  • Poly (3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3HB-co-4HB)] 미립구를 용매증발법으로 제조하였다. 3.9 mol% 4HB 조성의 P(3HB-co-4HB)를 Ralstonia eutropha의 유가식 배양으로부터 합성하였다. 계면활성제의 농도 및 종류(Tween 80, sodium dodecylsulfate, polyvinyl alcohol), 분산안정제 (Acacia)의 첨가, 고분자 및 모델 약물 (bovine serum albumin)의 농도 등이 미립구 입자 크기에 미치는 효과 및 in vitro 약물 방출 특성을 조사하였다. 평균 입자크기는 분산 안정제 첨가시 감소하였으며, 계면활성제, 약물 및 고분자의 농도가 증가할수록 증가하였다. 약물 방출량은 입자 크기가 감소할수록 증가하였다.

  • Spraque-Dawley계 수컷랫트에 계통분획한 단풍취의 각 분획물을 경구투여하고 혈청 ethanol농도와 간의 ADH활성에 미치는 효과를 검색 추적한 결과 알코올대사를 촉진시키는 성분은 주로 에탄올가용부에, 억제시키는 성분은 에탄올불용부에 주로 존재함을 추정할 수 있었고 현재 활성성분을 분리중에 있다.

한약재 조성물이 흰쥐의 알코올성 숙취 및 사염화탄소 유발 간 손상에 미치는 영향 (Effect of an Oriental Herbal Composition, Jang Baek Union (JBU), on Alcohol-Induced Hangover and CCl4-Induced Liver Injury in Rats)

  • 본 연구에서는 숙취제거 및 간기능 개선에 전통적으로 사용되어온 한약재로 구성된 조성물 JBU의 효능을 동물실험을 통하여 검증하였다. 숙취제거 효능을 확인하기 위하여 알코올과 그 대사산물인 아세트알데히드의 혈중 농도를 측정한 결과, 혈중 알코을 농도에서 JBU 추출물 처리군은 대조군에 비해 유의적인 감소를 보였고, 특히 숙취의 원인물질로 지목되는 아세트알데히드 감소에 있어서도 유의적인 효과를 보였다. 간기능개선 효능의 확인을 위하여서는 사염화탄소 유발 간손상 동물모델에서 GPT와 GOT의 활성도를 측정하여 간손상 정도를 확인하였고, 조직병리학적 관찰을 통하여 간의 병적 변화를 관찰하였다. 사염화탄소는 확연하게 간손상을 유발시켰고, JBU 추출물은 GPT 및 GOT의 증가를 유의적으로 억제하였으며 조직학적으로도 간조직의 손상을 뚜렷하게 억제하였다.

인삼의 해독작용에 관한 연구 급성 알코을 중독에 대한 인삼의 치료 효과 (Studies on the Antidotal Effect of Panax ginseng The Therapeutic Effect of Ginseng on the Acute Alcoholism)

  • 알코을 마취작용 및 독작용에 미치는 인삼(백삼 및 홍삼)의 영향을 보기 위하여 다음과 같은 실험을 시행하였다. 실험동물로는 백색 가토, 마우스, 및 잡견을 사용하였고, 백색 가토 및 잡견에 대한 알코올의 마취작용을 관찰하기 위하여 마취의 유도시간, 마취시간, 회복시간과 마취의 유도로부터 완전회복까지의 전시간을 각각 계정하였으며, 마우스에 대한 알코올의 독작용($LD_{50}$)을 측정하였다. 아울러 혈중 알코올 농도, 혈당량, 혈청 transaminase(GOT 및 GDT)활성도 및 혈청 alkaline phosphatase활성도를 각각 측정하였다. 그리고 건강한 대학생에 대한 임상실험으로는 code substitution, response time 및 muscle coordination을 각각 관찰하였다. 실험결과로는 1. 인삼 전처치로 알코올 마취의 유도시간은 지연되었고, 회복시간과 마취가 시작하 여 회복될때 까지의 전시간은 현저히 단축되었다. 2. 혈중 알코을 농도는 인삼 전처치로 백색 가로에 있어서는 감소되었으나. 잡견에서는 별 영향이 없었다. 3. 인삼 전처치로 헐당량, 혈청transaminase(GOT 및 GDT)활성도 및 쳔청 alkaline phosphatase활성도는 다소 변동이 있었으나, 각각 경상 범위 내에서의 변동이 였기 때문에 의의 있는 변화라고 할 수 없다. 4. 가토간 내 알코올 dehydrogenase활성도는 인삼 투여로 상승하였는데 특히 홍삼 투여로 더욱 현저하였다. 5. 마우스에 대한 알코올의$LD_{50}$은 인삼 전처치로 증가되었는데 홍삼 전처치로 더욱 현저히 증가되었다. 6. 학생에 대한 임상 실험에서는 알코올 투여 후 3시간에 측정하였는데 혈중 알코올 농도는 인삼 전처치로 변동이 없었으나 혈당량은 증가되었다. 7. 인삼 전처치로 알코을에 의한 response time은 현저히 단축되었으나 code substitution이나 muscle coordination에는 영향이 없었다.

