also increased in the treated group. 3. The biochemical parameters of serum, such as AST, ALT, LDH and TBA values, were significantly low in the paraquat plus glycyrrhizin group in comparison with that of the paraquat only. The above results suggest that the glycyrrhizin, somehow, detoxify the hepa to toxicity of paraquat in the experimenta-animals.

Dietary supplementation with different types of fiber in gestation and lactation: effects on sow serum biochemical values and performance

  • Objective: Three types of dietary fiber were fed to sows during gestation and lactation stages to monitor their physiological and metabolic adaptations during the pre-partum period and to determine how these effects may influence the lactation period and sow performance. Methods: Soon after breeding, 54 sows were selected and were fed with 20% supplementation as fed of wheat bran (WB), soya hulls (SH), or rice hulls (RH) in diets during gestation and lactation. Sows were weighed, backfat thickness was measured ultrasonically and jugular blood samples were collected from all sows. The litter size was equalized to 10, by fostering piglets from sows on the same treatment. Results: Sows gained 22.0, 21.8, and 25.5 kg of net maternal body weight during gestation (for WB, SH, and RH sows, respectively; p = 0.007). There was no treatment effect on the body weight change during lactation (p = 0.158), however RH sows consumed an average of 133.66 kg of feed, WB sows took 121.29 kg and SH sows took 126.77 kg during lactation (p<0.001). The SH litters gained an average of 59.34 kg of weight during lactation, while other litters gained 51.58 and 49.98 kg (for WB and RH litters, respectively; p<0.001). Exception for aspartate aminotransferase and alanine aminotransferase, measured serum biochemical values were broadly in agreement with earlier reports. Despite the use of additional vegetable oil to balance the energy level, RH sows still had lower concentrations of serum triglycerides in late gestation. Conclusion: Different types of fibrous ingredients in the gestation diet influenced most of the investigated reference values for sows. The values of serum biochemical parameters were generally not affected by fiber type during the lactation stage. The SH supplementation for sows is an effective approach to give heavier litters at birth and weaning and to increase voluntary feed intake in early lactation.

RELATIONSHIPS BETWEEN EGGSHELL QUALITY AND BIOCHEMICAL PARAMETERS OF CALCIUM METABOLISM

  • To determine relationships of biochemical parameters involved in Ca metabolism with eggshell quality, serum Ca level, duodenal and uterine calcium binding protein (CaBP) and uterine carbonic anhydrase (CA) activities were measured using 102-week old hens. Three groups of chickens were selected, those showing high quality (HQ) and low quality (LQ) eggshells and non-laying activity (NE). NE hens exhibited significantly (p < 0.05) lower serum Ca levels than laying hens. HQ and LQ hens were not different in the Ca level, indicating that serum Ca level was not good indicator of hen's ability to produce different quality eggshells. Duodenal CaBPs was highest in HQ and lowest in NE (p < 0.10). Uterine CaBPs of LQ and NE were not different and lower significantly (p<0.10) than that of HQ, suggesting that CaBP played an important role in determining eggshell quality. Uterine CA activities of the three groups were significantly different (p < 0.01) ; highest in HQ and lowest in NE, suggesting intimate relationship between CA and Eggshell quality.

일부 남자대학생의 비만지수에 따른 혈중 Leptin 농도 연구 (A Study on Serum Leptin Concentrations by Obesity Index in Male College Students in Korea)

  • This study was conducted to investigate the basal leptin concentrations in normal(n=17, BMI 20-25, obesity index 90-110%) and overweight(n=13, BMI > 25, obestity index > 120%) Korea young aldult males, and correlation between leptin concentrations, nutrients intake, anthropometry and other biochemical parameters. Nutritional status, serum leptin and biochemiccal parameters were evaluated based on 24hr-dietary records, anthropometric measurement and blood analysis. Obesity index were 138% and 101% in overweight and normal group, respectively. Serum leptin concentration was higher in overweight group than that in normal group (8.65$\pm$ 9.41 vs 2.06 $\pm$ 1.19, p<0.05). Serum triglyceride, HDL-cholesterol and insulin concentrations were higher in overweight group than in normal group(p<0.05). Nutrients intakes was not different between two group. The leptin concentrations were correlated with body weight(p<0.001), BMI(p<0.05), obesity index(p<0.05), waist circumference(p<0.05), animal lipid intake(p<0.05)in overweight group.

HEPATO-AND RENAL TOXICITY OF AQUEOUS EXTRACT OF A MUSHROOM, AMANITA VOLVATA IN MICE

  • Toxic effects of a mushroom, Amanita volvata, were studied in respect to biochemical and histological changes induced in the liver and kidneys of mice. The changes in biochemical parameters characteristically appeared 12 hrs after oral administration of an aqueous extract of the mushroom. The hepatic glycogen decreased markedly to 17% of the control level and a concemitant decrease in blood glucose was also observed. The activity of serum glutamic oxaloacetic transaminase (SGOT) was elevated by 2.5-fold and the level of blood urea nitrogen (BUN) increased by 3-fold, respectively, 12 hrs after administration of the mushroom, their levels being maintained up to 24 hrs.
