A Study on Effect of Soeuminsohabhyangwon on the Brain Serotonin contents of Stressed Mice (소음인(少陰人) 소합향원(蘇合香元)이 Stress 생쥐의 뇌(腦) Serotonin 함량(含量)에 미치는 영향(影響))

  • This study was aimed to evaluate the anti-stress effect of Soeuminsohabhyangwon on the mice in Cold and Swimming stress.In order to investigate the anti-stress effect of Soeuminsohabhyangwon in Cold and Swimming stressed mice, the serotonin contents were measured by HPLC method in various part of mouse brain The following results were observed. 1. In Cerebral Cortex of Frontal Lobe, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased in the SHW group as compared with Control group. 2. In hypothalamus, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with Control group. 3. In corpus striatum, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with Control group. 4. In hippocampus, the serotonin content was decreased in the Control group as compared with Normal group and the serotonin content was increased with statistical significance in the SHW group as compared with control group.Base on the above results, it may be concluded that Soeuminsohabhyangwon are effective to reduce stress.

Effect of Dietary Protein Level and Tryptophan Administration on Brain Serotonin Metabolism (식이 단백질 수준 및 Tryptophan 투여가 Serotonin 대사에 미치는 영향)

  • This study was designed to confirm the effect of dietary protein level and oral administration of tryptophan on brain serotonin metabolism. Two animal experiments were conducted. The objectives and results of research were as follows : In the first experiment, it was investigated whether administration of reserpine to Sprague-Dawley rats fed 6% or 20% casein diet induced decrease in serum tryptophan and large neutral amino acid(LNAA) concentrations, tryptophan/LNAA concentration ratio, brain tryptophan, serotonin and 5-hydroxyindoleacetic acid(5-HIAA) contents. Brain serotonin content of 6% casein diet group was lower than those of 20% casein diet group. Both 6% and 20% casein diet groups administered with reserpine to induce the analogous depression, showed the notable decrease in brain serotonin content when they were compared with 20% casein diet group not administered with reserpine. Serum tryptophan/LNAA ration and brain 5-HIAA content showed a tendency similar to the change of serotonin content, but the mean difference among all groups was not significant. From these results, it could be said that when the dietary protein level was low, brain serotonin content was decrease. The second experimnt was to see the change in serum tryptophan concentration and tryptophan/LNAA ratio and brain tryptophan, serotonin and 5-HIAA content when tryptophan was administered orally to the animals treated with reserpine. Serum tryptophan concentration tended to increase in both reserpine-treated 6% and 20% casein diet groups administered with tryptophan, especially in the 6% casein diet group. Serum tryptophan/LNAA concentration ratio tended to incrase in reserpine-tteated 6% casein diet group, while decrease in reserpine-treated 20% casein diet group. Brain tryptophan content was increased in both reserpine-treated 6% and 20% casein diet groups. However, brain serotonin content of reserpine-treated 6% casein diet group showed a tendency to decrease, while that of reserpine-treated 20% casein group increase. Consequently, the effect of tryptophan administration on increase of brain tryptophan and serotonin content in animals treated with reserpine was far more excellent in 20% casein diet groups. It was concluded that dietary protein intake and tryptophan administration increase brain serotonin level. Accordingly, it was possible to confirm that brain function, particularly in aspect of behavior related to the serotonin, was changed with manipulation of dietary composition.

Effects of Protoberberine Compounds on Serotonin Content in P815 Cells (Protoberberine 화합물이 P815 세포중의 serotonin 함량에 미치는 영향)

  • The effects of protoberberine compounds on serotonin biosynthesis in P815 cells were investigated. Protoberberine compounds such as berberine, palmatine and coralyne decreased serotonin content dose-dependently, but coptisine did not. The $IC_{50}$ values of berberine, palmatine and coralyne were $3.0\;{\mu}M,\;16.5\;{\mu}M\;and\;14.5\;{\mu}M$, respectively. Protoberberine compounds at concentrations up to $20\;{\mu}M$ were not cytotoxic towards P815 cells. The activity of tryptophan hydroxylase, a ratelimiting enzyme in serotonin biosynthesis, was inhibited by the exposure of berberine, palmatine and coralyne in P815 cells (14.9-19.3% inhibition at $2-15\;{\mu}M$), but that of aromatic L-amino acid decarboxylase was not. These results suggest that the inhibition of tryptophan hydroxylase activity by berberine, palmatine and coralyne might partially contribute to the decrease in serotonin content in P815 cells.

