A Study on the Extraction and Efficacy of active ingredient from Hovenia dulics

  • The water extracts of Hovenia dulcis Thunb. showed significant reducing alcohol concentration in blood and hepatoprotective acitivity against D-galactoseamine/LPS and CCl4-toxicity. The concentration of blood alcohol was decreased from 2hr after injected in rats with Hovenia dulcis extracts. The water extracts of Hovenia dulcis significantly inhibited the elevation of serum ALT, AST and LDH levels. In this study, we investigated the effect of Hovenia dulcis extract on reducing alcohol concentration in blood. Also the efficacy and toxicity of Hovenia dulcis extract were evaluated by in vivo and in vitro tests.

Effect of Zinc-Enriched Yeast Supplementation on Serum Zinc and Testosterone Concentrations in Ethanol Feeding Rats (알코올 급여 흰쥐의 혈중 성호르몬 및 아연 농도에 미치는 아연 고함유 효모 Saccharomyces cerevisiae 급여의 영향)

  • Zinc is an essential trace element participating in many physiological functions and notably an important function for sperm physiology. Zinc-enriched yeast strain, Saccharomyces cerevisiae, isolated from tropical fruit rambutan. This strain contained 306 ppm zinc concentration and other components contained by K 28,640 ppm, Mg 2,342 ppm, Na 1,048 ppm, Ca 366 ppm, Fe 236 ppm and Mn 4 ppm. The serum concentration of testosterone was decreased in ethanol treatment rats. As compared with ethanol treated control rats, the zinc-enriched yeast strain supplementation showed significantly increased the testosterone concentration in serum. In addition, zinc concentration in serum was decreased in alcohol treatment, but this reduction was significantly increased by zinc-enriched yeast strain supplementation in ethanol feeding rats. These results indicate that zinc-enriched yeast Saccharomyces cerevisiae strain could play an important role in the sperm physiology by the marked elevation of serum testosterone concentration.

Levels and Factors Affecting Serum Carotenoids Concentrations of Adults Living in Rural Area of Korea (한국농촌 성인의 혈청 Carotenoids 농도와 이에 영향을 미치는 요인에 관한 연구)

  • Recent researches suggest that carotenoids are important not only as provitamin A but also for prevention of chronic diseases. This study was conduction to determine levels and factors affecting serum levels of lutein + zeaxanthin, $\beta$-cryptoxanthin, and $\beta$-carotene in 93 adults living in rural area of Korea. Fasting blood samples were collected and serum carotenoid levels were measured by HPLC. Dietary intake was estimated by 24 hour recall method and frequency questionnare of major food groups. Mean serum concentration of lutein + zeaxanthin was 616.32 nmol/L, $\beta$-cryptoxanthin was 856.95nmol/L, and $\beta$-carotene was 242.90nmol/L. Serum $\beta$-carotene levels in study subjects were very low. Both $\beta$-cryptxanthin and $\beta$-carotene were negatively correlated with serum triglyceride and positively correlated with total-choesterol and LDL-cholesterol. Serum levels of female subjects were significantly higher than males in all carotenoids. For age groups, subjects in their 30's were shown to have the highest concentration of all carotenoids. Lutein + zeaxanthin were lowest in subjects in theri 40's while $\beta$-crytoxanthin and $\beta$-carotene levels were lowest in subjects in their 60's. The $\beta$-carotene levels in non-smokers were significantly higher than in drinkers. Lutein+zeaxanthin levels were significantly higher among subjects consuming more green and yellow vegetables by frequency questionnarie. In conclusion, serum carotenoids were affected by sex, age, serum lipids, smoking, and alcohol intake. Intake of vegetables and fruits could affect by sex, serum lipids, smoking, and alchol intake. Intake of vegetables and fruits could affect serum lutein+zeaxanthin level. This data indicated that compared to other studies, Korean adults in rural areas have high lutein+zeaxanthin concentratins and low $\beta$-carotene concentrations in serum. High lutein+zeaxanthin levels may be related to high consumption of vegetables in these subjects.