Effects of the Hovenia dulcis THUMBER var. Koreana Nakai Extracts on Hepatoprotective Activities and Improvement of Hepatofunction

  • The Hovenia extracts showed a significant hepato-protective activity against alcohol and CC4-induced hepatotoxicity. Injection of CC4 and alcohol induced liver injury and increased serum GOT (glutamic oxaloacetic transaminase) and GPT (glutamic pyuvic transaminase) levels in injured rats. The ADH (alcohol dehydrogenase)-like and ALDH (aldehyde dehydrogenase)-like activities in the Hovenia extracts were studied. Their activities in the extracts of Hovenia wood were higher than the extracts of other parts of Hovenia. And then Hovenia wood extracts have high effects on the alcohol and acetealdehyde degradation. The Hovenia extracts were decreased concentration of fusel oil. The ADH-like and ALDH-like activities in the Hovenia wood extract were 1.14 $unit/{\mu}l$, 0.33$unit/{\mu}l$ respectively.

Comparative Effects of Dietary Quercetin and Rutin in Rats Fed with the Lieber-DeCarli Ethanol Diet

  • Flavonoids including quercetin and rutin are a group of naturally occurring compounds widely distributed in plants, especially in buckwheat. Thus, cereal and the leaf of the plant have increasingly used as a source of nutritional and functional foods such as noodle, cake or soup in Korea, Japan and other countries. This study investigated comparative effects of dietary rutin rich in buckwheat and its aglycone, quercetin, on serum biomarkers and antioxidant parameters in rats treated with chronic ethanol. Rats were fed with the liquid diets prepared by the method of Lieber Decarli. Serum alanine transaminase (ALT) and aspartate transaminase (AST) activities increased significantly by alcohol feeding. Dietary flavonoids including rutin, quercetin and their mixtures (1/1, v/v) decreased significantly the activities of serum ALT whereas the feeding of quercetin decreased only the activity of serum AST. The concentration of serum malondialdehydes elevated by chronic alcohol feeding decreased markedly in all the experimental groups that were fed with the flavonoids; however, the combined administration of quercetin or rutin, but not that of rutin or quercetin alone decreased significantly the concentration of liver malondialdehydes to the normal range in rats fed without ethanol. Our results suggested that dietary combined mixture of rutin and quercetin might be effective in ameliorating adverse responses seen in rats exposed to ethanol chronically.