  • The effects of ethaverine on serotonin biosynthesis in murine mastocytoma P815 cells were investigated. Ethaverine at 2.5${\sim}$10 ${\mu}$M decreased serotonin content in a concentration-dependent manner. The IC$_{50}$ value of ethaverine was 6.1 ${\mu}$M. Ethaverine at concentrations up to 20 ${\mu}$M was not cytotoxic towards P815 cells. Under these conditions, tryptophan hydroxylase (EC 1.14.16.4; TPH), a rate-limiting enzyme in serotonin biosynthesis, was inhibited by ethaverine in P815 cells (15.3% inhibition at 7.5 ${\mu}$M), however, aromatic L-amino acid decarboxylase (EC 4.1.1.28) was not. These results indicate that ethaverine leads to a decrease in serotonin content by reducing TPH activity in P815 cells.

Inhibitory effects of tetrahydropapaverine on serotonin biosynthesis in murine mastocytoma P815 cells

  • The inhibitory effects of tetrahydropapaverine on serotonin biosynthesis in serotonin-producing murine mastocytoma P815 cells were investigated. Tetrahydropapaverine at concentration ranges of 5-20 M decreased serotonin content in a concentration-dependent manner in P815 cells and showed 42.1 % inhibition of serotonin content at 5.0 M for 24 hr. The value of 50 % inhibitory concentration, IC$\sub$50/, of tetrahydropapaverine was 6.2 M. Under these conditions, tryptophan hydroxylase (EC 1.14.16.4, TPH), was inhibited for 24-36 hr after treatment with tetrahydropapaverine in P815 cells(49.1 % inhibition at 7.5 M). (omitted)

  • This experiment was designed to investigate the effects of So-Oochim-tang-Gagam-bang (SOCT-G) on oxidative stress and serotonin metabolism in P815 Mast Cells The effects of SOCT-G on activity of 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging and Super Oxide Dismutase (SOD) in P815 mast cells were investigated. The effect of SOCT-G on content of serotonin in P815 mast cells was investigated. The effects of SOCT-G on expression of 5-hydroxytryptamine transporter (5-HTT), Tryptophan hydroxylase 1 (TPH-1) mRNA in P815 mast cells were investigated. The SOCT-G increased DPPH radical-scavenging activity in P815 mast cells. The SOCT-G increased SOD activity in P815 mast cells. The SOCT-G decreased the intracellular content of serotonin in P815 mast cells. The SOCT-G decreased 5-HTT and TPH-1 mRNA expression in P815 mast cells. This experiment shows that So-Ochm-Tang-Gagam-bang has a significant effect of oxidative stress that help prevent free radical damage. And So-Ochim-Tang-Gagam-bang decreased the intracellular content of serotonin and mRNA expression of 5-HTT and TPH-1. Therefore, further researches are suggested to reveal the anti-depressive effectiveness of So-Ochim-Tang-Gagam-bang.

  • Objectives: This study was perfomed to investigate the antioxidant activity and serotonin activity of Gami-guibi-tang on P815 Mast Cell. Methods: The effects of Gami-guibi-tang on activation of TPH-1 mRNA and AAADC mRNA in P815 mast cell were investigated. The effect of Gami-guibi-tang on content of serotonin in P815 mast cell was investigated. The effects of Gami-guibi-tang on activation of DPPH radical scavenging and SOD in P815 mast cell were investigated. Results: 1. The Gami-guibi-tang increased SOD activity and DPPH radical scavenging activity. 2. The Gami-guibi-tang increased the intracellular concentration of serotoninin 60 ${\mu}g/ml$, 80 ${\mu}g/ml$ experiment group. 3. The Gami-guibi-tang increased menaingful the manifestation TPH mRNA. 4. The manifestation of AAADC and MAO mRNA have not made menaingful changes on Gami-guibi-tang. Conclusions: This experiment shows that Gami-guibi-tang had significant anti-oxidative effect. And Gami-guibi-tang increased the intracellular concentration of serotonin. Therefore, Gami-guibi-tang can be used by the medication of major depression disorder. But Study on mechanism of increased serotonin and clinical research of Gami-guibi-tang is suggested for future research.

