The human semen ejaculated in a form of liquid state, coagulates immediately after ejaculation, and then liquefies again. However, the mechanisms of neither coagulation and liquefaction of semen have not been explained clearly so far, and very limited numbers of report are available, although the spermatology and andrology made rapid progress. This clinical study has been undertaken to investigate the liquefaction phenomena and practicability of the results might be applied to fertility and infertility problems. As a preliminary study, in this report the liquefaction time of various semen groups is measured and analysed. The following results are obtained: 1. An average liquefaction time of semen of a total of 60 subjects: 25 minutes. 2. An average liquefaction time of semen according to sperm count: 1) Normospermia group (20 cases): 34 minutes. 2) Oligospermia group (20 cases): 21 minutes. 3) Azoospermia group (20 cases): 20 minutes. 3. An average liquefaction time of semen according to abstinence period: 1) Less than 3 days group (30 cases): 22 minutes. 2) More then 5 days group (30 cases): 28 minutes. In conclusion: 1. The liquefaction time of semen of the normospermia group is longer than oligospermia group or azoosermia group. 2. The liquefaction time of semen may not be greatly influenced by the various factors such as abstinence period, semen volume, semen pH, age of the subjects and so on. 3. In routine semen analyses, it is recommended to begin the analysis at least 25 minutes after the ejaculation. 4. Further studies are required in conjunction with practical application of liquefaction mechanism in infertility and fertility control.
The aim of this experiments was to investigate the effect of Semen Plantaginis extract on. the renal function, plasma renin activity, plasma levels of atrial natriuretic peptide and aldosterone in rats. The results obtained were as follows: 1. Water balance decreased significantly after the administration of Semen Plantaginis extract. 2. Urine volume increased significantly after the administration of Semen Plantaginis extract, 0.1ml/200g. 3. Urinary excretion of sodium decreased significantly after the administration of Semen Plantaginis extract, 0.1 ml/200 g, 2week. 4. Urinary excretion of potassium decreased significantly after the administration of Semen Plantaginis extract, 0.1 ml/200 g, 2week. 5. Urinary excretion of chloride decreased significantly after the administration of Semen Plantaginis extract. 6. Free water clearance increased significantly after the administration of Semen Plantaginis extract 0.1 ml/200 g, 2 week and 0.2 ml/200 g. 7. Urinary excretion of creatinine increased significantly after the administration of Semen Plantaginis extract. 8. Plasma renin activity decreased significantly after the administration of Semen Plantaginis extract, 0.2 ml/200 g. 9. Plasma levels of aldosterone decreased significantly after the administration of Semen Plantaginis extract. 10. Plasma levels of atrial natriuretic peptide (ANP) decreased significantly after the administration of Semen Plantaginis extract.

Objectives : Armeniacae semen is the seed of Prunus armenica L. var. ansu MAXIM, and this is classified into Rosaceae. Armeniacae semen has been used for centuries in traditional oriental medicine for the treatment of pain and inflammatory diseases. Amygdalin is the major compound of Armeniacae semen, and it is now being used for the treatment of pain and cancer. Methods : In the present study, we compared the effects of an aqueous extract of Armeniacae semen and a solution of amygdalin extracted from Armeniacae semen on lipopolysaccharide(LPS)-stimulated prostaglandin E2 synthesis and nitric oxide production in mouse BV-2 microglial cells. For this study, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay, reverse transcription-polymerase chain reaction(RT-PCR), prostaglandin E2 immunoassay and nitric oxide detection were performed on mouse BV-2 microglial cells. Results : In the present study, an aqueous extract of Armeniacae semen and an amygdalin solution extracted from Armeniacae semen suppressed prostaglandin E2 synthesis and nitric oxide production by inhibiting the LPS-induced enhancement of cyclooxygenase-2(COX-2) mRNA and the inducible nitric oxide synthase mRNA in mouse BV-2 cells. For the cyclooxygenase-1(COX-1) expression, an aqueous extract of Armeniacae semen showed a more potent suppression effect compared to the amygdalin solution. However, the amygdalin solution more potently suppressed the LPS-induced COX-2 mRNA expression compared to the aqueous extract of Armeniacae semen. Conclusions : As a result, aqueous extract of Armeniacae semen and amygdalin exert anti-inflammatory and analgesic effects.
