PLANT BIOCHEMISTRY OF GINSENG SAPONINS (I) Saponins and Sapogenins from American Ginseng Plants

  • The sapogenins of two-and four-year-old A-merican ginseng plants (Panax quinquefolium L.) (Araliaceae) collected in July and September were studied. American ginseng saponins (panaquilins) differ from Korean ginseng (Panax ginseng C. A. Meyer) saponins (ginsenosides). The American ginseng saponins separated and named were panaquilins A, B, C, D, E-l, E-2, E-3, G-l, G-2, (c) and (d). One-dimensional thin-layer chromatography did not completely separate panaquilin mixture and were subject to misinterpretation. The panaquilins were more accurately separated and identified by the two-dimensional thin-layer method established. Some differences in American ginseng saponins were dependent upon the plant age, time of collection, and part extracted. The American ginseng sapogenin components are panxadiol (panaquilins B and C), oleanolic acid (panaquilin D) and panaxatriol (panaquilin G-l). The panaquilins E-l, E-2 and E-3 mixture contains both panaxadiol and panaxatriol. The genins of panaquilins A, (c), (d) and G-2 were not identified. In addition, ${\beta}-sitosterol$ and stigmasterol were identified from the root ether extracts.

Antimicrobial Effect of Coptidis rhizome Extract against Mutans Streptococci and Periodontopathogens

  • The purpose of the study was to investigate the antimicrobial activity of the methanol extract of Coptidis rhizome against the type strains of cariogenic bacteria, Streptococcus mutans and Streptococcus sobrinus, and the periodontopathogens, Porphyromonas gingivalis, Prevotella intermedia, Treponema denticola and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of the crude extract and the methanol extract fractions of Coptidis rhizome separated by silica gel chromatography were evaluated by determining the minimal bactericidal concentration (MBC) values, using the microdilution method. The cell viability test of the extracts of Coptidis rhizome on the KB cells was also studied by methyl thiazolyl tetrazolium (MTT) assay. Our results showed that the 11th fraction (F11) of the methanol extract had the greatest antimicrobial activity against the tested bacteria, with no associated cytotoxicity on the KB cells, upto a concentration of $50{\mu}g/ml$. These results suggest that the silica gel chromatography fraction F11 of the methanol extract of Coptidis rhizome, could be useful in the development of oral hygiene products as an antimicrobial agent for the prevention of dental caries and periodontal diseases.

  • A new cyclone system for the vacuum cleaner to collect dusts has been studied experimentally and numerically to meet the constant suction power, hygienic exhaust and a reduction of maintenance cost. The cyclone system of the vacuum cleaner consists of twin cyclones for improving dust collection efficiency. The first. cyclone catches large dust particles and the second one having two separated flows to decrease pressure drop collects small dust particles. The optimal design factors such as dust collection efficiency, pressure drop, and cut-size are investigated from the experimental results by the Taguchi method. Cyclone cleaner systems designed in this study has a good Performance taking into account the dust collection efficiency of 93% and the cut-size of $1.6{\mu}m$ in mass median diameter at the flow rate of 1 CMM. The cyclone vacuum cleaner showed the potential to be an effective method to collect dusts generated in the household.

  • This study aimed to apply a methodological approach, 'social network analysis' to a case study for the understanding of relational structure among stakeholders related to green tourism development. By doing so, this study argued that it is important to identify stakeholder's network structure to help green tourism planners develop collaborative relationship among stakeholders. This study identified the stakeholders regarding a community-based festival development in the southern area of Korea, and investigated two types of networks among them: decision-making power relational and intimate network. Interviewer-administrated survey and in-depth interview were employed for data collection. The data was analyzed by SPSS (version 10.0) and Net-MinerII (version 2.5.0), and by constant comparison method. The result revealed that low different groups of the stakeholders were separated in the intimate networt and that the festival organizational body was not connected with other stakeholders in the decision-making power relational network. The existence of separated groups and weak relationship among the stakeholders appeared to relate to age-group differences, and different views on the festival between the stakeholders.

Two new species of Trichoderma isolated from commercially grown oyster mushroom, Pleurotus ostreatus (oral)

  • We describe two new Trichoderma species associated with oyster mushroom in Korea. Trichoderma green mould has been one of the most serious diseases of oyster mushroom in Korea. Of these the predominant species are two unrecorded species. We designed as Trichoderma sp. Korean type 1 (Th K1) and Trichoderma sp. Korean type 2 (Th K2), respectively. Th K1 and Th K2 can be distinguished from previously reported Trichoderma species as well as each other in morphological characteristics including growth rate at 35$^{\circ}C$, colony morphology, conidia shape and branch pattern of phialides. Sequence of the ITS region of rDNA, the protein coding translation elongation factor gene(EF-1${\alpha}$), and RNA polymeraseII (RPB2) not only clearly separated Trichoderma sp. Korean types from their closely related T. harzianum biotype but also distinguished them from each other. Analyses of the EF-1${\alpha}$ and RPB2 sequences were found to be more useful for establishing systematic relationships among Trichoderma isolates than those of the ITS sequence. Characteristic Variations of H2O2 Concentrations Observed in Seoul (서울시 대기 중 과산화수소 농도 변화 특성)