Relationship of Serum Uric Acid to Cardiovascular Risk Factors in Korean Male Workers (일부 남자 근로자들의 혈청 요산과 심혈관질환 위험요인과의 관련성)

  • Objectives : To investigate the relationship of serum uric acid to cardiovascular risk factors in Korean male workers. Methods : We screened 522 male workers at an electrical manufacturing company by a periodic health examination and a questionnaire survey in 2000. We collected data relating to age, smoking status, alcohol drinking status, body mass index (BMI), blood pressure, serum lipid, fasting blood glucose and serum uric acid. The data were analyzed using a variety of methods, including ANOVA, Pearson's correlation, and multivariate regression analyses. to examine the association between uric acid and cardiovascular risk factors with a cross-sectional study design. Results : The concentration of serum uric acid showed positive associations with BMI, total cholesterol, triglyceride, and LDL-cholesterol; it was also significantly correlated to systolic and diastolic blood pressures. Life-style characteristics, such as smoking and alcohol drinking showed no significant association. From the multivariate regression analyses, BMI, total cholesterol and fasting blood glucose levels were found to be independent positive predictors of uric acid; while age, blood pressure and smoking status give no independent contribution explaining the variability of serum uric acid levels. Conclusions : This study demonstrates that serum uric acid level have a significant association with cardiovascular risk factors, such as BMI, total cholesterol and blood glucose in some Korean male workers.

Protein Binding of [S]-Perillyl Alcohol in HSA using High-Performance Frontal Analysis (HPFA를 이용한 HSA와 [S]-Perillyl Acohol의 단백질 결합력)

  • An on-line frontal analysis HPLC system was developed to determine the unbound concentration of (S)-perillyl alcohol, an potential anti-cancer agent, in human serum albumin (HSA) solution, The analysis was performed on a Develosil 100 Diol 5 (10 cm x 4.6 mm I.D.) high-performance frontal analysis (HPFA) column. Sodium phosphate solution was used as the mobile phase (pH 7.4, ionic strength 0.17) at a flow rate of 1 $m\ell$/min. UV wavelength was set at 205 nm. A injection volume of 600${mu}ell$ was chosen to ensure the compound eluted formed a zonal peak with a plateau. By Scatchard analysis, it was found that the binding constant(K) and binding number(n) of (S)-perillyl alcohol to molecular HSA were 2.05 x $10^6$ [$M{-1}$], 0.00428, respectively.

Serum Fatty Acids in Patients with Angiographically-Documented Coronary Artery Disease (관상동맥질환자의 혈청 지방산 조성에 관한 연구)

  • To study the relation between serum fatty acids and coronary artery disease(CAD), 194 subjects were randomly selected and divided into three groups(control, single vessel disease group(SVD), muliple vessel disease group(MVD)) according to the angiographic results. Total serum levels of fatty acids and serum phospholipid(PL)-fatty acids(FAs)were analysed using gas chromatography and their associations with CAD were examined. Different patterns of total serum fatty acid levels were found in men and women. Levels of most fatty acids of SVD and MVD were significantly lower in men, while those of MVD were significantly higher in women. In terms of PL-FAs in both men and women, the levels of PL-FAs follow the order of control < SVD < MVD and in women, the difference was significant. Various ratio(including ratios denoting the activites of desaturase and elongase) of total serum-and PL-FAs were similar in the three groups. In the relation of serum fatty acids to serum lipid profiles, PUFA & LDL showed a negative correlation, while, SFA & LDL-cholesterol and PUFA and HDL-cholesterol showed positive correlatons. correlations. Systolic blood pressure and alcohol intake levels negatively affected the levels of serum LA, AA, EPA and DHA in the risk factor analysis. These findings are consistent with other evidence indicating that fatty acid compositions are changed in CAD, especially on the concentration base and the change was related to the severity of the disease. Therefore, for the purpose of disease prevention and therapeutic use, balanced intakes of various fatty acids must be seriously considered.

Discrepancies and Validation of Ethanol Level Determination with Osmolar Gap Formula in Patients with Suspected Acute Poisoning (급성 중독환자에서 삼투압 계산식으로 추정된 에탄올 농도의 유효성 검증)

  • Purpose: Osmolar gap (OG) has been used for decades to screen for toxic alcohol levels. However, its reliability may vary due to several reasons. We validated the estimated ethanol concentration formula for patients with suspected poisoning and who visited the emergency department. We examined discrepancies in the ethanol level and patient characteristics by applying this formula when it was used to screen for intoxication due to toxic levels of alcohol. Methods: We retrospectively reviewed 153 emergency department cases to determine the measured levels of toxic ethanol ingestion and we calculated alcohol ingestion using a formula based on serum osmolality. Those patients who were subjected to simultaneous measurements of osmolality, sodium, urea, glucose, and ethanol were included in this study. Patients with exposure to other toxic alcohols (methanol, ethylene glycol, or isopropanol) or poisons that affect osmolality were excluded. OG (the measured-calculated serum osmolality) was used to determine the calculated ethanol concentration. Results: Among the 153 included cases, 114 had normal OGs (OG≤14 mOsm/kg), and 39 cases had elevated OGs (OG>14). The mean difference between the measured and estimated (calculated ethanol using OG) ethanol concentration was -9.8 mg/dL. The 95% limits of agreement were -121.1 and 101.5 mg/dL, and the correlation coefficient R was 0.7037. For the four subgroups stratified by comorbidities and poisoning, the correlation coefficients R were 0.692, 0.588, 0.835, and 0.412, respectively, and the mean differences in measurement between the measured and calculated ethanol levels were -2.4 mg/dL, -48.8 mg/dL, 9.4 mg/dL, and -4.7 mg/dL, respectively. The equation plots had wide limits of agreement. Conclusion: We found that there were some discrepancies between OGs and the calculated ethanol concentrations. Addition of a correction factor for unmeasured osmoles to the equation of the calculated serum osmolality would help mitigate these discrepancies.