The aims of this study were to measure the ultrasonographic biometry of genitalia of the indigenous rams and observe the relationship of biometry on semen parameters. The epididymal volume was significantly reduced (p < 0.01) after semen collection compared with before collection for both left and right part in all rams. The cumulative results showed that although there was no significant difference in length, width and volume of epididymis between before and after semen collection, however the values were lower after collection. The epididymal length was significantly correlated with epididymal volume (p < 0.01), semen motility (p < 0.05) and semen morphology (p < 0.01). Epididymal width was only significantly correlated with epididymal volume (p < 0.01) not with the semen parameters. Epididymal volume had a significant correlation only with semen morphology (p < 0.01).The scrotal circumference had the significant correlation with semen density, mass activity, concentration and motility (p < 0.01). The epididymis had the similar or slightly increased echogenicity as compared to the normal testis. During whole study, some white spots were found on testis which did not affect the semen quantity and quality. Significant variation was observed only for semen concentration and motility among the rams (p < 0.05). The overall normal morphology was 90.5 ± 4.6% with highest percentage of coiled tail abnormalities.
Rancidity changes were examined for 6 herbal medicines, namely Persicae Semen, Armeniacae Semen, Lini Semen, Trichosanthis Semen, Arecae Semen, Myristicae Semen known to have relatively high fat content. In order to reduce rancidity of herbal medicines, samples were stored at 3 different conditions of room, refrigerating and freezing temperatures, and the rancidity was measured for 10 months with every 2 month interval. Fat content was extracted by using ethyl ether, and acid values and peroxide values, which are generally accepted indicators of fat rancidity, were measured. When storing Persicae Semen, Lini Semen and Arecae Semen at room temperature, the acid values increased as the storage period increased, and it was higher than when stored in refrigeration or freezing. The measurement of peroxide value showed more significantly higher initial degree of rancidity when Persicae Semen, Trichosanthis Semen, Arecae Semen and Myristicae Semen were stored at room temperature. It was observed that storing herbal medicines in refrigeration or freezing inhibited their rancidity compared to storing them at room temperature. To investigate the quality changes according to rancidity, the analysis of aflatoxins and indicator components showed that aflatoxins B1 and B2 were detected in Armeniacae Semen, Arecae Semen and Myristicae Semen, and the amount of amygdalin was well maintained within the specification standard.
A study was conducted to determine the effect of prostaglandin $F_2$ alpha ($PGF_2$ alpha) on the reaction time and seminal characteristics of buffalo bulls. Semen was collected from three Murrah bulls in three periods: pre-treatment, treatment and post-treatment. During the treatment period each bull was administered 2 ml $PGF_2$ alpha (Synchrocept, Fenprostalene) im, 1 hour prior to semen collection. In the post-treatment, semen was collected 7 days after the last injection of $PGF_2$. Semen samples were evaluated immediately after collection. Pre-collection injection of $PGF_2$ alpha has no significant effect on reaction time, semen volume, percentage motility, sperm concentration and total number of sperms per ejaculate. Fluctuations in semen color and consistency were observed. There is a significant (p<0.05) increase in the mean percentage of normal spermatozoa during the treatment and post treatment periods. Likewise, administration of PG results into a significant (p<0.05) rise on the average percentage of live sperms but this effect was not manifested in the post-treatment period. Improvement in mass activity was observed during the treatment and post-treatment periods.
