혈액투석중인 만성신부전환자의 영양상태에 관한 연구 (Nutritional Assessment of the Hemodialysis Patients)

  • This study investigated the nutritional status of 33 hemodialysis patients. Their weight, height, triceps and mid-arm circumference were measured and their dietary intake and blood profiles evaluated. The subjects were 57.1$\pm$11.9 years old. The energy intakes of men and women were 61% and 68% of RDA, respectively. The men and women's intake of protein was 0.93g/IBWkg/d and 0.99g/IBWkg/d, respectively. According to the distribution of BMI, 22.2% of the men and 73.3% of the women were underweight. A total of 5.6% of the men were overweight, versus none of the women. The serum albumin levels of the men and women were 3.56 and 3.52g/dl, respectively. The serum cholesterol levels of the men and women were 134.1 and 148.5mg/dl, respectively. The subjects were divided into three groups according to the level of albumin, and their intakes of nutrients were compared with one another. The group with high levels of albumin did not show higher energy and protein intake than the other groups but the serum total protein level was significantly higher. When the nutritional status of the patients was evaluated by weight and serum albumin level, 6.0% of them showed kwashiorkor-type malnutrition and 75.8% of them showed mild malnutrition.

Ibuprofenlysine binding to human and bovine serum albumin using a fluorescence probe technique

  • The possibility of using a fluorescence probe technique for the study of ibuprofenlysine binding to human and bovine serum albumin was investigated. 1-anilino-8-naphalenesulfonate was used as the probe. The number of binding sites of human and bovine serum albumins for ibuprofenlysine appears to be 4 and 2, respectively. By using this technique, the association constants were found to be $1.533{\times}10^{4}M^{-1}$ and $2.238{\times}10^{4}M^{-1}$, respectively.

개구리目 혈색소와 혈청단백질의 전기영동 (Electrophoresis of the Hemoglobins and the Serum Proteins of Korean Anuran)

  • 韓國産 개구리 目의 8種에 대하여 血色素와 血淸 단백질의 cellulose acetate 電基泳動圖를 조사하였다. 혈색소의 전기영동도는 종에 따라 그 구성성분의 수와 이동도에 차이가 있었다. 두꺼비와 금개구리에서는 단일 밴드로 나타났고, 여타의 종에서는 2밴드로 분리되었다. 혈청단백질의 전기영동도는 종에 따른 특징을 잘 나타내고 있다. 이들 동물의 혈청단백질에서는 다형현상이 나타나지 않았다. 모든 실험재료에서 albumin은 뚜렷이 나타나지만 prealbumin은 나타나지 않았다. 두꺼비에서는 albumin과 postalbumin 사이가 분명하게 분리되지 않았다. 따라서 혈청단백질의 전기영동도에서 두꺼비와 다른 종들을 쉽게 구별할 수가 있었다.

청열막성방(淸熱膜性方)이 Cationized Bovine Serum Albumin투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향 (A Study on the Effects of Chungyeolmaksungbang on Mouse Model of Membranous Nephropathy Induced by Cationic Bovine Serum Albumin)

  • Objective : Membranous nephropathy (MN) is the most common cause of adult nephrotic syndrome worldwide. MN has been defined as granular subepithelial deposition of IgG immune complexes along the glomerular basement membrane (GBM). We aimed to identify the effects of Chungyeolmaksungbang (CYMSB) treatment on cBSA-induced in MN mouse model. Methods : The effect of Chungyeolmaksungbang treatment was studied on the morphology and protein excretion in the cationized bovine serum albumin (cBSA)induced mouse chronic serum sickness nephritis model. One group of mice was given intra-peritoneal (i.p.) immunizing doses of cBSA and complete Freund's adjuvant. One week later, these animals began a single i.p. injection of cBSA for 4 weeks. A second group followed the same injection protocol, but was given CYMSB p.o. Results : Proteinuria significantly was decreased and serum albumin was increased in the group treated with cBSA and CYMSB extract compared with the control. Serum BUN was significantly decreased on CYMSB compared with control. CD3e+/CD19 cells ratio of peripheral blood was decreased and CD4+/CD8 cells was increased. Level of $IL-1{\beta}$ was significantly decreased, and $IFN-{\gamma}$ was significantly increased. Concentration of IgG and IgM was significantly decreased compared with control. Thickness of GBM was decreased on histological analysis of kidney. Deposition of CD4 and CD8 was decreased on immunohistochemical staining of kidney. Conclusions : We conclude that CYMSB treatment may could be a useful remedy agents for treating the MN with cBSA.

Hansenula polymorpha DL-1이 생산하는 재조합 알부민의 정제 및 특성 (Purification and Characterizating of Recombinant Human Albumin from Hansenula polymorpha DL-1)

  • 메탄올 자화효모 Hansenula polymorpha 가 생산하는 재조합 인체 알부민 (human albumin)을 heat treatment ultrafiltration phenyl Sepharose CL-4B Mono Q 컬럼 크로마토그래피를 수행하여 수율 60%로 정제하였다. 정제된 재보합 알부민 SDS-PAGE를 수행한 결과 분자량이 65,000 Da으로 나타났고, 본 재조합 알부민의 N-말단 아미노산 서열은 Asp- Ala- His- Lys- Ser- Glu- Ala 으로 혈청유래 알부민 및 다른 효모 유래의 재조합 인체 알부민 과 동일함을 보였다. 정제한 재조합 인체 알부민의 아미노산 조성은 총 18종 의 아미노산이 검출되었고 아미노산 잔기 중 cysteine, arginine lysine 등이 이론치와 일치하였으며, 혈청 알부민과 동일하게 약 pI 4.8 정도 값을 나타내어 H. polymorpha 유래의 재조합 단백질의 전반적인 아미노산 구성이 혈청 알부민과 동일함을 확인하였다.

