Analysis of Genetic Diversity of Apple Cultivars Using RAPD and SSR Markers (RAPD와 SSR 마커를 이용한 사과 품종의 유전적 다양성 분석)

  • In this study, random amplified polymorphic DNA (RAPD) and simple sequence repeat (SSR) analyses were utilized for evaluation of genetic diversity of 34 Korean bred and introduced apple cultivars. Thirty-seven RAPD primers detected a total of 193 polymorphic bands (36.2%) with an average of 5.6. Twenty-six SSR markers generated a total of 112 alleles with an average 4.3 alleles per locus. Genetic diversity of 34 cultivars estimated by polymorphic information content (PIC) value ranged from 0.536 (CH03d12) to 0.952 (CH04c06) with an average of 0.843. By UPGMA (unweighted pair-group method arithmetic average) cluster analysis with 305 polymorphic bands, the apple cultivars were classified four groups by similarity index of 0.640. The 'Seokwang' was included in group I. Group II consisted of 12 cultivars which have 'Golden Delicious' in their pedigree, with the exception of 'Spur Earliblaze' and 'Jonathan'. Group III included 13 cultivars which have usually 'Fuji' in their ancestry and bud sport of 'Fuji' cultivars. Group IV consisted of 8 cultivars with 'Hongro', 'Gamhong', and 'Saenara'. Similarity values among the tested apple cultivars ranged from 0.529 to 0.987, and the average similarity value was 0.647. The similarity index was the highest (0.987) between 'Hwarang' and 'Danhong', and the lowest (0.529) between 'Seokwang' and 'Hwarang'. The genetic relationships among the 34 studied apple cultivars were basically consistent with the known pedigree.

Research on the Re-organization of the Administration of Labor's Records in the custody of the National Archives (노동청 기록의 재조직에 관한 연구 - 국가기록원 소장 기록을 중심으로 -)

  • The Administration of Labor was responsible for the technical and practical functions like policy-making of labor matters and implementing the relevant laws. However, there has been a few record transferred to the National Archives to help find out the labor policy-making process. This is one of the typical examples that shows the discontinuity and unbalance, and disorderly filing of the administrative records in Korea. Naturally it is almost impossible to retrieve the appropriate content through the records file-name. Users should be at the trouble to compare the record items and their content one by one. For the re-organization of the Administration of Labor' records, this research suggests the four-level analysis of functions of the Administration. The Administration of Labor' survived records could be linked to each level function. And the publication of the 'Records Abstract Catalog' providing users with more information about the records would pave the way for easier access to the records. In addition, it also suggests the logical re-filing of the survived records of which we cannot find the order or sequence. This re-organization of the survived records would help to establish the acquisition and appraisal policy of the labor records as well as the new way of description and finding tool hereafter. Drawing up labor history map is a starting point for the acquisition strategy of the labor records, which could allow users to gain systematic access on the survived records. Of course, extensive investigation and research on the survived records is a prerequisite for the map. It would be required to research on the survived records of the other government agencies, including economic-social area ministries and investigation agencies and the National Assembly as well. It is also needed to arrange and typify the significant incidents and activities on thematic and periodic frames in the labor history. If possible to understand or connect the survived records and these accomplishments comprehensively, it would be of great help for the acquisition of the labor records and the related oral records projects.

Electroencephalographic Changes Induced by a Neurofeedback Training : A Preliminary Study in Primary Insomniac Patients (뉴로피드백 훈련에 의한 뇌파 변화 연구 : 일차성 불면증 환자에 대한 예비 연구)

