The Changes of Serum Angiotensin Converting Enzyme Activity in Lung Cancer Patients (폐암 환자의 혈청 Angiotensin Converting Enzyme 활성도의 변화)

  • Background: Angiotensin converting enzyme is a glycoprotein peptidyldipeptide hydrolase which cleaves the c-terminal dipeptides of several oligopeptides. It is a menbrane-bound protein mainly synthesized by the endothelial cells. Since the lung has the largest capillary bed of any organ in the body, it is here that ACE acts on circulating substrates like angiotensin I and bradykinin. It is well known that ACE correlates with disease activity in sarcoidosis and also there are reports that changes in serum ACE activity are found in many acute and chronic lung diseases. So we planned this study to see if serum ACE activity can act as a prognostic factor in lung cancer. Methods: Forty-one newly diagnosed lung cancer patients were included in the study group. There were 19 patients with squamous cell lung cancer, 13 with adenocarcinoma, and 9 with small cell carcinoma. Patients were excluded from the study if they had high blood pressure, heart disease, liver disease, renal disease, or other lung disease. Serum ACE activity was analyzed according to cell type, staging, mode of treatment, and clinical response to treatment. Results: 1) There was no difference in serum ACE activity between lung cancer patients and the control group. Also no difference in serum ACE activity was found according to cancer cell type or staging. 2) In patients who underwent curative resection of lung cancer, serum ACE activity was decreased significantly after the operation. 3) In patients who were diagnosed as non-small cell lung cancer and were treated with 4 cycles of anti-cancer chemotherapy without clinical improvement, changes in serum ACE activity were not seen after the treatment. 4) In patients diagnosed as small cell lung cancer treated with 4 cycles of anti-cancer chemotherapy with clinical improvement, changes in serum ACE activity were also not observed. Conclusion: Serum ACE activity was decreased after lung resection but had no relation to cell type, staging, or clinical response to treatment in lung cancer patients. Therefore, serum ACE activity is not suitable in predicting clinical outcome of lung cancer patients.

Effects of Green Tea Catechin on Serum Lipid Composition and Angiotensin- Converting Enzyme Activity in Rats Exposed to Microwave

  • This study aims to investigate the changes in the lipid composition and angiotensin-converting enzyme activity in serum of rats exposed to microwave radiation, and to examine improving effects of green tea catechin to the lipid composition. The microwave-exposed rats received the normal and one of 3 diets: catechin free (MW-0C), 0.25% catechin (MW-0.25C) or 0.5% catechin (MW-0.5C). Rats were sacrificed 6th day after microwave radiation (2.45 ㎓, 15 min). The concentration of serum triglyceridein MW-0C group was increased by 85%, compared with the normal group, but that of MW-0.25C group with 0.25% catechin supplementation was to 17% lower, compared with the MW-0C group. There was no significant difference between normal group and MW-0.5C group. Total-cholesterol and LDL-cholesterol concentrations were increased by 21.4% and 38.6%, respectively, by microwave irradiation. The concentration of HDL-cholesterol in MW-0C group was lower to 29.3%, but it was maintained at the normal level by catechin supplementation. There was no significant difference among four groups in HLD-cholesterol/LDL-cholesterol. Atherogenic index in MW-0C group was increased by 56%, compared with the normal group. Angiotensin-converting enzyme (ACE) activity in lung tissue of MW-0C group was increased by 59 %, while that of MW -0.25C and MW -0.5C group were maintained at the normal level. ACE activity of MW-0C group in serum was increased by 122.8%, compared with the normal group. Catechin supplementation group was significantly reduced, compared with the MW-0C group ACE activity. In conclusion, microwave irradiation increased the serum triglyceride and cholesterol concentrations and ACE activity which are considered as the blood pressure increasing agents. However, catechin supplementation decreased the level of triglyceride, total cholesterol, LDL cholesterol, and ACE activity, which maybe consider catechin as being agent of lowering effect for blood lipid profile for athero-genesis.

Effect of Soybean Hydrolysate on Hypertension in Spontaneously Hypertensive Rats (대두 가수분해물에서 분리한 UF-peptide가 in vivo에서 자발성 고혈압 흰쥐의 혈압 강하에 미치는 영향)

  • Effect of soybean hydrolysate prepared by ultrafiltration(UF-peptide) was evaluated regarding its action on blood pressure, selected biochemical changes in spontaneously hypertensive rats(SHR). Male SHR were divided into three groups and fed diet with 0, 3, 10% UF-peptide for 6 weeks. Development of hypertension was determined by tail cuff method during the experimental period. After the experimental period, angiotension I converting enzyme(ACE) activity in serum and vascular tissue, and lipids level in serum were measured. Spontaneous elevation of blood pressure was suppressed by the UF-peptide supplementation after two weeks of feeding. After five weeks of feeding the development of hypertension was significantly inhibited in the UF-peptide administered groups compared to the control. In addition, the UF-peptide was effective for lowering serum lipids level such as triglyceride and total cholesterol, as well as ameliorating some pathological changes in SHR.