Studies on Lipid Intake, Serum Lipid Profiles and Serum Fatty Acid Composition in College Women Who Smoke (흡연여대생의 지질섭취와 혈중지질 및 지방산 조성에 관한 연구)

  • Cigarette smoking has long been recognized as a major risk factor in the development of coronary heart disease. Several investigators have reported the strong association between smoking and high serum cholesterol, triglyceride concentration, SFA and low HDL cholesterol, PUFA and $\omega$6 concentrations. Therefore, this study was done to investigate the effect of smoking on the serum lipid profile and fatty acid composition of college women. Sixty-one non-smokers and twenty-seven smokers were selected from college women students in the Seoul area. Their lipid intake, serum lipid concentration and fatty acid composition were examined. There were no differences in the general characteristics and anthropometric indices between the smokers and non-smokers. However, alcohol consumption was significantly higher in smokers than non-smokers (p<0.001). The daily caloric intake of smokers and non-smokers were 1875.84 kcal and 1915.53 kacl, respectively. On the other hand, the mean daily intake of lipids and cholesterol were significantly lower in smokers (p<0.05). In smokers, the mean concentrations of serum total cholesterol, LDL-cholesterol and the LDL-C/HDL-C ratio were higher, and the compositions of EPA and DHA were lowe than in non-smokers. There was a negative correlation between the serum triglyceride and PUFA levels in the two groups. Also, serum HDL-C correlated negatively with MUFA in smokers and non-smokers (p<0.01). These results suggest that smoking cause inadequate changes in serum lipid profile and serum fatty acid composition, thereby increasing the tendency for coronary heart disease.

Serum Lipid and Lipoperoxide levels and Their Related Factors in Middle-Aged Men in Teagu (대구지역 중년 남성의 혈청 지질과 혈청 과산화지질의 관련 인자 연구)

  • The present study was aimed to examine various factors related to serum total cholesterol, triglyceride, atherogenic index, lipoperoxides and vitamin E in 174 middle-aged men residing in Taegu. Body mass index was positively correlated with serum triglyceride, and waist-hip circumference ratio was positively correlated with serum total cholesterol, triglyceride, atherogenic index and vitamin E concentration. Total cholesterol, triglyceride and atherogenic index of the subjects who smoked more than 20 cigarettes per day were significantly higher than those with smoking less or non-smokers. drinking alcohol more than 5 times per week showed higher atherogenic index than drinking less frequently of non-drinking. Subjects doing exercise had higher HDL-cholesterol level than those with no exercise. The intakes of all nutrients were above 90% of RDA and the subjects with good food habit had lower of total cholesterol, triglyceride, atherogenic index and lipoperoxides. The study indicates that avoiding heavy smoking and drinking as well as obesity and keeping regular exercise, good food habit and diversity of foods will reduce risk of cardiovascular diseases in middle-aged men.

Synthesis of Size Controllable Silk Fibroin Microparticles and Their Stability on Different Solutions

  • Silk fibroin microparticles were fabricated using a phase separation technique between silk fibroin solution and polyvinyl alcohol. We found that the concentration of polyvinyl alcohol determines the size of microparticles. The mean diameter of the silk fibroin microparticles varied from 3.48 ㎛ to 4.05 ㎛. The silk fibroin microparticle size increased as a function of the concentration of PVA in aqueous silk solution. The resulting silk fibroin microparticles have narrow size distribution (i.e. monodisperse) and smooth/spherical surface. Also, we studied the effects of mouse serum, sodium phosphate buffer (PBS), and pH on the stability of the silk fibroin microparticles. Overall, we demonstrated the simple method to fabricate and to control the silk fibroin microparticles that makes our silk microparticles to be usable for a potential drug delivery carrier.