Objectives:The present study purposed to evaluate the effects of Leonuri Herba and Leonuri Semen on hypercholesterolemia and provide data for the appropriate clinical application of Leonuri Herba and Leonuri Semen. Methods:We applied the water extracts of Leonuri Herba and Leonuri Semen to rats with hypercholesterolemia induced by high-cholesteroldiet, and examined their effects. Results:The levels of the total cholesterol and LDL-cholesterol were reduced by the uptake of Leonuri Herba to rats with hypercholesterolemia, but there was no significant effect on the level of TG(triglyceride), HDLc(HDL-cholesterol), FFA(free fatty acid), and Fc(free cholesterol), while the amount of AST and ALT was inhibited. The application of Leonuri Semen brought about the decrease of the total cholesterol and Fc, the increase of TG, HDLc, and FFA, and the significant decrease of the activation of AST. Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia. Conclusion:Based on the fact that Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia, it is concluded that Leonuri Semen is a better solution for hypercholesterolemia than Leonuri Herba.
In the oriental medicine, the Persicae Semen is a useful material for promoting blood circulation through removement of the extravasated blood in a blood vessel. This study is based on Dongueibogam, which has been widely used for herbal clinic in Korea. And this study is investigated to make sure of the range of Persicae Semen treatment, the nature of disease, the pathology and the dosage of it in Dongueibogam. The following conclusions were reached through investigations on the main prescriptions that used in Dongueibogam as a key ingredient. 1. The prescriptions of blending with the Persicae Semen are utilized in 25 fields of treatments such as feces, uterus, blood diseases, etc. 2. The Persicae Semen is used in the treatment of constipation, removement of blood stasis of gynecology. 3. The average mass of the Persicae Semen is 0.352g, the standard limited range of adding the Persicae Semen to medical prescription is 0.5$\sim$40g or 6$\sim$40pieces per dose. But, in most case of treatment medicines, an typical and regular adding dose is the range of 2$\sim$4g. 4. The Persicae Semen is one of basic materials for a fundamental prescription among several herbal remedies for the pathology.
A study was conducted at Ijok, Malaysia, to determine the relationship of testicular measurements with libido and semen quality in tropical and imported temperate breeds of sheep. Ten rams each of Malin (M), Siamese Longtail (L), Cross of Merino with Border Leicester (C), Dorset (D) and Suffolk (S) were used for the study. Libido, semen volume and semen quality were recorded monthly for a year together with testicular length, width and circumference. The results showed that there were breed differences in volume and quality of semen where the tropical breeds had better semen compared to the temperate breeds. There was positive and significant correlation between testicle length and semen volume in all the breeds. Testicular length was found to be positively and significantly correlated with motility and sperm concentration in the tropical breeds (L and M). The relationship between libido and testicle measurements in the tropical breeds was not significant (p<0.05). There was variable relationship between the testicular measurements and libido in the temperate breeds where the relationship was significant and negative in breeds C and D and highly significant and positive in S. It was evident that the long testicles influenced the quality of the semen whereas testicles with greater circumference influenced the libido of the rams.
Currently, boars selected for commercial use as AI sires are evaluated on grow-finish performance and carcass characteristics. If AI sires were also evaluated and selected on semen production, it may be possible to reduce the number of boars required to service sows, thereby improving the productivity and profitability of the boar stud. The objective of this study was to estimate genetic correlations between production and semen traits in the boar: average daily gain (ADG), backfat thickness (BF) and muscle depth (MD) as production traits, and total sperm cells (TSC), total concentration (TC), volume collected (SV), number of extended doses (ND), and acceptance rate of ejaculates (AR) as semen traits. Semen collection records and performance data for 843 boars and two generations of pedigree data were provided by Smithfield Premium Genetics. Backfat thickness and MD were measured by real-time ultrasound. Genetic parameters were estimated from five four-trait and one five-trait animal models using MTDFREML. Average heritability estimates were 0.39 for ADG, 0.32 for BF, 0.15 for MD, and repeatability estimates were 0.38 for SV, 0.37 for TSC, 0.09 for TC, 0.39 for ND, and 0.16 for AR. Semen traits showed a strong negative genetic correlation with MD and positive genetic correlation with BF. Genetic correlations between semen traits and ADG were low. Therefore, current AI boar selection practices may be having a detrimental effect on semen production.