Effects of Captopril on the Active Angiotensin Converting Enzyme at the Pulmonary Endothelial Cells (Captopril에 의한 폐동맥 내피세포중 활성형 Angiotensin 전환효소의 변화)

  • The effect of captopril on the lung angiotensin converting enzyme (ACE) was investigated after 3 weeks oral administration (120~160 mg/kg/day) through drinking water in SpragueDawley rats. On the $^{125}$I-351A, an ACE inhibitor, binding assay in the isolated perpused lungs, the number of ACE molecules at the intrapulmonary endothelial cell surface was significantly decreased (p<0.001), and recovered to the normal level 7 days after discontinuation of captopril treatment. Intrapulmonary conversion ratio of Al to All was also significantly decreased (p<0.05) in the isolated perpused lungs. Bolus intravenous injection of angiotensin I did not showed pressor response in the both of systemic and pulmonary blood pressure of the anesthetized rats. ACE activity of the lung homogenates was also significantly reduced. These data consistently indicate the decrease of functionally active ACE molecule at the pulmonary artery after chronic captopril treatment. However, serum ACE activity was increased three fold in captopril treated rats compared to the normal rats. So, these results suggest that the functionally active ACE molecule at the pulmonary artery was still inhibited, which is directly associated with the antihypertensive effects, even if the total angiotensn converting enzyme induction was resulted after chronic captopril treatment.

The Probiotic Characteristics of Lactobacillus acidophilus Isolated from Infant Feces (신생아분변 유래 Lactobacillus acidophilus의 프로바이오틱으로서의 특성)

  • This study was conducted to obtain a good probiotic strain of L. acidophilus from infant feces which have the acid and bile tolerance. The selection criteria for the strain included antimicrobial activity, serum cholesterol reduction, resistance to the hydrogen peroxide, angiotensin converting enzyme (ACE) inhibition activity and iron solubility. To this end, five probiotic Lactobacillus strains have been isolated from infant feces. Especially, L. acidophilus SD 105 had strong antimicrobial activity against Listeria sp., high deconjugation activity in the medium which contained 0.5% of glycocholate (GCA) and high resistance to the hydrogen peroxide. L. acidophilus SD 102 showed the highest ACE inhibition activity among the tested cultures and L. acidophilus SD 103 showed iron solubility of more than 70%.

Antihypertensive and ACE Inhibitory Effects of Novel Isoflavone-free Black Soy Peptide Mixture (Isoflavone-Free 검은콩펩타이드의 항고혈압 및 ACE 활성 억제효과)

  • Antiobesity, hypotriglyceridemic and antihypertensive activities of isoflavone-free peptide mixture (black soybean peptide, BSP) were reported in our previous experiments. In the present study, angiotensin converting enzyme inhibitory (ACEi) activity was decreased in the aorta tissues of spontaneously hypertensive rats (SHRs) treated with BSP (1% in drink water) for 4 weeks, but not in serum. BSP administration significantly decreased ACE activity by 17.5% (from $33.2{\pm}4.5$ to $27.4{\pm}1.96$ mUnit/mg, p=0.0013) in aorta tissue hydrolysate. BSP treatment also decreased significantly mean blood pressure (BP) (from $213.0{\pm}16.96$ to $184.0{\pm}6.53$ mmHg, p<0.0001) as expected. These results indicate that BSP has antihypertensive activity as well as ACEi activity.

The antihypertensive effect of ethyl acetate extract of radish leaves in spontaneously hypertensive rats

  • Radish (Raphanus sativus L.) is a cruciferous vegetable, and its leaves have antioxidant and anticancer properties. This study was conducted to evaluate the effects of ethyl acetate extracts from radish leaves on hypertension in 11-week-old spontaneously hypertensive rats (SHRs). The SHRs were randomly divided into 3 groups of 6 rats each on the basis of initial systolic blood pressure (SBP) and were treated with oral administration of radish leaf extract (0, 30, or 90 mg/kg body weight [bw], respectively) for 5 weeks. Six Wistar rats were used as normotensive controls. The amount of the radish leaf extract had no effect on body weight. The SBP of the SHRs showed a decreasing trend with the consumption of the radish leaf extract. In the third week, the SBP of the group fed 90 mg extract/kg bw reduced from 214 mmHg to 166 mmHg and was significantly lower than that of the normotensive and hypertensive controls. The extract did not show a significant effect on the angiotensin-converting enzyme (ACE) activity in the serum, kidney, and lung. The extract increased the concentration of NO in serum and the activities of antioxidant enzymes such as glutathione peroxidase and catalase in red blood cells (RBCs). The serum concentrations of $Na^+$ and $K^+$ were not significantly different between all groups. However, the fecal concentrations of $Na^+$ and $K^+$ increased; the fecal concentrations of $Na^+$ and$K^+$for the normotensive and hypertensive controls were not different. Urinary excretion of $Na^+$ was higher in the normotensive Wistar rats than in the SHRs, while that of $K^+$ was not significantly different. These findings indicate that consumption of radish leaves might have had antihypertensive effects in SHRs by increasing the serum concentration of NO and fecal concentration of $Na^+$ and enhancing antioxidant activities.

