The Screening of Marine Birnavirus (MABV) Infected in Brood Stocks of Flounder, Paralichthys olivaceus (친어용 넙치 성어에 있어 Marine Birnavirus (MABV) 감염에 관한 검색)

  • Presence of marine birnavirus (MABV) was examined against egg and ovarian fluid, and seminal fluid from the brood stocks of flounder, Paralichthys olivaceus collected from 9 different stations around Korean peninsula. The detection rate of MABV in brood stocks flounder was observed to 34% by PCR. The mean virus titer of the PCR positive fish was $10^{2.30}$ to $10^{4.30}$ $TCID_{50}$/g(ml). By a neutralization test, all of the isolated virus were ascertained to be closely related to marine birnavirus (MABV).

Morphological and Absorptive Studies on Canal Epithelium of the Various Ducts in Rooster Epididymal Region (닭 부고환부(副睾丸部)의 각종(各種) 관상피(管上皮)의 형태(形態) 및 흡수(吸收)에 관한 연구(硏究))

  • Histological and histochemical studies were made on the lining epithelia of the various ducts in epidymis of the Rooster and absorptive function of the canal epithelial cells in the Rooster epididymis were also investigated after administration of India ink. The results obtained were summerized as follows; 1. Epihtelium lining the rate testis was mainaly composed of single later of cuboidal cells, and was partially composed of flattened squamous or low columnar cells. Efferential ductules were characterized by having many villous projections orrfolds which extened into the lumen, and were lined by stereociliated pseudostratified epithelium which consisted of manily ciliated columnar cells, a few scattered clear cells and basal cells. Connecting ductules were lined by ciliated pseudostriatified colummnar epithelium in which ciliated columnar cells, clear cells and basal cells were noted. Epididymal ducts were lined by pseudostratified epidhelium in which columnar and basal cells were noted. 2. PAS-granules, saliva resistant were noted mainly in the epithelial cells of efferential and connecting ductules. 3. Sudan black B stained heavily the granules in the epithelial cells of sufferential and connecting ductules. 4. The granules reactive to acid phosphatase most abundant in the epithelial cells of efferential ductules and were lesser amount in the epithelial cells of connecting ductules where as very few or no granules were seen in the rest of the ducts. 5. Alkaline phosphatase activity was most prominent but discontinuous in the luminal surface of the epithelium of efferential ductules and less marked in the connecting ductless. No enzyme activity was noted in the canal epithelium of epididyml duct. 6. India ink granules were most numerous in the epithlial cells of efferential ductules and were a few in connecting ductules. Very few or no granules of India ink were noted in the other types of the ducts. India ink granules in the epithelium increased gradually as the time after the administration of India ink (one up to twenty-nine hours) has proceeded. From those results it is suggested that epithelial cells of efferential and connecting ductules have active absorptive function, whereas the rest of duct system in the epididymis of the Rooster may be the mere pathway of the seminal fluid without significant modification of its constituents.

The reactive oxygen species-total antioxidant capacity score is a new measure of oxidative stress to predict male infertility

  • The imbalance between reactive oxygen species (ROS) production and total antioxidant capacity (TAC) in seminal fluid indicates oxidative stress and is correlated with male infertility. A composite ROS-TAC score may be more strongly correlated with infertility than ROS or TAC alone. We measured ROS, TAC, and ROS-TAC scores in semen from 127 patients and 24 healthy controls. Of the patients, 56 had varicocele, eight had varicocele with prostatitis, 35 had vasectomy reversals, and 28 had Idiopathic infertility. ROS levels were higher among infertile men, especially those with varicocele with prostatitis (mean ${\pm}$ SE, 3.25 ${\pm}$ 0.89) and vasectomy reversals (2.65 ${\pm}$ 1.01). All infertility groups had significantly lower ROS-TAC scores than control. ROS-TAC score identified 80% of patients and was significantly better than ROS at identifying varicocele and idiopathic infertility. The 13 patients whose partners later achieved pregnancies had a mean ROS-TAC score of 47.7 ${\pm}$ 13.2, similar to controls but significantly higher than the 39 patients who remained infertile (35.8 ${\pm}$ 15.0; P < 0.01). ROS-TAC score is a novel measure of oxidative stress and Is superior to ROS or TAC alone in discriminating between fertile and infertile men. Infertile men with male factor or idiopathic diagnoses had significantly lower ROS-TAC scores than controls, and men with male factor diagnoses that eventually were able to initiate a successful pregnancy had significantly higher ROS-TAC scores than those who failed.

  • Semen preparation for Assisted Reproductive Technology(ART) involves the separation of the motile sperm from non-motile, dead sperm, cellular debris and seminal fluid. The aim of this study is to compare the motile sperm recovery rate and motility index of different sperm preparation method(swim-up method, 80% isotonic continuous percoll method, two-layer discontinuous percoll method, mini-percoll method). In Mini-Percoll method, pellets from patients were suspended in 0.3ml of medium and layerd on a discontinuous percoll gradient consisting of 0.3ml each of 50, 70, 95% isotonic percoll. All semen samples are divided into normal and subnormal sperm group(oligo-, astheno-, oligoasthenozoospermia). Especially, we evaluated the effect of mini-percoll method in subnormal sperm group. In normal sperm group, mini-percoll method and two layer discontinuous percoll method (40%/80%) allowed increasing of motile sperm recovery rate. But motility index was higher in swimup method than the other methods. In subnormal sperm group, mini-percoll method has advantages as compare with the other methods in motile sperm recovery rate and motility index. These results suggest that modified mini-percoll method could be certainly a valuable tool in some cases of severe male factor sperm.