음주에 따른 음성 변화에 대한 고찰 (Studies on Voice Changes Associated with Alcohol Intake)

  • Background and Objectives : It has been pointed out that alcohol intake in human beings induces changes in voice register and maximum phonation time. These changes supposedly result from injection of the vibratory vocal folds. The purpose of this study was to clarify the voice changes associated with alcohol intake and the changes of laryngeal mucosa. Materials and Methods : The subjects included 29 volunteers, including 20 men and 9 women ranging in age from 22 to 31 years. Alcohol intake was accomplished by oral administration of 23% soju 1 bottle (255cc). Serum alcohol concentration levels were evaluated hourly for 3 h after ingestion of alcohol. Seven measurements were performed at pre-alcohol intake and post-alcohol intake hourly : fundamental frequency, jitter, shimmer, noise to harmonic ratio as the acoustic analysis, maximal phonation time, mean flow rate, and subglottal pressure as the aerodynamic analysis. The changes of laryngeal mucosa were evaluated by flexible laryngoscope at each measurement. Results : By comparing the acoustic and aerodynamic data and laryngeal mucosa before and after alcohol intake, there were not remarkable changes (p>0.05). Conclusion : The voice and laryngeal mucosa have not remarkably changed according to alcohol concentration in this study. Furthermore studies on the voice change induced by multiple alcohol concentrations are required.

에탄올을 투여한 흰쥐에서 식이 단백질 섭취 수준이 혈청중 지방 및 효소활성에 미치는 영향 (Effects of Dietary Protein Levels on the Serum Lipids and Enzyme Activities of Ethanol-Administered Rats)

  • This study was undertaken to investigate effects of alcohol and dietary protein levels on serum lipids and enzyme activities in 15 week-old male rats given a normal diet. Rats were divided into 8 groups : control group (16% protein 16PC) and 8%(8PE) 16%(16PE) and 24% protein groups(24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight organ weight and various blood components were determined at 4 and 10 weeks. Body weight gain organ weight hemoglobin concentration and hematocrit value were not influenced by ethanol and dietary protein levels. The levels of total cholesterol HDL-cholesterol and phospholipid in serum were not affected by ethanol consumption. Serum triglyceride concen-trations after 10 weeks were significnatly increased ethanol-treated group compared with that of control group and the effect was greater in low protein group than control group. Serum ALP activity was significantly higher in 8PE group than other group but there was no influence by ethanol consumption.

  • As an initial step for evaluating hepatoprotective components against alcohol-induced toxicity, the effect of various fractions from Aloe vera on alcohol metabolism in rats were examined and the results were as follows: Water soluble fraction, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic ADH activity. On the other hand, the fractions soluble in organic solvent was found to cause an increase in the blood ethanol concentration and inhibit ADH activity. Further fractionation of the water soluble fraction by ultrafiltration system gave four subfractions corresponding to molecular weight and treatment of them in rats demonstrated that subfraction of M.W. > 30,000 exhibited the most potent enhancing activity of ethanol methabolism.

Association between serum 25-hydroxyvitamin D levels and adiposity measurements in the general Korean population

  • BACKGROUND/OBJECTIVES: Obesity, which is a known risk factor for many chronic diseases, has also been associated with vitamin D deficiency. This study explored the relationship between serum 25-hydroxy-vitamin D [25(OH)D] concentrations and adiposity measures in a general Korean population using the most recent, nationally representative survey data. SUBJECTS/METHODS: The study sample consisted of 4,771 Korean adults (${\geq}19years$) who participated in the fifth Korean National Health and Nutrition Examination Surveys. Serum 25(OH)D was determined by radioimmunoassay. Body mass index (BMI), waist circumference (WC) and total body fat content were measured as adiposity measurements. Total body fat content was measured by dual-energy X-ray absorptiometry. RESULTS: The serum 25(OH)D level was significantly higher in men than in women. Serum 25(OH)D concentration was positively correlated with energy intake, and it was negatively correlated with total body fat content (P < 0.0001) and percentage body fat (P < 0.0001) after adjustment for age in both sexes, while was inversely correlated with BMI only in women. In multivariable regression analysis, serum 25(OH)D was inversely associated with the total body fat content after adjustment for age, BMI, education, region, smoking, alcohol consumption, physical activity, and energy intake only in men (P = 0.0047). However, the serum 25(OH)D concentration was not associated with WC or BMI, indicators of adiposity after adjustment for potential risk factors. CONCLUSIONS: Serum 25(OH)D concentration was independently associated with the total body fat content in a general Korean population, but it may be not associated with the indicators for estimating adiposity, such as WC or BMI.