  • Objectives: Insomnia is one of the most prevalent sleep disorders. Recent studies suggest that cognitive and physical arousal play an important role in the generation of primary insomnia. Studies have also shown that information processing disorders due to cortical hyperactivity might interfere with normal sleep onset and sleep continuity. Therefore, focusing on central nervous system arousal and normalizing the information process have become current topics of interest. It has been well known that neurofeedback can reduce the brain hyperarousal by modulating patients' brain waves during a sequence of behavior therapy. The purpose of this study was to investigate effects of neurofeedback therapy on electroencephalography (EEG) characteristics in patients with primary insomnia. Methods: Thirteen subjects who met the criteria for an insomnia diagnosis and 14 control subjects who were matched on sex and age were included. Neurofeedback and sham treatments were performed in a random order for 30 minutes, respectively. EEG spectral power analyses were performed to quantify effects of the neurofeedback therapy on brain wave forms. Results: In patients with primary insomnia, relative spectral theta and sigma power during a therapeutic neurofeedback session were significantly lower than during a sham session ($13.9{\pm}2.6$ vs. $12.2{\pm}3.8$ and $3.6{\pm}0.9$ vs. $3.2{\pm}1.0$ in %, respectively; p < 0.05). There were no statistically significant changes in other EEG spectral bands. Conclusion: For the first time in Korea, EEG spectral power in the theta band was found to increase when a neurofeedback session was applied to patients with insomnia. This outcome might provide some insight into new interventions for improving sleep onset. However, the treatment response of insomniacs was not precisely evaluated due to limitations of the current pilot study, which requires follow-up studies with larger samples in the future.

Somatic Cell Analysis and Cobalamin Responsiveness Study in Ten Korean Patients with Methylmalonic Aciduria (한국 메틸말로닌산혈증 환아 10례에서 Somatic Cell 분석과 cobalamin 반응성 연구)

  • Purpose: Isolated methylmalonic acidemia (MMA) is an autosomal recessive inherited disorder of propionate metabolism. There are two subtypes of MMUT gene defects. $Mut^0$ represents complete loss of methylmalonyl-CoA mutase (MCM) activity while mut- is associated with residual MCM activity, which can be stimulated by hydroxocobalamin (OHCbl) supplementation. The objective of this study is to investigate cobalamin responsiveness and mutations present in Korean MMA population. Methods: We evaluated 10 MMA patients using somatic cell complementation analysis on their fibroblasts to measure MCM activity and vitamin B12 responsiveness for the optimal treatment. MMUT gene was sequenced to identify the MMA mutations. Results: For all patients, the incorporation of $[^{14}C]-propionate$ was low, and there was no response to OHCbl. The incorporation of $[^{14}C]-methyltetrahydrofolate$ and $[^{57}Co]-CNCbl$ fell within the normal range. There was adequate synthesis of methylcobalamin while the synthesis of adenosylcobalamin was low. The complementation analysis showed all patients were $mut^0$. The sequence analysis identified 12 different MMUT mutations, including 2 novel mutations, p.Gln267Ter and p.Ile697Phe, were identified. All the patients in this study had neonatal onset of symptoms, belonged to $mut^0$ complementation class, and as a result, showed no cobalamin responsiveness. Conclusion: No Korean MMA patient showed cobalamin responsiveness.

Geological Structures of the Taean Formation in the Gomseom Area, Southwestern Gyeonggi Massif (경기육괴 남서부 곰섬 일대 태안층의 지질구조)

  • The western Gyeonggi Massif, where records evidence of Phanerozoic subduction/collision tectonics, is an important area to understand the crustal evolutionary history of the Korean Peninsula. This study presents geometric and kinematic characteristics of the geological structures of the Taean Formation in the Gomseom area, southwestern Gyeonggi Massif. We interpreted the geometric relationships between structural elements, and conducted stereographic and down-plunge projections for structural domains. As a result, at least three different deformational events ($D_1$, $D_2$ and $D_3$) are recognized in the study area. In the first deformational event ($D_1$), regional foliations being well defined by the preferred orientation of muscovite and biotite were formed. In the second deformational event ($D_2$), NNE-trending low-angle contractional faults and related crenulation lineations/cleavages were formed. The crenulation lineations shallowly plunge toward SSW~SSE or NNW~NNE. In the third deformational event ($D_3$), SE-plunging folds and NE-trending high-angle faults were formed as 'fault-related fold' and 'fold-accommodation fault', indicating that the $D_3$ folds and faults are genetically linked to each other. This contribution provides important insights into the structural evolution of the Taean Formation along western Gyeonggi Massif, where had evolved as subduction/collisional orogenic belts in the East Asia.