In Vitro Studies on the Release of Intracelluar Prolactin from Lymphocytes Using Strees Related Amines and Hormones

  • Circulating lymphocytes collected from control and heat-stressed buffaloes were subjected to in vitro culture with glucocorticoids, epinephrine or serotonin and their effect, if any, on the release of intracellular prolactin (PRL) was studied using ELISA and C-ELISA techniques. It was noted from the study that PRL level was higher in lymphocytes than in plasma of the control and heat-stressed animals, and that the PRL levels increased in the plasma of heat-stressed animals compared to that of non stressed animals with a significant decrease in lymphocytic PRL content by heat stress. Epinephrine and serotonin significantly increased the release of intracellular PRL from the lymphocytes of both in the control and the heat-stressed buffaloes but release of PRL from lymphocyte was not significantly changed by cortisol treatment in both control and heat-stressed buffaloes as compared to epinephrine and serotonin in vitro. When lympocytes were incubated with serotonin, it caused drastic lysis of the lymphocytes but epinephirine and cortisol did not show any lysis. It may be concluded from this study that hormones like epinephrine or serotonin known to increase during stress, release intracellular PRL from lymphocytes, the satellite PRL storage/synthesizing organ of blood, although the mechanism of the release is different.

Experimental Effects of Aucklandiae Radix and Cyperi Rhizoma Extract on Chronic Stress in Rats (목향과 향부자 추출물이 흰쥐의 만성 스트레스에 미치는 실험적 효과)

  • This study aimed to investigate the anti-stress function of Aucklandiae Radix (AR) and Cyperi Rhizoma (CR). The essential oils used in the experiment were extracted from AR and CR using Steam Distillation Extraction and Super critical CO2 extraction. To observe the effects of sample administration, we measured feed intake, leukocytes, red blood cells, hemoglobin, platelets, serum serotonin content, immobility time, climbing time, and swimming time in mice subjected to chronic restraint stress as behavioral changes. The average body weight of all experimental groups increased than the average body weight of the control group. The immobility and climbing times of experimental groups A and B administered with supercritical extraction samples were shorter than those of the other experimental groups and the control group, and the swimming time was longer. The serotonin content in the blood of all experimental groups decreased compared to the normal group, and the serotonin content of the control group was increased. The authors suggest that Korean herbal medicines AR and CR may be utilized as anti-stress flavoring agents based on the above results.

The Effects of OnDam-tang-Kami-bang (ODK) in Antioxidant and Serotonin Metabolism Testing on P815 Cell (온담탕가미방(溫膽湯加味方)의 항산화와 Serotonin 대사 과정에 미치는 영향)

  • Objectives : The purpose of this study is to examine the effects of antioxidant activities and serotonin activities of OnDam-tang-Kami-bang (ODK) on P815 Mast Cell. Methods : The effects of ODK on the activation of DPPH radical-scavenging and SOD in P815 mast cell were investigated. The effect of ODK on the content of serotonin in P815 mast cell was investigated. The effects of ODK on the activation of 5-HTT, TPH-1 mRNA in P815 mast cell were investigated. Results : It was found that the ODK increased SOD activities and DPPH radical-scavenging activities in the P815 mast cell. Also, the ODK decreased the intracellular concentration of serotonin in the P815 mast cell. Further, the ODK decreased 5-HTT and TPH-1 mRNA expression in the P815 mast cell. Conclusions : The results of this experiment reveal that ODK has significant antioxidative effects. However, ODK decreased the intracellular concentration of serotonin and mRNA expression of 5-HTT and TPH-1, which implies that ODK might not be effective for treating depression. Further research exploring the positive aspects of ODK is suggested such that ODK could adequately target symptoms that are to be treated.