혈청 알부민 나노입자를 이용한 항생제 흡착 (Adsorption of Antibiotics on Serum Albumin Nanoparticle)

  • 항생제는 감염병 환자의 치료, 농수축산업의 생산성 향상을 위한 목적으로 광범위하게 사용되는 약물이다. 그러나 항생제 과용 및 낮은 생분해성으로 인해 상당량이 하수로 누출되어 환경오염을 유발하며 내성 박테리아 출현을 촉진하고 있다. 본 연구에서는 생분해성 혈청 단백질인 알부민을 항생제 흡착제로 사용하기 위한 가능성을 탐구하였다. 혈청 알부민은 다양한 대사 산물과 호르몬을 모든 조직의 혈관 외 공간으로 운반하는 천연 혈액 단백질이다. 혈청 알부민은 수용성이 높지만 이온성, 친수성 또는 소수성 분자를 쉽게 수용하는 고유 결합 부위를 가지고 있어 나노 흡착제로 유망한 물질이다. 코아세르베이션(coacervation)을 유도하기 위해 탈용매제인 에탄올을 알부민 수용액에 적가하여 150 ~ 170 nm 크기 범위의 알부민 나노 입자로 탈수화 및 액-액분리 하였다. 글루타르알데히드 가교제를 첨가할 경우 알부민 나노입자의 크기 안정성 및 동질성이 증가하였다. 항생제 아목시실린에 대한 알부민 나노입자의 흡착능 평가에서 가교제 사용 농도, pH에 따른 흡착능의 차이가 관찰되었다. 분광광도법으로 측정한 알부민 나노입자의 단위질량당(mg) 최대 흡착능은 pH 4.0 수용액에서 아목시실린 12.4 마이크로그램(㎍)이다. 이러한 결과는 물에서 항생제를 제거하는 천연 나노 흡착제를 제조하기 위한 구성 물질로서 혈청 알부민의 잠재력을 보여준다.

Bovine Serum Albumin 수용액의 초음파 측정 (Ultrasonic Measurement in Bovine Serum Albumin Solution)

  • 단백질 수용액 중의 초음파흡수 mechanism을 규명하기 위하여 pH 7의 소혈청 albumin(BSA)수용액의 초음파흡수 측정을 주파수 100 kHz에서 1600 MHz에 걸쳐 행하였다. 측정방법은 plano-concave 공명법(0.1${\sim}$10 MHz)과 고분해능 Bragg 반사법(120${\sim}$1600 MHz)을 사용하였다. pH7에서의 초음파흡수스펙트럼은 Davidson-Cole 분포식의 거울상인 분포함수를 사용한 완화곡선과 일치하였다. 이 완화현상을 BSA 분자의 수화평행에 의한 것으로 해석하였다.

  • Binding of six cephalosporins (cefotaxime, cefuroxime, cefazoline, cephalothin, cephaloridine, cephacetrile) to bovine serum albumin was studied. Fluorescence probe technique and difference spectrophotometry were employed to evaluate the nature and degree of association of cephalosporin albumin complex. 1-Anilinonaphthalene-8-sulfonate (ANS) was used as the fluorescence probe. 2-(4'-hydroxybenzeneazo) benzoic acid(HBAB) was employed as the UV spectrophotometric probe. Compentitive bindings between cephalosporins and probes were observed. The number of binding sites of bovine serum albumin for each cephalcsporin is 2. Among six cephaloporins, cefotaxime has the highest binding constant followed by cafazoline, cefuroxime, cephalothin, cephaloridine and cephacetrile.

지속성 외래 복막투석환자의 영양상태에 관한 연구 (Nutritional Assessment of the Continuous Ambulatory Peritoneal Dialysis Patients)

  • This study investigated the nutritional status of 32 CAPD patients. Their weight, height, triceps, mid arm circumference were measured and their dietary intake and the blood profiles were evaluated. They were 56.0${\pm}$11.8 years old. Their intake of energy and protein were 78% and 93% of RDA respectively. The energy from dextrose was 376.1${\pm}$83.2kcal. The men's and women's intake of protein was 84.6% and 102.1% of RDA respectively, According to the distribution of BMI, 23.5% of the men and 6.1% of the women were underweight and 5.9% of the men and 20% of the women were overweight. The serum albumin levels of the men and women were 3.30 and 3.71g/41 respectively. However, the average amount of serum ferritin, as a whole, comes within the normal range, which shows that stored iron was not decreased and that their anemia was not caused by iron deficiency. The subjects were divided into three groups according to the level of albumin, and their intakes of nutrients were compared with one another, The group with the high level of albumin showed that energy and protein intake was significantly larger and that BMI was also significantly higher than the other groups. There was a positive correlation between BMI and energy intake. Serum total protein had a positive correlation to energy intake ; hematocrit, to carbohydrate intake. BMI had a positive correlation to energy intake. A relative magnitude of factors affecting albumin level was analyzed by Stepwise multiple regression analysis. Overall results about relative influence of independent variables to dependent variable(albumin) indicated that the blood total protein(p <0.0001) was the most significantly correlated with serum albumin level in all subjects,1311owe4 by creatinine and total cholesterol.