Studies on Biological Activity from Antler extract added Medical plants (한약재를 가미한 녹용추출물의 생리활성에 관한 연구)

  • This study was designed to investigate biological activity of antler extract added medical plants. The scavenging activity of DPPH radical was low scavenging activity at 0.01% concentration. But in the 0.05% and higher concentration, electron donating ability(EDA) is above 50% except Kongindangagam(48.5%) and significantly good above 70% in the 4 extracts. Superoxide dismutase(SOD)-like activity was 44.3% and 45.1% extracts of Ohjayenjongwhangami and M(market sample), Inhibition of xanthine oxidase were above 50% at 0.5% concentration except Boshingiwhangwhangami and from 62.4% to 84.9% in the 4 extracts. Inhibition rate of boshingiwhangwhangami was hasty increased from 33.5% to 77.5% at 1.0% concentration and others the higher concentration, the more increasing inhibition. Angiotension I-converting enzyme(ACE) inhibitory activities were high activity all of extracts. In the 0.5% concentration, ACE inhibition was above 80%. Especially 0.01% concentration of M was presented 81.8%. The study which conducted to investigate the effect of feeding antler extract group for 50 days on sperm concentration, Ca contents of serum, kidney and femur in rats was higher than that saline group.

The Effect of Makgeolli on Blood Flow, Serum Lipid Improvement and Inhibition of ACE in vitro (막걸리의 혈행, 지질개선 효과 및 in vitro에서의 ACE 저해 효과)

  • This study was conducted to investigate the effect of Makgeolli (MG) and Makgeolli GiGemi (MGG) on blood flow, serum lipid improvement in vivo, and inhibition of angiotensin converting enzyme (ACE) in vitro. The activities of serum AST and ALT were increased by ovariectomy. Serum AST levels were decreased to $77.71{\pm}13.97$ and $74.57{\pm}14.90\;unit/ml$ in the OVX-MG50 and OVX-MGG50 groups compared to the OVX-control group ($91.14{\pm}12.02\;unit/ml$). Serum ALT levels were decreased to $34.00{\pm}8.41$ and $30.43{\pm}3.60\;unit/ml$ in OVX-MG50 and OVX-MGG50 groups compared to the OVX-control group ($37.14{\pm}5.40\;unit/ml$). Serum total cholesterol and triglyceride contents decreased in the sham group compared with OVX-control group by ovariectomy. Six weeks feeding of MG and MGG resulted in a decrease to $116.14{\pm}36.02$ and $109.14{\pm}11.55\;mg/dl$ compared to the OVX-control group ($120.43{\pm}8.36\;mg/dl$) in serum total cholesterol, and triglyceride levels were decreased to $52.43{\pm}12.41$ and $47.29{\pm}12.08\;mg/dl$ in the OVX-MG50 and OVX-MGG50 groups compared to the OVX-control group ($58.57{\pm}5.47\;mg/dl$). The level of HDL-cholesterol in the OVX-control group was significantly reduced to $51.29{\pm}20.49\;mg/dl$ compared to the sham group ($72.29{\pm}10.29\;mg/dl$), but it was increased to $70.71{\pm}19.53$ and $62.00{\pm}20.20\;mg/dl$ with MG and MGG supplementation. Furthermore, the effect of the MG group was higher than the MGG group. Microscopic observation showed that whole blood passed smoothly through the micro channels in the MG and MGG supplemented groups. The platelet aggregation ability of the groups treated with MG and MGG was less than that of the OVX-control group. In vitro assay, the angiotensin converting enzyme (ACE) activity was significantly inhibited by MG and MGG (82.6% and 68.9% inhibition at 0.4 g/ml). These results suggest that the beneficial effects of MG and MGG may be used to improve on the lipid metabolic syndrome of menopausal women. In addition, MG and MGG might improve blood homeostasis mediated activities via antiplatelets and MG and MGG may be used as antihypertensive functional foods and nutraceuticals.

Effect of Biologically Active Fractions from Onion on Physiological Activity and Lipid Metabolism (양파로부터 분리한 생리활성분획이 생리활성 및 지질대사에 미치는 영향)

  • From the 80% ethanol extraction of onion flesh, fraction I,II and III were separated by Sephadex LH-20 column chromatography. Fraction II and III had a high inhibitory effect on ACE and xanthine oxidase. The ACE was about 80% at 50 ppm of fraction IIand III. The xanthine oxidase was about 82.5% at 100 ppm of fraction In. This research was studied to investigate the effects of the fraction In isolated from onion on the reduction of fat accumulation in rats fed high fat diet for 2 months. The metabolism was examined by analyzing the fat in serum and liver. It was shown that cholesterol in serum was remarkably reduced in onion fraction In as compared with those of control.