The impact of two doses of coenzyme Q10 on semen parameters and antioxidant status in men with idiopathic oligoasthenoteratozoospermia

  • Objective: Oxidative stress contributes to male infertility, and antioxidants have been recommended for treating idiopathic oligoasthenoteratozoospermia (OAT). There is, however, a lack of agreement on the type, dosing, and use of individual antioxidants or combinations thereof. The purpose of this study was to compare the effects of two doses of coenzyme Q10 (CoQ10) on semen parameters and antioxidant status in men with idiopathic OAT. Methods: In this prospective study, patients with idiopathic OAT received 200 mg/day (n = 35) or 400 mg/day (n = 30) of CoQ10 orally for 3 months. All patients underwent semen analysis according to the fifth editions of the World Health Organization criteria. Total antioxidant capacity (TAC), catalase (CAT) activity, and superoxide dismutase (SOD) activity were measured both before and after treatment. Results: Treatment with CoQ10 (200 mg/day or 400 mg/day) resulted in a significant increase in sperm concentration from baseline ($8.22{\pm}6.88$ to $12.53{\pm}8.11million/mL$, p= 0.019; $7.58{\pm}5.41$ to $12.33{\pm}6.1million/mL$, p= 0.002, respectively), progressive motility ($16.54%{\pm}9.26%$ to $22.58%{\pm}10.15%$, p=0.011; $14.22%{\pm}12.85%$ to $26.1%{\pm}14.52%$, p= 0.001, respectively), and total motility ($25.68%{\pm}6.41%$ to $29.96%{\pm}8.09%$, p= 0.016; $23.46%{\pm}12.59%$ to $34.82%{\pm}14.17%$, p= 0.001, respectively). CoQ10 therapy also increased TAC (p= 0.009, p= 0.001, respectively), SOD activity (p= 0.004, p= 0.001, respectively), and CAT activity (p= 0.039, p= 0.024, respectively). Furthermore, antioxidant measures correlated significantly with seminal fluid parameters (r = 0.36-0.76). Conclusion: CoQ10 supplementation improved semen parameters and antioxidant status in men with idiopathic OAT, with a greater improvement shown in men who took 400 mg/day than in those who took 200 mg/day.

  • The angiotensin-converting enzyme 2 receptor (ACE2) appears to be widely expressed in cells in the testes, predominantly in spermatogonia, Sertoli cells, and Leydig cells, and its co-expression with transmembrane protease serine 2 (TMPRSS2) is essential for the entry of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). For this reason, the male reproductive system could be considered a potential target for SARS-CoV-2, as well as a possible reservoir of infection. However, to date, there is very little evidence about the presence of SARS-CoV-2 in semen and testicular samples. The aim of this paper was to review the current evidence regarding the impact of SARS-CoV-2 on male fertility and sexual health, with a particular focus on reproductive hormones, the presence of the virus in seminal fluid and testis, and its impact on fertility parameters. We found very limited evidence reporting the presence of SARS-CoV-2 in semen and testicular samples, and the impact of SARS-CoV-2 on reproductive hormones and fertility parameters is unclear. The quality of the examined studies was poor due to the small sample size and several selection biases, precluding definitive conclusions. Hence, future well-designed prospective studies are needed to assess the real impact of SARS-CoV-2 on male reproductive function.

Properties of Semen and Sperm Motility in Black Seabream, Acanthopagrus schlegeli (감성돔, Acanthopagrus schlegeli 정액의 특성과 정자의 운동성)

  • In order to obtain the basic knowledges concerned to the semen preservation of aquacultural fishes, studies on the physical and chemical properties of semen, and sperm motility with the different osmotic pressures making by adding $Na^+,\;K^+,\; Mg^{++},\;and\;Ca^{++}$ to artificial seawater (ASW) were conducted in black seabream, Acanthopagrus schlegeli. Average semen volume per fish in one strip was 1.97ml and sperm concentration was $2.33\pm1.30\times10^{10}$ sperm/ml. Spermatocrit and pH of semen were $90.6\pm5.0\;and\;8.3\pm0.1$, respectively, Osmotic pressures of rearing seawater, seminal fluid and plasma were $939\pm24,382\pm70\;and\;342\pm77$ mOsm/l, and $Na^+,\;K^+$ and $Cl^-$ concentrations of seminal fluid were $169.5\pm4.5,\;4.9\pm2.2,\;156.0\pm2.0\;mM/l$, respectively. When semen were diluted by using $Na^+,\;K^+,\;Mg^{++}\;and\;Ca^{++}$ free ASW, only $Na^+$ free ASW had no sperm motility. As raising osmotic pressure graduary by addition of 1M NaCl to the $Na^+$ free ASW, spermatozoa showed the high motilities in 457-1128 mOsm/l, but the low motilities in 1398-1736 mOsm/l. In the case of same treatments with 1M of KCl, $MgC1_2\;and\;CaC1_2$ to the $K^+,\;Mg^{++}\;and\;Ca^{++}$ free ASW, spermatozoa revealed the high motilities in $904\~1434,\;818\~1175\;and\;956\~1343$ mOsm/l, respectively.