Functional Expression of an Anti-GFP Camel Heavy Chain Antibody Fused to Streptavidin (Streptavidin이 융합된 GFP항원 특이적인 VHH 항체의 기능적 발현)

  • With strong biotin binding affinity ($K_D=10^{-14}M$), the tetrameric feature of streptavidin could be used to increase the antigen binding activity of a camel heavy chain (VHH) antibody through their fusion, here stained with biotinylated horseradish peroxidase and subsequent immunoassays ELISA and Western blot analysis. For this application, we cloned the streptavidin gene amplified from the Streptomyces avidinii chromosome by PCR, and this was fused to the gene of the 8B9 VHH antibody which is specific to green fluorescent protein (GFP) antigens. To express a soluble fusion protein in Escherichia coli, we used the pUC119 plasmid-based expression system which uses the lacZ promoter for induction by IPTG, the pelB leader sequence at the N-terminus for secretion into the periplasmic space, and six polyhistidine tags at the C-terminus for purification of the expressed proteins using an $Ni^+$-NTA-agarose column. Although streptavidin is toxic to E. coli because of its strong biotin binding property, this soluble fusion protein was expressed successfully. In SDS-PAGE, the size of the purified fusion protein was 122.4 kDa in its native condition and 30.6 kDa once denatured by boiling, suggesting the tetramerization of the monomeric subunit by non-covalent association through the streptavidin moiety fusing to the 8B9 VHH antibody. In addition, this fusion protein showed biotin binding activity similar to streptavidin as well as GFP antigen binding activity through both ELISA and Western blot analysis. In conclusion, the protein resulting from the fusion of an 8B9 VHH antibody with streptavidin was successfully expressed and purified as a soluble tetramer in E. coli; it showed both biotin and GFP antigen binding activity suggesting the possible production of a tetrameric and bifunctional VHH antibody.

Quality and genetic relationship on dehulling characteristic in oat variety (국내 귀리품종 탈부특성에 따른 품질 및 유연관계 분석)

  • The consumption of oats (Avena sativa L.) with high nutritional utility is accelerating due to the increased consumers' demand for functional foods. In Korea, naked oats are used as food, while covered oats are used for animal feed. However, it is difficult to distinguish naked oats from covered oats when the husk is removed from the grains by a special process. The present study was carried out to investigate experimental methods that would be beneficial in the segregation of different types of oats after husk removal. Grain quality-related biochemical compounds were analyzed in a bid to differentiate the oat dehulling characteristics. In addition, 61 SSR markers were examined for genetic relationship and variety identification of oats using five naked and seven covered oat varieties. Results showed that, the contents of protein, lipid, and ${\beta}-glucan$ were not significantly different among the oat varieties and this could not be used as an index for distinguishing oats husk character. However, in the fatty acid composition ratio,, naked oats had a higher ratio of stearic acid (C18:0) and oleic acid (C18:1) than covered oats, and covered oats had a higher ratio of linoleic acid (C18:2) and linoleic acid (C18:3) than naked oats. The assessment of SSR marker genotype revealed that 33 polymorphic bands among 12 oat varieties and 1 variety could be distinguished through the combination of polymorphic markers thus indicating the usability of these markers for variety identification in oats.

Hydrothermal Evolution for the Inseong Au-Ag Deposit in the Hwanggangri Metallogenic Region, Korea (황강리 광화대 인성 금-은 광상의 광화 유체 진화)

  • The Inseong Au-Ag and base metal deposit, located in Chungchengbuk-do, Korea, consists of series of quartz veins filling fissures. The deposit occurs in Hwanggangri meta-sediment formation, a lime pebble-bearing phyllite, in the Okcheon Supergroup. Abundant ore minerals in the deposit are pyrite, arsenopyrite, sphalerite, chalcopyrite and galena. The gangue minerals are quartz, calcite and chlorite. Hydrothermal alteration such as chlorization, silicitication, sericitization and carbonitization can be observed around the quartz veins. 4 vein stages can be distinguished based on its paragenetic sequence, vein structure, alteration features and ore minerals. Microthermometry of the fluid inclusion assemblages occur in the veins are conducted to reconstruct a hydrothermal P-T evolution. Fluid inclusions in clean and barren quartz vein in stage 1 have Th of $270{\sim}342^{\circ}C$ and salinity of 1.7~6.4 (NaCl eqiv.) wt%. Euhedral quartz crystal in stage 2 have Th of $108{\sim}350^{\circ}C$ and salinity of 0.5~7.5 wt%. Barren milky quartz vein in stage 3 have Th of $174{\sim}380^{\circ}C$ and salinity of 0.8~7.5 wt%. Calcite vein in stage 4 have Th of $103{\sim}265^{\circ}C$ and salinity of 0.7~6.4 wt%. Calculated paleodepth about 0.5~1.5 km (hydrostatic pressure) indicate epithermal ore-forming condition. Shallow depth but relatively high-T hydrothermal fluids possibly create a steep geothermal gradient, sufficient for base metal precipitation in the Inseong deposit.