  • During January $2002{\sim}April\;2004$, hydrogen peroxide ($H_{2}O_{2}$) measurements were performed at the campus of Korea University, which is located in the northeastern part of Seoul. Gas phase hydroperoxide was collected in aqueous solution and separated by HPLC. Concentrations were determined by fluorescence using postcolumn enzyme derivatization. This measurement system was improved to be run automatically from sample collection at every 10 minutes through chemical analysis for data collection. Detection limits of $H_{2}O_{2}$ is $10{\sim}17\;pptv$, and the overall uncertainty of the measurements is better than 8%. Two-year measurements of $H_{2}O_{2}$ show typical seasonal variations. Concentrations of $H_{2}O_{2}$ were higher during $June{\sim}October$ and lower during $January{\sim}February$. Maximum concentration of 1-hour averaged $H_{2}O_{2}$ was 6.5 ppbv, which was observed in August and September. In general $H_{2}O_{2}$ concentrations were well correlated with $O_{3}$ concentrations and largely affected by meteorological factors such as temperature and wind direction.

Ecogeological Description of Sanyang Gotjawal, Jeju Island, Korea

  • BACKGROUND: Gotjawal forest on Jeju Island is characterized by uneven topography that developed as a result of freezing-thawing weathering process and irregular substrates caused by numerous lava-flow collapses. Sanyang Gotjawal, located in southwest of Jeju Island, is a well-developed forest with a long history. In addition to photographs, there is a need for a good way to describe the unique features of Gotjawal, including its geology and vegetation. METHODS AND RESULTS: We illustrated the area's natural features using Clip Studio Paint 1.12.0. To reveal its complexity, we separated the vegetation and geological features in the illustrations. CONCLUSION(S): We drew a cross-section of Gotjawal's unique layers, including lava flows. In addition to an eco-geological approach, we described the dominant vegetation and geological characteristics of the three forest layers (trees, shrubs, and herbs) in Gotjawal.

The Regulatory Framework of Taiwan′s Municipal Waste Recycling

  • In addition to landfilling and incineration, waste minimization and recycling have become priorities in Taiwan's municipal waste management strategies over the past 10 years. Major recycling initiatives being implemented in Taiwan include producer responsibility, "pay-as-you-throw, " and mandatory sorting provisions. Currently, the producer responsibility regime established under the Waste Disposal Act and administered by the Environmental Protection Administration (EPA) requires that manufacturers or importers of listed items, including containers, batteries. cars, motorcycles ("scooters"), tires, oil, televisions, refrigerators, air conditioners, washing machines, computers, and printers, pay recycling fees to government recycling funds. EPA then uses the recycling funds to subsidize collection and recycling. The 2001 recycling fund budget totals NT$57 billion (NT$35 = US$l). Under the producer responsibility regime, EPA-designated retailers ("sellers") must accept end-of-life items returned to them by the consumers. At the local level, Taipei City implements a pay-as-you-throw program, whereby citizens pay waste collection and treatment fees through the purchase of special trash bags approved by the Taipei City Government. However. recyclables that are separated by citizens are collected free-of-charge by the City. Taichung City and Kaohsiung City, on the other hand, enforce mandatory sorting schemes, whereby citizens face penalties if they don't separate recyclables from the trash before pick-up. These programs have resulted in a significant reduction in municipal waste. Per capita waste collected per day has dropped from 1.143 kg in 1997 to 0.978 kg in 2000. Targeting a 10% recycling rate for municipal waste in 2001. EPA plans to research and develop new recycling techniques, expand the scope of producer responsibilities, and strengthen existing municipal recyclable collection programs. To this end, among other initiatives, EPA has prepared a draft "Resource Recycling and Reuse Act" that would complement the existing producer responsibility programs by incorporating into the current recycling framework the concept of product life-cycle management.

A Study on the Requirements for Building of the Regional Library Portal (지역기반 도서관 포털구축의 요건에 관한 연구)

  • The purpose of this study is to analyze the political and functional requirements of the regional library portal and to evaluate such requirements. A literature review was carried out to discuss library portal, and introduce the best practices of the regional library portal. After analyzing the political and functional requirements of the regional library portal, AHP was applied to evaluate requirements. In the political requirements, standards, digital collection development, operation and evaluation were ranked according to importance, and copyright & licence management which belongs to digital collection development was underrated. There were differences in the importance among the functional requirements of the regional library portal, integrated search, integrated interface, information service, separated digital collection management and access management were ranked according to importance.