  • In order to observe the antigenic localization in the tissues of the adult Clonorchis sinensis, immunogold labeling method was applied using serum immunoglobulins (IgG) of either worm·infected rabbits (group I) or antigen-immunized rabbits (group II) (by the body quid obtained from the adult worms). The electron micrographs of the sectioned worm tissue antigens, embedded in Lowicryl HM 20 medium and stained with protein A-gold complex (particle sixte: 12 nm), were compared between the group I and group II. The gold particles were observed in the interstitial matrix of the worm parenchyma, the epithelial lamellae of the cecum, and the cecal lumen both in group I and II. But the particles were in general more concentrated in group II. The gold particles were not observed on the basal lamina of the tegument or on vitelline glands in group I, while they were highly concentrated on those areas in group II. There were also differences in the antigenicity of interstitial matrix(reacted with group I IgG) and head part(reacted with group II IgG) of the sperm cells in the seminal receptacle. Conclusively, it is suggested that the substances comprising the basal lamina of the tegument or vitelline glands act as specific antigens reacting with antigen(body quid) immunized rabbit IgG. On the other hand, the substances in the cecal lumen and cecal epithelial lamellae are thought to be the specific antigen that react with the worm-infected rabbit IgG.

Pre- and postnatal development study of amitraz in rats (랫드에서 amitraz의 출생 전후 발생 시험)

  • This study investigated the potential effects of amitraz on the pre- and postnatal development, behavior, and reproductive performance of offspring of parent rats given amitraz during pre-mating, gestation, and lactation. The test chemical was administered via the drinking water containing 0, 40, 120, and 360 ppm to male rats from 2 weeks before mating to the end of 14-day mating period and to females from 2 weeks before mating, throughout mating, gestation and lactation up to weaning. Based on fluid consumption, the male rats received an average of $0,\;5.7{\pm}1.33,\;13.2{\pm}2.08,$ and $35.8{\pm}3.42$ mg/kg/day amitraz, and the female rats received an average of $0,8.7{\pm}4.42,\;20.1{\pm}9.60,\;and\;47.6{\pm}22.38$ mg/kg/day amitraz, respectively. At 360 ppm, an increase in the incidence of abnormal clinical signs, a suppression in the body weight gain, a decrease in the food consumption and litter size, an increase in the post-implantation loss, and a decrease in the seminal vesicle weight were observed in the parent animals. In addition, a suppression in the body weight gain, a decrease in the grip strength, a delay in the negative geotaxis, an increase in the pre- and post-implantation loss, and a decrease in the number of live embryos were observed in the offspring. At 120 ppm, suppressed body weight gain and reduced food consumption were observed in the parent rats. Suppressed body weight gain and decreased grip strength were also observed in the offspring. There were no signs of either reproductive or developmental toxicity at 40 ppm. Under these experimental conditions, the no-observed-adverse-effect level of amitraz for parent rats and their offspring was estimated to be 40 ppm in rats.

The Effects of Flutamide and Diethylstilbestrol Exposure on the Reproductive Organs and Thyroid of Male Rats by the Rodent 20-day Thyroid/Pubertal Assay

  • To establish a test protocol for the rodent 20-day thyroid/pubertal assay, flutamide and diethylstilbestrol (DES) were administered to intact male Sprague-Dawley rats from postnatal day 33 for 20 days. Flutamide (1, 5, and 25 mg/kg/day) or DES (10, 20, and 40 ug/kg/day) was given once daily by oral gavage to immature male rats. Prepuce separation was significantly delayed in flutamide group and in DES group. One day after the last dose, the rats were killed and pituitary, thyroid, and reproductive organs were removed and weighed. Flutamide treatment resulted in a significant reduction in the weights of epididymides, ventral prostate, seminal vesicles plus coagulating glands and fluid (SVCGF), levator ani. bulbocarvenus muscles (LABC), Cowper's glands, and glans penis. The weight of adrenal glands decreased at % mg/kg/day, while testes and any other organ weights were unaffected. No microscopic changes were observed in the thyroid glands. Serum levels of testosterone wert significantly increased in the flutamide-treated groups and serum levels of estradiol were also increased. A significant reduction in the weights of testes, epididymides, ventral prostate, SVCGF, LABC, Cowpers glands, and glans penis of DES treated group. Serum testosterone and LH decreased significantly in DES group. Decrease of estradiol was observed, but not significant. These results indicate that flutamide and DES delay puberty in the male rat and its mode of action appears to be via altered secretion of steroids, which subsequently affect the development of the reproductive tract.