A Study on Technological Thinking Disposition of the Specialized and Meister High School Students (특성화 및 마이스터 고등학교 학생들의 기술적 사고성향 연구)

  • This study has been performed for the purpose of providing the basic pedagogical resources to the specialized and meister high school related educational settings in either constructing educational contents suitable for the students'technological thinking disposition or fortifying the disposition required for the students.. A tool of secured validity and reliability has been used to test technological thinking disposition for the specialized and Meister high school students. Followings are the major results of the survey analysis for the subjects. 1. Technological thinking disposition with the most component ratio for the specialized & meister high school was turned out to be Technological Operating Disposition(TOD). The second most was Technological Planning and Reflecting Disposition(TPRD). Technological Curiosity Disposition(TCD), Technological Problem Identifying and Resolving Disposition (TPIRD), Technological Analyzing Disposition(TAD), Technological Creativity and Expressing Disposition(TCED) were in sequence. 2. In comparing the technological disposition of specialized high school students with that of meister high school students, the statistical test showed no evidence for the difference between the two group. 3. Statistical comparison test for the gender difference in technological thinking disposition has also been performed for the students of specialized and meister high school. The result showed that the magnitude of difference between the component ratio of TCD and TPIRD for the boy students was bigger than that for the girl students. In the TPRD, the component ratio for the disposition of the girl students was bigger than that of the boy students on the other hand. 4. For the comparison test of the technological thinking disposition between boys and girls only for the specialized high school students, the results showed the same different component ratio results as the results of the test for the specialized and meister high school students. 5. For the gender difference of meister high school student for the technological thinking disposition, there was no statistical evidence supporting the difference.

Hydrolysis of Non-digestible Components of Soybean Meal by α-Galactosidase from Bacillus coagulans NRR1207 (Bacillus coagulans NRR1207이 생산하는 α-galactosidase에 의한 대두박 비소화성분의 가수분해)

  • The fermentation of non-digestible soy meal can convert polysaccharides into many compounds that have a wide variety of biological functions. Bacillus strains are capable of hydrolyzing non-digestible saccharides, such as melibiose, raffinose, and stachyose, found in soy meal components. A highly active ${\alpha}$-galactosidase (${\alpha}$-d-galactoside galactohydrolase, EC 3.2.1.22) was isolated from a bacterium in a traditional Korean fermented medicinal herb preparation. The isolate, T2-16, was identified as Bacillus coagulans based on its 16S rRNA sequence and biochemical properties, and the strain was named Bacillus coagulans NRR-1207. When incubated in 10%(w/v) skim milk, Bacillus coagulans NRR1207 caused a decrease in the pH of the culture medium, as well as an increase in titratable acidity and viable cell counts. This strain also showed higher activities of ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\alpha}$-glucosidase, naphthol-AS-BO-phosphohydrolase, and acid phosphatase when compared to other enzymes. It hydrolyzed oligomeric substrates, such as raffinose and stachyose, and liberated galactose, indicating that the Bacillus coagulans NRR1207 ${\alpha}$-galactosidase hydrolyzed the ${\alpha}$-1,6 glycoside linkage. These results suggest that the decreased stachyose and raffinose contents observed in fermented soy meal are due to this ${\alpha}$-galactosidase activity. Bacillus coagulans NRR1207 therefore has potential probiotic activity and could be utilized in feed manufacturing, as well as for hydrolyzing non-digestible soy meal components.
